Regulation und Funktionalität des Hitzeschockproteins HSP60 in Prostatakarzinomzellen

  • Tumorbiologie und molekulare Mechanismen der Progression des Prostatakarzinoms (PCa) sind komplex und bisher nur unzureichend verstanden. Trotz einiger medikamentöser Ansätze zur Therapie des PCa mit teilweise neuartigen Wirkstoffen spielen primäre und sekundäre Therapie-Resistenzen bei der Behandlung eine große Rolle. Ziel dieser Arbeit war die Untersuchung des apoptotischen Faktors Hitzeschockprotein HSP60 in PCa-Zellen, um potentielle Signalkaskaden und Effektormoleküle einer HSP60-abhängigen PCa-Progression aufzudecken. Durch massenspektrometrische Analysen von potentiellen HSP60-Bindungspartnern wurden mit den Proteinen Peroxiredoxin-1 (Prx-1), Peroxiredoxin-2 (Prx-2), Hitzeschockprotein HSP70, Poly(ADP-ribose)-Polymerase (PARP) sowie Prohibitin (PHB) Kandidaten identifiziert, die bei Chemoresistenz wie auch bei proliferativen und apoptotischen Prozessen eine Rolle spielen können. Jedoch konnten durch Western Blot Analysen der Bindungs-Experimente nur Prx-1 und PHB als HSP60-Bindungspartner bestätigt werden. Die Modulation der HSP60-Expression mittels klonierter Überexpressions- und Knock-Down-Vektoren schlug aufgrund einer mutmaßlichen starken und sehr raschen zellulären Gegenregulation fehl. Dies kann als Hinweis darauf gedeutet werden, dass es sich bei HSP60 um einen essentiellen zellulären Faktor mit einer sehr stringenten Expressions-Kontrolle handeln könnte. Analysen der HSP60-Expression nach Modulation der Syntheseraten von miR-1 zeigten eine deutliche Induktion des Hitzeschockproteins durch die MikroRNA. Die Frage, ob HSP60 in PCa-Zellen pro- oder anti-apoptotisch wirkt, konnte mangels effizienter Modulation der HSP60-Expression nicht ausführlicher untersucht werden. Da der positive HSP60-Regulatior miR-1 jedoch als Tumorsuppressor beschrieben wird und eine Protein-Protein-Interaktion von HSP60 mit den pro-apoptotischen Proteinen Prx-1 und PHB nachgewiesen werden konnte, kann man die begründete Annahme formulieren, dass HSP60 in PCa-Zellen pro-apoptotische Funktionen ausführt.
  • Tumor biology and molecular mechanisms of progression of prostate cancer (PCa) are complex and not yet adequately understood. Despite some pharmacological approaches to the treatment of PCa with partially novel drugs primary and secondary therapy-resistance play a major role in the treatment. The aim of this work was to investigate the apoptotic factor heat shock protein 60 (HSP60) in PCa-cells to reveal potential signaling pathways and effector molecules of HSP60-dependent PCa progression. By mass spectrometric analysis of potential HSP60 binding partners were the proteins Peroxiredoxin-1 (PRX 1) Peroxiredoxin-2 (Prx-2), heat shock protein HSP70, poly (ADP-ribose) polymerase (PARP) and prohibitin (PHB) identified as candidates that may play a role in chemoresistance as well as in proliferative and apoptotic processes. However only Prx-1 and PHB could be confirmed by Western blot analysis as HSP60-binding partners. The modulation of HSP60 expression by cloned overexpression and knockdown vectors failed because of a suspected strong and very rapid cellular counter-regulation. This can be interpreted as an indication that HSP60 might be an essential cellular factor with a very stringent expression control. Analysis of HSP60 expression by modulating the synthesis rate of miR-1 showed a significant induction of the heat shock protein by the microRNA. The question of whether HSP60 acts pro- or anti-apoptotic in PCa-cells, could not be examined in detail for lack of efficient modulation of HSP60 expression. Since the positive HSP60-Regulatior miR-1 is described as a tumor suppressor and a protein-protein interaction of HSP60 with the pro-apoptotic proteins Prx-1 and PHB could be detected, one can formulate a founded assumption that HSP60 performs pro-apoptotic features in PCa-cells.
