Untersuchungen zur Angiotensin II-vermittelten Tissue Factor-Induktion an einem In-vitro-Monozytenmodell

  • Tissue Factor (TF) ist eine gerinnungsaktive Substanz. Eine erhöhte monozytäre TF - Aktivität ist in arteriosklerotischen Plaques sowie auf zirkulierenden Blutmonozyten (MO) im akuten Myokardinfarkt nachgewiesen. ACE - Hemmer hemmen beides. Ziel dieser Arbeit ist die Evaluierung eines Effektes von Angiotensin II (All) und dem selektiven A II -Rezeptorantagonisten Losartan auf die monozytäre TF - Expression in vitro. Menschliche MO werden über einen Ficoll - Dichtegradienten und eine Fibronektinadhärenzmethode aus Buffy Coats isoliert. Diese werden mit rekombinanten Angiotensin II und Losartan stimuliert und maximal 48 Stunden kultiviert. Bestimmt werden die Prokoagulatorische Aktivität, die TF -Konzentration mit einem Faktor Xa sensitiven Assay und eine spezifische TF - mRNA mittels reverse Transkriptase - PCR. Mit allen drei Methoden ist ein signifikanter Effekt von AII auf die monozytäre TF - Synthese nachweisbar, wobei das Maximum der TF - mRNA bei 3 Std und der TF - Aktivität sowie des TF - Proteins bei 6 Std nachweisbar ist. Losartan zeigt keinen Einfluss auf TF, hemmt aber die A II - Wirkung. Die hier untersuchte monozytäre TF - Expression ist geringer als die in der Literatur beschriebene von Makrophagen und zeigt einen anderen zeitlichen Verlauf. Während ihrer Differenzierung regulieren MO ihr Renin - Angiotensin -System herauf. Die A II - vermittelte TF - Synthese nicht - differenzierter MO dürfte somit bei der Arteriosklerose und der akuten Gerinnungsstörung im akuten Myokardinfarkt eine eher geringere Bedeutung als die von Makrophagen oder die von anderen Botenstoffen haben.
  • Tissue Factor (TF) is a clotting protein. Elevated levels of monocytal TF - activity are found in arteriosclerotic plaques and circulating blood monocytes (MO) during acute myocardial infarction. ACE - Inhibitors diminish both. Our target was to evaluate a possible effect of Angiotensin II (All) and Losartan, a selective Angiotensin II - receptor antagonist, on monocytal TF - expression in vitro. Human blood monocytes are isolated by a Ficoll - density gradient and a Fibronectin adherence method from buffy coats. MO are stimulated with a recombinant AII and Losartan and kept in culture for at last 48 hours. Procoagulatory Activity, TF - concentration with a Factor Xa sensitive Assay and a specific TF - mRNA by reverse Transkriptase - PCR are determined. All three methods show a significant effect of AII on monocytal TF - synthesis, with maximum TF - mRNA at 3 hours and TF - activity and protein at 6 hours. Losartan has no influence on TF, but inhibits the AII - effect. This monocytal TF -expression is lower than that described in macrophages and has another time course. While differentiation MO rule up its Renin - Angiotensin - System. AII - mediated TF - synthesis of none - differentiated MO is suspected to be of minor relevance in arteriosclerosis and acute clotting disorders during acute myocardial infarction than that mediated by macrophages or other mediators.
