N2 - The topic of this dissertation was to better characterize known inhibitors of the glutathione peroxidase as well as identify novel compounds that can inhibit the enzyme and to characterize their biological effects in various cancer cell lines. Beginning with the well-known inhibitors mercaptosuccinic acid and tiopronin, data is presented for the inhibition of the bovine enzyme in terms of IC50-values and inhibition constants (Ki). After modification of the glutathione peroxidase assay to detect activity of glutathione peroxidase in lysates of human cancer cells, we found that mercaptosuccic acid but not tiopronin was able to inhibit human enzyme activity. In cancer cells the combination of mercaptosuccic acid with hydrogen peroxide resulted in a greater accumulation of reactive oxygen species compared to cells incubated with just hydrogen peroxide. That led to the presumption that mercaptosuccinic acid acts to inhibit the glutathione peroxidase in cells, thus decreasing its destruction. In contrast, these same experiments with tiopronin showed the opposite effect, indicating that the thiol compound acts instead as an antioxidant. In support of this hypothesis, combinations of tiopronin with cisplatin, doxorubicine and methotrexate resulted in an attenuated cytostatic activity of the anticancer drugs. A highlight of this work was the discovery of a novel class of inhibitors of the glutathione peroxidase, the pentathiepins. These compounds, possessing varying heteroaromatic structures and various substituents showed interesting structure-activity relationships. The mode of inhibition appears to be irreversible and competitive. These investigations showed that pentathiepins have a greater enzyme inhibiting potency and selectivity compared to all previously known inhibitors. In addition, in cellular systems pentathiepins showing interesting biological activities. In a broad range of human cancer cell lines we were able to detect IC50-values for inactivation of the viability and inhibition of proliferation in low micro molar concentrations. Statistical correlation analysis between potency of cell growth inhibition (IC50) and potency towards enzyme inhibition (IC50) of the bovine enzyme, a significant correlation between these two properties was observed. In addition, pentathiepins cause a large burst of reactive oxygen species in cancer cells. Pentathipins also induced apoptotic death in cells, evidenced by the Annexin-V/propidium iodide assay, the detection of PARP-cleavage and loss of mitochondria polarization. With the human leukemia cell line HAP-1 and its glutathione peroxidase knockdown variant, different cytotoxic potencies were identified for several anticancer drugs, namely: cisplatin, lomustine and temozolomide. In combination with cisplatin, the inhibitors of the glutathione peroxidase were found to attenuated the potency of cisplatin in different cancer cell lines. On the other hand, lomustine or temozolomide combined with either pentathiepins or mercaptosuccinic acid, combination results in significant increases in cytotoxic potency in various cancer cell lines. In summary these results show that the new inhibitors of the glutathione peroxidase can be a useful tools for better understanding of biological processes in case of cancer cells and their handling with oxidative stress. Combinations of glutathione peroxidase inhibitors with different anticancer agents results in a greater potency of some of the drugs but not for others. Due to the often poor prognosis of cancer, combination of pentathiepins to the chemotherapy might improve the outcome of some treatment regimes.
