Der Einfluss von "acute rapid pacing" und Irbesartan auf den TGF-beta-Signalweg am Modell von HL-1-Zellen

  • Zusammenfassung: Zielstellung dieser Arbeit war es, den Einfluss von „lone atrial fibrillation“ auf die extrazelluläre und intrazelluläre Signaltransduktion des TGF-beta1-Signalweges zu untersuchen. Dazu wurde das Modell des „acute rapid pacing“ unter Verwendung muriner HL-1-Zellen genutzt. Weiterhin wurde die Einflussnahme von Irbesartan auf die festgestellten Veränderungen geprüft. „Acute rapid pacing“ führte zu einer erhöhten mRNA-Expression profibrotischer Faktoren wie CTGF, SGK1 und TGF-beta1. Marker für kardiale Schädigung wie MSTN und FSTL3 zeigten ebenfalls eine Erhöhung des mRNA-Gehaltes nach „acute rapid pacing“. Kardial protektive Faktoren wie FSTL1 fanden sich im mRNA-Gehalt dagegen erniedrigt. Auf Proteinebene zeigte sich eine Mehrexpression des Stressmarkers GSK. Die Verwendung von Irbesartan beim „acute rapid pacing“ führte zu einer Reduktion der elevierten mRNA-Gehalte der profibrotischen Faktoren CTGF, SGK1 und TGF-beta1. Gleichfalls sank durch Irbesartan der erhöhte mRNA-Gehalt der kardialen Schädigungsmarker MSTN und FSTL3. Auf den mRNA-Gehalt des kardioprotektiven FSTL1 hatte Irbesartan keinen bedeutenden Einfluss. Auf Proteinebene konnte eine Minderexpression des Stressmarkers GSK festgestellt werden, wenn „rapidly paced“ Zellen mit Irbesartan inkubiert worden waren. Für andere untersuchte, mutmaßliche Modifikatoren wie Endoglin und FSTL5 lassen sich keine relevanten Aussagen ableiten. In Bezug auf den weiteren Signalweg sprechen die Ergebnisse der mRNA-Werte von ALK1 (Acvrl1), ALK2 (Acvr1) und ALK5 (Tgfbr1) nach „acute rapid pacing“ für eine Aktivierung des Phospho-Smad-1/-5/-8-Schenkels. Die Ergebnisse der Proteinexpression von Phospho-Smad-1/-5/-8 und phospho-Smad-2 nach „acute rapid pacing“ bzw. Inkubation mit TGF-beta1 stützen diese These. Tgfbr2-mRNA konnte in HL-1-Zellen wiederholt nicht nachgewiesen werden, wurde jedoch in Kardiozyten von Mus musculus detektiert. Dies spricht für relevante Unterschiede im kanonischen TGF-beta-Signalweg zwischen HL-1-Zellen und nativem Mausgewebe. Die untersuchten Zielgene des TGF-beta-Signalwegs – ID1, ID2 und ID3 – zeigten in Bezug auf ihre mRNA nach „acute rapid pacing“ ein differenziertes Verhalten mit Anstieg von ID1 und Absenkung von ID2 und ID3, was zum Prozess eines Remodelings passt. Irbesartan führte in Bezug auf die mRNA der genannten Zielgene nach „acute rapid pacing“ zu keiner signifikanten Änderung.
  • summary: The aim of this work was to investigate the influence of “lone atrial fibrillation” on the extracellular and intracellular signal transduction of the TGF-beta1 signaling pathway. For this purpose, the model of "acute rapid pacing" using murine HL-1 cells was used. The influence of irbesartan on the changes identified was also examined. "Acute rapid pacing" led to an increased mRNA expression of profibrotic factors such as CTGF, SGK1 and TGF-beta1. Cardiac damage markers such as MSTN and FSTL3 also showed an increase in the mRNA content after "acute rapid pacing". Cardiac protective factors such as FSTL1, on the other hand, were found to be reduced in the mRNA content. At the protein level, an elevated expression of the stress marker GSK was shown. The use of irbesartan in "acute rapid pacing" led to a reduction in the elevated mRNA levels of the profibrotic factors CTGF, SGK1 and TGF-beta1. Likewise, the increased mRNA content of the cardiac damage markers MSTN and FSTL3 decreased due to irbesartan. Irbesartan had no significant influence on the mRNA content of the cardioprotective FSTL1. At the protein level, a reduced expression of the stress marker GSK was found when "rapidly paced" cells were incubated with irbesartan. No relevant statements can be made for other investigated, putative modifiers such as Endoglin and FSTL5. Regarding the intracellular signaling pathway, the results of the mRNA level ​​of ALK1 (Acvrl1), ALK2 (Acvr1) and ALK5 (Tgfbr1) after "acute rapid pacing" indicate an activation of the Phospho-Smad-1/-5/-8 pathway. The results of the protein expression of Phospho-Smad-1/-5/-8 and phospho-Smad-2 after "acute rapid pacing" or incubation with TGF-beta1 support this thesis. Tgfbr2 mRNA was not once detected in HL-1 cells, but was detected in Mus musculus cardiocytes. This indicates relevant differences in the canonical TGF-beta signaling path between HL-1 cells and native mouse tissue. The investigated target genes of the TGF-beta signaling pathway - ID1, ID2 and ID3 - showed a differentiated behavior with regard to their mRNA after "acute rapid pacing" with an increase in ID1 and a decrease in ID2 and ID3, which fits the process of remodeling. Irbesartan did not significantly change the mRNA of the target genes mentioned after acute rapid pacing.
