Etablierung eines Expressionsmodels für cKir3.4 in E.coli zur Analyse der Rolle von cKir3.4 bei der ischaemischen Praekonditionierung

  • Die ischämische Präkonditionierung ist ein endogener kardioprotektiver Schutzmechanismus, vermittelt über kurze Phasen von Ischämie und Reperfusion. Dieser wird unter anderem durch die Öffnung von ATP-sensitiven Kaliumkanälen der inneren Mitochondrienmembran vermittelt. In der vorliegenden Arbeit wurde die zytosolische Domäne (cKir3.4) der Untereinheit Kir3.4 des kardialen, ATP-sensitiven Kaliumkanals untersucht. Ausgehend von der cDNA für die zytosolische Domäne der Untereinheit Kir3.4 (cKir3.4) wurden Protokolle zur Expression als 6xHis-tag-Fusionsproteinsowie als GST-Fusionsprotein inEscherichia coli etabliert.Es zeigte sich eine Lokalisation von über 90% des rekombinanten Proteins in Einschlusskörpern, nur 10% lagen in den löslichen Fraktionen vor. Nach Modifikation der Aufreinigungsbedingungen und der Proteingröße konnte via Affinitätschromatographie rekombinantes Protein in quantitativem Maßstab gewonnen werden. Ferner wurde im Labor ein Präkonditionierungsmodell für Rattenherzen etabliert und im anschluss für die Phase der Präkonditionierung spezifische Proteine isoliert. Zudem wurde ein polyklonaler Antikörper im Kaninchen gegen cKir3.4 generiert.
  • Ischemic preconditioning is an important endogenous mechanism of cardioprotection, transmitted by short phases if ischemia and reperfusion. One of the milesotnes of the transmission of this protection mechanism is the opening of ATP-sensitive potassium channels in the inner mitochondrial membrane. Thus thesis analyses the cytosolic domain (cKir3.4) of the ATP-sensitive potassium channel in the heart. STarting with the cDNA of cKir3.4 a protocoll for the expression as a fusionprotein with 6xHis-tag-Fusionprotein and GST in Escherichia coli was established. We find a localisation of more then 90% of the recombinant protein in inclusion bodies, just 10% were soluble. After modification of purification-protocolls and the proteinsize it was possible to achieve via affinity-chromatografie recombinant protein in quantitative measures. Further we established a modell for preconditioning of rat hearts in the laboratory and isolated proteins for each phase of preconditioning. Also we generated a polyclonal antibody in rabbits.
