Zelluläre und molekulare Charakterisierung der Docetaxel-Wirkung auf Prostatakarzinomzellen

  • Docetaxel ist ein halbsynthetisches Zytostatikum aus der Klasse der Taxane, welches den intrazellulären Mikrotubulusapparat stabilisiert und so den gezielten Zelltod einleitet. Dieser Mechanismus wird zur Behandlung von schnell proliferierenden Tumorzellen genutzt, weshalb Docetaxel bis dato als eine First-line Medikation des kastrationsresisten Prostatakarzinoms (CRPC) gilt. Sowohl ein negatives Ansprechen der Therapie als auch die Entstehung zahlreicher Resistenzmechanismen können zur Verzögerung einer adäquaten Behandlung der malignen Erkrankung führen. Besonders Expressionänderungen von HSP27 werden in diesem Zusammenhang als Ursache für Zytostatikaresistenzen in diversen Tumorarten angegeben. In dieser Arbeit sollten die zellulären sowie molekularen Wirkungsweisen von Docetaxel auf PCa-Zellen verschiedener Dignität charakterisiert werden. Dabei wurde gezeigt, dass Docetaxel in allen Zelllinien einen negativen Einfluss auf die Vitalität und Zellzahl ausübt. Über die Aktivierung des Tumorsuppressors p53 führt Docetaxel in LNCaP-Zellen zur Fragmentierung der Poly-(ADP-Ribose)-Polymerase, was den Eintritt in die Apoptose charakterisiert. Wir demonstrierten, dass unter Docetaxel-Behandlung die Expression von HSP27 in LNCaP-, PC-3-Zellen sowie in PC-3-Zellen mit überexprimiertem HSP27 sowohl auf mRNA- als auch auf Proteinebene steigt. Weiterhin wurde bewiesen, dass eine erhöhte HSP27-Expression mit einer intrazellulären Chemoresistenz einhergeht und dies sowohl in hormonsensitiven als auch in hormoninsensitiven PCa-Zellen den vitalitäts- und proliferationshemmenden Effekt von Docetaxel verringert. Die Blockierung von HSP27, wie es derzeit mit dem HSP27-Inhibitor OGX-427 in Phase II-Studien getestet wird, könnte somit einen geeigneten Angriffspunkt zur Verhinderung einer Resistenzentwicklung im CRPC darstellen und eine adäquate Therapie ermöglichen.
  • Docetaxel is a semisynthetic cytostatic drug from the class of taxanes, which stabilizes microtubules intracellular and initiates the targeted cell death. This mechanism is used for the treatment of rapidly proliferating tumor cells. This is the reason why docetaxel is still considered as a first-line medication of castration resistant prostate cancer (CRPC). A negative response to treatment and the development of numerous resistance mechanisms may result in a delay of appropriate tumor treatment. Especially changes in the expression of HSP27 are reported in this context as a cause of cytostatic drug resistance in various tumor types. In this study the cellular and molecular mechanisms of docetaxel were characterized in different prostate cancer cells (PCa cells). We could show that docetaxel influences negatively the viability and number of cells in all cell lines. Via activation of the tumor suppressor p53, docetaxel leads to fragmentation of the poly-(ADP-ribose)-polymerase in LNCaP cells, which characterizes the entry into apoptosis. Further it has been demonstrated that docetaxel treatment increases the mRNA and protein expression of HSP27 in LNCaP, PC-3 cells as well as in PC-3 cells with overexpressed HSP27. We proved that an increased HSP27 expression is associated with intracellular chemoresistance in hormone-sensitive and in hormon-insensitive PCa cells and impairs the vitality- and proliferation-inhibiting effects of docetaxel. Blocking HSP27, as currently being tested in Phase II trialswith the HSP27 inhibitor OGX-427, could thus constitute a suitable point of attack, prevent the development of resistance in CRPC and might allow an adequate therapy.
