Proteininteraktionen von thrombozytären Transportproteinen

  • Das MRP4 ist ein Mitglied der ABCC-Subfamilie der ATP-binding cassette transporters. Es transportiert eine große Vielfalt an endogenen und xenobiotischen Verbindungen aus der Zelle. Des Weiteren vermittelt MRP4 den Transport von Signalmolekülen wie z.B. zyklische Nukleotide. Das einzigartige Substratspektrum, die Regulation und die zelluläre Lokalisation des MRP4 stehen in Verbindung mit seiner möglichen Funktion beim Zell-Schutz und dem zellulären Signaling. MRP4 ist abhängig vom Zelltyp entweder in der basolateralen (Prostata, Leber) oder apikalen (Nieren, Kapillaren des Gehirns) Membran von polarisierten Zellen lokalisiert. Des Weiteren wird MRP4 auch in Thrombozyten und Erythrozyten exprimiert. Protein-Protein-Interaktionen können die Funktion, Lokalisation und Expression von Transportern in der Plasmamembran beeinflussen. Viele Interaktionen beinhalten die stabile Assoziation von Proteinen innerhalb von Multi-Untereinheitskomplexen sowie die vorübergehende Assoziation von regulatorischen Proteinen. MRP4 weist u.a. ein sogenanntes PDZ-Bindemotiv auf, welches durch die Aminosäuren ETAL charakterisiert wird und die Interaktion mit PDZ-Adaptorproteinen vermitteln kann. Speziell in Thrombozyten ist MRP4 neben der Plasmamembran auch in den δ-Granula lokalisiert. Hier könnten interagierende Proteine eine wichtige Rolle spielen. Änderungen in diesen Proteinen könnten eine Ursache für Störungen der Thrombozytenfunktion mit einer Fehllokalisation von MRP4 sein. Ziel der vorliegenden Arbeit war die Identifizierung möglicher Interaktionspartner von MRP4, insbesondere in Thrombozyten. Dafür wurde ein Fusionsprotein aus einem c-terminalen Fragment (117 Aminosäuren) des MRP4 mit der Glutathion-S-Transferase (GST) generiert. Nach Aufreinigung des Fusionsproteins (GST-MRP4) mittels Glutathion-Sepharose wurden pull-down-Experimente mit Thrombozytenlysat durchgeführt. Mittels Western Blot und LC-MS/MS-Analyse konnten als mögliche Interaktionspartner das EBP50/NHERF1, das PSD95, SNX27, die β-Untereinheit des AP3B1 und das HSP90 identifiziert werden. Durch indirekte Immunfluoreszenz konnte außerdem eine partielle Ko-Lokalisation der genannten möglichen Interaktionspartner und MRP4 in den Thrombozyten dargestellt werden. Ein weiterer Ansatz zur Untersuchung der Protein-Interaktion war die Ko-Präzipitation des MRP4 und der daran gebundenen Proteine mittels eines anti-MRP4-Antikörpers, der an magnetische beads gebunden war. Mithilfe der Ko-Immunpräzipitation konnten SNX27, HSP90, AP3B1, EBP50 und PSD95 unterschiedlich stark ko-präzipitiert werden. Es wurde untersucht, inwiefern das PDZ-Bindemotiv des MRP4 für die Interaktion der detektierten Interaktionsproteine essentiell ist. Dafür wurde ein GST-MRP4-Konstrukt ohne das C-terminale PDZ-Motiv generiert. Damit konnte gezeigt werden, dass das PDZ-Bindemotiv nur für die Interaktion mit SNX27, EBP50 und PSD95 notwendig ist, während die Bindung von AP3B1 und HSP90 unabhängig davon erfolgte. Nachdem auf Proteinebene mit verschiedenen Versuchen dargestellt werden konnte, welche Adaptorproteine mit dem MRP4 interagieren, sollte im letzten Abschnitt auf funktioneller Ebene gezeigt werden, inwiefern sich die Lokalisation des MRP4 verändert, sofern das Bindemotiv des MRP4 nicht mehr vorhanden ist bzw. die Adaptorproteine herunterreguliert werden. In Bezug auf das Herunterregulieren der Adaptorproteine wurde die megakaryoblastische Leukämie-Zelllinie M07e als Modell für Thrombozyten-Vorläuferzellen verwendet. Des Weiteren lag das Interesse bei den Interaktionsproteinen AP3, PSD95 und SNX27. Nach Transfektion der M07e-Zellen mit der entsprechenden siRNA und der sich anschließenden Immunfluoreszenz konnte gezeigt werden, dass die Ko-Lokalisation des MRP4 mit den Adaptorproteinen nach knock-down verringert und die Plasmamembran-Lokalisation von MRP4 signifikant gesteigert war. Umgekehrt sollte die Überexpression dieser Adaptorproteine die Plasmamembran-Lokalisation verringern. Dies wurde exemplarisch für SNX27 in MDCK-Zellen untersucht. Dabei sollte auch nochmals die Rolle des PDZ-Bindungsmotivs für die MRP4-Lokalisation gezeigt werden. Dafür wurden die Fluoreszenz-markierten Fusionsproteine CFP-MRP4, SNX27-YFP und das CFP-MRP4(-PDZ) synthetisiert, in MDCK-Zellen transfiziert und ihre Lokalisation mittels konfokaler Fluoreszenzmikroskopie analysiert. Es zeigte sich, dass nach Ko-Transfektion des CFP-MRP4 mit SNX27-YFP das MRP4 hauptsächlich im Zell-Inneren lokalisiert war. Außerdem wurde verdeutlicht, dass das PDZ-Bindemotiv für die Internalisierung des MRP4 in das Innere der Zelle durch das Interaktionsprotein SNX27 essentiell ist. Zusammenfassend liefert diese Arbeit wichtige grundlegende Erkenntnisse zu möglichen Protein-Interaktionen von MRP4 und deren Einfluss auf die Lokalisation des Transporters. Deren mögliche physiologische und pathophysiologische Rolle für die MRP4-Funktion in Thrombozyten sollte in weiterführenden Studien näher untersucht werden.
