FRET-Interaktionsstudie zum Androgenrezeptor in Prostatakarzinomzellen

  • Das Prostatakarzinom ist bei Männern in Deutschland die häufigste Krebserkrankungen und die dritthäufigste Krebstodesursache. Aufgrund der steigenden Inzidenz mit fortschreitendem Alter und der parallel dazu immer älter werdenden Bevölkerung ist diese Erkrankung von großer sozioökonomischer Bedeutung. Nach einer antiandrogenen Therapie im fortgeschrittenen Tumorstadium kommt es häufig zur erneuten Progression des Tumorwachstums. In diesem kastrationsresistenten Stadium kommt es zur hormonunabhängigen Aktivierung und Aufrechterhaltung von Signalwegen des Androgenrezeptors. Ein in diesem Zusammenhang postulierter Mechanismus besteht in der direkten Wechselwirkung des Rezeptors mit dem Tumorprotein PC1 (prostate and colon gene-1), welches auch als Tumorprotein D52 bezeichnet. Es liegt in Prostatakarzinomzellen häufig überexprimiert vor und steht daher im Verdacht, maßgeblich zur Progression des Tumorwachstums beizutragen, insbesondere im kastrationsresistenten Stadium. In der vorliegenden Arbeit sollten mit Hilfe von FRET-basierten Interaktionsstudien mögliche Wechselwirkungen von PC1 mit dem Androgenrezeptor untersucht werden. Da es bereits Hinweise für eine Interaktion von PC1 mit Prdx1 gab, wurde dieses Protein in die Analyse mit einbezogen. Die Untersuchungen wurden an den Prostatakarzinom- zelllinien LNCaP und PC-3 unter verschiedenen Kulturbedingungen durchgeführt. Die verwendeten Methoden umfassten die Klonierung von Expressionsvektoren, Transfektion von Plasmid-DNA in Säugerzellen, indirekte Immunfluoreszenz, Westernblot-Analysen, Durchflusszytometrie sowie konfokale Mikroskopie. In dieser Arbeit konnte eine Kolokalisation zwischen PC1 und dem Androgenrezeptor sowie zwischen PC1 und Prdx1 im Zytoplasma von androgenabhängig wachsenden LNCaP-Zellen gezeigt werden. Auch in PC-3-Zellen, welche ähnlich wie kastrationsresistente Zellen androgenunabhängig wachsen, gelang es, eine Kolokalisation zwischen PC1 und dem Androgenrezeptor im Zytoplasma nachzuweisen. Bei Kultivierung dieser Zellen in hormonfreiem Medium waren PC1 und der Androgenrezeptor sogar im Zellkern von PC-3-Zellen kolokalisiert. Die Ergebnisse dieser Arbeit deuten auf die besondere und zentrale Funktion des PC1 bei der Aufrechterhaltung des Tumorwachstums hin. Somit wäre das prostataspezifisch vorkommende PC1 eine durchaus interessante Zielstruktur für die Entwicklung neuer Therapiestrategien beim fortgeschrittenen Prostatakarzinom.
  • Prostate carcinoma is the most common cancer in men in Germany and the third most frequent cause of cancer death. Due to the increasing incidence with advancing age and the parallel ageing of the population, this disease has great socio-economic importance. After antiandrogenic therapy in the advanced tumor stage, tumor growth often regresses. In this castration-resistant stage, hormone-independent activation and maintenance of signaling pathways of the androgen receptor occurs. A mechanism postulated in this context is the direct interaction of the receptor with the tumor protein PC1 (prostate and colon gene-1), also known as tumor protein D52. It is often overexpressed in prostate cancer cells and is therefore suspected to contribute significantly to the progression of tumor growth, especially in the castration-resistant stage. In the present study, FRET-based interaction studies were conducted to investigate possible interactions of PC1 with the androgen receptor. There was already evidence for an interaction of PC1 with Prdx1, therefore this protein was included in the analysis. The investigations were performed on the prostate carcinoma cell lines LNCaP and PC-3 in different culture conditions. The used methods included cloning of expression vectors, transfection of plasmid DNA into mammalian cells, indirect immunofluorescence, western blot analysis, flow cytometry and confocal microscopy. In this work colocalization between PC1 and the androgen receptor and between PC1 and Prdx1 in the cytoplasm of androgen-dependent growing LNCaP cells could be shown. Also in PC-3 cells, which behave like castration-resistant cells and grow androgen-independently, colocalization between PC1 and the androgen receptor in the cytoplasm could be detected. When these cells were cultivated in hormone-free medium, PC1 and the androgen receptor were even colocalized in the nucleus of PC-3 cells. The results of this work indicate the special and central function of PC1 in the maintenance of tumor growth. Thus, the prostate specific PC1 would be a very interesting target structure for the development of new therapeutic strategies in advanced prostate cancer.
