Einsatz von Laccasen aus Pilzen zur Herstellung von Hybridmolekülen unter besonderer Berücksichtigung der Synthese von antifungalen Substanzen und Aminosäurederivaten

  • Im Rahmen dieser Dissertation konnten zahlreiche unterschiedliche Substanzklassen mit Hilfe der Laccase verknüpft werden. Die dabei synthetisierten Verbindungen können nicht nur als Feinchemikalien in der organischen Synthese sondern auch für die Herstellung von antimikrobiellen und antikanzerogenen Medikamenten oder funktionellen Biomaterialien genutzt werden. Potential der Laccase zur Herstellung von Wirkstoffen: Die Synthese von antimikrobiellen Wirkstoffen durch die laccasekatalysierte Derivatisierung konzentrierte sich vor allem auf verschiedene Azole und Morpholine. Die isolierten Derivate wurden hinsichtlich ihrer biologischen Wirkung untersucht. Azole und Morpholine sind Bestandteil verschiedener antifungaler Medikamente und Fungizide. Eine Derivatisierung dieser Substanzklassen könnte neue Verbindungen mit neuen Eigenschaften, wie z.B. einem erweiterten Wirkungsspektrum, führen. Synthese der potentiellen Wirkstoffe 1. Für die laccasevermittelte Derivatisierung der Azole und Morpholine wurden para-dihydroxylierte aromatische Verbindungen als Ausgangsstoffe eingesetzt. Im Verlauf der Reaktion erfolgte eine laccasekatalysierte Oxidation der jeweiligen para-dihydroxylierten aromatischen Verbindung zum Chinon, welches dann einer Aminierung durch eine intermolekulare Michael-Addition (1,4-Addition) unterlag und in der Bildung von C N verknüpften monoaminierten Produkten resultierte. Bei einer erneuten laccasevermittelten Oxidation des gebildeten monoaminierten Produktes und anschließender Aminierung entstanden diaminierte Produkte. Die gebildeten Hybridmoleküle bestanden folglich aus einem Molekül der para-dihydroxylierten aromatischen Verbindungen in chinoider Form und einem (monoaminiertes Produkt; Dimer) bzw. zwei (diaminiertes Produkt; Trimer) Molekül/en des Morpholins bzw. Azols. 2. Darüber hinaus wurde die laccasekatalysierte Synthese von neuartigen Zyklisierungsprodukten nachgewiesen, deren Bildung weder mittels Laccase noch mit chemischen Syntheseverfahren bislang beschrieben wurde. Die wichtigsten Gruppen dieser neuen Zyklisierungsprodukte sind Cycloheptene und Cyclooctene. Testung der potentiellen Wirkstoffe hinsichtlich ihrer biologischen Wirkung 3. Zur Prüfung der antimikrobiellen Aktivität gegenüber Candida maltosa und bei ausgewählten Verbindungen gegenüber Candida albicans wurde die Methode des EUCAST discussion document E.Dis 7.1 über die MHK-Bestimmung in Mikrotiterplatten modifiziert. Ein Vergleich der hemmenden Wirkung von ausgewählten Produkten und der für deren Synthese genutzten Ausgangsstoffe gegenüber dem Testorganismus Candida maltosa mit Candida albicans ergab eine weitestgehende Übereinstimmung der MHK-Werte und ermöglicht in hohem Maße die Übertragung der ermittelten MHK-Werte mit Candida maltosa auf die medizinisch relevante Hefe Candida albicans. Zur Bestimmung der antimikrobiellen Aktivität wurde ein Agardiffusionstest eingesetzt und die Testung der zytotoxischen Aktivität erfolgte gegenüber einer humanen Blasenkrebszelllinie. 4. Die neu synthetisierten Verbindungen wiesen sowohl eine antibakterielle und antifungale Aktivität als auch eine zytotoxische Wirkung auf. Die Dimere zeigten z.B. eine antibakterielle und antifungale Wirkung während die Trimere keine oder nur vereinzelt eine antimikrobielle Wirkung aufwiesen. Die getesteten Zyklisierungsprodukte zeigten meist nur geringe hemmende Wirkung auf Candida maltosa. Potential der Laccase zur Herstellung von Biomaterialien: Als Vorbild für die Herstellung von Biomaterialien diente der von marinen Muscheln zur Anheftung an den Untergrund gebildete Klebstoff, der durch Polymerisationsreaktionen von Peptiden entsteht. Derartige Polymere könnten zur Herstellung von bioresorbierbaren Klebstoffen, z.B. für die Gesichtschirurgie zur Klebung von Knochensplittern, genutzt werden. 1. Die Derivatisierung von Aminosäuren und Peptiden mit para- und ortho-dihydroxylierten aromatischen Substanzen führte zur Bildung sowohl von di- und trimeren als auch polymeren Reaktionsprodukten. Die Laccase ermöglichte die Derivatisierung von geschützten aber auch ungeschützen Aminosäuren und Peptiden in einer sogenannten "one pot" Reaktion. 