Der Einsatz von atmosphärischem Niedertemperaturplasma zur Behandlung chronischer Wunden stellt eine erfolgversprechende, in Entwicklung befindliche Therapieoption dar. Dabei wird von der Hypothese ausgegangen, dass chronische Wunden durch die Stimulation mittels Plasma aktiviert werden können, wodurch die über die Stufe der resorptiven Inflammation die physiologische Heilung wieder in Gang gesetzt werden kann. Daher wurde zur Identifizierung des Reiz- und Inflammationspotential des Plasmas und zur Ableitung geeigneter Behandlungsparameter Plasma im HET-CAM geprüft. Zunächst wurden mit einem Atmosphärendruck Plasma-Jet unter Verwendung von Argongas und einer Dielectric-Barrier Discharge (DBD)-Plasmaquelle die Reizwirkungen an der CAM durch mäanderförmige Plasmabehandlung und punktuelle Anwendung getestet. Dabei wurden neben mäanderförmigen und punktuellen Applikationsmustern unterschiedliche Anwendungsfrequenzen und damit unterschiedliche Plasmadosen untersucht. Die Temperatur des Plasmas nahm v. a. bei längeren Behandlungszeiten Einfluss und führte zu einer Wirkungsverstärkung bis hin zu thermischen Schädigungen. Der Gasfluss erwies sich als limitierend für das geprüfte Modell. Der gepulste Modus war bei mäanderförmiger Anwendung am verträglichsten. Die Überprüfung auf Reversibilität der Effekte ergab, dass geringe Reizungen vollständig reversibel waren, währen schwere Reizungen zu nicht reversiblen Koagulationen führten. Aufgrund der unbefriedigenden Ergebnisse mit sehr schneller Bildung von Thrombosen, geringem Arbeitsabstand und fehlender Potentialfreiheit wurde die DBD-Elektrode von der weiteren Untersuchung ausgenommen und der kINPen09 als Weiterentwicklung des Plasma-Jets mit geringerer Plasmatemperatur auf inflammatorische Wirkung bei punktueller Anwendung mit verschiedenen Expositionszeiten im kontinuierlichen Modus und im gepulsten Modus mit Argon als Trägergas, sowie im kontinuierlichen Modus unter Zusatz von 0,1 % Sauerstoff, getestet. In dieser Testreihe war ebenfalls das gepulste Plasma am verträglichsten, das mit Sauerstoff angereicherte Plasma führte zu den schwersten Inflammationen. Alle Reaktionen wurden durch die kombinierte Anwendung mit Hydrocortison deutlich abgeschwächt. Mit der Inflammationsinduktion konnten Gewebeprozesse in Form von Kontraktion, Koagulation und inflammationsassoziierter Angiogenese auf der CAM erzeugt werden, die für Aktivierung der sekundären Wundheilung relevant sein dürften. Ableitend aus den Untersuchungen kann für die Anwendung an chronischen Wunden eine Behandlungszeit von maximal 5 s pro qcm Wundfläche für den kontinuierlichen Modus empfohlen werden. Dabei sollte wegen der besseren Verträglichkeit der mäanderförmigen Behandlung gegenüber der punktuellen Anwendung der Vorzug gegeben werden. Unter Berücksichtigung der vorliegenden Untersuchung zur Reiz- und Inflammationswirkung von Plasma im HET-CAM und weiteren Prüfungen zur Wirksamkeit und Verträglichkeit in vivo und in vitro erweist sich die Plasmaapplikation mit dem kINPen09 als vielversprechende Option zur Behandlung chronischer Wunden.
