Regulation von Peroxiredoxinen in aktivierten und infizierten murinen Makrophagen

  • Makrophagen spielen eine essentielle Rolle bei Entzündungsprozessen sowie bei der Aktivierung von Abwehrmechanismen gegenüber bakteriellen Infektionen. Als Antwort auf inflammatorische Cytokine und bakterielle Produkte setzen Makrophagen Stickstoffmonoxid (NO) und reaktive Sauerstoffverbindungen (ROS) frei, die sowohl redox-sensitive Signaltransduktionswege vermitteln als auch Zellschäden induzieren. Ein wichtiger Bestandteil des zellulären Abwehrsystems stellen unter anderem die ubiquitär exprimierten Peroxiredoxine (Prxs) dar. Sie bilden eine Familie von sechs Thiol-abhängigen Peroxidasen, die Wasserstoffperoxid, organische Peroxide sowie reaktive Stickstoffverbindungen reduzieren können. Neben ihrer antioxidativen Funktion sind sie in die Regulation der Zellproliferation, Signaltransduktion sowie Apoptose involviert. In dieser Arbeit wurde gezeigt, dass Peroxiredoxine in Knochenmarkmakrophagen (bone marrow-derived macrophages; BMM) von BALB/c- und C57BL/6-Mäusen konstitutiv exprimiert werden und die Stimulation mit Lipopolysaccharid (LPS) und Interferon γ (IFNγ) zu einer Änderung der Genexpression von Prxs führt. Während in C57BL/6 BMM die Induktion der Genexpression von Prx 1, 2, 4 und 6 durch LPS und IFNγ von der induzierbaren NO-Synthase (iNOS) abhängig war, konnte in BALB/c BMM nur eine iNOS-abhängige Induktion der Prx 1- und Prx 6-Genexpression nachgewiesen werden. Des Weiteren wurde untersucht, ob die Tyrosinkinase JAK2, Tyrosinkinasen der Src-Familie, die Phosphatidylinositol-3-Kinase (PI3K), Proteinkinase C-Isoenzyme sowie p44/42 MAPK, p38 MAPK und c-Jun-N-terminalen Kinase (JNK) an der LPS- und IFNγ-induzierten Genexpression von Prx 1, 5 und 6 beteiligt sind. Außerdem konnte erstmals gezeigt werden, dass die Genexpression von Prx 6 durch Nrf2- (nuclear factor-erythroid 2p45 (NF-E2)-related factor 2-) Aktivatoren induzierbar ist und der Genknockout von Nrf2 die LPS- und IFNγ-vermittelte Induktion von Prx 6 in Makrophagen herabsetzt. Des Weiteren war die cytosolische Phospholipase A(2) (cPLA(2)) in die Regulation der LPS- und IFNγ-induzierten Transkription von Prx 5 und Prx 6 involviert. Die Hemmung der stromabwärts der cPLA(2)-gelegenen Cyclooxygenase-Enzyme (COX) führte zu einer signifikanten Abnahme der Prostaglandin (PG) E2-Sekretion sowie der Genexpression von Prx 6. Im Gegensatz dazu resultierte exogen zugeführtes PGD(2)-, PGE(1)-, PGE(2)-, PGF(2α), PGA(1), PGA(2) oder 15-deoxy-Δ12,14-PGJ(2) in einer konzentrations- und zeitabhängigen Zunahme des Prx 6-Transkriptes. Es konnte festgestellt werden, dass an der Regulation der PGD2- und PGE2-induzierten Prx 6-Genexpression die Adenylylzyklase und durch sie generiertes cAMP beteiligt sind, aber die durch cAMP-aktivierbare Proteinkinase A sowie Epac (exchange protein directly activated by cAMP) keine essentielle Bedeutung für die Prx 6-Genregulation besitzen. Die Untersuchungen der Regulation der PGE2- oder PGD2-induzierten Prx 6-Transkription zeigten weiterhin die Beteiligung der JAK2, PI3K, p38 MAPK und einiger Isoenzyme der PKC sowie die Bedeutung von Nrf2 für die Induktion der Genexpression von Prx 6 durch konventionelle und Cyclopentenon-Prostaglandine. In dieser Arbeit wurde zudem der Nachweis erbracht, dass durch die Infektion mit dem Pathogen Burkholderia pseudomallei, das als Modellorganismus Gram-negativer, intrazellulärer Erreger dient, die Genexpression von Prx 1, 5 und 6 in IFNγ-aktivierten Makrophagen von C57BL/6- und BALB/c-Mäusen induziert wird, wobei die mRNA-Induktion von Prx 1 und Prx 6 stärker in C57BL/6 als in BALB/cBMM erhöht wird. In IFNγ-aktivierten und B. pseudomallei-infizierten Makrophagen zeigte sich sowohl eine NO-abhängige Induktion der Prx 1- und Prx 6-Transkription und eine Abhängigkeit der Prx 5- und Prx 6-Genexpression von der NADPH-Oxidase-gebildeten ROS als auch eine Beteiligung von COX-1 und COX-2 an der durch B. pseudomallei-erhöhten mRNA-Expression von Prx 6. IFN&gamma-aktivierte Makrophagen, die mit Stämmen der virulenten Burkholderia-Spezies pseudomallei infiziert wurden, verfügten in den meisten Fällen über eine verstärkte Geninduktion von Prx 6, sowie iNOS- und COX-2-Proteinexpression gegenüber Makrophagen, die mit Stämmen der avirulenten Burkholderia-Spezies thailandensis infiziert wurden. Darüber hinaus zeichneten sich B. thailandensis- im Vergleich zu B. pseudomallei-infizierte BMM überwiegend durch eine geringere Genexpression und Sekretion verschiedener proinflammatorischer Cytokine wie IL-1beta, IL-6 und TNFalpha aus.
