Proteine im Liquor cerebrospinalis von Patienten mit entzündlichen Erkrankungen des Nervensystems

  • In der vorliegenden Arbeit wurden Liquorproben von Patienten mit neurologischen Erkrankungen auf ihre Proteinzusammensetzung hin untersucht und die Proteine quantitativ analysiert. Es wurde versucht, Proteine zu identifizieren, die als Krankheits-Marker dienen könnten.Untersucht wurden zwei immunvermittelte neurologische Erkrankungen, die Multiple Sklerose (MS), sowie das Guillain-Barré-Synrom (GBS). Die Liquorproteine von MS- und GBS-Patienten wurden mit Hilfe der 2D-Gelelektrophorese aufgetrennt und die Proteine quantitativ analysiert. Nach der Quantifizierung konnten 15 Proteine identifiziert werden, die erhöht auftraten und 11 Proteine die vermindert im MS-Liquor auftraten. Im Liquor von den GBS-Patienten konnten 4 Proteine erhöht und 7 Proteine vermindert detektiert werden. Im MS Liquor handelt es sich bei den erhöhten Proteine um Komplement Faktor B, Beta-2-Glykoprotein, Protaglandin-H2-Isomerase und Cystatin C und bei den verminderten Proteine um Antithrombin III, Protaglandin-H2-Isomerase und Transthyretin. Im GBS Liquor konnten verstärkt Haptoglobin und Coeruloplasmin identifiziert werden und vermindert Serotransferrin, Protaglandin-H2-Isomerase, Cystatin C und Transthyretin.
  • In the present work the protein composition of liquor from patients with neurological disease was analyzed and the proteins were investigated quantitatively. The aim of this study was the identification of proteins that could serve as disease markers. Two immune-mediated neurological diseases, multiple sclerosis (MS) and Guillain-Barré Synrom (GBS) were tested. The liquorproteins of MS and GBS patients were precipitated using 2D-gel electrophoresis and analyzed quantitatively. Quantification revealed a decrease in 11 proteins and an increase in 15 proteins in the MS-liquor, whereas in the liquor of the GBS-patients 7 proteins were decreased and 4 proteins were increased. Amounts of antithrombin III, protaglandin-H2-isomerase and transthyretin proteins were reduced, while complement B, beta-2-glykoprotein, protaglandin-H2-isomerase and cystatin C proteins were found in high amounts in MS-patients. Concerning the GBS-patients, the proteins haptoglobin und ceruloplasmin were detected in elevated, and serotransferrin, protaglandin-H2-isomerase, cystatin C und transthyretin proteins in reduced quantities.
