TY - THES U1 - Dissertation / Habilitation A1 - Frenzel, Marie Louise T1 - Weiterentwicklung eines Zellmodells zur Untersuchung der ischämischen Prä- und Postkonditionierung N2 - In der westlichen Welt zählen der Myokardinfarkt, bzw. die kardiovaskulären Erkrankungen, zu den häufigsten Todesursachen. Dies ist einer der Gründe, warum in den vergangenen Jahrzehnten intensiv nach Möglichkeiten gesucht wurde, die Folgen von Herzinfarkten zu minimieren. Grundlage für viele neuere Versuche war die Entdeckung, dass die Wiederdurchblutung des ischämischen Gewebes eine weitere Zellschädigung verursacht, genannt Reperfusionsschaden. Diese Zellen sterben nicht durch die Ischämie, sondern durch ein intrazelluläres Ungleichgewicht der Elektrolytkonzentrationen und einem ATP-Mangel. Es wurde gezeigt, dass man mit Hilfe von Präkonditionierung, später auch Postkonditionierung, diese Gruppe von Zellen vor dem Untergang schützen und somit das verbleibende Infarktareal verkleinern kann. Im Rahmen dieser experimentellen Arbeit wurde ein zellbasiertes Modell zur Erforschung der Postkonditionierung an Herzzellen so weiterentwickelt, dass Bestandteile der postulierten Signalkaskade der Postkonditionierung auch auf zellulärer Ebene erforscht werden können. Darüber hinaus ist es möglich, verschiedene Substanzen in ihrer Fähigkeit diesen Schutz auszulösen, zu untersuchen. Mit Hilfe einer immortalen Tumorzelllinie der Maus (HL-1-Zellen), die herzzellähnliche Eigenschaften besitzt, konnten Zellen so kultiviert werden, dass eine Vielzahl von Experimenten möglich wurde. Durch den Farbstoff TMRE, ein Marker für das mitochondriale Membranpotential, konnte bei Messungen mit der FACS-Technik eine gute Diskriminierung zwischen den toten und vitalen Zellen erreicht werden. Der Austausch des Nährmediums mit einem speziellen Puffer während der Experimente bewirkte, dass die Konzentration von Calcimycin, bei gleicher Wirkung, herabgesetzt werden konnte. Calcimycin ist ein Kalziumionophor, der den tödlichen Reperfusionsschaden auf der Basis einer Kalziumüberladung auslöst. So konnte anhand dieses Zellmodels die schützende Wirkung von BAY 58-2667 und Eplerenon auf ischämiegeschädigte Zellen bestätigt werden. Es konnte gezeigt werden, dass es ein Modell auf dieser Basis ermöglicht, Bestandteile der Signalkaskade zu identifizieren und Pharmaka auf ihren infarktgrößenreduzierenden Effekt zu testen. Die Methode stellt allerdings keinen Ersatz zu Tierexperimenten und klinischen Forschungen dar, dennoch ist sie eine gute Möglichkeit die Substanzen vorab auf ihre Tauglichkeit zu testen und somit gezielter Tierversuche durchzuführen. N2 - In the Western world myocardial infarction or cardiovascular disease is one of the leading cause of death. This is one of the reasons, why there has been an intensive search for investigations to minimize the consequences of heart attacks in the past decades. Background for many recent experiments was the discovery of the fact, that reperfusion of ischemic tissue causes further cell damage, so called reperfusion injury. These cells do not die due to ischemia, but in consequence of an intracellular imbalance of electrolyte concentrations and ATP depletion. Brief episodes of ischemia and reperfusion applied before or after the indexischemia could reduce the infarction. This so called ischemic pre- and postcondioning indicates high potential for investigation against reperfusion injury. The aim of this study was to develop a cell-based model to study the post-conditioning on heart cells, so that the components of the postulated signaling cascade of postconditioning can also be explored at cellular level. Moreover, it is possible to investigate different compounds in their ability to trigger this protection. By using an immortal tumor cell line of mice (HL-1 cells), which have the same properties as heart cells, it was possible to culture cells, compared to primare cellculture, and do a variety of experiments. The dye TMRE, a marker of the mitochondrial membrane potential, measured with the FACS technology, allowed a good discrimination between the dead and living cells. Furthermore, with the exchange of the culture medium through a special buffer during the experiments lead to lower concentration of Calcimycin, that caused the same effect. Calcimycin is a calcium ionophore, which triggers the lethal reperfusion injury based on a calcium overload. With these cell models could be shown that BAY 58-2667, an activator of soluble guanylyl cyclase, and eplerenone protect ischemia. Taken together, the results of the present study are showing properties of a cell model to identify components of the signaling cascade and to test drugs in their infarct size reducing effect. The method is, however, not a substitute for animal experiments and clinical studies, but it is still a good way to test the potential substances in advance to their suitability to protect and thus perform more targeted experiments on animals.
