N2  - Im Rahmen des Verbund-Forschungsprojektes KOKON wurde nach systematischer Literaturrecherche eine Datenbank entwickelt; in der KOKONbase sind sowohl die Interaktionsprofile als auch die Interaktionsmatrix die wesentlichen Elemente der Vorhaltung von primärem und bewertetem Wissen. In der Interaktionsmatrix wird mit Hilfe eines Ampelschemas die Möglichkeit der Beeinflussung der Pharmakokinetik von Arzneistoffen durch ausgewählte Drogen dargestellt. Die Droge und die Arzneistoffe werden paarweise abgebildet. Die Interaktionsmatrix wird durch praktisch tätige Onkologen als sehr wertvolles Instrument in der onkologischen Beratungspraxis angesehen, um schnell einen Überblick über das von einer Droge ausgehende Gefahrenpotential bzgl. der Beeinflussung der Wirksamkeit eines in der Onkologie genutzten Arzneistoffs zu bekommen.
N2  - A grading system that prioritizes the type of study, consistency of study results, and interaction strength was developed that generated risk categories for herb]drug interactions. Few clinical trials assessed herb]drug interactions directly. For the majority of risk assessments, risk was extrapolated from metabolic and in vitro studies. Risk categories were assigned to and dynamically visualised for the five plants and 161 antitumor drugs. As expected, St. Johnfs Wort was the herb with the highest risk for herb]drug interactions due to both interacting with highly relevant metabolising enzymes and drug transporters and a high coverage by herb]drug interaction literature. In contrast, mistletoe was the herb that was covered the least by literature data on pharmacokinetic herb]drug interactions.The developed algorithm allows to transparently generate and Display interaction risk grading from datasets extracted from drug interaction literature. This tool can therefore help to provide easy access to herb]drug interaction risk information and may support clinicians advising their patients.