  • Macrophages play a decisive role during inflammation as well as for the activation of defense mechanisms against bacterial infections. In response to inflammatory cytokines and bacterial products macrophages generate nitric oxide (NO) and reactive oxygen species (ROS), that not only transduce redox-sensitive signalling but also induce cell damage. Amongst others, the ubiquitously expressed peroxiredoxins (Prxs) are important components of the cellular defense system. They constitute a family of six thiol-dependent peroxidases, which are able to reduce hydrogen peroxide, organic peroxides and reactive nitrogen compounds. Next to their antioxidant capacity, peroxiredoxins are involved in the regulation of cell proliferation, signal transduction as well as apoptosis. This thesis investigated the constitutive expression of peroxiredoxins in bone marrow-derived macrophages (BMM) of C57BL/6 and BALB/c mice and the alteration of the gene expression of Prxs by treatment with interferon γ (IFNγ) and lipopolysaccharide (LPS). While in C57BL/6 BMM the mRNA induction of Prx 1, 2, 4 and 6 was dependent on the inducible NO-synthase (iNOS), only the LPS- and IFNγ-increased gene expression of Prx 1 and Prx 6 was mediated by iNOS-derived NO. Moreover the influence of tyrosine kinase JAK2 and Src family tyrosine kinases, phosphatidylinositol 3-kinase (PI3K) and protein kinase C as well as p44/42 MAPK, p38 MAPK and c-Jun NH2-terminal kinase (JNK) on the enhanced gene expression of Prx 1, 5 und 6 was analysed after stimulation with LPS and IFNγ. For the first time it could be shown that gene expression of Prx 6 is inducible by several Nrf2- (nuclear factor-erythroid 2p45 (NF-E2)-related factor 2-) activators and LPS- and IFNγ-induced Prx 6 mRNA expression is partially mediated by Nrf2 in primary macrophages. In addition, the cytosolic phospholipase A2 (cPLA2) was involved in the regulation of the LPS- and IFNγ-induced transcription of Prx 5 and Prx 6. The inhibition of cyclooxygenase enzymes (COX) downstream of cPLA2 led to a significant decrease of prostaglandin (PG) E2 secretion as well as Prx 6 gene expression. In contrast, exogenously given PGD2, PGE1, PGE2, PGF2α, PGA1, PGA2 or 15-desoxy-Δ12,14-PGJ2 resulted in a dose- and time-dependent increase of the Prx 6 mRNA expression. It could be detected that adenylyl cyclase and adenylyl cyclase-generated cAMP are involved in the regulation of PGD2- and PGE2-induced Prx 6 transcription. Contrary to expectations, cAMP-dependent protein kinase A and Epac (exchange protein directly activated by cAMP) were not determined as key mediators of the PGD2- or PGE2-induced upregulation of Prx 6 gene expression. Further investigations of the regulation controlling the induced mRNA expression of Prx 6 by PGD2 or PGE2 revealed the involvement of JAK2, PI3K, p38 MAPK and specific PKC isozymes as well as the importance of Nrf2 for the enhanced Prx 6 gene expression by conventional or cyclopentenone prostaglandins. In addition this work also proved that infection with the bacterial pathogen Burkholderia pseudomallei, which was used as Gram-negative model organism, leads to increased gene expression of Prx 1, 5 and 6 in IFNγ activated macrophages of C57BL/6 and BALB/c mice, although a higher gene expression of Prx 1 and Prx 6 was observed in BMM of C57BL/6 than BALB/c mice. IFNγ-activated und B. pseudomallei-infected macrophages showed both a NO-dependent induction of Prx 1 and Prx 6 gene expression and a dependence of Prx 5 and Prx 6 transcription on the NADPH oxidase-derived ROS as well as the attendance of COX-1 and COX-2 at the B. pseudomallei-increased mRNA expression of Prx 6. IFNγ activated macrophages that were infected with strains of the virulent Burkholderia species pseudomallei possessed a higher induction of Prx 6 mRNA expression as well as iNOS and COX-2 protein expression than macrophages that were infected with strains of the avirulent Burkholderia species thailandensis. Furthermore B. thailandensis-infected BMM predominantly exhibited a lower gene expression and secretion of several proinflammatory cytokines, e.g. IL-1β, IL-6 and TNFα.
