Assoziation von genetischen Polymorphismen in der Promotorregion des Endothelin-Converting Enzymes-1a zum Risiko der koronaren Herzkrankheit

  • Die koronare Herzkrankheit ist die häufigste Todesursache in den Ländern der westlichen Welt. Dem Endothelin-System wird zunehmend eine Rolle in der Pathogenese der Atherosklerose und der koronaren Herzkrankheit (KHK) zugesprochen. Das Endothelin-Converting Enzyme (ECE) katalysiert im letzten Schritt des Endothelin-Systems die Bildung des aktiven Endothelins aus seinem inaktiven Vorläufer big-Endothelin. In dieser Arbeit wurde die Assoziation der beiden Polymorphismen G-377A und C-854T in der Promotorregion des ECE-1a, einer Isoform des ECE, zum Risiko der KHK in einem großen Kollektiv von 1000 Patienten mit einer angiografisch verifizierten KHK und 1000 Kontroll-Patienten untersucht. Die Untersuchung ergab für die Variante G-377A eine Häufigkeit des A-Allels von 10,44% in der KHK-Gruppe gegenüber 10,69% in der Kontrollgruppe. Für die Variante C-854T ergab sich eine Häufigkeit des T-Allels von 8,05% in der KHK-Gruppe gegenüber 7,24% in der Kontrollgruppe. Beim Vergleich der Genotypenverteilung mittels x2-Test ergab sich für beide Varianten kein signifikanter Unterschied (p=0.71 bzw. p=0.63). Für beide Varianten erfolgten Stratifizierungen nach allgemeinen Faktoren wie Alter und Geschlecht und nach den verschiedenen Risikofaktoren einer KHK, die insgesamt keine signifikanten Verteilungsunterschiede ergaben. Zusammenfassend konnte kein Beweis einer Assoziation der beiden Polymorphismen zum Risiko der koronaren Herzkrankheit erbracht werden
  • Coronary artery disease (CAD) is the main cause of death in the Western world. The Endothelin-system is increasingly supposed to play a role in the pathogenesis of atherosclerosis and coronary artery disease. The Endothelin-Converting Enzyme (ECE) catalyses the creation of the active Endothelin from its inactive precursor big-Endothelin in the final step of Endothelin-processing. In this study we examined the association of the two polymorphisms G-377A and C-854T in the promoter region of ECE-1a, an isoform of ECE, to the risk of CAD in a big study population of 1000 patients with an angiographically verified CAD und 1000 controls. For the variant G-377A we found a frequency of 10,44% for the A-allel in the CAD-group vs. 10,69% in the control group. For the variant C-854T we found a frequency of 8,05% of the T-allel in the CAD-group vs. 7,24% in the control group. The comparison of the distribution of both genotypes by x2test showed no significant difference. For both variants statistic stratifications were carried out for common factors like gender and age and for the different risk factors of CAD which altogether showed no significant differences. Briefly no proof of an association of both examined polymorphisms to the risk of CAD was found.
