Untersuchungen zur oralen Inokulation und Kolonisation mit parodontal-pathogenen Bakterien im murinen Modell.

  • In der vorliegenden Arbeit wurden die nicht-invasiven Inokulationsmethoden Gavage und Inokulation über das Futter zur Induktion einer Parodontitis im murinen Modell untersucht und verglichen. Für die experimentelle Infektion wurden A. actinomycetemcomitans und P. gingivalis eingesetzt. Untersucht wurde, ob diese Keime die Maulhöhle von BALB/c Mäusen über einen längeren Zeitraum kolonisieren können und wann sie einen Knochenabbau induzieren. Es konnte eine gute Kolonisation mit dem Stamm A. actinomycetemcomitans 1005 erzielt werden, wenn die Infektion über Futtergabe erfolgte. Auch kam es zu einem signifikant erhöhten Knochenabbau bei der Infektion über Futtergabe. Ein deutlich erhöhter Antikörpertiter gegenüber A. actinomycetemcomitans bei den infizierten Tieren zeigt an, dass die Infektion auch eine systemische humorale Immunantwort auslöste. Im Gegensatz zu der Inokulation über das Futter konnte bei der Infektion mittels Gavage jedoch keine befriedigende Kolonisation der Maulhöhle erreicht werden. Es kam unter dieser Infektionsmethode zwar tendenziell zu einem Knochenabbau, der aber statistisch nicht signifikant war. Nach P. gingivalis Infektion konnte zwar eine Kolonisation nach der Infektion nachgewiesen werden. Allerdings konnte nach Infektion per Gavage kein signifikanter Knochenbau im Vergleich zu der Kontrollgruppe erzeugt werden. Die erhobenen Daten zeigen, dass die Induktion einer Parodontitis in weiblichen BALB/c Mäusen mit dem Stamm A. actinomycetemcomitans 1005 mittels Inokulation über das Futter eine vielversprechende Methode darstellt.
  • In the present study, the non-invasive methods of inoculation, gavage and inoculation over the feed, were compared and examined for the induction of periodontal disease in the mouse model. A. actinomycetemcomitans and P. gingivalis were used for experimental infection. Whether these bacteria can colonize the oral cavity of BALB / c mice over a longer period of time and the induction bone loss was investigated. Using the strain A. actinomycetemcomitans 1005, by inoculation via feed, a solid oral colonization and a significantly increased bone resorption were observed. A significantly increased antibody titer against A. actinomycetemcomitans in infected animals indicates that the infection caused a systemic humoral immune response. In contrast to the inoculation with the feed the infection by gavage however showed no satisfactory colonization of the oral cavity. The bone resorption using the gavage method showed a tendency to more bone loss but the results weren’t statistically significant. The P. gingivalis infection via gavage showed a colonization of the oral cavity. However, there was no significant bone loss in comparison to the control group. The collected data show that the induction of periodontitis in female BALB / c mice with the strain A. actinomycetemcomitans 1005 by inoculation via the feed is a promising method.
