Nachwirkung von in die Subkultur verschleppten Wirkstoffspuren ausgeschaltet werden, weil andernfalls eine zu hohe Wirksamkeit vorgetäuscht wird. Deshalb wurde die Neutralisierung der oberflächenaktiven kationischen Antiseptika Chlorhexidindigluconat, Octenidindihydrochlorid und Polyhexamethylenbiguanidinhydrochlorid durch Lipofundin MCT 20 % bei Einsatz von E. coli, S. aureus, P. aeruginosa, E. hirae und C. albicans gemäß DIN EN 13727 untersucht. Zu 8 Volumenanteilen Inaktivator (Lipofundin) wurden zunächst 1 Volumenanteil Wirkstofflösung hinzugefügt. Nach 5 min Kontakt bei Raumtemperatur wurde dann die Mischung mit 1 Volumenanteil Prüfmikroorganismen mit einer Dichte von 104 KbE/ml versetzt, gründlich gemischt und anschließend die vorhandenen KbE quantifiziert. Um die Neutralisierungskapazität von Lipofundin genauer zu bestimmen, wurde abweichend von der Empfehlung der Norm, wonach die KbE-Anzahl nach Neutralisierung unter der Hälfte der KbE-Anzahl in der Kontrolle betragen muss, als Maßstab ein fehlender signifikanter Unterschied zwischen beiden Ansätzen zugrunde gelegt. Die Ergebnisse zeigen, dass CHX, OCT und PHMB durch die Bindung an die Phospholipidschicht des Lipofundins konzentrationsabhängig neutralisiert wurden, was zu einem Weiterwachstum von E. coli, S. aureus, E. hirae, P. aeruginosa und C. albicans geführt hat. Damit erwies sich Lipofundin als effektiver Neutralisator. Im Vergleich zu CHX und OCT war der Konzentrationsbereich für die Neutralisierung bei PHMB allerdings kleiner. Daraus lässt sich umgekehrt ableiten, dass PHMB in Gegenwart von Phosphatidylcholin weniger an mikrobiozider Wirkung einbüßt als OCT und CHX, was für die antiseptische Anwendung vorteilhaft sein könnte. N2 - While examining microbiocidal agents in a quantitative suspension test, the so-called “after-effect” of contamination by traces of agents introduced into the subculture has to be eliminated; otherwise, the test will indicate an effectiveness higher than it really is. Therefore, the neutralizing effects of Lipofundin MCT 20 % on the cationic surfactant antiseptics chlorhexidine digluconate (CHX), octenidine dihydrochloride (OCT), and poly hexamethylene biguanide hydrochloride (PHMB) were examined with E. coli, S. aureus, P. aeruginosa, E. hirae, and C. albicans in accordance with DIN EN 13727. First, 1 volume fraction of active agent solution was added to 8 volume fractions of the inactivator (Lipofundin). After 5 min of contact at room temperature, the solution was mixed with 1 volume fraction of diagnostic microorganisms with a concentration of 104 CFU/ml; then, the existing CFU were quantified. In order to determine the neutralizing capacity of Lipofundin more precisely, and deviating from the recommendation of DIN EN 13727 that the number of CFU after neutralization should be less than half of the number of CFU in the control probe, a missing significant difference between both approaches has been chosen as standard. The results show that CHX, OCT, and PHMB were neutralized irrespective of their concentration by bonding to the phospholipid layer of the Lipofundin, which led to a continued growth of E. coli, S. aureus, E. hirae, P. aeruginosa, and C. albicans. Thus, Lipofundin proved to be an effective neutralizing agent. The concentration range for neutralizing PHMB was smaller than that for CHX and OCT. This indicates that in the presence of phosphatidylcholine, PHMB loses less of its microbiocidal effectiveness than OCT and CHX, which could be of advantage for antiseptic application.
