Etablierung eines auf Hefezellen basierenden Assays zur Erfassung estrogener Wirkungen im Rahmen der Umwelt-, Nahrungs- und Futtermittelanalytik

  • Ziel der Arbeiten war es, ein Hefe basiertes Testsystem zu entwickeln, mit dem in komplexen Proben (Urin, Milch, Ab-, Brack-, See-, Mineralwasser, Lebensmittel, Kosmetika und pharmazeutische Formulierungen) mit möglichst geringem Aufwand/Probevolumen/Kosten estrogene Aktivitäten be-stimmbar sind. Der entwickelte neue Arxula adeninivorans Estrogen-Screen (nAES-Assay) ermöglicht die Detektion estrogen-wirksamer Substanzen als Summenparameter. Der In vitro-Assay basiert auf transgenen A. adeninivorans-Zellen mit zwei Expressionsmodulen (Rezeptorgenmodul mit TEF1-Promotor – hERa-Gen – PHO5-Terminator, Reportergenmodul mit Arxula eigenen GAA-Promotor – phyK/ATAN1-Gen – PHO5-Terminator). Durch die Insertion zweier "Estrogen-Response-Elemente" (EREs) in die GAA-Promotorregion wurde diese zum Estrogen induzierbaren Promotor GAA2xERE–107 modifiziert. Als Reporter wurden zwei nicht-konventionelle Gene benutzt, die Klebsiella sp. ASR1 abgeleitete PhytaseK (phyK-Gen) und die Tannase (ATAN1-Gen) aus Arxula. Um die Genmodule in das Arxula Genom zu integrieren wurde die Transformations-/Expressionsplattform Xplor® 2 mit den Selektionsmarkermodulen ALEU2-/delALEU2-Promotor – ATRP1-Gen verwendet. Vorteil dieser Plattform ist, dass keine dominanten Resistenzmarker (HPH1(r)) in die Hefe übertragen werden und sich mitotisch stabile Hefetransformanten selektieren lassen. Xplor® 2 ermöglicht zudem die komplette Eliminierung aller E. coli-Plasmidbestandteile einschließlich Resistenzgene (Kan(r), Amp(r)). Die im Rahmen der Arbeit selektierten Transformanten wurden bezüglich ihrer Robustheit und Anwendbarkeit als biologische Komponente für den nAES-Assay geprüft. Anhand der auf PhytaseK und Tannase als Reporter basierenden zwei Varianten des nAES-Assays (nAES-P, nAES-T), wurde eine "Standard Operation Procedure – SOP" erstellt, was die Nutzbarkeit des Assays vereinfacht. Die Testplattform umfasst 96-well Arbeitsstandard, lyophilisierten Hefezellen und der validierten Testprozedur mit passender, von der quo data GmbH Dresden entwickelen Auswertesoftware Bioval®. Die Verwendung von A. adeninivorans G1212 [aleu2 atrp1::ALEU2] mit unikal integrierter Kassette in Verbindung mit dem Selektionsprotokoll ermöglichte eine ~2-fache Verbesserung der Messparameter des nAES im Vergleich zum konventionellen A-YES. Die zwei nAES Assay genutzten Reportervarianten wurden hinsichtlich Temperatur-, pH-Optimum und Anwendbarkeit in verschiedenen Proben charakterisiert. So eignet sich nAES-P besser für die Messung von Brack- und Seewasserproben, während der nAES-T die höhere Robustheit gegenüber NaCl aufweist. nAES-T und nAES-P erreichen bei der Bestimmung von estrogenwirksamen Substanzen in Urin und Abwasserproben mit 6–25 h Assaydauer, Nachweis-, Bestimmungsgrenze und EC50-Wert für 17b-Estradiol von 2,8; 5,9; 33,2 ng/l (nAES-P) bzw. 3,1; 6,7; 39,4 ng/l (nAES-T) ähnliche Charakteristika. Dem gegenüber sind die Substratspezifität und der dynamische Messbereich innerhalb der beiden Varianten annähernd gleich. Daraus ergibt sich, dass sich der nAES-Assay basierend auf der nicht-konventionellen Hefe A. adeninivorans besonders für die Analyse von komplexen Umweltproben und im regulatorischen Sektor (Abwasserkontrolle, Steroidanalytik, REACh) eignet. In ersten Versuchen mit Realproben, wie Abwasser zeigten sich durchschnittliche estrogene Belastungen des Abwassers von < 6 ng/l. In Direkteinleitern ohne jegliche Behandlung konnten 17b-Estradiol estrogenequivalente-Aktivitäten (nAES-EEQ) von 8–70 ng/l detektiert werden. Die zusätzliche Messung von 47 Rinderurinen mit dem nAES-Assay auf estrogen-wirksame Substanzen ergab eine gute Korrelation zur parallel durchgeführten chemischen Analysen (ana-EEQ) mit GC/MS. Dies unterstreicht den praktischen Nutzen der nAES-EEQ Ergebnisse. In Kälberurin wurde ein durchschnittlicher nAES-EEQ von 800 ng/l und in Rinderurinen (älter als 24 Wochen) von 13000 ng/l bestimmt. Auch Testserien mit Mineralwasser und Eluaten aus dazugehörigen Verpackungsbestandteilen dokumentierten mit nAES-EEQs von < 6 ng/l die Praxistauglichkeit des nAES Assays. Damit hat sich dieser Assay als ein relativ schneller Assay zu Messung estrogener Aktivitäten in komplexen Probenmatrices (inklusive inhibitorischer Bestandteile), wie Urin und Abwasser, ohne aufwendige Aufkonzentrierungen und Vorbehandlungsschritte erwiesen. Die Vorteile zu vergleichbaren Assays und zelllinienbasierten Testsystemen sind seine leichte Handhabung und das Vorhandensein einer bereits erprobten/validierten Testprozedur.
