TY - THES U1 - Dissertation / Habilitation A1 - Bertram, Jonathan T1 - Einfluss der Deletion von srGAP3 auf den Hippocampus bei Mäusen N2 - Im Jahr 2002 wurde bei einer stark mental retardierten Patientin im Rahmen einer Genanalyse ein Strangbruch am kurzen Arm des dritten Chromosoms festgestellt. Das davon betroffene Gen codiert für ein bis dahin unbekanntes Protein, welches als srGAP3 / MEGAP / WRP benannt wurde. Es gehört zur Familie der Rho-GTPasen aktivierenden Proteine. Diese Rho-GTPasen nehmen über verschiedene Signaltransduktionsketten Einfluss auf die Wegfindung, Differenzierung und Verknüpfung von Neuronen während ihrer Entwicklung. Mit Hilfe eines Knockoutmausmodells konnten in vorangegangenen Forschungsarbeiten starke Veränderungen der Gehirnarchitektur, sowie Schichtverdickungen im Bereich des Hippocampus‘ festgestellt werden. Die Beziehung von srGAP3 zu den Rho-Proteinen und den damit entstehenden Einfluss auf die neuronale Entwicklung ließ den Hippocampus als Region der adulten Neurogenese in den Fokus der Forschungsarbeit rücken. Die durchgeführten Untersuchungen erfolgten im srGAP3-Knockoutmausmodell mittels Kleintier-MRT, Golgi-Imprägnierung und immunhistochemischen Färbungen (gegen Doublecortin und Phosphohiston 3). In den Ergebnissen konnte durch den srGAP3-Knockout zwar eine Volumenzunahme des Hippocampus‘, jedoch weder eine signifikante Veränderung der Neuronenanzahl, der Neuronenmorphidität, noch der Spinedichte oder der Spinelänge im Hippocampus festgestellt werden. Als Erklärung der präsentierten Ergebnisse und möglicher neuer Forschungsansatz wäre die These einer Zunahme an kortikalen Neuronen, welche in den Hippocampus projizieren, denkbar. Dieser Anstieg könnte sowohl eine faserbedingte Volumenzunahme, die gleichzeitig fehlenden neurostrukturellen Veränderungen im Hippocampus, als auch die milden verhaltensbiologischen Auffälligkeiten in den bisher durchgeführten Tests erklären. Die Untersuchung der Tiere in komplexeren Verhaltenstests könnte dahingehend wegweisend sein. N2 - In 2002 a genetic analysis of a girl with profound mental retardation showed a translocation of the short arm of the third chromosome. The breaking point led to the disruption of a gene, which was unknown at that time. The gene was named srGAP3 / MEGAP / WRP. It is part of the family of Rho-GTPases activating proteins which play an important role in the pathfinding, differentiation and connection of developing neurons. For further research, a knockout mouse model was established. The mice showed enormous changes in the architecture of the brain, especially enlarged ventricles, increased wet weight of the brain and thicker hippocampal layers. The analysis of the expression of srGAP3-mRNA led to the hypothesis that the protein concentrates its effect on the hippocampus, where the expression was most evident. The hippocampus plays an important role in the process of learning and memory-forming which was considerably reduced in the presented patient. Interestingly, the srGAP3-related Rho-Proteins are essential factors in neuronal development and the hippocampal gyrus dentatus is one of the few parts of the brain where the learning and memory-related neurogenesis takes place. The study focused on three main points: the hippocampal size, the neurogenesis and the neuronal network. The hippocampal size was measured with a MRE specialized for small animals. The neurogenesis was analysed with immunohistochemical staining against Doublecortin and Phosphohistone H3. The neuronal network was visualized with golgi staining which allowed the evaluation of the quantity and morphology of neurons and the density and length of spines. The results showed an increased hippocampal volume but neither the neuronal network nor the neurogenesis in the hippocampal gyrus dentatus was altered. An increased number of projecting cortical neurons could be an explanation of the existing results and may become the focus of further research. It can be presumed that they lead to the enlargement of the hippocampus without changing the neuronal network in the gyrus dentatus. Moreover this hypothesis could explain the small changes in simple cortical tests which may differ in cortical tests with a higher level of complexity.
