Die Expression des Membrantransporter ABCA1 im Hirngewebe des Menschen – ein Beitrag zur Pathogenese der Alzheimer-Demenz

  • Zahlreiche Studien bestätigen einen Zusammenhang zwischen dem Cholesterolstoffwechsel des Gehirns und der Pathogenese der Alzheimer-Demenz (AD). Eine besondere Rolle soll dabei dem ABCA1-Transporter zukommen. Im Rahmen unserer Studie untersuchten wir an 79 Gehirnproben nichtdementer Probanden die Expression des ABCA1-Transporters und korrelierten dies mit Faktoren, die in der Pathogenese der AD eine Rolle spielen sollen: Alter, Geschlecht, Plaqueanzahl, ApoE-Genotyp, insbesondere des ApoE4-Allels, P-Glykoprotein sowie Abeta1-40 und Abeat1-42. Dabei wurden gesondert untersucht Hippocampus, Mark, Kortex und Leptomeninx. Wir detektierten eine signifikante Abnahme ABCA1-Expression beim Vergleich der Gruppe der 55-64jährigen mit der Altersgruppe der 65-74jährigen Probanden in der Leptomeninx bei höherem Lebensalter. Desweiteren konnte für den Kortex gezeigt werden, dass Träger des ApoE 2/4-Genotyps signifikant weniger ABCA1 exprimieren als Träger des ApoE 3/3 und ApoE 4/4- Allels. Auch konnte für die Leptomeninx gezeigt werden, dass ein signifikanter Zusammenhang besteht zwischen der Ablagerung von Abeta1-42 in den Gefässen und der ABCA1-Expression in den Gefässen.
  • Numerous studies confirm a connection between the metabolism of Cholesterol of the brain and the pathogenesis of the Alzheimer dementia (AD). It seems like a special role comes up to the ABCA1 transporter. In the context of our study we examined the expression of the ABCA1-Transporter at 79 brain samples of nondemented test persons and correlated this with factors, which are to play a role in the pathogenesis of the AD: Age, gender, numbers of plaques, ApoE Genotype, in particular the ApoE4-Allele, P-Glycoprotein as well as Abeta1-40 and Abeta1-42. We seperatly examinend Hippocampus, brain medulla, Cortex and Leptomeninx. By comparing the age groups of the 55-64 and the 65-74 years old test persons we detected a significant decreased expression of ABCA1 in the Leptomeninx in higher age. Further it could be shown for the cortex that carriers of the ApoE 2/4-Genotype express significantly less ABCA1 carriers compared to the ApoE 3/3 and of ApoE 4/4 allele. Also it could be shown for the Leptomeninx that a significant connection exists between the deposit of Abeta1-42 in the vessels and the expression of ABCA1 transporter in the vessels.