Auf dem Teilgebiet der Plasmamedizin, welches sich mit der Grundlagenforschung der Plasma-Zielstruktur-Interaktion beschäftigt, haben sich in den letzten Jahren zwei grundlegende Erkenntnisse ergeben: (1) Biologische Effekte kalter Atmosphärendruckplasmen (CAP) werden maßgeblich über die Flüssigkeitsphasen vermittelt. (2) Diese Effekte basieren auf den Reaktionen redoxaktiver Spezies. Darüberhinaus wurde festgestellt, dass Flüssigkeiten durch die Behandlung mit CAP "aktiviert" werden können und selbst einen biologischen Effekt aufweisen. Auf dieser Grundlage beschäftigt sich nun ein Großteil der Untersuchungen mit der Identifikation der biologisch relevanten redoxaktiven Spezies in Flüssigkeiten. In der Redoxbiologie sind verschiedene Reaktionsketten bekannt, welche reversible Oxidations- und Reduktionsreaktionen beinhalten. Diese redoxbasierten Interaktionen sind nicht stets liganden- bzw. rezeptorspezifisch, so dass sich folgende Hypothese formulieren lässt: Der biologische Plasmaeffekt von CAP wird nicht durch einzelne, rezeptorspezifische Reaktionen ausgelöst, sondern ist vielmehr von dem sich in der flüssigen Umgebung der behandelten Zielstruktur herrschenden Redoxpotential abhängig. Für die Untersuchung der Hypothese wurde physiologische Kochsalzlösung in Luft mit einer Plasmaquelle, die nach dem Prinzip einer dielektrisch behinderten Oberflächenentladung funktioniert, behandelt und der antibakterieller Effekt der plasmabehandelten physiologischen Kochsalzlösung gegen E. coli erfasst. Parallel dazu wurde das Redoxpotential der plasmabehandelten physiologischen Kochsalzlösung mittels einer Goldelektrode erfasst. 5 ml physiologische Kochsalzlösung, welche 5 min mit CAP behandelt wurde, führte zu einer kompletten Inaktivierung von E. coli mit einer Reduktion der Bakterienanzahl > 6 Log-Stufen, wohingegen eine 3 minütige Plasmabehandlung nur eine Reduktion der Bakterienzahl < 1 Log-Stufe zur Folge hatte. Die komplette Inaktivierung von E. coli entsprach einer Redoxspannung von 414 mV (nach 3 min CAP-Behandlung: 376 mV). Bei Untersuchungen mit einem Vergleichssystem, welches aus verschieden konzentrierten Wasserstoffperoxidlösungen bestand, konnte gezeigt werden, dass mit einer Wasserstoffperoxidkonzentration von 30 000 mg/l eine komplette Inaktivierung von E. coli erreicht werden konnte. Die dabei herrschende Redoxspannung betrug ebenfalls 414 mV. Jedoch konnte auch gezeigt werden, dass die in plasmabehandelter physiologischer Kochsalzlösung herrschende Redoxspannung nicht allein durch die dabei herrschende Wasserstoffperoxidkonzentration ausgelöst wird, da die Wasserstoffperoxidkonzentration in 5 ml CAP-behandelter physiologischer Kochsalzlösung nur 5 mg/l betrug. Anhand dieser ersten Untersuchungsergebnisse lässt sich zusammenfassen, dass der antibakterielle Effekt, sowohl von CAP-behandelter physiologischer Kochsalzlösung, als auch von Wasserstoffperoxidlösungen, mit der in der jeweiligen Flüssigkeit herrschenden Redoxspannung korreliert. Inwieweit sich dies für andere biologische Plasmaeffekte nutzen lässt, ist noch nicht absehbar. Dafür bedarf es weiterführender Untersuchungen.
