Evaluation eines orthotopen Pankreastumormodells in der Maus durch in vivo Fluoreszenz-Imaging und MRT

  • Das Pankreasadenokarzinom zeichnet sich durch aggressives Wachstum, frühe Metastasierung, späte Diagnosestellung und vermindertes Ansprechen auf zytostatische Therapien aus. Ziel der vorliegenden Arbeit ist zum Einen die Etablierung und Validierung eines Ganzkörper-Fluoreszenz-basierten Nachweises von Tumorwachstum und Metastasierung im Mausmodell als mögliche Basis für darauf aufbauende präklinische Pankreaskarzinom-Studien. Zum Anderen werden Vor- und Nachteile im Vergleich zum Schnittbildverfahren eines 7-Tesla-Kleintier-MRT untersucht. Nach stabiler Transfektion der murinen Adenokarzinom-Zelllinie Panc 02 mit eGFP-, RFP- und Lumineszenz-Expressionsvektoren wurde deren Fluoreszenz-/Lumineszenz-, Proliferations- und Apoptoseverhalten in vitro evaluiert. Nach orthotoper Injektion in den Pankreaskopf wurde das Tumorwachstum und Metastasierungsverhalten in vivo über einen Zeitverlauf von 21 Tagen mittels Ganzkörper-Fluoreszenz-Imaging und MRT untersucht. Ergänzend erfolgten im Anschluss entsprechende Analysen zu Proliferation und Apoptose in situ anhand der explantierten histologisch und immunhistochemisch aufgearbeiteten Organe. Die murine Panc 02 Adenokarzinom-Zelllinie konnte mit einer Proliferationsrate von 88,4 % bei 0 % Apoptose in vitro als besonders aggressiv wachsende Adenokarzinom-Zelllinie charakterisiert werden. Die Ergebnisse änderten sich auch in situ kaum. Während das Fluoreszenz-Imaging hochspezifisch, kostengünstig, untersucherunabhängig und einfach auswertbar progredientes lokales Tumorwachstum mit deutlicher peritonealer Metastasierung nachweisen konnte, zeigte das MRT eine bessere anatomisch-topographische Auflösung ohne Interferenz mit Haut, Haaren und Narbengewebe bei jedoch geringerer Sensitivität und Spezifität. Mit Hilfe der Fluoreszenz-Kryo-Histologie konnte anschließend ein Tumornachweis bis auf Einzelzellebene erbracht werden. Zwischen den beiden bildgebenden Verfahren des Ganzkörper-Fluoreszenz-Imagings und MRTs konnte ein komplementäres Bild aufgezeigt werden.
  • Pancreatic cancer is characterized by aggressive growth, early metastases, late diagnosis and reduced response to cytostatic therapies. The aim of this thesis is on one hand to establish and validate a detection of tumor growth and metastases based on whole-body-fluorescence in a mouse model serving as a possible foundation for constitutive preclinical pancreatic cancer studies. On the other hand, this thesis should depict advantages and disadvantages in comparison to the sectional images of a 7-tesla-small-animal-MRI. After stable transfection of the murine adenocarcinom cell line Panc 02 with eGFP, RFP and luminescence expression vectors, the fluorescence, luminescence , proliferation and apoptosis in vitro were evaluated. After orthotopic injection in the head of the pancreas, the tumor growth and the metastatic process in vivo over a time period of 21 days were examined by whole-body-fluorescence-imaging and MRI. In addition, explanted organs, which were histologically and immunhistochemically processed, were analysed regarding their proliferation and apoptosis in situ. The murine adenocarcinom cell line Panc 02 was characterized as a very aggressive adenocarcinom cell line with a proliferating rate of 88,4 % with 0 % apoptosis in vitro. The in situ results were nearly the same. While fluorescence imaging detects local tumor growth and obvious peritoneal metastasis highly specific, low prized, examiner-independent and allows easy analysis; MRI shows a better anatomic-topographical resolution without interference of skin, hairs or scar tissue but with less sensitivity and specificity. With fluorscence kryo histology, tumor detection of single cells is possible afterwards. Thus, whole-body-fluorescence-imaging and MRI are two different imaging techniques of complementary use.
