TY - THES U1 - Dissertation / Habilitation A1 - Lange, Thom T1 - Zum Einfluss des Kationenkanals TRPV4 auf Kontraktionsverhalten und Ermüdung von Maus-Skelettmuskeln N2 - Diejenigen Mechanismen, welche innerhalb der skeletalen Myozyten zur Kontraktion und Kraftentfaltung führen, sind heute, bis auf wenige verbleibende Mysterien, sehr gut verstanden. In der Hauptsache werden zu den relevanten Membranproteinen, die im Exzitations- und Kontraktionsgeschehen der Myozyten von Bedeutung sind, der sarkolemmale Dihydropyridinrezeptor sowie der sarkoplasmatische Ryanodinrezeptor gezählt - nicht aber TRP-Ionenkanäle. Diese werden hingegen u.a. mit der Sensorik von Geschmack, Temperatur, Osmolarität, Nozizeption sowie taktiler Reize in Verbindung gebracht. TRP-Ionenkanäle werden ubiquitär exprimiert. Ihre Existenz innerhalb des Sarkolemms von Myozyten, sowohl vom glatten als auch vom quergestreiften Typus, ist belegt. Die belgische Gruppe um Nadège Zanou, Georges Shapovalov und Phillip Gailly publizierten Hinweise, die darauf hindeuten, dass ein spezieller kanonischer TRP-Ionenkanal, der TRPC1, möglicherweise eine Rolle im Kontraktionsgeschehen der quergestreiften Myozyten spielt. Solche Beobachtungen werfen unter anderem die Frage auf, ob es weitere Kandidaten der TRP-Proteinfamilie gibt, die in die myozytären Kontraktionsprozesse involviert sind. Es ist derzeit teilweise geklärt, welche Funktionen TRP-Ionenkanäle der TRPV-Subfamilie innerhalb glatter Muskelzellen übernehmen. Welche Bedeutung Vertreter der TRPV-Subfamilie für die quergestreiften Myozyten haben, ist aktuell aber noch nicht hinreichend geklärt. Die vorliegende Dissertation thematisiert die wissenschaftliche Frage nach der funktionellen Bedeutung von TRPV4-Ionenkanälen für die Kontraktions- und Ermüdungsvorgänge innerhalb der quergestreiften Muskulatur der Maus. Um die Frage beantworten zu können, ob TRPV4-Kationenkanäle innerhalb der quergestreiften Myozyten funktional sind, führten wir In-vitro-Kraftmessungen mit isolierten Mm. solei der Wildtypmäuse C57Bl/10Sc/J und C57Bl/6 sowie der TRPV4-defizienten Maus durch. Darüber hinaus haben wir den Einfluss von 4aPDD, ein Phorbolesterderivat und selektiver TRPV4-Aktivator, auf Kontraktions- und Relaxationszeiten, die maximalen Kraftentwicklungen sowie die Muskelermüdung (Fatigue) untersucht. Im Rahmen unserer Untersuchungen konnten wir zeigen, dass sich der quergestreifte Muskel über eine TRPV4-Stimulation im Hinblick auf seine Maximalkraftentwicklung und Ermüdungserscheinungen positiv beeinflussen lässt, wohingegen dabei sowohl die Kontraktions- als auch die Relaxationskinetiken unbeeinflusst blieben. Unsere Resultate und Beobachtungen stellen somit ein deutliches Plädoyer für die Funktionalität der TRPV4-Ionenkanäle innerhalb der quergestreiften Myozyten dar. N2 - Except for a few mysteries, muscle contraction mechanisms leading to force production at the level of myocytes are well understood. Variations in calcium concentrations modulate muscle mechanisms. An increased concentration in the sarcoplasm yields muscular contraction through actin and myosin interaction, a decreased calcium concentration allows muscular relaxation. To the major relevant proteins in the process of calcium concentration regulation counts on the one hand the dihydropyridine receptor and on the other hand the sarcoplasmic ryanodine receptor – not as yet TRP-channels. TRP-channels are widely expressed on myocyte sarcolemma of smooth and skeletal muscles. These channels are currently known to be important functional proteins regulating nociception, temperature and osmolality. Their role in skeletal muscle is currently unclear. Nadege Zanou and colleagues have shown that TRPC1 could play a role in muscle contraction. Hence, additional TRP-channels, such as, the TRPV subfamily may help regulate the process of muscular contraction. Krüger et al. has shown that TRPV4 is expressed in mice skeletal muscle. Even though TRV4- channels are existent in the sarcolemma, the function of TRPV4 within the skeletal myocyte is still unknown. Therefore our study focused on TRPV4 function in the skeletal muscle. Isolated soleus muscles of C57B1/10Sc/J, C57B1/6 (“black six”) and TRPV4 knockout mice were used in the study. A force measurement method allowed the examination of force production and fatigue of the selected muscles. A TRPV4 selective activator, 4aPDD, was used to test its effect on the TRPV4 channels during contraction with force production and fatigue. Time to peak and half relaxation time, indirect parameters of contraction kinetics, were also evaluated. The study results reveal that 4aPDD increases muscular tetanic force by at least 10% and attenuates muscle fatigue. These effects were absent in the soleus muscles of the TRP-V4 knockout mice. These findings suggest that TRPV4 is important in the functional process of mice skeletal muscle force production and fatigue.