Die Zahl von Parodontitispatienten steigt jährlich an. Außerdem wurden vermehrt Implantate insertiert, die analog zur Parodontitis von Periimplantitis betroffen sind. Ursächlich für beide Erkrankungen sind Biofilme. Es gibt keine befriedigenden Methoden zur Biofilmentfernung, die außerdem eine wundheilungsfördernde Oberfläche erzeugen. Daher werden neue Behandlungsmethoden benötigt. In dieser Arbeit wurde drei Biofilmmodelle mit C. albicans, S. mutans und Speichelmikroorganismen mit drei verschiedenen Plasmaquellen (kINPen09, Hohlelektroden-DBD, Volumen-DBD) sowie zwei verschiedenen Gasmischungen (Argon und Argon+1% O2) jeweils 1, 2, 5 und 10 min mit Plasma behandelt. Als Positivkontrolle wurde Chlorhexidin mitgeführt. Außerdem wurden verschiedene Titanbearbeitungsformen (maschiniert, diamantbearbeitet, pulverbestrahlt sowie geätzt und gestrahlt) mit Argon+1%O2-Plasma mittels kINPen09 behandelt. Anschließend wurden die Elementzusammensetzung, der Kontaktwinkel sowie die Ausbreitung von osteoblastenartigen Zellen MG-63 auf diesen Oberflächen bestimmt. SLactive􀂓 wurde hierbei als Positivkontrolle verwendet. Um eine potentielle Anwendung in der Parodontologie zu prüfen, wurden diese Untersuchungen auch auf Dentin durchgeführt. Alle Plasmaquellen und –parameter wirkten antimikrobiell. Die Zerstörung der Zellen wurde im Rasterelektronenmikroskop deutlich. Hierbei reduzierte die Volumen-DBD die Koloniebildenden Einheiten um circa 5 log-Stufen und wies damit die höchste antimikrobielle Wirksamkeit auf. Sauerstoffzumischung führte nur bei der Hohlelektroden-DBD zu einer erhöhten antimikrobiellen Wirksamkeit. Die Plasmabehandlung reduzierte die Kontaktwinkel auf allen Oberflächen teilweise bis in den superhydrophilen Bereich. EDX-Analysen zeigten eine Reduktion der Masseprozent von Kohlenstoff sowie eine Erhöhung des Sauerstoffgehalts aller Oberflächen nach Plasmabehandlung. Die Ausbreitung der Osteoblasten war auf den plasmabehandelten Oberflächen signifikant höher als auf den unbehandelten Oberflächen und konnte sogar die Werte der hydrophilen SLactive􀂓-Oberfläche übersteigen. Diese Effekte konnten sowohl auf Titan als auch auf Dentin nachgewiesen werden. Da Plasma antimikrobiell wirkt und, wie in weiterführenden Versuchen gezeigt werden konnte, auch Biofilm entfernt, eignet es sich zur Therapie der Periimplantitis und Parodontitis. Außerdem wird die Oberfläche biokompatibler, wodurch die Wundheilung gefördert werden könnte. Da Plasma weitere wundheilungsstimulierende Faktoren beinhaltet, stellt es in Zukunft eine Erfolg versprechende Therapieoption für die Behandlung von Parodontitis und Periimplantits dar.
In der vorliegenden Arbeit wurde der Einfluss von kaltem Atmosphärendruckplasma (CAP) auf die Oberfläche des dentalen Restaurationsmaterials Amalgam untersucht.
Bisher liegen in der Literatur noch keine vergleichbaren Untersuchungen vor.
Die Amalgamprüfkörper wurden aus dem kupferhaltigen Non-Gamma-2-Amalgam Amalcap plus (Ivoclar Vivadent, Schaan, Liechtenstein) hergestellt, welches in zylinderförmige Polyoxymethylenformen kondensiert wurde. Die Kaltplasma-Behandlung erfolgte mit dem kINPen® (neoplas GmbH, Greifswald, Deutschland) in 3 Behandlungsgruppen. Gruppe A wurde 60 Sekunden rasternd, Gruppe B 30 Sekunden stationär und Gruppe C 5 Minuten stationär mit CAP behandelt.
Die Amalgamprüfkörperoberflächen wurden jeweils vor und nach der Plasmabehandlung untersucht. Die mikroskopische Dokumentation erfolgte hierbei mittels Lichtmikroskopie und Rasterelektronenmikroskopie (REM), die chemische Analyse durch die Röntgenphotoelektronenspektroskopie (XPS). Für die Beurteilung der Oberflächenrauheit wurden Weißlichtinterferometer und Profilometer eingesetzt. Zur Ermittlung der Hydrophilität kam die Kontaktwinkelmessung zum Einsatz.
