Identifizierung und Charakterisierung einer Lipase (ALIP2) und zweier Carboxylesterasen (BEST1 und BEST2) aus der Hefe Blastobotrys raffinosifermentans LS3

  • In unserem Alltag sind Polymere weit verbreitet. In Form von funktionellen Polymeren werden sie u.a. als Wirk- oder Effektstoff eingesetzt. Sie bestehen aus einem Träger, an welchen über einen Spacer eine funktionelle Gruppe gebunden ist. Die Spacergruppen beeinflussen die chemischen, physikalischen und biologischen Eigenschaften der Polymere bzw. ermöglichen diese erst. Dadurch stellen sie in der pharmazeutischen Industrie und der medizinischen Chemie Schlüsselbausteine dar. Auch Monoester von symmetrischen Dicarbonsäuren oder symmetrischen Diolen werden für die Einführung von Spacergruppen verwendet. Sie können durch die Hydrolyse von Diestern oder Dioldiestern chemisch synthetisiert werden. Da diese Reaktion nicht selektiv erfolgt, entstehen Nebenprodukte wie Disäuren oder Diole, die die Ausbeuten schmälern und eine aufwändige Aufarbeitung notwendig machen. Selektive enzymatische Verfahren stellen eine echte Alternative dar, denn eine Trennung des Produkts vom Nebenprodukt ist nicht notwendig. Bisher sind nur wenige Enzyme bekannt und verfügbar, die zur Synthese von Monoestern verwendet werden können. Ziel dieser Arbeit ist die Entdeckung neuer Lipasen und Carboxylesterasen als Biokatalysatoren zur Synthese von Monoestern, die zudem in ausreichender Verfügbarkeit generiert werden sollen. Als Gendonor und Expressionssystem diente hierfür die Hefe Blastobotrys raffinosifermentans. Die nicht-konventionelle, nicht-pathogene und thermotolerante Hefe B. raffinosifermentans weist ein breites Kohlenstoff- und Stickstoff-Quellen Spektrum auf, was sie für industrielle Anwendungen interessant macht. Aufgrund einer bereits vielfach eingesetzten, effizienten Transformationsmethode wurde die Hefe bereits zur Produktion verschiedener Proteine wie humanem Serumalbumin, Interleukin-6, Phosphatasen mit Phytase-Aktivität, Tannasen und einer Lipase eingesetzt. Die exzellenten Wachstumsparameter garantieren hohe Enzymausbeuten. Insgesamt wurden in dieser Arbeit 30 putative Lipase- und Carboxylesterase-Gene in ihrem Genom durch Annotationsanalysen identifiziert. Diese Gene wurden isoliert, amplifiziert und in der Hefe selbst überexprimiert. Die Proteinextrakte der erzeugten Stämme wurden anschließend auf Esteraseaktivität getestet, wovon sieben Kandidaten das Substrat p-Nitrophenylbutyrat (pNP-Butyrat) hydrolysierten. Anschließend wurde mittels eines Assays untersucht, ob die Enzyme die Hydrolyse der Substrate Adipinsäurediethylester (DEA), Dimethyl trans-1,4-cyclohexandicarboxylat (DMCH), Terephthalsäurediethylester (DETS) und Decandiol-dimethacrylsäureester (DDMAE) katalysieren. Vier Kandidaten hydrolysierten DEA und DMCH und ein Extrakt eignete sich zur Hydrolyse von DETS. Es folgten eine Testung auf Selektivität mittels Gaschromatographie mit gekoppeltem Flammenionisationsdetektor und eine affinitätschromatographische Reinigung der fünf Proteine. Dabei stellten sich die drei Kandidaten Alip2-6hp, 6h-Best1p und 6h-Best2p, eine putative Lipase und zwei putative Carboxylesterasen, als potenziell geeignete Kandidaten heraus. Anschließend erfolgte die biochemische Charakterisierung der drei Proteine. Das Temperatur-Optimum der Enzyme lag zwischen 31 °C und 41 °C und das pH-Optimum zwischen 6,6 und 7,0. Die Metallionen Fe2+, Fe3+ und Cu2+ inhibierten alle drei Biokatalysatoren und auch die Zugabe verschiedener Lösungsmittel verringerte ihre Aktivität. Die Untersuchung des Substratspektrums mit p-Nitrophenylestern mit Kettenlängen von C2 bis C18 zeigte eine Präferenz von Alip2-6hp für mittelkettige pNP-Ester mit einem Maximum bei pNP-Caproat und von 6h-Best1p und 6h-Best2p für kurzkettige pNP-Ester mit einem Maximum bei pNP-Acetat. 6h-Best1p und 6h-Best2p zeigen damit das für Carboxylhydrolasen typische Substratspektrum. Da Lipasen üblicherweise langkettige Substrate bevorzugen, wurde die Klassifizierung für Alip2-6hp mittels Tween 20- und Olivenöl-Agarplattentest weiter untersucht. Das positive Ergebnis dieser Untersuchung lässt auf eine Lipase schließen. Zur Bestimmung der Selektivität der Enzyme wurde die Hydrolyse von DEA und DMCH zeitlich per GC-FID verfolgt. Nach Derivatisierung der Carboxylgruppen war die quantitative Auswertung zum Gehalt an Monoester, Diester und Disäure möglich. Es ließ sich damit die Hydrolyse von DEA mit 6h-Best1p bestätigen. Bessere Ergebnisse wurden mit Alip2-6hp für das Substrat DMCH erzielt und mit Abstand die schnellste Hydrolyse wurde mit DEA als Substrat erreicht. In gereinigter Form hydrolysierte Alip2-6hp das Substrat DEA selektiv zu MEA, sodass bis zu 96 % Monoester synthetisiert werden konnten. Im Vergleich dazu wird MEA deutlich langsamer hydrolysiert. Zusätzlich wurden fünf unterschiedliche Formulierungen des Enzyms Alip2-6hp mit dem Substrat DEA getestet: (1) Rohextrakt, (2) freies, gereinigtes Enzym, (3) immobilisiertes, gereinigtes Enzym (Beads) und als Ganzzellkatalysatoren (4) permeabilisierte (Triton-) Zellen und (5) permeabilisierte, immobilisierte (Triton-) Zellen. Die vielversprechendsten Ergebnisse wurden mit isoliertem gereinigtem Enzym erzielt. DEA wurde vollständig und spezifisch zu MEA umgesetzt. Zur Gewährleistung einer ausreichenden Verfügbarkeit der Enzyme erfolgte die Kultivierung der Überexpressionsstämme im Fermenter im Fed-batch. Der Alip2-6hp produzierende Hefestamm erbrachte Aktivitäten von 674 U L-1, während der 6h-Best2p Überexpressionsstamm 2239 U L-1 produzierte.