  • To overcome the problem of testing thousands of substances and their mixtures with a minimal cleaning set up and sample amount in different complex samples (like urine, milk, wastewater, brackish water, seawater, nutriments and pharmaceutical formulations) with minimal laboratory effort/sample-volume and costs we have developed the new Arxula adeninivorans Estrogen-Screen (nAES-Assay) towards detection of estrogenic activities as a environmental relevant sum parameter. The in-vitro-assay based on transgenic yeast A. adeninivorans-cells contains a receptor gene cassette consisting of: TEF1-promotor, hERa-gene, PHO5-terminator and a reporter gene cassette under the control of an Arxula-derived glucoamylase promotor (GAA-promotor). This promoter was modified by the insertion of two estrogen-responsive-elements (EREs) to obtain the estrogen inducible promotor GAA2xERE–107. As reporters two non-conventional gens were used the Klebsiella sp. ASR1 derived phytaseK (phyK) and an Arxula-derived tannase (ATAN1-gene). To integrated the gene modules in the Arxula genome the transformation-/expression platform Xplor® 2, with the selectable marker modules ALEU2-/delALEU2-promotor infront of the selection marker gene ATRP1 was used. Advantage of this system is that it works without dominat resistance markers (HPH1(r)), and allows us the selection of mitotic stable yeast transformants. Xplor® 2 moreover enables the elimination of complete E. coli-elements (Kan(r), Amp(r)) before transformation in Arxula. The transformants were analyzed for robustness and suitability to be the bio-component for the nAES-assay. The two types of the nAES-assays based on the reporter phytase and tannase (nAES-P, nAES-T) were used to evaluate a Standard operation procedure – SOP to minimize the test effort. Around the yeast bio-component a assay platform was established consisting of full 96-well standard, lyophilized yeast cells and a validated procedure for the test with matching analysis software Bioval® developed by quo data GmbH Dresden.Overall the usage of G1212 [aleu2 atrp1::ALEU2] in combination with the linear YRC transformation cassetts and the improved selection protocol enabled a nearly two-fold better measurement parameters (e.g. EC50-value) of the nAES- compared to the A-YES-assay. The two reporter variants were characterized with reference to temperature, pH-optimum and suitability within different samples in 96well test format with colorometric substrates. The nAES-P type is more suitable for the analysis of seawater and brackish water whereas the nAES-T type exhibited higher robustness to NaCl. Both assay types have similar characteristics for the determination of es-trogen active substances in water, sewage and urine samples e.g. 6–25 h assay period with detection-/determination-limits (LoD, LoQ) and EC50-values for 17b-estradiol of 2.8, 5.9, 33.2 ng L–1 (nAES-P) and 3.1, 6.7 and 39.4 ng L–1 (nAES-T). Substrate specificity among the reporter variants as well as the analytical measurement range (AMR) for both assay types are similar. These characteristics show that the nAES-assay based on a non-conventional salt tolerant yeast A. adeninivorans is applicable for a high throughput estrogen analysis in the environmental and regulatory control sectors (wastewater control, misuse of steroids, consumer protection and food safety issues, REACh). First measurements with real life samples were made, e.g. wastewater. The average estrogenic effects in the wastewater treatment plant effluent is therefor < 6 ng L–1 but in direct wastewater influents without any cleaning steps it range from 8–70 ng L–1 equivalents to 17b-estradiol (nAES-EEQ). The measurement of 47 different bovine urine samples with nAES-assay showed a high correlation to the GC/MS single substance determination (ana-EEQ) and underlined the practicalness of nAES-EEQ results. In calf urine average equivalent levels of ~800 ng L–1 were determined compared to ~13.000 ng L–1 once in mature cattle (older than 24 weeks). In series of tests with mineral water alone and elutions of belonging packaging materials no 17b-estradiol-equivalents above the determination-limits of < 6 ng L–1 was determined. The nAES-assay offers a rapid tool for urine estrogen determination as it does not require any kind of sample concentration steps. Overall the nAES-assay is one of the most sensitive yeast based estrogen assays known and particularly suitable to measure complex samples (including inhibitory substances) which other yeast and cell line based assays lag. Another advantage especially to cell line based assays are the easy usage and existence of an proven/valid method procedure.
