Die vorliegenden Daten gestatteten keine Aussagen zu den beeinflussten CYP-Subfamilien und zur Dosisabhängigkeit der Induktion. In dieser Studie wurde die Beeinflussung der mikrosomalen Ethylresorufin-0- Deethylase (EROD), Ethoxycumarin-0-Deethylase (ECOD), Pentylresorufin-0- Depentylase (PROD), Diazepam-N-Demethylase (DNDM), Dextromethorphan-0- Demethylase (DXDM), p-Nitrophenolhydroxylase (NPH) und Erythromycin-N- Demethylase (ERDM) in der Leber männlicher Ratten nach Behandlung mit 0,6-60 mg/kg Propiverin über 5 Tage im Vergleich mit den bekannten CYP- Induktoren Phenobarbital, ß-Naphthoflavon und Dexamethason untersucht. Es zeigte sich, dass Propiverin in einer Dosierung bis 2 mg/kg weder die untersuchten Monooxygenaseaktivitäten noch den Gesamt-CYP-Gehalt beeinflusst. Das Induktionsmuster nach der Behandlung mit Propiverin hatte mit einer herausragenden Induktion der PROD (33fach) sowie einer geringeren Erhöhung der EROD, ECOD, DNDM und ERDM den Charakter einer Phenobarbital-Typ-lnduktion. Die vermehrte CYP2B-Expression nach Gabe von 60 mg/kg Propiverin konnte mittels Western Blot nachgewiesen werden. Die DXDM als Maß für die CYP2D-Aktivität änderte sich durch Propiverin- behandlung im untersuchten Dosisbereich nicht. Zusätzlich zeigten Propiverin und Despropylpropiverin in vitro eine Hemmung der PROD (halbmaximale Hemmkonzentration IC50 ca. 0,5 µmol/l Propiverin); die EROD und ERDM blieben in vitro unbeeinflusst. Zur Induktion der untersuchten Enzyme kam es erst nach Gabe des Vielfachen der empfohlenen Menge für den therapeutischen Gebrauch. N2 - The urinary bladder anti-spasmodic Propiverine was known from literature as an inducer of CYP enzymes in the rat liver. The data in hand did not allow a statement about the involved CYP subfamilies or the dosage-dependency of the induction. In the study we investigated the influence of a 5-day treatment with 0,6-60 mg/kg Propiverine on the activity of microsomal ethylresorufin-0-deethylase (EROD), ethoxycoumarin-0-deethylase (ECOD), pentylresorufin-0-depentylase (PROD), diazepam-N-demthylase (DNDM), dextromethorphan-0-demethylase (DXDM), p- nitrophenolhydroxylase (NPH), and erythromycin-N-demethylase (ERDM) in the liver of male rats in comparison to the CYP-inducers phenobarbital, ß-naphthoflavone, and dexamethasone. Propiverine in doses up to 2 mg/kg neither influences the tested monooxygenase activities nor the total CYP content. The induction pattern after treatment with Propiverine had the character of a phenobarbital type induction, with eminent induction of PROD (33-fold) and slow increase of EROD, ECOD, DNDM, and ERDM. The increased CYP2B expression after 60 mg/kg Propiverine was proved by means of western blot. The DXDM as measure for CYP2D activity was unchanged after Propiverine treatment in this study. In vitro Propiverine and Despropylpropiverine inhibited the PROD activity (IC50 about 0,5 µmol/l Propiverine). EROD and ERDM were not effected on in vitro. The induction of the tested enzymes was observed only after treatment with doses several times the therapeutical dosage in man, therefore drug-interaction by CYP induction is not to be expected in clinical use.
