  • The androgen receptor is the central regulator of the PCa cell and plays a key role in the development of a castration-resistant tumor stage. Therefore, possible interaction partners of the androgen receptor axis should be identified and their regulatory influence investigated. In the first part of this work a positive correlation between the AR and the HSP27 in PCa cells as well as a direct influence of the HSP27 on the AR could be shown. In the androgen-dependent LNCaP cells a higher endogenous HSP27 level was detectable than in the androgen-independent PC3 cells. Targeted HSP27 knock-down in LNCaP cells resulted in a significant reduction in AR mRNA as well as the AR protein. In addition, there was a reduction in the transcriptional activity of AR in the form of a reduced number of PSA transcripts. Consistent with this, HSP27 overexpression in PC3 cells resulted in re-expression of AR mRNA, although no functional AR protein was detectable. The transcriptional activity was also unaffected. As a potential modulator of this AR-HSP27 interaction, the microRNA miR-1 could be identified. This was in negative correlation to AR and HSP27 in the PCa cell lines used. Thus, a lower miR-1 detection rate was detected in LNCaP cells with high endogenous HSP27 levels than in the less HSP27-expressing PC3 cells. Evidence for the direct interaction of HSP27 with miR-1 is the fact that in PC3-HSP27 cells a significantly lower miR-1 expression was detectable than in PC3 cells. In conclusion, these data confirm the direct involvement of HSP27 and miR-1 in the AR signaling cascade and the direct affectibility of AR by HSP27. This underlines the potential pharmacological utility of both targets in the treatment of prostate cancer.
