Modelle des Molybdän-Cofaktors mit von Pterin abgeleiteten Dithiolen-Liganden und Untersuchungen zum Ersatz von Molybdän durch Rhenium

  • Aufgrund der extremen Instabilität des Molybdän Cofaktors (MoCo) ist eine genauere Untersuchung der aktiven Zentren der lebenswichtigen MoCo-abhängigen Enzyme allein durch biochemische Methoden fast unmöglich. Hierfür liefert eine chemische Modellierung des Cofaktors die einzige Möglichkeit einen tieferen Einblick in seine Struktur und Funktion. Die vorliegende Dissertation ermöglicht einen weitaus tieferen Einblick in Struktur-Funktionsbeziehung des Molybdän-Cofaktors hinsichtlich des zentralen Metalls und des Molybdopterin-Liganden. Zunächst wurde die Rolle des Molybdänzentrums in den Modellverbindungen detailliert analysiert. Hierfür wurde in den synthetisierten Modellen Molybdän mit Rhenium, ausgetauscht. Die erhaltenen Komplexe wurden zuerst umfangreichend durch verschiedene Methoden Kristallstrukturanalyse, IR-, Raman-, NMR-, 2D-NMR-Spektroskopie, temperaturabhängige Elektrochemie und quantenchemischen Berechnungen analysiert und auf Analogien und Unterschiede verglichen. Dabei wurde auf der Suche eines MoCo-Modells, das die richtige Balance zwischen katalytischer Aktivität und Stabilität besitzt, untersucht, ob Rhenium eine potenzielle Alternative zu Molybdän darstellen kann. Um einen tieferen Einblick in die Chemie des Pterin-Strukturabschnitts von MoCo zu erschaffen, beschäftigt sich diese Arbeit mit der Feinabstimmung von Chinoxalin- und Pterin-Dithiolen-Liganden sowie mit der Entwicklung deren Molybdän-Komplexen. Dazu konnten neuartige Chinoxalin- und Pterin-Dithiolen-Liganden synthetisiert werden, die als Modell-Liganden für die Erforschung der Biosynthese des MoCos fungieren können. Hierin wird die Synthese und die vollständige Charakterisierung eines neuartigen Oxo-Bis(pterin)dithiolen-Molybdän-Komplexes beschrieben. Durch 2D-NMR Spektroskopie konnte die Struktur des erhaltenen Komplexes in Lösung detailliert analysiert werden. Schließlich wurden im Rahmen der vorliegenden Arbeit erstmals durchgeführte Untersuchungen zur Bindung von chemisch synthetisierten MoCo-Modellen mit dem Apoenzym der Trimethylamin-N-Oxid-Reduktase unternommen. Dabei konnte die essenzielle Rolle des Pterin-Gerüstes für die richtige Platzierung des Cofaktors in der Bindungstasche des Apoenzyms etwas näher aufgeklärt werden. Zukünftig könnten noch strukturell genauere MoCo-Modelle den Weg für die Synthese einer semi-artifiziellen Sulfitoxidase, die als eine Behandlungsmöglichkeit der Molybdän-Cofaktor-Defizienz (MoCoD) und der isolierten Sulfitoxidase-Defizienz (iSOD) eingesetzt werden, eröffnen.
  • Due to the oxygen-sensitive nature of the molybdenum cofactor (MoCo), a comprehensive investigation of the physiologically essential MoCo-dependent enzymes by pure biochemical methods is nearly impossible. The field of bioinorganic modeling represents the only way to gain a deeper insight into the structure and function of MoCo. Aim of the present dissertation was to provide a deeper insight into the structure-function relationship of the molybdenum cofactor with a focus on the metal center and the molybdopterin ligand. In order to investigate the role of the natural cofactor´s metal center, molybdenum was replaced by rhenium in the synthesized models. The obtained Mo/Re complex pairs were comprehensively studied and investigated comparatively by X-Ray crystallography, IR-, Raman-, NMR, 2D NMR spectroscopy, temperature-dependent cyclic voltammetry and quantum chemical calculations with a focus on the analogy and differences between both metals. In the search for a structural and functional MoCo model, with optimum balance between stability and catalytic activity, it was examined whether rhenium could potentially be a suitable alternative to molybdenum. In order to better understand the role of the pterin unit in the natural cofactor, this work deals with the fine-tuning of quinoxaline- and pterin-dithiolene ligands and with the development of their molybdenum complexes. Novel quinoxaline- and pterin-dithiolene ligands were synthesized and completely characterized. These ligands are appropriate models for the investigation of the MoCo-biosynthesis. More importantly, this work describes the synthesis and full characterization of a novel oxo-bis(pterin)dithiolene-molybdenum complex. The structure of the synthesized complex could extensively be analyzed in solution by 2D-NMR spectroscopy. Finally, for the very first time, studies on the binding of chemically synthesized MoCo models with the apoenzyme of trimethylamine N-oxide reductase were carried out. The essential role of the pterin moiety for the correct positioning of the cofactor into apoenzyme´s binding pocket was closer elucidated. In the future, more structurally accurate MoCo models could open the way for the development of a semi-artificial sulfite oxidase, which could be applied in the treatment for the molybdenum cofactor deficiency (MoCoD) and the isolated sulfite oxidase deficiency (iSOD).
