TY - THES U1 - Dissertation / Habilitation A1 - Persike, Maria T1 - Die Aktivität, Aktivierung und Verteilung der Proteinmenge von Cathepsin B in subzellulären Kompartimenten der Azinuszelle während der akuten experimentellen Pankreatitis N2 - Die lysosomale Hydrolase Cathepsin B (CTSB) spielt in der Pathogenese der akuten Pankreatitis eine zentrale Rolle. Sie gilt als wichtiges Aktivatorenzym von Trypsinogen, welches einen kaskadenartigen Aktivierungsprozess weiterer Zymogene (inaktiver Vor- stufen von Proteasen) auslöst. Seit 1987 geht man davon aus, dass es in der frühen Phase der Pankreatitis zu einer Umverteilung der CTSB-Aktivität von den Lysosomen in das sekretorische Kompartiment der Azinuszelle kommt. Ziel meiner Arbeit war es, diese Umverteilung am Modell der Caerulein-induzierten Pankreatitis in FVB-Mäusen näher zu untersuchen. Dafür nutzte ich die Methode der subzellulären Fraktionierung, die mittels Sucrosegradient und verschiedener Zentrifugierungsschritte die Komparti- mente (Zymogene, Lysosomen und Cytosol) der Azinuszelle ihrer Dichte nach trennt. In den einzelnen Kompartimenten habe ich sowohl fluorometrisch die Aktivität, als auch mittels Western Blot den Proteingehalt von CTSB im Verlauf der Pankreatitis gemes- sen. Ergänzend untersuchte ich die Prozessierung der Hydrolase unter Verwendung eines neuen hochspezifischen Inhibitors (NS-196-CTSB-Inhibitor) von Cathepsin B. Meine Er- gebnisse bestätigen die Aktivitätsumverteilung des CTSB von der lysosomalen Fraktion unter physiologischen Bedingungen in die Zymogengranula-angereicherte Fraktion in der Pankreatitis bereits nach 1 h. Darüber hinaus zeigte sich eine signifikante Erhöhung der Gesamtaktivität in der frühen Phase der Erkrankung. Der CTSB-Proteingehalt blieb allerdings in der genannten Zeitspanne in den drei Kompartimenten mehr oder weniger unverändert und stieg erst nach 8 h an. Weiterhin konnte ich feststellen, dass die Pro- zessierung von CTSB hauptsächlich im lysosomalen und sekretorischen Kompartiment der Azinuszelle stattfindet und sich während der akuten experimentellen Pankreatitis nicht erhöht. Im Cytosol fand sich trotz erheblicher Proteinmengen nur wenig prozes- siertes CTSB und kaum messbare Aktivität. Diese Ergebnisse lassen daher den Schluss zu, dass die Umverteilung der CTSB-Aktivität weder auf einer Kompartimentverschie- bung von Protein noch auf einer veränderten Prozessierung des Enzyms basiert, sondern auf einer Zunahme der Aktivität der gleichen Menge von prozessiertem CTSB. Insofern erfolgt keine Umverteilung des Proteins CTSB in das sekretorische Kompartiment, noch eine veränderte Prozessierung von CTSB im sekretorischen Kompartiment, sondern eine Zunahme der Aktivität von CTSB in diesem Kompartiment. Mögliche Ursachen hier- für wären eine Änderung der biophysikalischen Umgebung (z.B. pH, Ca++) oder eine Veränderung im Gehalt physiologischer CTSB-Inhibitoren (z. B. Cystatine). N2 - The lysosomal hydrolase cathepsin B (CTSB) plays a central role in the pathogenesis of acute pancreatitis. It is supposed to be an important activator enzyme of trypsinogen, which causes a cascade of activation process further zymogens (inactive precursor proteases). Since 1987 a redistribution of CTSB activity from lysosomal into sectretory compartment of the acinar cell during the early time of pancreatitis is considered. Aim of my work was to study this redistribution by the model of caerulin induced pancreatitis in fvb mice. Therefore I was using the method of subcellular fractionation, which separates the three compartments (zymogenes, lysosomes and cytosol) in relation to their density by the help of a sucrose gradient and different centrifugation steps. Within those compartments I was measuring both flourometric the activity and in western blot the protein content of CTSB during pancreatitis. In addition I analysed the processing of the hydrolase by using a new highspecific inhibitor (NS-196-CTSB-inhibitor) of cathepsin B. My results confirm the redistribution of CTSB from the lysosomal fraction under physiological conditions in to the zymogen-granule enriched fraction already after 1h of pancreatitis. Furthermore a significant increase of total activity in the early time of the disease was shown. In contrary CTSB protein content remained more or less unchanged in those three subcellular compartments within the same period of time and increased only after 8 h. Besides I found that the processing of CTSB mostly takes place in the lysosomal and secretory compartment and that it does not increase during acute experimental pancreatitis. In the cytosol almost no activity and a few processed CTSB was measured in spite of relevant amounts of protein content. These findings lead to conclude that the redistribution of CTSB activity is nether based on a shift of protein in the compartments nor on a different processing of the enzyme, but on an increase of activity in relation to the same amount of processed CTSB. In so far there is no redistribution of protein content into the secretory compartment and no different processing in the same, but an increase of activity. Possible reasons for this could be a change of biophysical conditions (e.g. pH, Ca++) or a modification in the content of physiological CTSB inhibitors (e.g. cystatins).
