Ribozymes for Aminoacylation

  • Aminoacyl-tRNA synthetases (aaRS) are at the heart of modern translation, catalyzing the accurate biosynthesis of aminoacyl-tRNAs. According to the RNA world hypothesis, the early translation system should have aminoacylation ribozymes for RNA aminoacylation. For this, an aaRS ribozyme system, consisting of the KK13 ribozyme and the C3a ribozyme was successfully designed, which can perform both amino acid activation and aminoacyl transfer reaction. Generation of such aminoacylation ribozyme system would fill up the gap between the RNA world and the modern biological world. In addition, two types of diversified aminoacylation ribozymes, symmetrical ribozymes and self-assembling ribozymes were successfully developed, which may have great meaning in the origin of life.
  • Aminoacyl-tRNA-Synthetasen (aaRS) könnten auch als das Herz der biologischen Translation bezeichnet werden: Sie katalysieren die akkurate Biosynthese der Aminoacyl-tRNAs. Laut RNA-Welt-Hypothese wurde die Aminoacylierung von RNAs zuvor von Ribozymen übernommen. Aus diesem Grund wurde ein System, bestehend aus dem KK13- und dem C3a-Ribozym, entwickelt. Dieses System kann sowohl die Aminosäure-Aktivierung als auch den Aminoacyltransfer katalysieren. Die Herstellung solcher Ribozymsysteme könnte ein Brücke zwischen RNA-Welt und der modernen biologischen Welt darstellen. Darüber hinaus wurden zwei weitere Arten von Aminoacylierungsribozymen, symmetrische Ribozyme und selbst-organisierende Ribozyme, erfolgreich entwickelt. Diese haben möglicherweise eine große Bedeutung in der Theorie zur Entstehung des Lebens – dem "Origin of Life".
