Untersuchungen zur gemeinsamen zytotoxischen Wirkung von TRAIL und Docetaxel in humanen endometrialen Adenokarzinomzellen

  • In dieser Arbeit konnte gezeigt werden, dass die zytotoxischen Effekte von TRAIL und Docetaxel auf endometriale Adenokarzinomzellen unter kombinierter Stimulation stärker waren, als unter Einzelstimulation. TRAIL ist für seine selektive Induktion von Apoptose in Tumorzellen bekannt, ohne dem normalen Gewebe zu schaden. Doch obwohl für alle fünf untersuchten Zelllinien eine Expression der Todesrezeptoren DR4 und 5 nachgewiesen werden konnte, induzierte TRAIL allein kaum Apoptose. In Kombination mit Docetaxel, dem halbsynthetischen Analogon von Paclitaxel, zeigte sich eine größere Zytotoxizität als unter Docetaxel und TRAIL allein. Es kam zu einer signifikanten Reduktion der Zellzahl sowie zu einem Anstieg der Apoptose- und Nekroserate. Hierbei deuteten sich synergistische Wirkweisen an. Eine ursächliche Veränderung des Rezeptorprofils durch Docetaxel war nicht zu erkennen. Die Schnittstelle liegt möglicherweise auf einer anderen Ebene. Die Freisetzung der Chemokine IL-8, MCP-1 und RANTES wurde durch Docetaxel, TRAIL und besonders stark durch die Kombination von beiden stimuliert. Dies könnte sich sowohl positiv als auch negativ auf die Tumorprogression auswirken, insbesondere im inflammatorischen Kontext und in vivo. Hier wären weitere Untersuchungen indiziert. Insgesamt unterstreichen diese Ergebnisse, dass eine Kombination von TRAIL bzw. agonistischen TRAIL-Rezeptorantikörpern mit Taxanen das Potential hat, in der Therapie von Endometriumkarzinomen eine Rolle zu spielen.
  • In this paper we show that the cytotoxic effects on endometrial adenocarcinoma cells triggered by a combination of Trail and Docetaxel were stronger than those in the case of stimulation with each one separately. TRAIL is known for its selective induction of apoptosis in cancer cells while not negatively affecting normal tissue. However, even though expressions of the death receptors DR4 and 5 were proven in all of the five investigated cell lines, TRAIL alone did rarely induce apoptosis in these cells. In combination with Docetaxel, being the semisynthetic analogue to Paclitaxel, more cell death occurred than under the treatment with Docetaxel or TRAIL alone. We could demonstrate a significant reduction in the number of vital cells, as well as an increase in apoptosis and necrosis, which may indicate synergistic effects. There was no causal change in the TRAIL receptors’ expression due to Docetaxel detectable; the cause might be found on a different stage. The release of the chemokines IL-8, MCP-1 and RANTES was stimulated by TRAIL, Docetaxel and even more by a combination of both. This could have positive as well as negative effects on the tumors´ progression, especially in vivo and with regard to the inflammatory context. These impacts demand further investigation. All in all, these results show that the combination of TRAIL or agonistic TRAIL receptor antibodies and taxanes might have the potential to play an important role in endometrial cancer therapy.
