Bestimmung der Häufigkeit und Isolierung des Granulozytenantigens HNA-3a

  • Einführung Das humane Neutrophilen-Antigen HNA-3a steht in ursächlichem Zusammenhang mit der Entstehung transfusions- assoziierter Lungeninsuffizienz und weiteren Erkrankungen. Die Isolierung des Antigens ermöglichte eine Identifizierung und den Aufbau von routinemäßig durchführbaren Testverfahren. Zielsetzung 1. Etablieren eines Nachweisverfahrens und Screening von Blutspendern auf HNA-3a. 2. Aufreinigung der aHNA-3a-Antikörper aus dem Plasma immunisierter Blutspender. 3. Biotinylierung des Antigens und Koppelung an verschiedene Festphasen. 4. Immunpräzipitation. 5. Detektion mittels Gelelektrophorese und Immunoblotting. Material und Methode Mit Agglutinationstests und Durchflußzytometrie wurde das Screening durchgeführt. Um das Antigen nach einer Isolierung nachweisen zu können, erfolgte eine Biotinylierung der Oberflächen-Proteine. Die Isolierung erfolgte mittels Immunpräzipitation, 1-D–Gel- elektrophorese und einen Nachweis im Western-Blot mit konjugiertem Streptavidin. Ergebnisse Im Screening wurden mehrere HNA-3a-negative Spender identifiziert. Die Häufigkeit des Antigens lag bei 98,5 %. Die Biotinylierung beeinflusste die verwendeten Verfahren nicht bemerkbar. Bei der Immunpräzipitation mit HNA-3a-positiven Granulozyten, die mit aHNA- 3a-Plasma inkubiert wurden, stellte sich reproduzierbar eine Bande zwischen 90 und 105 kDa dar. Diskussion Da die Ergebnisse reproduzierbar waren, konnte davon ausgegangen werden, dass es sich bei der Bande, die bei der Immunpräzipitation mit aHNA-3a Plasma und HNA-3a positiven Lysaten spezifisch ist, um das das Epitop HNA-3a tragende Protein handelt. Für eine Identifizierung des Antigens konnte die Bande aus dem Gel ausgeschnitten werden und mit weitergehenden Methoden charakterisiert werden. Exakt dieses Vorgehen führte zu der erfolgreichen Identifizierung des Epitops HNA-3a.

The Isolation of the antigen allowed the identification and the establishment of routine testing Methods. Objective: First Establishing a detection Method and Screening of blood Donors for HNA-3a. Second Purification of the Ahna-3a antibodies from the plasma of immunized blood donors. Third Biotinylation of the antigen and coupling to various solid phases. 4th Immunoprecipitation. 5th Detection by gel electrophoresis and immunoblotting. Materials and Methods: With agglutination test and flow cytometry screening was performed. To demonstrate the antigen on a Western-Blot after Immuneprecitpitation Biotinylation of surface Proteins was performed. Results: In screening several HNA-3a-negative donors were identified. The frequency of Antigen was 98.5%. Biotinylation did not noticeably affect the method used. For Immunoprecipitation HNA-3a-positive granulocytes, incubated with aHNA-3a-plasma stood reproducibly a band ranging from 90-105 kDa. Discussion: Since the results were reproducible, it could be assumed that the ptotein isolated with immunoprecipitation using aHNA-3a plasma and HNA-3a positive lysates is the HNA-3a-bearing protein. For Identification of the Band was cut out the Western-Blot and characterized by further methods. Exactly this Approach led to the successful Identification of the Epitope HNA-3a.