  • The Multidrug resistance protein 4 (MRP4 / ABCC4) is a member of the ABCC-Subfamily of the ATP-binding cassette transporters. It transports a wide range of endogenous and xenobiotic organic anionic compounds out of cells and mediates the transport of signal molecules e.g. cyclic nucleotides and eicosanoids. The unique spectrum of substrates, the regulation and the cellular localization of MRP4 connect to its possible function in cell protection and cellular signaling. The localization of MRP4 is dependent on the cell type. MRP4 is either in the basolateral (prostate, liver) or apical (kidneys, capillaries of the brain) membrane of polarized cells localized. In addition MRP4 is also expressed in platelets, erythrocytes, astrocytes and dendritic cells. Protein-protein interactions can influence the function, localization and expression of transporters in the plasma membrane. Many interactions involve a stable association of proteins within a multi-subunit complexes as well as the temporary association of regulatory proteins. MRP4 exhibit a PDZ-binding motif which is characterized by the C-terminale amino acids (ETAL) and mediate the interaction with PDZ-adaptor proteins. In platelets MRP4 is localized in the plasma membrane as well as in intracellular δ-granules where interacting proteins can play an important role. Modifications in these proteins could be a reason for platelet function disorders with a changed localization of MRP4. Therefore, the aim of this work was the identification of possible interaction partners of MRP4, especially in platelets. For this purpose, a recombinant fusion protein was generated which consists a carboxy-terminal fragment (117 amino acids) of the MRP4 with the glutathione-S-transferase (GST). Pull-down experiments with platelets lysate were executed after purification of the fusion protein (GST-MRP4) using glutathione-(GSH)-sepharose. By Western Blot and LC-MS/MS-analysis the interaction proteins ezrin-binding protein 50 (EBP50/NHERF1), postsynaptic density protein 95 (PSD95) and the sorting nexin 27 (SNX27) as well as the β-subunit of adaptor protein complex 3 (AP3B1) and the heat shock protein 90 (HSP90) could be identified. Furthermore a partial co-localization of the possible interaction proteins and MRP4 in platelets could be described by indirect immunofluorescence. An alternative approach to investigate the protein interaction was the co-precipitation of MRP4 and the binding proteins by anti-MRP4 antibody which was coupled to magnetic beads. SNX27, HSP90, AP3B1, EBP50 and PSD95 were co-precipitated differently. Subsequently we investigate if the PDZ-binding motif of MRP4 is essential for interaction of the detected interacting proteins by generating the GST-MRP4 construct lacking the C-terminale PDZ-motif. It could be demonstrated that the PDZ-binding motif is essential for interaction of SNX27, EBP50 and PSD95. Binding of AP3B1 and HSP90 occur without PDZ-binding motif. After demonstrating on protein level which adaptor proteins interact with MRP4 on functional level could be shown that the MPR4 localization is changed if the binding motif of MRP4 isnt available and respectively the adaptor proteins are down regulated. The megakaryoblastic leukemia cells M07e were used as model of platelets progenitor cells. Furthermore we are interested in the interacting proteins AP3, PSD95 and SNX27. The co-localization of MRP4 with the adaptor proteins was reduced after knock-down and the plasma membrane localization of MRP4 was significantly increased after transfection of M07e cells with siRNA. In contrast overexpression of these adaptor proteins should decrease the plasma membrane localization. This prediction was examined exemplarily for SNX27 in MDCK cells and the importance of the PDZ-binding motif for the MRP4 localization should also be demonstrated. Fluorescence-labeled fusion proteins CFP-MRP4, SNX27-YFP and CFP-MRP4(-PDZ) were synthesized, transfected in MDCK cells and their localization were analyzed by confocal fluorescence microscopy. When CFP-MRP4 was co-transfected with SNX27-YFP MRP4 was localized on intracellular structures. In addition the PDZ-binding motif is essential for the internalization of MRP4 by the interacting protein SNX27. In conclusion this study provides important evidences for possible protein interactions of MRP4 and the effect of transporter localization. Further studies are required to investigate their physiological and pathophysiological role for the MRP4 function in platelets.