  • Polymers are widely used in our everyday lives. In the form of functional polymers, they are used, among other things, as active ingredients or effect substances. They consist of a carrier to which a functional group is bonded via a spacer. The spacer groups influence the chemical, physical and biological properties of the polymers or make them possible in the first place. As a result, they are key building blocks in the pharmaceutical industry and medicinal chemistry. Monoesters of symmetrical dicarboxylic acids or symmetrical diols are also used for the introduction of spacer groups. They can be chemically synthesized by the hydrolysis of diesters or diol diesters. Since this reaction is not selective, by-products such as diacids or diols are formed, which reduce the yields and require expensive work-up. Selective enzymatic processes represent a real alternative, because it is not necessary to separate the product from the by-product. To date, only a few enzymes are known and available that can be used for the synthesis of monoesters. The aim of this work is to discover new lipases and carboxylesterases as biocatalysts for the synthesis of monoesters, which should also be generated in sufficient availability. The yeast Blastobotrys raffinosifermentans served as gene donor and expression system for this purpose. The non-conventional, non-pathogenic and thermotolerant yeast B. raffinosifermentans has a broad carbon and nitrogen source spectrum, which makes it interesting for industrial applications. Based on a widely used, efficient transformation method, the yeast has already been used to produce various proteins such as human serum albumin, interleukin-6, phosphatases with phytase activity, tannases and a lipase. The excellent growth parameters guarantee high enzyme yields. In total, 30 putative lipase and carboxylesterase genes were identified in its genome by annotation analysis in this work. These genes were isolated, amplified and overexpressed in the yeast itself. Protein extracts of the generated strains were then tested for esterase activity, of which seven candidates hydrolyzed the substrate p-nitrophenylbutyrate (pNP-butyrate). An assay was then performed to determine whether the enzymes catalyzed the hydrolysis of the substrates adipic acid diethyl ester (DEA), dimethyl trans-1,4-cyclohexanedicarboxylate (DMCH), terephthalic acid diethyl ester (DETS), and decanediol dimethacrylic acid ester (DDMAE). Four candidates hydrolyzed DEA and DMCH and one extract was suitable for hydrolysis of DETS. This was followed by testing for selectivity by gas chromatography with coupled flame ionization detector and affinity chromatographic purification of the five proteins. Three candidates, Alip2-6hp, 6h-Best1p, and 6h-Best2p, a putative lipase and two putative carboxylesterases, were found to be potentially suitable candidates. Subsequently, biochemical characterization of the three proteins was performed. The temperature optimum of the enzymes was between 31 °C and 41 °C and the pH optimum was between 6.6 and 7.0. Metal ions Fe2+, Fe3+ and Cu2+ inhibited all three biocatalysts, and the addition of various solvents also decreased their activity. Examination of the substrate spectrum with p-nitrophenyl esters with chain lengths from C2 to C18 showed a preference of Alip2-6hp for medium-chain pNP esters with a maximum at pNP-caproate and of 6h-Best1p and 6h-Best2p for short-chain pNP esters with a maximum at pNP-acetate. Thus, 6h-Best1p and 6h-Best2p show the substrate spectrum typical for carboxyl hydrolases. Since lipases usually prefer long-chain substrates, the classification for Alip2-6hp was further investigated using Tween 20 and olive oil agar plate assays. The positive result of this assay suggests a lipase. To determine the selectivity of the enzymes, the hydrolysis of DEA and DMCH was followed temporally by GC-FID. After derivatization of the carboxyl groups, quantitative evaluation for the content of monoester, diester and diacid was possible. Hydrolysis of DEA with 6h-Best1p could be confirmed. Better results were obtained with Alip2-6hp for the substrate DMCH and by far the fastest hydrolysis was achieved with DEA as substrate. In purified form, Alip2-6hp selectively hydrolyzed the substrate DEA to MEA, allowing synthesis of up to 96 % monoesters. In comparison, MEA is hydrolyzed at a much slower rate. In addition, five different formulations of Alip2-6hp enzyme were tested with the substrate DEA: (1) crude extract, (2) free purified enzyme, (3) immobilized purified enzyme (beads) and as whole cell catalysts (4) permeabilized (Triton) cells and (5) permeabilized immobilized (Triton) cells. The most promising results were obtained with isolated purified enzyme. DEA was completely and specifically converted to MEA. To ensure sufficient availability of the enzymes, cultivation of the overexpression strains was performed in the fermenter in fed-batch. The Alip2-6hp producing yeast strain yielded activities of 674 U L-1, while the 6h-Best2p overexpression strain produced 2239 U L-1.
