Der Zebrafisch als Modellorganismus für den Knockdown von Palladin

  • Palladin, ein Aktin-assoziiertes Protein, beeinflusst die Morphologie, Migration, Adhäsion und Polarität von Zellen maßgeblich. Zudem wurde bereits in klinischen Studien gezeigt, dass ein Zusammenhang zwischen der Expression von Palladin und der Metastasierungsfähigkeit von Tumoren besteht. Das Studium der Funktion von Palladin in vivo ist aufgrund der bereits intrauterinen Letalität eines Palladin-Knockouts in der Maus nicht möglich. Daher wurde im Rahmen der vorliegenden Arbeit der Zebrafisch als Modellorganismus für die Untersuchung der Funktion von Palladin verwendet. Durch Mikroinjektion von sogenannten Morpholinos in die befruchteten Zebrafischeier konnte Palladin herunterreguliert werden. Mittels Western Blot und RT-PCR wurde diese Abnahme von Palladin bestätigt. In Analogie zur Maus führte der Kockdown von Palladin zu einem letalen Phänotyp zum Zeitpunkt 11-18 hpf, das auf schwere Entwicklungsschäden zurück zu führen ist. Um Zebrafischlarven mit einem Palladin Knockdown dennoch histologisch genauer untersuchen zu können, wurde ein sogenannter Mosaikphänotyp erzeugt. Hierbei zeigte sich, dass die Ausbildung geordneter Aktin-Filamente in den Myotomen gestört war. Mit Hilfe der in vivo Mikroskopie konnte ferner an lebenden Palladin-Knockdown Zebrafischlarven zum Zeitpunkt 6 und 8.5 hpf erstmals gezeigt werden, dass der Verlust von Palladin zu einer veränderten Migration von Zellen und zur Instabilität von Zell-Zell bzw. Zell-Matrix Kontakten führt. Durch eine Gene Array Analyse der Palladin-Knockdown Larven (10 hpf) konnte gezeigt werden, dass 1335 von 8200 Proteinen in Abhängigkeit der Palladin-Expression signifikant in ihrer Regulation verändert waren. Dabei sind unter den hoch- bzw. herunterregulierten Proteinen auch solche Proteine vertreten, die einen entscheidenden Einfluss auf das Aktin-Zytoskelett und auf Zell-Matrix- bzw. Zell-Zell-Kontakte haben. Zusammenfassend zeigte sich, dass der Zebrafisch ein ideales Tiermodel ist, um die Rolle von Palladin in vivo zu untersuchen.
  • Palladin, an actin associated protein that plays an important role for cell morphology, migration, adhesion and polarity. Palladin could be associated with the ability of malign tumors to metastasize in recent studies. The investigation of the role of palladin in vivo within the mouse model is not possible because of its intrauterine lethal phenotype. That is why we studied the zebrafish as a new model organism for the investigation of palladin in vivo in this publication. We created a knockdown due to Morpholino antisense technology. The down regulation of palladin was proofed via Western Blot and RT-PCR. Analogues to the mouse model a knockdown of palladin led to a lethal phenotype at 11-18 hours post fertilization due to severe impairment of development. For further examinations we created a so called mosaic phenotype. Within those animals we could show a disruption in actin in the Myotoms of the larvae. We could show that at a time point of 6-8.5 hpf the migration of cells was altered and cell-cell and cell-matrix contacts were instable. Through a Gene Array analysis of the knockdown larvae at 10 hpf we could demonstrate that 1335 of 8200 proteins were altered significantly depending on the level of palladin expression. Within these altered proteins there were important proteins for regulation of actin cytoskeleton, cell-cell and cell-matrix contacts. Summarizing we could show that the zebrafish presents as an ideal model for the investigation of palladins role in vivo.
