Zelluläre Lokalisation und Translokation der Kationenkanäle TRPC3 und TRPC6 in isolierten Skelettmuskelfasern von mdx- und Kontrollmäusen

  • TRPC3 und TRPC6 sind Mitglieder der nicht selektiven Kationenkanäle aus der Superfamilie der „Transient Receptor Potential“-Kanäle (TRP-Kanäle). In der Erforschung der patho-physiologischen Abläufe der Duchenne Muskeldystrophie und des damit einhergehenden gestörten intrazellulären Kalziumhaushalts rückten besagte Kanäle als mögliche verursachende Kandidaten in den Fokus der Aufmerksamkeit. TRP-Kanäle weisen eine große Zahl unterschiedlichster Aktivierungsmechanismen auf, einer davon ist die Translokation. Diesen Mechanismus hat man für TRPC3 und TRPC6 bereits an verschiedenen Zellkulturen und Zelllinien feststellen können. Den Nachweis einer Translokation in ausdifferenzierten Skelettmuskelzellen gab es bisher nicht und war Gegenstand dieser Arbeit. An Skelettmuskelzellen von mdx-Mäusen sowie an solchen von Kontroll-Mäusen wurden geeignete Experimente zur Translokation der Kanäle durchgeführt. Dazu wurden die Zellen mit schon bekannten Stimulantien wie Gadolinium, EGF und Thapsigargin inkubiert. Immunfluoreszenzfärbungen der Kanalproteine ergaben interessante Neuigkeiten zur zellulären Lokalisation und Translokation von TRPC3 und TRPC6 in normalen und Dystrophin-defizienten (mdx) Muskelfasern.
  • TRPC3 and TRPC6 constitute non-selective cation-permeable ion channels of the plasma membrane and are members of the TRP (Transient Receptor Potential) superfamily of channels. The two channel proteins appear to be possible candidates causing the defective calcium regulation in dystrophin-deficient muscle fibres in Duchenne muscular dystrophy. TRP channels show a remarkable diversity in their activation mechanisms and regulation. Translocation to the plasma membrane seems to be one important mechanism. For TRPC3 and TRPC6, this mechanism has already been shown in a variety of cell cultures and cell lines. In differentiated skeletal muscle fibres translocation of the above mentioned cation channels has not been observed before and were investigated in this study, both for normal and mdx muscle fibres. To this end muscle fibres were incubated with well known stimulators of translocation to the plasma membrane, such as gadolinium, EGF and thapsigargin. Immunofluorescence staining of the channel proteins resulted in interesting new findings on the cellular localization and translocation of TRPC3 and TRPC6 in normal and dystrophin-deficient (mdx) muscle fibres.
