Untersuchungen zu Tetrazolato-Pt Komplexen und zur Photoaktivierbarkeit von Diiodido-Pt(IV) diaminen mit upconversion-lumineszierenden Nanopartikeln

  • Trotz aller Erfolge in der Chemotherapie mit platinhaltigen Antitumormitteln bestehen dennoch viele Nachteile. Ein Fortschritt brachte die Entwicklung von lichtempfindlichen Pt(IV)-Verbindungen, die sich selektiv in Tumorzellen aktivieren lassen. Durch die geringe Eindringtiefe von kurzwelligem Licht wäre das Anwendungsgebiet auf oberflächige Tumore limitiert. Durch die Kopplung von Pt(IV)-Prodrugs mit upconversion-lumineszierende Nanopartikel (UCNPs) könnte die aktivierte Pt(II)-Spezies durch Bestrahlung mit NIR-Licht, welches durch UCNPs aufwärtskonvertiert wird, freigesetzt werden. Daher wurden sechs Bistetrazolatoplatin(II)-Komplexverbindungen sowie zwölf Bistetrazolato-Pt(IV) Komplexe synthetisiert und charakterisiert. Die Bistetrazolato-Pt(II) Komplexe stellen die ersten Beispiele für mononukleare Diamminbistetrazolato-Pt(II) Komplexe dar. Untersuchungen der Synthese ließen vermuten, dass es während der Synthese zur cis/trans-Isomerie kommt. Durch die Röntgenkristallstruktur konnte eine N2-Koordination des Tetrazolatliganden nachgewiesen werden. Entgegen der Erwartung, konnte keine eine lichtinduzierte Aktivierung der hergestellten Bistetrazolato-Pt(IV) Komplexe nachgewiesen werden. Trotzdem wurden die Eigenschaften der synthetisierten Verbindungen im Hinblick auf eine potenzielle Anwendung als Pt-basierte Zytostatika weiter untersucht wurde. Es zeigte sich, dass cis/trans-[Pt(OH)2(5-phenyltetrazolato)2(NH3)2] durch Ascorbinsäure in eine Spezies, die irreversibel an DNA bindet, umgewandelt werden kann. Die Bistetrazolato-Pt(II) Komplexe waren gegen adhärente Zelllinien nicht sonderlich aktiv, wohingegen nahezu alle der getesteten Bistetrazolato-Pt(IV) diammine antiproliferierende Aktivität gegen humane adhärente Krebszelllinien zeigten und dabei (IC50-Werte < 20 µM) gefunden wurden. Auch wenn einige der vorgestellten Syntheserouten eine Optimierung bedürfen und die gefundene Zytotoxizität durch die Einführung anderer Tetrazolderivate in Pt(II)-Komplexverbindung sicherlich weiter gesteigert werden kann, konnten die Befunde einen relevanten Beitrag zum Verständnis der Komplexierung von Tetrazolatanionen an ein Pt-Zentrum leisten. Aufgrund dessen, dass die Lichtempfindlichkeit von Diiodido-Pt(IV) Komplexen bekannt war, wurden fünf neuartige Diiodido-Pt(IV) diamine (25, 26, 27, 28 und 29) synthetisiert. Daraufhin wurde sich mit der Herstellung von seltenerd-dotierten NaYF4/NaGdF4-UCNPs sowie mit der Kopplung dieser mit Diiodido-Pt(IV) Komplexen beschäftigt. Zunächst wurde überprüft, ob die Bestrahlung mit einem NIR-Laser (&lambda; = 980 nm) in Gegenwart von UCNPs tatsächlich zu einem Abbau der Diiodido-Pt(IV) Komplexe sowie zu einer erhöhten DNA-Platinierung führt. Nachdem dies bestätigt war, wurden drei verschiedene Kopplungsmethoden angewandt. Für die kovalente Kopplung (Konjugatherstellung) wurden Diiodido-Pt(IV) Komplexe mit freier Carboxygruppe über eine Amidbindung mit amino-modifizierten NaGdF4:Yb,Er/Tm-UCNPs verknüpft. Der zweite Kopplungsansatz beinhaltet den Austausch der an der Nanopartikeloberfläche koordinierten Oleatliganden mit Diiodido-Pt(IV) Komplexen (Herstellung von Konstrukten). Durch die XPS-Analyse der Kopplungsprodukte kann gefolgert werden, dass beide Strategien eine Kopplung zwischen Pt Komplex und UCNPs ermöglichen, da Platin auf der Nanopartikeloberfläche nachgewiesen werden konnte. Allerdings liegt, unabhängig von der angewandten Kopplungsmethode, Pt zu 70–80 % im Oxidationszustand II vor. Die Verbindungen wiesen ohnehin eine gewisse Instabilität in Lösung auf. Die dritte Methode umfasst den Einschub des hydrophoben Pt(IV) Komplexes 29 in die Oleathülle. Der Nachweis von Pt auf diesem Konstrukt ist noch nicht gelungen, in vitro-Testungen ließen jedoch das Vorhandensein einer aktiven Pt-Spezies auf der Oberfläche vermuten. Für die Evaluierung der Zytotoxizität der NIR-Licht-vermittelten Antitumor-Aktivierungssysteme (Konjugate/Konstrukte) wurden diverse Methoden mit Variation im Bestrahlungsablauf vorgestellt. Hierbei stellte sich heraus, dass die Viabilität bei Bestrahlung der Konjugate/Konstrukte in Anwesenheit von Zellen im Vergleich zu Dunkelkontrollen signifikant herabgesenkt werden konnte. Allerdings besaß der NIR-Laser selbst einen toxischen Effekt gegenüber HL-60-Zellen. Die zunehmende Pt-Freisetzung der Konjugate mit der Bestrahlungsdauer und die geringe Pt-Freisetzung von Konstrukten, macht die Konjugate, insbesondere NaGdF4:Yb,Er-OA-SiO2-NH-28, zu besseren Kandidaten für die PACT. Dies wird bekräftigt durch die leicht höhere gefundene Toxizität der Konjugate im Vergleich zu den entsprechenden Konstrukten. Die dargestellten Konjugate/Konstrukte sind die ersten Beispiele für eine Kopplung von Diiodido-Pt(IV) Komplexen mit NaGdF4-UCNPs. Damit sich allerdings das vorgestellte Konzept der NIR-Aktivierung von lichtempfindlichen Diiodido-Pt(IV) Komplexen durchzusetzen kann, bedarf es noch Optimierungen hinsichtlich der Kopplungsstrategien.