2. Ein Vergleich der Reaktionen mit Laccase und dem chemischen Katalysator Natriumiodat ergab für die Reaktionen der Aminosäuren L-Phenylalanin und L-Tryptophan mit 2,5-Dihydroxyacetophenon einen klaren Vorteil der Laccase. Die Ergebnisse der einzelnen Reaktionen belegen, dass eine effektive Derivatisierung von verschiedenen Substanzklassen mit Hilfe der Laccase als einem biologischen Katalysator möglich ist. Die relativ einfach durchführbare und effiziente laccasevermittelte Reaktion kann dabei auch für die Herstellung von neuartigen Verbindungen mit biologischer Wirkung genutzt werden. Die weitere Charakterisierung der Wirkstoffe wird zeigen, ob und inwieweit diese Verbindungen konkurrenzfähig gegenüber Standardverbindungen sind, die in der Human- und Veterinärmedizin aber auch der Landwirtschaft eingesetzt werden.
  • This thesis describes the cross-linking of different substance classes by laccase. The synthesized compounds can be used not only as fine chemicals in organic synthesis but also for the production of antimicrobial and anticancer drugs or functional biomaterials. Potential of laccase for the production of bioactive agents: The synthesis of antibacterial agents through laccase-catalyzed derivatization focused mainly on different azoles and morpholines. The isolated derivatives were studied for their biological effects. Azoles and morpholines are part of various antifungal drugs and fungicides. A derivatization of these compounds could result in new substances with new properties, e.g. a broader antifungal spectrum. Synthesis of potential drugs 1. For laccase-mediated derivatization of azoles and morpholines para-dihydroxylated aromatic compounds were used as starting materials. The first step is the laccase-catalyzed oxidation of the respective para-dihydroxylated aromatic substance with oxygen, resulting in quinonoid derivatives which then undergo an amination by intermolecular Michael addition(1,4-addition). The formed C-N monoaminated products may undergo a further laccase-mediated oxidation and subsequent amination resulting in diaminated products. 2. In addition, the synthesis of novel cyclic products with laccase was demonstrated for the first time. The most important of these new groups are cycloheptenes and cyclooctenes. Testing of the potential drugs for their biological effects 3. A modified method of EUCAST discussion document E. Dis 7.1 was used to determine the antimicrobial activity of the synthesized products against Candida maltosa and Candida albicans. The MIC values of selected products and the reactants used for their synthesis against the test organisms Candida albicans and Candida maltosa were comparable. Additionally, the agar disc diffusion method was chosen for the determination of antibacterial and antifungal activities. The test for the evaluation of cytotoxic activity was performed with human urinary bladder carcinoma cell line. 4. The newly synthesized compounds exhibited both antibacterial and antifungal activity and cytotoxic effect. The dimers showed an antibacterial and antifungal effect while the trimers showed no or only sporadic antimicrobial effect. The tested cyclic products generally showed only slight inhibitory effect on Candida maltosa. Potential of laccase for the production of biomaterials: The model for the production of biomaterials was the adhesive from marine mussels which is formed by the polymerization of peptides. Such polymers could be the basis for new adhesives, e.g. for the use in medicine, surgery, ophthalmology or dentistry. 1. The derivatization of amino acids and peptides with ortho- and para-dihydroxylated aromatic compounds led to the formation of di- and trimers as well as polymers. The laccase allowed the derivatization of unprotected but also protected amino acids and peptides in a so-called "one pot" reaction. 2. The reactions with laccase and the chemical catalyst sodium iodate were compared. A clear advantage of the laccase-catalyzed reactions of the amino acids L-phenylalanine or L-tryptophan with 2.5-dihydroxyacetophenone was determined. The results of the experiments demonstrate that an effective derivatization of various classes of compounds using laccase as a biological catalyst is possible. The laccase-mediated reaction is easy, efficient and can also be used for the production of novel substances with biological activity. The further characterization of the active ingredients will show whether and to what extent these compounds are competitive with standard compounds used in human and veterinary medicine but also in agriculture.
