Die Absorption von Propiverin ist abhängig von einer circardialen prä-systemischen Eliminierung, welche gekennzeichnet ist durch Biotransformation und aktivem Transport. Die Konzentrations-Zeit-Kurven des Arzneimittels wurden ermittelt nach der Gabe von oralen Dosen von 10, 15 und 30 mg Propiverinhydrochlorid in Drageeform und 15 mg Trinkflüssigkeit im Vergleich zu 15 mg intravenöser Gabe in Rahmen einer randomisierten, offenen, 5fach change-over Studie an 10 gesunden Probanden ( 4 männliche, 6 weibliche, Alter19-29 Jahre, Körpergewicht: 50-94 kg), um eine Dosisproportionalität zu untersuchen. Weiterhin wurden die pharmakodynamischen Auswirkungen von Propiverin auf Salivation, Akkommodation und Pupillenreaktion untersucht. Der gemessene Anstieg von AUC und Cmax war proportional zum Anstieg der oralen Dosis. Eine Auswirkung des Arzneimittels auf Akkommodation und Pupillenreaktion waren nicht messbar. Die Salivation wurde nach 8 Stunden in jeder Dosis signifikant beeinflusst. Die Pharmakokinetik des oral applizierten Proiverins in Dosen zwischen 10 und 30 mg ist nicht dosisabhängig. Das Arzneimittel ist als sicher und gut verträglich einzustufen. N2 - DOSE RELATED PHARMACOKINETICS OF ORAL PROPIVERINE Propiverine (PROP) is widely used in the treatment of urinary bladder instability. Its absorption is limited by circadian time dependent pre-systemic elimination most likely caused by biotransformation and active transport. Concentrations-time profiles of PROP were measured (LC-MS) after oral administration of 10, 15 and 30 mg propiverine hydrochloride in coated tablets and 15 mg in solution relative to 15 mg given iv in a randomised, open, change-over study with 10 healthy subjects (4 male, 6 female, age 19-29 years, body weight 50-94 kg) to verify dose-linearity of its pharmacokinetics. Furthermore, the influence of PROP on salivation, accommodation and pupil response was assessed. The increase of AUC and Cmax was proportionally related to the increase of the oral dose. PROP lacked effects on accommodation und pupil function. Salivation was significantly reduced 8 h after each administration. Pharmacokinetics of oral propiverine between 10 and 30 mg is not dose dependent. The drug was found safe and well tolerated.
