N2  - Es wurden die Lokalisation des (P)RR/ATP6ap2 in PC12 Tumorzellen neuronalen Ursprungs, kardialen und renalen Zellen und Geweben sowie die funktionellen Folgen nach Downregulation oder Überexpression (paradoxe Downregulation) des Rezeptors untersucht. Er weist sowohl eine ER-spezifische, Membran-assoziierte als auch eine Lokalisation in der löslichen Fraktion auf.
Als akzessorische Untereinheit der vakuolären H+ ATPase beeinflusst der Rezeptor ihre Integrität und Aktivität. Die Vielzahl differenter Daten zwischen den mit einem V ATPase-Inhibitor behandelten und den (P)RR/ATP6ap2-downregulierten PC12 Zellen verdeutlicht, dass die Funktion des (P)RR/ATP6ap2 insbesondere im Metabolismus über eine bloße Regulation der V ATPase-Aktivität hinausgeht und möglicherweise durch die Integration des (P)RR/ATP6ap2 im Wnt-Pathway bestimmt wird.
N2  - The localization of the (P)RR/ATP6ap2 in PC12 cancer cells of neuronal origin, cardial and renal cells and tissues as well as the functional consequences after downregulation or overexpression (paradox downregulation) of the receptor has been examined. It shows both an ER-specific, membrane-associated localisation and a localization in the soluble fraction.
As an accessory subunit of the vacuolar H+‑ATPase influences the receptor its integrity and activity. The diversity of different data between the PC12 cells treated with a V-ATPase-inhibitor and downregulated (P)RR/ATP6ap2 elucidates that the function of the (P)RR/ATP6ap2 exceeds a mere regulation of the V-ATPase-activity, especially in metabolism, and is probably determined through the integration of the (P)RR/ATP6ap2 in the Wnt-pathway.
