Der Einfluss von cGMP-Modulatoren auf die Kontraktilität und Sensitivität glatter Gefäßmuskulatur in Rattenaorten

  • In vielen wissenschaftlichen Studien konnte die Bedeutung des NO-cGMP-PKG-Signalweges für die Regulation der Spannung von glatten Muskelzellen gezeigt werden. Im Mittelpunkt standen dabei oftmals die Guanylatzyklase, die Proteinkinase G oder zelluläre Kalziumkanäle und deren Bedeutung für die Muskeltätigkeit. In der vorliegenden Arbeit sollte über Modulatoren der zelluläre cGMP-Spiegel in glatten Gefäßmuskelzellen von Ratten verändert und durch Messung der Kontraktionsstärken dessen Einfluss auf das Kontraktionsverhalten nachgewiesen werden. Dabei sollte sowohl die Auswirkung von cGMP-Veränderungen auf die Spannung bei maximaler Stimulation als auch auf die Sensitivität für Phenylephrin untersucht werden. Nach Inkubation der Aorten mit einem cGMP-Modulator und Stimulation mit Phenylephrin wurden die Kontraktionen von Aortenringen über Schreiber direkt aufgezeichnet. Als Modulatoren wurden das Urikosurikum Probenecid, der Phosphodiesterasehemmer Sildenafil und der Inhibitor der löslichen Guanylatzyklase ODQ eingesetzt. Ein besonderer Fokus sollte darauf gelegt werden, inwiefern die Hemmung der MRP-Transportproteine 4 und 5 durch Probenecid- und Sildenafilinkubation auf den cGMP-Spiegel und damit die Spannung der glatten Muskelzellen auswirkt. In den Aufzeichnungen der Kontraktionen zeigte sich unter ODQ-Einfluss eine deutlich gesteigerte Spannung in g pro g Aortengewebe (Mw=2761,44*; SEM=236,4; n=9) bei maximaler Stimulation mit Phenylephrin im Vergleich zur Kontrolle (1515,15; 186; n=9). Unter Vorinkubation mit Sildenafil konnten sowohl mit der geringeren Konzentration von 1 µM (689,15*; 163,3; n=5) als auch bei 50 µM (187,17*; 154,5; n=5) signifikant niedrigere Spannungen festgestellt werden. In höherer Konzentration kommt neben der Inhibition der PDE auch der hemmende Einfluss von Sildenafil auf die MRP-Transporter zum Tragen. Auch nach der Inkubation mit Probenecid konnte sowohl mit 10 µM (783,70; 150,7; n=5) als auch mit 100 µM (693,64*; 128,4; n=5) eine verminderte Kontraktion der Gefäße festgestellt werden. Die Untersuchung der Sensitivität der Aorten auf Phenylephrin über Vergleiche der EC50-Werte ergab, dass insgesamt durch den Einsatz von cGMP-Modulatoren keine Veränderungen der Sensitivität verursacht werden. Schlussfolgernd ergibt sich aus den Ergebnissen dieser Arbeit, dass glatte Gefäßmuskulatur über cGMP-Modulatoren hinsichtlich der Maximalspannungen beeinflussbar ist und dass der cGMP-Transport über MRP 4 und 5 für die Kontraktion von glatten Muskelzellen physiologisch bedeutsam zu sein scheint, was neue Möglichkeiten des medikamentösen Eingreifens in die Gefäßregulation eröffnen könnte.
  • Many scientific studies showed the importance of the NO-cGMP-PKG-pathway for the regulation of tension in smooth muscle cells. The focus was often on the guanylate cyclases, protein kinase G or cellular calcium channels and their significance for muscle activity. In the present study the cellular cGMP levels in vascular smooth muscle cells of rats were modulated and the effect on contraction and sensitivity for stimulation with phenylephrine meassured. After incubation with a cGMP modulator the contraction of the aortic rings was induced by phenylephrine and recorded. As modulators the uricosuric drug probenecid, the PDE- inhibitor sildenafil and the inhibitor of soluble guanylate cyclase ODQ were used. A particular focus should be placed on the inhibition of MRP transport proteins 4 and 5 by probenecid and sildenafil incubation, their influence on the cGMP levels and thus effect on the tension in smooth muscles. The records of the contractions showed significantly increased tension in g per g aortic tissue (SD = 2761.44 *, SEM = 236.4, n = 9) by maximum stimulation with phenylephrine under influence of ODQ compared with the control group (1515.15, 186, n = 9). In pre-incubation with sildenafil significantly lower tensions in both groups at concentration of 1 µM (689.15 *, 163.3, n = 5) and 50 µM (187.17 *, 154.5, n = 5) sildenafil were found. In addition to the PDE inhibition should the inhibitory effect of sildenafil on the MRP transporters increase at higher concentration. Even after incubation with probenecid, both with 10 µM (783.70, 150.7, n = 5) and 100 µM (693.64 *, 128.4, n = 5) reduced contraction of the vessels were found. The investigation of the sensitivity of the aortic rings to phenylephrine by comparing EC50 values showed that none of the used cGMP modulators had influenced the sensitivity. In conclusion, the results of this study show that vascular smooth muscles can be influenced by cGMP modulators with regard to the maximum tension and that the cGMP transporters MRP 4 and 5 seem to be physiologically important for the contraction of smooth muscle cells. These findings could possibly open new opportunities for drug intervention in the vascular regulation.
