TY - THES U1 - Dissertation / Habilitation A1 - Tscheche, Sandra T1 - Der Einfluss von Leptin und Resistin auf die postnatale Entwicklung des Nebenhodenepithels der Ratte N2 - Die vorliegende Arbeit befasst sich mit dem Nachweis von Leptin und Resistin mittels immunhistochemischer Methoden. Im Vordergrund stand dabei die Frage einer positiven Leptin- und Resistinreaktion im periepididymalen Fettgewebe zum Zeitpunkt der Ausdifferenzierung des Nebenhodenepithels sowie einer gleichzeitigen positiven Rezeptorreaktion im Hypothalamus. Dazu erfolgte der immunhistochemische Nachweis von Leptin, Resistin, Ob- R und Insulinrezeptor im Bereich des Nebenhodens, des Hypothalamus, des Pankreas und der Lunge männlicher Ratten im Alter von 35- 65 d. p. p. Die Untersuchungen ergaben eine positive Reaktion der Hormone und Rezeptoren in allen Organen, außer beim Nachweis von Resistin im Bereich des Hypothalamus. Es fanden sich nur gerinfügige Intensitätsunterschiede zwischen dem 35. und 65. d. p. p., ein zeitlicher Peak der Hormonkonzentration zum Zeitpunkt der Ausdifferenzierung des Nebenhodenepithels war nicht erkennbar. Somit ist eine regulatorische Funktion von Leptin und Resistin bezüglich der männlichen Reproduktion zwar anzunehmen, jedoch ist diese sicher nicht von herausragender Bedeutung. N2 - This thesis with the evidence of the hormones leptin and resistin making use of immunohistochemical methods. We mainly focused on a positive reaction of leptin and resistin within the periepidiymal adipose tissue at the time of differentiation and morphological development of the epididymal epithelium. In addition we examed a simultainlous positive reaction of the leptin receptor (ob-r) in certain hypothalamical nuclei. Therefore we ran an immunohistochemical test of leptin, resistin, ob- r and insulin receptor in the epididymis, as well as in the hypothalamus, the pancreas and the lungs of male rats ages 35- 65 d. p. p. From the test followed a positive reaction of the hormones and receptors in all investigates tissues, except for resistin in the hypothalamical nuclei. Only minor differences were observed concerning the reaction´s intensity between the 35th and the 65th d. p. p. There was no climax in the hormone concentrations at the time of epididymal differentiation. Therefore we assume leptin and resistin to be part of the regulatory circle of male fertility and reproduction, however it isn´t of great importance.
