Die Wirkung von Coelomflüssigkeit und Coelomozytenlysat der Art Eisenia fetida (Sav.) auf humane Tumorzellen unter in vitro Kulturbedingungen

  • Die vorliegende Arbeit befasst sich damit, ob und wenn ja in welcher Konzentration, die bekannten lytischen Proteine aus E. fetida zytolytisch auf humane Tumorzellen wirken (IGR-1, Caki-1 und 2, RCC-EW, Du-145). Es wurden reine Coelomflüssigkeit (CF), Coelomozytenlysat und mittels PAGE isolierte Hämolysine eingesetzt, um den zytotoxischen Effekt auf die Zelllinien zu untersuchen. Weiterhin sollte untersucht werden, ob CF und die isolierten Proteine in der Lage sind, in den Tumorzellen Apoptose auszulösen. Als Positivkontrolle für Apoptoseinduktion und zytomorphologische Vergleichsstudien wurde Camptothecin eingesetzt. Hierbei kamen die Verfahren des MTT-Testes, des Trypanblautestes, das Annexin V Fluos Staining Kit, sowie der PARP Western Blot und die RT-MPCR unter Verwendung der „human Apoptose Sets 5 und 7“ zum Einsatz. Zur Isolation der Proteine wurde die Polyacrylamidgelelektrophorese verwand. In den Untersuchungen konnte ein eindeutiger zytotoxischer Effekt der Testsubstanzen gezeigt werden. Lichtmikroskopische Untersuchungen deuteten auf Apoptoseinduktion hin, die durch Ergebnisses des Annexin V Tests bestätigt wurden. PARP Western Blot und die RT-MPCR erbrachten keine weiteren Resultate. Durch die PAGE wurde ein neues hämolytisches Protein isoliert, welches als new Protein bezeichnet wurde.
  • This thesis deals with the question if lytic proteins of the species E. fetida have a cytolytic effect on human tumor cells (IGR-1, Caki-1 and 2, RCC-EW, Du-124) in vitro and if that is the case, which concentration is required. The cytotoxic effect was examed by the use of coelomic fluid (CF), coelomocyte lysat and haemolysins which were isolated by the PAGE method. In addition we focused on the question, if CF and the isolated proteins are able to induce apoptosis in the tumor cells. Camptothecin was chosen as positive control concerning the induction of apoptosis and to compare cytomorphological changes. Therefore we ran different tests including MTT-test, trypanblue test, annexinVfluos staining kit, PARP western blot and RT-MPCR ( using the human apoptosis sets 5 and 7).The isolation of the proteins was done by the use of polyacrylamidgelelectophoresis. The tests showed a clear zytotoxic effect of the examined substances on tumor cells. Examinations with the microscop let us assume an induction of apoptosis. This was confirmed by the annexin V staining test. The PARP western blot and RT-MPCR had no further results. Using the PAGE we were also able to isolate a new haemolytic protein, which we referred to as “new protein”.
