  • The relationship between the activity of different epigenetic-associated enzymes and neoplastic diseases can be regarded as experimentally confirmed. In addition, it was also proven causally with approval of inhibitors of DNA methyltransferase and histone deacetylases. The less well studied 2 oxoglutarate and iron(II)-dependent JumonjiC domain-containing histone demethylases (KDMs) are also considered as a possible epigenetic target in tumor therapy due to overexpression in some tumor cells. One aim of this work was the synthesis of KDM4 inhibitors with tetrazole- and hydrazide motifs. In further investigations of a known inhibitor, synthetic routes were developed to modify the aliphatic backbone with aromatic residues or to completely replace the backbone with an aromatic structure based on anthranilic acid. Moreover, synthetic routes for various (1H-tetrazol-5-yl) pyridines were evaluated. For this purpose, a tetrazole moiety was inserted in position four of the pyridine ring and systematic investigations of the side chain in position two of the pyridine ring were carried out. Besides direct heteroatom coupling, carbonyl compounds such as carboxylic acid amides or a hydroxamic acid were also prepared. Additionally, methylene group containing substances in position two of the pyridine ring were successfully synthesized. Biological testing of the compounds was performed with an antibody-based LANCE assay with KDM4A. In addition, four of the substances were measured by X-ray crystallography bound to the isoform KDM4D. A hydroxamic acid inserted in position two of the pyridine ring showed relevant interactions in the enzyme and demonstrated that (1H-tetrazol-5-yl) pyridines are able to exert high affinity towards certain isoforms of histone demethylases. The relatively well decoded relationships between epigenetics and neoplastic diseases were extended with experiments in rodents which demonstrated that epigenetically modifying enzymes are also involved in the development of depression-like phenotypes. However, the research for new therapeutic options apart from the monoamine hypothesis for depressed patients is of major interest because approved drugs are known to have strong side effects and delayed onset. In the clinic, depressed patients who were treated with the NMDA receptor antagonist ketamine showed rapid symptom relief with a comparably favorable side-effect profile. The responsible molecules for this mood improvement are still the subject of current research, because in addition to ketamine itself some metabolites such as (2R,6R)-hydroxynorketamine are associated with antidepressant effects. For the qualitative and quantitative analysis of (R)- and (S)-ketamine as well as the metabolites (R)- and (S)-norketamine, (R)- and (S)-dehydronorketamine and (2R,6R)- and (2S,6S)-hydroxynorketamine, respectively, in human urine a chromatographic method was developed and validated. Therefore, supercritical fluid chromatography and mass spectrometry were applied (SFC-MS). The inherent features of SFC such as high flow rates, excellent separation efficiency and the utilization of carbon dioxide as an eco-friendly mobile phase component could be used to solve the separation problem in just one time-saving method. In addition, the consumption of organic solvents was reduced. Thus, the developed method may have an impact on future ketamine research.
