Neurotoxizitätsprüfung ausgewählter Desinfektionsmittel und Antiseptika im Labyrinthversuch mit Ratten

  • Das medizinische Personal, speziell das Chirurgische, kommt kontinuierlich mit Desinfektionsmitteln und Antiseptika in Kontakt. In bisherigen Untersuchungen werden Langzeitwirkungen vernachlässigt. Deshalb haben wir die neurotoxischen Nebenwirkungen handelsüblicher Desinfektionsmittel und Antiseptika bei dermaler Exposition an Ratten untersucht. Die bestimmten Parameter waren die Fehleranzahl und die Laufzeit im Labyrinthversuch. Wir exponierten die Tiere mit Wasser, Hexachlorophen, Spitaderm (Chlorhexidin), Betaisodona (Polyvidon-Iod), Octenisept (Octenidin), Poly Alcohol Hände Antisepticum (Iso-Propanol). Die Tiere sind 11d konditioniert worden, 90 d exponiert und 35 d nachbeobachtet worden. Die Exposition der Tiere ist 20 mal Höher als die Klinikexposition. Die Untersuchungen ergeben Ratten sind ein geeignetes Modell für verhaltensbiologische Analysen. Im Labyrinthversuch sind Lernverhalten und Gedächtnisfunktionen gut untersuchbar. Die pathohistologische Untersuchung ergibt Leberschäden, eine pathologische Veränderung im ZNS tritt nicht auf. Die Konditionierungsphase ergibt, daß die Leistungen der Tiere gleich waren, das heißt das psychomotorische Muster funktioniert. Die Nachbeobachtungszeit zeigt einen Rückgang der Fehleranzahl und der Laufzeit. Die Tiere zeigten während der Exposition eine Zunahme der Fehleranzahl und der Laufzeit. Eine neurotoxische Schädigung zeigt die Hexachlorophen -Gruppe, die Spitaderm - Gruppe zeigt eine Beeinflussung, Octenisept und Betaisodona- Gruppen zeigen eine geringe Zunahme von Fehlern und Laufzeit. Die Poly Alcohol Hände Antisepticum – Gruppe ähnelt der Wassergruppe.
  • Medical stuff, in particular surgical stuff, often has contact with desinfections and antiseptics. Since long term effects often are not taken into consideration in former studies, we examine the neurological side effects of common desinfectants and antiseptics with dermal exposition in rats. The following parameter are examined: running time and errors in a maze. We expone the rats with water (control), Hexachlorophene, Spitaderm (chlorhexidine), Octenisept (octenidine), Betaisodona (polyvidoniode) and Poly Alcohol Hände Antisepticum (iso-propanol). The exposition of the animals was about twenty times higher than the clinical exposition. The rats are conditioned during 11, exponed during 90 and observed for 35 days. The study shows that rats are an appropriate model for behavioural studies. The maze offers an excellent opportunity for examining learning and memorising. The patho-histological examination shows hepatic damages. Pathological modifications in the CNS, however, can not be determined. The conditioning phase shows that the capacity of all animals is equal. In other words, the psycho-motorical scheme is stable and reliable. The observing phase shows a decrease in running time and errors. During exposition the animals show an increase in running time and errors. The Hexaclorophene group shows a neurotoxical damage, the Spitaderm group shows an influence, witch needs an intensive control to exclude neurotoxical damages. The Octenisept, Betaisodona and Poly Alcohol Hände Antisepticum groups show the same results as in the water groups. Consequently there is no reference to neurotoxical risks.
