Charakterisierung des Abbaus von Bisphenol A durch Cupriavidus basilensis und Bewertung der cytotoxischen Wirkung dieses Schadstoffs anhand von Membranlipidanalysen in Pseudomonas putida

  • Bisphenol A (BPA) ist ein Umweltschadstoff, der für die Produktion von Polykarbonatplastik und Epoxydharzen verwendet wird. Aufgrund seiner weltweiten Verbreitung, seiner Persistenz in der Umwelt und vor allen Dingen wegen seiner endokrinen Wirkung, stellt diese Verbindung eine große Gefahr für die Tier- und Pflanzenwelt sowie für den Menschen dar. Deshalb wurde aus einem bepflanzten Festbettreaktor, welcher mit BPA behandelt wurde, der Bakterienstamm RW4 isoliert, der fähig ist Bisphenol A als alleinige Kohlenstoff- und Energiequelle zu nutzen. Das Gram-negative Bakterium wurde mittels 16S rRNA-Analyse als Cupriavidus basilensis identifiziert und mittel der BIOLOG-Methode charakterisiert. Der Bakterienstamm kann ein sehr großes Spektrum an aliphatischen Stoffen wie Aceton, Ethanol, 1-Oktanol und aromatischen Verbindungen wie Phenol, Mono- und Dihydroxybenzoesäuren für das Wachstum nutzen. C. basilensis RW4 baut Bisphenol A nur sehr langsam ab. Es ist jedoch möglich den BPA-Abbau sowohl in Schüttelkulturen und als auch in kontinuierlich laufenden Sandsäulen durch Zugabe von Phenol als Co-Substrat und Wachstumsstimulanz deutlich zu steigern. Die erfassten Abbauintermediate wie 4-Hydroxyacetophenon und 2-(4-Propanol)-phenol wiesen darauf hin, dass C. basilensis RW4 einen ähnlichen Abbauweg nutzt wie Sphingomonas sp. TTNP3 von Kolvenbach et al. (2007) nutzt. In weiteren Untersuchungen, wurde die toxische Wirkung von Bisphenol A auf den Standardmikroorganismus Pseudomonas putida P8 in Schüttelkulturen und in kontinuierlich laufenden Sandsäulen analysiert. Es zeigte sich, dass BPA das Wachstum von P. putida P8 hemmt und Modifikationen in der Zusammensetzung der Phospholipide in der Membran hervorruft. P. putida P8 reagierte unmittelbar auf das Einwirken von BPA auf der Membranebene durch Veränderung des Verhältnisses der trans-ungesättigten Fettsäuren zu den cis-ungesättigten Fettsäuren, was schon in früheren Untersuchungen für andere organische Lösungsmittel beschrieben wurde. Dementsprechend wurde eine Korrelation zwischen der BPA-Konzentration und dem Anstieg des trans-cis-Verhältnisses ermittelt. Weil diese Anpassungsreaktion eine kurzfristige Antwort auf den einwirkenden Stressor ist, konnte der Anstieg des trans-cis-Verhältnisses nur kurz nach BPA-Zugabe erfasst werden. Somit kann das trans-cis-Verhältnis nur als temporärer Biomarker für die Toxizität von BPA in Schüttelkulturen genutzt werden, jedoch nicht in langfristig laufenden Sandsäulen.
  • Bisphenol A (BPA) is an organic pollutant, which is used for the production of polycarbonate plastics and resigns. Because of its ubiquitous distribution, its persistence in the environment and especially its endocrine effects, BPA poses a high risk on plants, animals and even human. Therefore the bacterial strain RW4, which is able to utilize bisphenol A as sole carbon and energy source was isolated from a planted fixed bed reactor. The Gram-negative bacterium was identified as Cupriavidus basilensis via 16S rRNA-analysis and characterized via the BIOLOG method. Next to BPA the strain utilizes a wide range of aliphatic compounds e.g. acetone, ethanol, 1-octanol and aromatic compounds e.g. phenol, mono- and dihydroxybenzoic acid for growth. C. basilensis RW4 degraded BPA very slow. But it was possible to enhance the BPA degradation in batch cultures and in continuously running sand columns by applying phenol as co-substrate and growth stimulant. Detection of various degradation intermediates as 4-(2-propanol)-phenol and 4-hydroxyphenone indicated that C. basilensis RW4 uses a degradation pathway similar to those postulated for the strain Sphingomonas sp. TTNP3 (Kolvenbach et al., 2007). In further investigations, the toxic effects of Bisphenol A were studied on a standard microorganism Pseudomonas putida P8 in batch cultures and continuously running sand columns. It was shown that BPA inhibited growth of P. putida P8 and induced changes in the membrane phospholipid fatty acid composition as adaptation reactions. Cells of P. putida P8, without the capability for BPA-degradation, immediately responded to BPA via altering the trans-cis ratio of their unsaturated fatty acids, as already described for other organic solvents in former studies. Thus, a correlation between the BPA concentration and the increase in the ratio of trans unsaturated to cis unsaturated fatty acids could be ascertained. Since the increase of the trans-cis ratio was only detectable briefly after BPA-addition; this adaptation reaction is a short-term response to toxic compounds. Therefore, the trans-cis ratio can only be used as temporary marker for the toxicity of BPA in batch cultures but not in long-term continuously running sand columns.
