Sensibilisierung von Tumorzellen für immunologische Effektormechanismen durch Histondeacetylase-Inhibitoren

  • Histondeacetylase-Inhibitoren (HDI) wirken toxisch auf verschiedene Tumortypen. Sie lockern durch Hemmung von Histondeacetylasen die Chromatinstruktur maligner Zellen. Dadurch können sie Apoptose, Zellzyklusarrest und Differenzierung auslösen. Weiterhin ermöglichen sie die Wiederaufnahme der Transkription von Tumorsuppressorgenen sowie die verstärkte Expression proapoptotischer Proteine, und sie führen zu einer Aktivitätserhöhung von Caspasen. Werden Tumorzellen mit HDI inkubiert, erhöht sich deren Empfindlichkeit gegenüber dem tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), so dass in Tumorzellen, die vorher nahezu resistent gegen TRAIL waren, sehr effektiv Apoptose ausgelöst wird. Natural killer cells (NK-Zellen) sezernieren nach Stimulation mit Interleukin 2 (IL-2) TRAIL und exprimieren das Molekül auf ihrer Zelloberfläche. Deshalb sollte in dieser Arbeit untersucht werden, ob eine Vorbehandlung mit HDI die Empfindlichkeit von Tumorzellen für zytolytische Effekte von Immunzellen erhöht. Dafür wurden Prostatakarzinomzellen (PC3) und Medulloblastomzellen (DAOY) 24 Stunden mit verschiedenen HDI vorinkubiert und dann mit IL-2 stimulierten peripheral blood mononuclear cells (PBMC) konfrontiert. IL-2 stimulierte PBMC enthalten aktivierte cytotoxische T-Lymphozyten (CTL) und aktivierte NK-Zellen. Beide Zellpopulationen können prinzipiell in Tumorzellen Apoptose auslösen. Verwendete HDI waren suberoyl anilide hydroxamic acid (SAHA), Natriumbutyrat (NaB) und das Benzamid MS-275. Um deren Wirkungen mit einem etablierten Zytostatikum zu vergleichen, wurde Vincristin als Kontrolle eingesetzt. Die quantitative Bestimmung der Zahl überlebender Tumorzellen erfolgte 24 bzw. 48 Stunden nach der Konfrontation mit den Zytokin-aktivierten Blutzellen in einem durchflusszytometrischen Verfahren. Propidiumjodid wurde zur Identifizierung toter Tumorzellen verwendet. Die HDI SAHA, NaB und MS-275 hatten allein eine dosisabhängige zytotoxische Wirkung auf die getesteten Tumorzelllinien. In Kombination mit den aktivierten PBMC zeigten die HDI überadditive und manchmal sogar synergistische zytolytische Effekte. Dagegen wirkten Vincristin und aktivierte PBMC stets nur additiv. Die Ergebnisse dieser Arbeit weisen darauf hin, dass die in klinischen Studien beobachteten tumortherapeutischen Wirkungen von HDI neben ihrer direkten zytotoxischen Wirkung teilweise auf einer synergistischen Wirkung mit Effektormechanismen des Immunsystems beruhen könnten.
  • Histone deacetylase inhibitors (HDI) have cytotoxic effects on different tumor cell types. By inhibiting histone deacetylases they change the chromatin structure of malign cells. In tumor cells they induce apoptosis, cell cycle arrest and differentiation. Furthermore they induce transcription of tumor suppressor genes, expression of proapoptotic proteins and reactivate caspases. HDI pre-incubated tumor cells are rendered susceptible to tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), despite being almost resistant to TRAIL before treatment with HDI. After stimulation with interleukin 2 (IL-2) the expression of TRAIL on natural killer cells (NK cells) is upregulated. In this study we investigated the susceptibility of tumor cells after pretreatment with HDI to cytolytic effects of immune cells. Prostatic cancer cells (PC3) and medulloblastoma cells (DAOY) were pre-incubated with different HDI for 24 hours and then confronted with IL-2 stimulated peripheral blood mononuclear cells (PBMC). IL-2 stimulated PBMC feature activated cytotoxic T-lymphocytes (CTL) and activated NK cells. We used three different HDI: suberoyl anilide hydroxamic acid (SAHA), sodiumbutyrate (NaB) and the benzamide MS-275. To compare their effects to an established cytostatic drug, we used vincristin. 24 and 48 hours after incubation with IL-2 activated PBMC tumor cell death was measured with a cytofluorometric assay. To identify dead tumor cells we used a propidiumiodide staining. HDI SAHA, NaB and MS-275 exerted dose-dependent cytotoxic effects on both tumor cell lines, as well as PBMC. In combination, activated PBMC and HDI showed over-additve or even synergistic cytotoxic effects. Vincristin and activated PBMC had only additive effects on both tumor cell lines. The results of this study could explain the tumor-toxic effects of HDI seen in clinical trials. Besides their direct cytotoxic effects, they could eventually facilitate cytolytic effects of the immune system. This could be an enormous effect in the treatment of cancer.
