Analyse von kälteregulierten DNA-Sequenzen auf ihre Eignung für ein Bacillus subtilis Niedrigtemperatur-Expressionssystem

  • Im Rahmen der vorliegenden Arbeit wurde ein auf dem Promotor Pdes basierendes Kälte-induzierbares Expressionssystem für B. subtilis konstruiert und sukzessive optimiert. Dazu wurden verschiedene Kälte-regulatorische DNA-Sequenzen aus B. subtilis an das entsprechende Zielgen fusioniert, was neben der Kälte-Induzierbarkeit in einem positiven Einfluss auf die Expressionsstärke durch eine effizientere Translation bzw. Stabilisierung der mRNA resultierte. Vorausgehend wurde in vergleichenden Versuchen die Eignung unterschiedlicher Galaktosidasen zur Verwendung als Reporterenzyme für B. subtilis untersucht. Hierbei wurde erstmals die heterologe Expression einer Kälte-angepassten β-Galaktosidase aus P. haloplanktis TAE79 in B. subtilis durchgeführt und diese durch die Integration der DB-Sequenz sowie einer stem-loop-Struktur aus der 5‘-UTR des B. subtilis cspB-Gens gesteigert. Somit konnte nachgewiesen werden dass sowohl die additiven Sequenzen der cspB-DB und der cspB-sl-UTR als auch des bkdB-Terminators zu einer deutlich erhöhten Synthese der entsprechenden Zielproteine führt. Anhand der Überexpression einer Xylanase aus B. subtilis sowie einer α-Glucosidase aus S. cerevisiae wurde abschließend die Eignung des konstruierten Systems für die sekretorische und intrazelluläre Proteinsynthese in B. subtilis demonstriert. Diese Ergebnisse bestätigen die Eignung von B. subtilis als Wirtsorganismus auch für die Überproduktion kritischer, schwer zu faltender Proteine.
  • The present thesis focuses on the construction and successive optimization of a cold-inducible expression system for B. subtilis, which bases on the promoter of the desaturase (des) gene. For this purpose, several cold-regulatory DNA sequences from B. subtilis were fused to the corresponding target gene, resulting in increased expression levels due to more efficient translation or mRNA-stabilization. The suitability of galactosidases from different organisms as reporter enzymes for B. subtilis was investigated in advance by preparatory comparative analyses. Thereby, the cold-adapted β-galactosidase from P. haloplanktis TAE79 could be expressed in B. subtilis for the first time and its expression was shown to be significantly increased by integrating the DB sequence and a stem-loop structure from the 5'-UTR of the B. subtilis cspB gene. Consequently, it could be demonstrated that both additive sequences, cspB-DB and cspB-DB-UTR, and the terminator-sequence of the bkdB gene facilitate a significant increase the synthesis of the corresponding target proteins. Over-expression of a xylanase from B. subtilis and a α-glucosidase from S. cerevisiae finally verified the applicability of the engineered expression system for the extra- and intracellular protein synthesis in B. subtilis. The results obtained in this thesis thus clearly confirm the suitability of B. subtilis as a host organism for the overproduction of critical, poorly soluble proteins.