Optisch traten in Behandlungsgruppe A und B keine Veränderungen auf. Lediglich bei den 5 Minuten plasmabehandelten Amalgamproben waren leichte Veränderungen erkennbar, die vor allem im Bereich der Poren und der optisch dunkleren Cu6Sn5-Phase lokalisiert waren und sich mikroskopisch braun-orange beziehungsweise dunkelgrau bis schwarz darstellten. Mittels Focused Ion Beam (FIB) präparierte Querschnitte zeigten hierbei keine Veränderungen in der Tiefe, sodass anzunehmen ist, dass die Effekte sich auf die direkt mit dem Plasma in Kontakt stehende Oberfläche beschränken.
Eine Veränderung der Oberflächenrauheit konnte in keiner Behandlungsgruppe festgestellt werden.
Direkt nach der Plasmabehandlung zeigte sich in allen Behandlungsgruppen eine Hydrophilisierung der Oberfläche. Diese war in Gruppe C am stärksten ausgeprägt. Allerdings haben sich bereits nach 2 Tagen die Ausgangswerte wieder eingestellt, der Effekt war also reversibel. Elementar hat durch die Plasmabehandlung im Rahmen eines Reinigungseffekts der oberflächliche Kohlenstoffanteil deutlich abgenommen, der Sauerstoffanteil hingegen zugenommen. Gleichzeitig stieg das Zinn/Quecksilberverhältnis zugunsten des Zinns. Zusätzlich ist es in Gruppe A zu einer Verschiebung des Cu 2p-Peaks in Richtung einer geringeren Bindungsenergie gekommen, das Kupfer lag also nicht mehr einwertig, sondern zunehmend zweiwertig vor.
Mittels energiedispersiver Röntgenspektroskopie (EDX) war es zudem möglich, die an der Oberfläche optisch differenzierbaren Phasen des Amalgams zu charakterisieren.
Insgesamt kann festgestellt werden, dass es in durchgeführten Untersuchungen keine Hinweise gab, die darauf schließen lassen, dass es durch die Einwirkung von CAP auf Amalgamrestaurationen zu unerwünschten Effekten bezüglich der Oberflächeneigenschaften kommt, die sich negativ auf Patienten oder Behandler auswirken könnten. Um die Übertragbarkeit der Ergebnisse auf die klinische Praxis sicherzustellen, sind allerdings noch weitere Untersuchungen notwendig.
Pharmaceutical residues are found in increasing concentrations in the environment and in potable water where they have verifiable effects on aquatic life. Conventional methods for water treatment are not able to sufficiently abate these generally stable compounds. It was found that physical plasma generated directly in water can degrade several of these recalcitrant organic pollutants. Studies on the basic plasma chemical processes for the model system of phenol showed that the degradation is primarily caused by hydroxyl radicals. This was confirmed by reaction chemistry and spin trap enhanced electron paramagnetic resonance spectroscopy (EPR). The degradation of diclofenac and its by-products were investigated in detail to perform a first risk-assessment of the new technology. Findings are not limited to the application of plasma but applicable to other advanced oxidation processes (AOP) that are based on the generation of hydroxyl radicals as well. Additionally, pulsed corona plasma and pulsed electric fields were assessed for their capacity to kill Legionella pneumophila in water. Whereas it was possible to kill L. Pneumophila with both methods, plasma treatment resulted in an enhanced bacterial killing. Therefore, advanced oxidation processes (AOP) and plasma treatment in particular are some of the few feasible approaches to decompose recalcitrant compounds in water.
