Expression der MikroRNA (miR-1) bei Patienten mit uterinen Leiomyosarkomen

  • Die uterine Leiomyosarkomen sind eine Gruppe von seltenen und aggressiven mesenchymalen Tumoren des Uterus. Die Diagnose dieser Tumoren wird oftmals erst zum Zeitpunkt der histo-pathologischen Befundung im Rahmen einer Hysterektomie gestellt. Klinisch ist es bisher nicht möglich eine Verdachtsdiagnose eines U-LMS zu stellen, da spezifische Symptome in den frühen Tumorstadien fehlen und keine geeigneten diagnostischen Methoden zur Verfügung stehen. Andererseits wird bei frühzeitiger Diagnose die Therapiewahl signifikant zum verbesserten Überleben der Patientin führen. Die komplette chirurgische Resektion des Uterus, d.h. Hysterektomie ohne Morcellment stellt die erste Therapiewahl dar und vermindert entscheidend das Rezidiv Risiko. Trotz einer erzielbaren kompletten Tumorresektion der U-LMS im frühen Tumorstadium, bleibt das Rezidiv Risiko hoch. Die Therapie der inoperablen Tumoren beinhaltet die systemische palliative Chemotherapie, die Target-Agents Therapie, Hormontherapie und die Radiotherapie. Das Ziel dieser Arbeit war den Stellenwert von nicht-kodierenden RNA-Moleküle, beim U-LMS zu unteruschen. In der Zelllinie SK-UT-1, die ähnlichen Charakteristika wie die U-LMS aufweist, wurde die Expression der microRNA miR-1 analysiert. Im Vergleich mit anderen 4-Zelllinien, zeigte die SK-UT-1 Zelllinie eine niedrigere miR-1 Expression. Die weiteren Untersuchungen der miR-1 Expression im gesundem Uterus-Geweben der Menschen, Gewebe des Herzmuskels von Mäusen und Gewebe der U-LMS ergab die höchste miR-1 Expression im Herzmuskel-Gewebe, die sich durch eine große Affinität der miR-1 für Muskel-Gewebe erklären lässt. Im Vergleich zwischen gesundem und U-LMS-Gewebe zeigte sich eine Überexpression in gesunden Geweben und eine erniedrigte Expression der miR-1 in U-LMS Gewebe. Diese Ergebnisse unterstreichen die Hypothese, dass die miR-1 Moleküle eine Rolle als Tumor-Inhibitoren bei der Genregulation der U-LMS spielen könnten. Des Weiteren zeigte, dass miR-1 keinen wesentlichen Einfluss auf die p38 und ERK-1,2 Expression zu haben scheint. Diese Proteine spielen eine wichtige Rolle bei der Apoptose und Stress-Antwort der Zellen auf externe Stimuli. Zusammenfassend, lässt sich festhalten, dass die miR-1-Molkülen vermutlich eine bedeutende Rolle bei der Entwicklung der U-LMS insbesondere bei der Transkription und Gen-Regulation zu haben scheinen.
  • The uterine leiomyosarcomas are a group of rare and aggressive mesenchymal tumors of the uterus. The diagnosis of these tumors is often made at the time of the histopathologic examination in the context of a hysterectomy. Clinically, it is not possible to diagnose a U-LMS, since specific symptoms are missing in the early tumor stages and no suitable diagnostic methods are available. On the other hand, in the event of an early diagnosis, the therapy will significantly improve the survival of the patient. Complete surgical resection of the uterus, is the Hysterectomy without morphology and is the first therapy option and significantly reduces the risk of recurrence. Despite the achievable complete tumor resection of the U-LMS in the early stage of the tumor, the recurrence risk remains high. The therapy of inoperable tumors includes systemic palliative chemotherapy, target agent therapy, hormone therapy and radiotherapy. The aim of this study was to evaluate the importance of non-coding RNA molecules in U-LMS. The expression of the microRNA miR-1 was analyzed in the cell line SK-UT-1, which has similar characteristics as the U-LMS. In comparison with other 4-cell lines, the SK-UT-1 cell line showed a lower miR-1 expression. Further studies of miR-1 expression in healthy uterine tissues of humans, tissues of the heart muscle of mice and tissues of U-LMS revealed the highest miR-1 expression in the heart muscle tissue, which is characterized by a great affinity of the miR-1 for Muscle tissue. In comparison to healthy and U-LMS tissue, overexpression in healthy tissues was observed and decreased expression of miR-1 in U-LMS tissues. These results underline the hypothesis that the miR-1 molecules might play a role as tumor inhibitors in gene regulation of the U-LMS. Furthermore, miR-1 does not appear to have a significant effect on p38 and ERK-1,2 expression. These proteins play an important role in the apoptosis and stress response of the cells to external stimuli. In summary, it can be said that the miR-1 molecules seem to have a significant role in the development of U-LMS, especially in transcription and gene regulation.
