TY - THES U1 - Dissertation / Habilitation A1 - Bock, Christian T1 - Synthese und Charakterisierung von Liganden spannungsabhängiger Kv7-Kaliumkanäle auf Grundlage der Arzneistoffe Flupirtin und Retigabin N2 - Die Arzneistoffe Flupirtin und Retigabin wurden über viele Jahre hinweg erfolgreich als Analgetikum bzw. Antiepileptikum eingesetzt. Vor allem aufgrund ihres einzigartigen Wirkmechanismus, welcher in der Öffnung spannungsabhängiger Kv7-Kaliumkanäle liegt, konnte eine weitgehend nebenwirkungsfreie Therapie ermöglicht werden. Innerhalb der letzten drei Jahre wurden allerdings sowohl Flupirtin als auch Retigabin aufgrund von seltenen, aber schwerwiegenden unerwünschten Arzneimittelwirkungen (UAWs) vom Markt genommen. Man geht davon aus, dass die Lebertoxizität von Flupirtin ebenso über eine oxidative Verstoffwechslung zu instabilen Metaboliten vermittelt wird, wie die reversiblen Blauverfärbungen von bestimmten Geweben unter Retigabin-Therapie. Im Rahmen dieser Promotionsarbeit wurden verschiedene Modifikationen am Triamino-Aromaten-Motiv der Arzneistoffe vorgenommen und deren Einfluss auf verschiedene Eigenschaften untersucht. So wurde die Oxidierbarkeit von 55 Verbindungen cyclovoltammetrisch bestimmt und der Aktivität und Toxizität gegenübergestellt. Die beste Substanz 3-(3,5-Difluorphenyl)-N-(6-[isobutylthio)-2-(pyrrolidin-1-yl)pyridin-3-yl]propanamid konnte dabei eine 918-fach höhere Aktivität als Flupirtin, bei gleichzeitig gesteigerter oxidativer Stabilität aufweisen. Zusätzlich wurden durch die schrittweise Derivatisierung von Flupirtin Struktur-Aktivitäts-Beziehungen für Kv7.2/3-Heterotetramere erhalten. N2 - The former drugs flupirtine and retigabine have been used for years in the treatment of pain and epilepsy, respectively. Especially their unique mode of action, the opening of voltage gated Kv7 potassium channels, resulted in a well-tolerated treatment of impairments. Within the last three years, both drugs were withdrawn from the market due to rare but severe side effects. The biggest drawbacks, flupirtines liver toxicity and the blue discoloration of various tissues under retigabine therapy, seem to be mediated via oxidation to instable metabolites. Therefore, it seemed worthwhile to change the oxidative properties of the drugs by modifying the triamino-aromatic core-motif. We measured the oxidizability of 55 compounds in cyclic voltammetry experiments and correlated it with their activity and toxicity, resulting in the compound 3-(3,5-difluorophenyl)-N-(6-(isobutylthio)-2-(pyrrolidin-1-yl)pyridin-3-yl)propenamide being 918-times more active than flupirtine, while being more resistant to electrochemical oxidation. Furthermore, structure-activity-relationships for Kv7.2/3 channels were established in the present work.
