Dipeptidyl-Peptidase I, CTSC) ist eine lysosomale Cysteinprotease, die in zahlreichen Geweben verschiedener Spezies exprimiert wird und der eine wichtige Role bei entzündlichen Erkrankungen zugeschrieben wird. Bislang ist nicht bekannt, ob und inwieweit dies auch Bedeutung in der akuten Pankreatitis hat. Von anderen lysosomalen Enzymen wie Cathepsin B und L ist bekannt, dass sie die Aktivierung von Trypsinogen regulieren. So spaltet Cathepsin B Trypsinogen zu aktivem Trypsin, während mit Cathepsin L ein enzymatisch inaktives trunkiertes Trypsin resultiert. Das Vorhandensein von aktivem Trypsin ist eine entscheidende Voraussetzung für die kaskadenartige Aktivierung weiterer Proteasen, welche den pankreatischen Schaden bei der akuten Pankreatitis bedingen. Anhand von immunhistochemischen Färbungen, enzymatischer Messungen und Western Blot-Analysen konnte gezeigt werden, dass Cathepsin C in Azinuszellen exprimiert und ähnlich wie Cathepsin B in der Frühphase der akuten Pankreatitis intrazellulär in die Zymogenfraktion umverteilt wird. In isolierten und mit supramaximalen Konzentrationen von Cholezystokinin (1 µM) stimulierten Azinuszellen fand sich eine Zunahme der intrazellulären Aktivierung von Cathepsin C, die vergleichbar mit der Aktivität von Cathepsin B und Trypsin war. In vorbereitenden Versuchen konnten für ex-vivo Versuche in pankreatischen Azinuszellen optimale Konzentrationen eines für Cathepsin C spezifischen Substrates (Gly-Arg-AMC) und Inhibitors (Gly-Phe-CHN2) etabliert werden. Durch den Inhibitor konnte eine intrazelluläre Cathepsin C Reduktion um ca. 25% erreicht werden. Diese war begleitet mit einer Verminderung der intrazellulären Cathepsin B-Aktivität, jedoch nicht mit einer Verminderung von Trypsin oder Elastase. Auch blieb die azinäre Nekrose unverändert. Ebenso konnte ein direkter Effekt von Cathepsin C auf die Trypsinogenaktivierung in-vitro ausgeschlossen werden. Diese Ergebnisse lassen vermuten, dass Cathepsin C an der Aktivierung von Pro-Cathepsin B beteiligt ist. Die unveränderte Aktivierung der Zymogene sowie die nicht veränderte Nekrose der Azinuszellen deuten darauf hin, dass der pankreatische Schaden unabhängig vom in Azinuszellen exprimierten Cathepsin C sein muss. Die Ergebnisse lassen ferner vermuten, dass diese lysosomale Protease zusätzliche Effekte auf weitere Proteasen haben muss, da trotz Reduktion von Cathepsin B eine unveränderte Trypsinogenaktivierung beobachtet wurde. Diese Analysen sollen Gegenstand zukünftiger Untersuchungen sein. N2 - Acute pancreatitis is a complex disorder that involves both premature protease activation and recruitment of inflammtory cells in the organ. Intracellular activation of the zymogen protease trypsinogen by limited proteolysis is one of the earliest moments in early acute pancreatitis which is driven by the lysosomal protease cathepsin B. Meanwhile other lysosomal proteases were identified in regulation of trypsinogen, either by a direct or an indirect way. Besides intraacinar localized lysosomal enzymes that account for intracellular trypsinogen activation and thus development of acute pancreatitis other cell types of the body also contain lysosomal hydrolases. These enzymes might be relevant for initiation of acute pancreatitis as well, despite its mainly extrapancreatic expression. Cathepsin C (CTSC, syn. Dipeptidyl-peptidase I or DPPI, EC 3.4.14.1) is one of these lysosomal proteases which is mainly expressed in lungs, kidney, placenta and in inflammatory cells, i.e. polymorphonuclear leukocytes and alveolar macrophages and to a lesser extent in other organs. Cathepsin C is capable of activating many serine proteases contained in immune cells and thus might be of potential interest for pancreatic digestive enzymes, too. Moreover, there is a complex interaction of cathepsin C with other lysosomal hydrolases as autoactivation of cathespin C is dependent on endopeptidases such as cathepsin L and to a lesser extent cathepsin S, while cathepsin D degrades both inactive and mature cathepsin C. Similarly, cathepsin C in combination with cathepsin B appears to process N-termini of cathepsin B yielding the correct NH2-terminus of mature cathepsin B. In view of the different role of cathepsin C here was investigated its potential function in acute pancreatitis using immunhistochemical coloring, enzymatic assays and western blot-analysis in a mouse model. It is shown that cathepsin C is expressed in acinar cells and move like cathepsin b during acute pancreatitis in the zymogens. In with cholezystokinin activated acinar cells there was an increase of the intracellular activation from cathepsin C like cathepsin B and trypsin. Using an inhibitor for cathespin C (Gly-Phe-CHN2) there was an complete inhibition in vitro and an smaller inhibition of 25% in acinar cells. Using this inhibitor there was also an inhibition of cathepsin B but not of trypsin and elastase. Necrosis was not effected. In vitro experiments couöd exclude a direct inhibition of cathepsin B with the cathepsin C inhibitor.
