Auswirkung von TRAIL auf die Resistenz, die Sensitivität und das Wachstum von Pankreaskarzinomzelllinien

  • TRAIL ist ein Mitglied der TNF-Familie und kann Apoptose in vielen Tumorzellen auslösen, ohne dabei nicht transformierte Zellen zu schädigen. Leider haben viele Tumorzellen Mechnismen entwickelt, sich der TRAIL induzierten Apoptose zu entziehen. Es war Gegenstand dieser Arbeit die Rolle von TRAIL, seinen Rezeptoren den Bindungspartnern OPG und RANKL, sowie innerhalb chronischer Inflammation und Stress zu untersuchen. Vier verschiedene Pankreaskarzinomzelllinien, MiaPaCa2, Panc1, BXPC3 und Colo357 wurden dabei in vitro auf die oben genannten Parameter untersucht. Es konnte gezeigt werden, dass TRAIL Apoptose in MiaPaca2 um bis zu 27% steigert, Colo357 bis zu 35%. Panc1(<6%) und BXPC3(14%) zeigten sich nahezu resistent. Die Resistenz gegenüber TRAIL scheint mit der Abnahme funktioneller TRAIL-Rezeptoren DR4 und DR5 sowie der Zunahme nicht funktioneller Decoyrezeptoren DcR1/2 in Zusammenhang zu stehen. Interessanterweise zeigte sich keine Korrelation zwischen der Genexpression der TRAIL-Rezeptoren und der auf Proteinebene gemessenen TRAIL-Rezeptoren. OPG scheint ebenfalls die Resistenz gegenüber TRAIL zu fördern (BxPc3>Panc1>Colo357), es konnte bei MiaPaCa nicht gemessen werden. RANKL konnte nur in geringen Konzentrationen bei der Zelllinie Panc1 nachgewiesen werden. LPS, welches ein inflammatorisches Molekül darstellt zeigte insgesamt keinen Effekt, bzw. nur einen moderaten Anstieg von OPG bei Colo357. Chronischer Stress scheint ebenfalls keinen Effekt auf die TRAIL Wirkung zu haben.
  • TRAIL is a member of the TNF-family inducing apoptosis in many tumour cells whereas untransformed cells are usually not altered. However, many tumour cells have developed mechanisms enabling them to escape TRAIL-mediated apoptosis. Aims/Objektives: To asses the role of TRAIL, its receptors DR4, DR5, DcR1 and DcR2, their binding partners OPG and RANKL and inflammatory and stress stimuli in pancreatic cancer. Methods: Four different pancreatic cancer cell lines, MiaPaCa2, Panc1, BXPC3 und Colo357, were analyzed to investigate the above aims in vitro. and/or DR5 and an increase of non-functional decoy receptors DcR1/2. TRAIL-binding proteins as well as by reducing the inflammatory micro milieu in pancreatic cancer. Results: TRAIL could increase apoptosis in MiaPaCa2 (27%) and Colo357 (35%); BxPc3 (14%) was only moderately inducible, Panc1 (<6%) was TRAIL-resistant. The gain of TRAIL-resistance correlated with a decrease of functional TRAIL receptors DR4 and/or DR5 (p<0.01=Panc1, p<0.001=BxPc3) and an increase of non-functional decoy receptors DcR1/2 (p<0.01=BxPc3). Interestingly, the level of expression of TRAIL receptor genes and expression and function of TRAIL-receptors did not correlate well. OPG may also confer TRAIL-resistance (BxPc3>Panc1>Colo357) but was not present in MiaPaca2. RANKL was detected in minor concentrations only in PANC1 but did not have an impact on TRAIL-apoptosis (p>0.05). LPS, representing an inflammatory molecule, may moderately increase OPG levels (p<0.05=Colo357) whereas the stress-inducing catecholamine analogue isoproterenol did not have any significant impact on the actions of TRAIL.