On the aqueous phase chemistry of atmospheric-pressure plasma jets for biomedical applications

Cold atmospheric-pressure plasmas are candidate biomedical tools proposed for various applications, such as biological decontamination, cancer regression, and promotion of wound healing. Plasmas, which are in the fourth state of matter, can be generated using inert gases (e.g., argon, helium, ambient air) and different source concepts. Together with the applied parameters, the source design defines the chemical-physical characteristics of the resulting plasma, leading in turn to variable biochemical effects on biological matter. The medical effectiveness of cold plasmas has been proven in vitro and in vivo, also in clinical trials for wound healing in patients using two certified plasmas sources, the kINPen MED and the PlasmaDerm. However, molecular mechanisms leading to those effects are unclear. In the same way, it must be studied if the modulation of plasma properties could improve the specificity of biological effects. These findings are needed to define the concept of plasma dose to be optimized in targeting peculiar pathologic conditions. The present thesis consisting of five peer-reviewed publications has investigated these aspects of plasma research.
In the gaseous phase of cold plasmas, various components with biological activity are produced, such as radiation (e.g., vacuum UV, UV) and reactive species (e.g., •O, 1O2, •OH, •NO, •NO2, O3). As most gaseous species are short-lived, liquid compartments surrounding cells and molecular structures could mediate their transformation and/or the production of other aqueous species. For this reason, plasma-induced aqueous chemistry has been mainly investigated in this thesis. The reaction pathways of reactive oxygen and nitrogen species in liquid were analyzed by monitoring the oxidative modifications induced on tyrosine and cysteine, which are biological structures essential in cellular protein functioning. Liquid chromatography and mass spectrometry-based strategies have been elaborated to elucidate structural changes and characterize the oxidative pattern occurring on the tracers after treatment with plasmas.
As a first result, it could be shown that the oxidative pattern induced on tyrosine or cysteine variated qualitatively and quantitatively with the applied conditions, reflecting the action of differently produced/deposited species in liquid. Biologically relevant structures were identified and in part quantified (e.g., cystine, sulfonic acid, sulfinic acid, S-sulfonate, S-nitrosocysteine, nitrotyrosine, nitrosotyrosine). By using isotopically labeled oxygen or nitrogen in the gas plasma, or labeled oxygen in the target liquid, the incorporation of gaseous or aqueous species in the tracer’s structures was monitored via mass spectrometry. With this strategy, the reaction mechanisms involving gaseous oxygen and nitrogen species at the liquid interface were clarified, as well as the de novo production of reactive species in liquid. Short-lived gaseous oxygen species such as atomic and singlet oxygen (•O, 1O2), predominantly formed in conditions with oxygen in the plasma gas, were able to modify the cysteine structures in highly oxidized derivatives, such as cysteine sulfonic acid. Due to their half-life, however, their activity occurred mainly at the interface. Vacuum UV radiation and •O also led to the formation in liquid of hydroxyl radicals (•OH) and hydrogen peroxide (H2O2), due to water photolysis and homolysis. Water-derived species were responsible for the formation of reversible modifications, such as cysteine S-sulfonate, cystine, and cystine sulfoxides. Nitrosative modifications (e.g., S-nitrosocysteine, nitrosotyrosine, nitrotyrosine) could be observed only in conditions with both nitrogen and oxygen in the plasma gas, and further optimization occurred in presence of water molecules in the gas. In this case, the formation and action of peroxynitrite (ONOO-) in generating nitrotyrosine was proven by using a scavenger molecule for ONOO-.
Finally, the cysteine product pattern was applied as a tool to characterize and compare the overall chemistry generated in liquid by different plasma sources and applied parameters. These findings aim to support and contribute to the definition of plasma dose for plasma medicine, through the standardization, control, tuning, and optimization of plasma parameters and plasma liquid chemistry. These results may be applied in the future to improve the specificity and selectivity of the biological effects generated by the described atmospheric-pressure plasma jets.