  • Despite successes in chemotherapy with platinum-based anti-tumor agents, there are still many drawbacks. The development of light-sensitive Pt(IV) compounds, which can be selectively activated in tumor cells by irradiation, affords a valuable approach. Due to the low penetration into tissue of short wavelength light the application would be limited to superficial tumors. By the use of upconversion luminescent nanoparticles (UCNPs) in the photoactivated chemotherapy (PACT) deeper tissue layers may be addressed. The coupling of UCNPs to Pt(IV) prodrugs allows the delivery of Pt(II) species upon excitation with NIR light, which is up-converted by UCNPs. Consequently, six bistetrazolatoplatinum(II) complex and twelve bistetrazolato-platinum(IV) complexes were synthesized and characterized. The bistetrazolato-Pt(II) complexes represent the first examples of mononuclear diamminebistetrazolato-Pt(II) complexes. Studies gave rise to the presumption that cis/trans isomerism occurs during the synthesis. The X-ray crystal structure confirmed an N2 coordination of both tetrazolate ligands. Contrary to expectation, a light-induced activation of the synthesized bistetrazolato-Pt(IV) complexes could not be detected. Nevertheless, the properties of the synthesized compounds were further investigated with regard to a potential application as Pt-based cytotoxic agents. It was found that cis/trans-[Pt(OH)2(5-phenyltetrazolato)2(NH3)2] was reduced by ascorbic acid to a species which bind irreversibly to DNA. The bistetrazolato-Pt(II) complexes were more or less active against adherent humane cell lines, whereas almost all of the tested bistetrazolato-Pt(IV) diammines showed antiproliferative activity against cancer cell lines (IC50 values < 20 uM). Although some of the presented synthesis routes require optimization and the cytotoxicity can certainly be increased further by the introduction of other tetrazole derivatives in Pt (II) complexes, the findings were a significant contribution to the understanding of complexing tetrazolate anions to a Pt center. Due to the fact that the light sensitivity of diiodido-Pt(IV) complexes has been known, five novel diiodido-Pt(IV) diamines were synthesized (25, 26, 27, 28 and 29). Afterwards the focus was on the preparation of rare earth-doped NaYF4/NaGdF4-UCNPs and on the coupling of these with diiodido-Pt(IV) complexes. First, it was investigated whether the irradiation with an NIR laser (&lambda; = 980 nm) in the presence of UCNPs actually degrade the diiodido-Pt (IV) complexes. Moreover, it was proved that NIR-mediated activation of the complexes leads to increased DNA platination. Subsequently, three different coupling methods have been applied. For the covalent attachment diiodido-Pt(IV) complexes (26 and 28) exhibiting a free carboxyl group in the axial ligands were coupled via an amide bond with amino-modified NaGdF4:Yb,Er/Tm-UCNPs. Two different conjugates were formed. The second approach involves coupling by the exchange of the coordinated oleate on the nanoparticle surface with diiodido-Pt(IV) complexes (26 and 28) for producing two UCNP constructs. By the XPS analysis of coupling products can be concluded that both strategies enable a coupling between Pt complex and UCNPs as platinum could be detected on the nanoparticle surface. However, independently from the applied coupling method, 70-80 % of Pt was in oxidation state II. Additionally, the compounds had shown instability in solution. The third method involves the insertion of the hydrophobic Pt(IV) complex 29 into the oleate shell by hydrophobic interactions. Evidence of Pt on this construct has not yet succeeded, but in vitro testings suggest the presence of an active Pt species on the surface. For the evaluation of the cytotoxicity of the NIR light-mediated anti-tumor activation systems (conjugates/constructs), diverse methods were presented with variation in the irradiation sequence. Here, it was found that the viability of HL-60 leukemia cells could be significantly decreased upon irradiation of the conjugates/constructs in the presence of cells compared with dark controls. However, the NIR laser alone possessed even a toxic (thermic) effect against HL-60 cells. The increasing Pt release of the conjugates with the irradiation time and the low Pt release of constructs, makes the conjugates, particularly NaGdF4:Yb,Er-OA-SiO2-NH-28 to more useful candidates for PACT. This is confirmed by the slightly higher toxicity of the conjugates found, when compared to the corresponding constructs. The presented conjugates/constructs are the first examples of a nanohybrid of diiodido-Pt(IV) complexes with NaGdF4-UCNPs. The proposed concept of the NIR-activation of photosensitive diiodido-Pt(IV) complexes can become established in PACT, if improvements in terms of coupling strategies will be undertaken.
