N2  - Mit dieser Arbeit sollte die Auswirkung eines P-gp knockouts auf das Genom des Gehirns von Mäusen (P-gp „knockout“ Mäuse) im Alter von 3, 6, 9 sowie 12 Monaten im Vergleich zu Wildtyptieren untersucht werden. Die vorgelegte Arbeit konnte Hinweise auf einen unmittelbaren Zusammenhang zwischen P-gp knockout und neuronaler Dysfunktion geben. Dabei fanden sich insbesondere bei den jungen Tieren signifikante Unterschiede in der Expression von Genen, die für entzündliche Prozesse oder auch strukturelle und funktionelle Eigenschaften der verschiedenen Zellen des Gehirns eine wichtige Rolle spielen.
N2  - An important research focus of the Neuropathology work of the Institute of Pathology of the University of Greifswald is the investigation of the importance of multidrug transports, in particular the P-glycoprotein, in the pathogenesis of Alzheimers disease. The aim of this work was to investigate the effect of P-gp knockout on the genome of the brain of mice (P-gp "knockout" mice) at 3, 6, 9 and 12 months compared to wildtype. The presented work could indicate a direct relationship between P-gp knockout and neuronal dysfunction. Particularly in the young animals there were significant differences in the expression of genes, which play an important role for inflammatory processes or also structural and functional properties of the different cells of the brain. This can serve as an approach for further studies of P-gp knockout on gene expression on cell plane. In summary, the results support the hypothesis that induction of P-gp is an approach for new innovative therapy options in neurodegenerative diseases such as Alzheimers disease.