Ziel dieser Arbeit war die Darstellung der physikalischen Charakteristik der zu untersuchenden DBD-Plasmaquelle. Außerdem sollte orientierend gezeigt werden, dass die entwickelte und geprüfte Plasmaquelle in dieser Konzeption biologisch verträglich ist und eine klinisch relevante Bakterienreduktion in vitro (RF) ermöglicht. Für die Risikobewertung wurden Untersuchungen der zu erwartenden UV-Belastung nach gültiger Referenzierung (ICNIRP), Ex-vivo-Studien mit Behandlung von Hautbiopsien sowie In-vivo-Studien am Mausmodell durchgeführt. Es konnte gezeigt werden, dass bei keimwirksamen Behandlungsdosen mit großer therapeutischer Breite keine schädigungsrelevanten UV-Dosen appliziert wurden und weder ex vivo noch in vivo mikroskopische Schädigungen im Hautzellverband auftraten. Daraus lässt sich schlussfolgern, dass die charakterisierte und geprüfte DBD-Plasmaquelle für die Behandlung humaner Haut geeignet erscheint, was durch weiterführende Untersuchungen in vitro, ex vivo und in vivo in größeren Untersuchungsserien abzusichern ist. Anhand der Daten kann als potenzielle Behandlungsindikation die antimikrobielle Hautbehandlung abgeleitet werden, z.B. MRSA-Dekontamination oder die Behandlung von oberflächlichen Hautinfektionen.
In the last decade a new domain has developed in plasma physics: plasma medicine. Despite the successes that have already been achieved in this exciting new field, the interaction of plasmas with “biological materials” is not yet fully understood. Further investigations in particular with respect to the properties of the applied plasmas sources are therefore essential in order to decode this complex interaction process. Currently, a great variety of different discharge types are used in plasma medical investigation which are generally are operated in noble gases like helium and argon or with dry air. In the present work, the main focuses is on the diagnostics of reactive oxygen and nitrogen species (RONS) resulting from the plasma chemistry of an argon radio-frequency (RF) atmospheric pressure plasma jet (APPJ) and its interaction with the ambient atmosphere. To conduct this study, a commercially available plasma device, so-called kinpen is used due to its technical development maturity and its accessibility on the market. As a method of choice, diagnostic techniques are based on optical spectroscopy known to be a reliable tool to investigate plasmas. Consequently, three complementary optical laser diagnostics, namely quantum cascade laser absorption spectroscopy (QCLAS), laser induced fluorescence (LIF) and planar single shot LIF (PLIF), have been successfully applied to the plasma jet itself or its effluent. All of these diagnostics offer a high species selectivity and an excellent spatial and temporal resolution. They are used in this work for i) the characterization of the plasma chemical dynamics with respect to the generation of biological active RONS – in particular for the case of N2 and O2 admixtures. ii) the measurement of the NO density profile in the plasma effluent iii) the investigation of the flow characteristics of the neutral gas component (laminar vs. turbulent) and its influence on the plasma chemistry. Numerical analysis have been carried out in collaboration with PLASMANT (University of Antwerp) via kinetic simulations of the entire plasma chemistry. Expectingly, atomic oxygen (O) and nitric oxide (NO) turn out to be precursors of ozone (O3) and nitric dioxide (NO2). However, it was intriguing to unveil that atomic oxygen and nitrogen metastable (N2(A)) play together a key part --as intermediate species-- in the generation of more stable RONS, e.g. NO. The absolute density of NO space resolved was measured by LIF and absolutely calibrated molecular beam mass spectrometer. LIF was used to determine relative density of OH radical in the plasma plume. 2D-LIF was used to investigate the gas flow pattern with OH as a flow tracer. The results are discussed in details and show different operating mode of the jet, e.g. laminar or turbulent and that the plasma influences these regimes. The first detection and relative measurement by LIF of nitrogen metastable (N2(A)) produced by an argon APPJ is also shortly reported in this work. The outcome of this thesis will bring new insights in the field of argon APPJs chemistry and its interaction with the ambient atmosphere which can be valuable to support plasma modelling and to consider for the applications in plasma medicine.
In this work the mechanisms leading to the generation of the various reactive oxygen and nitrogen species (RONS) in a cold atmospheric plasma (CAP) jet and means to control their composition were studied. The investigated CAP jet kinpen is typically operated with Ar feed gas (pure or with molecular admixtures), driven at a frequency of approximately 1 MHz and features fast ionization waves or guided streamers, traveling at velocities of several km/s. The complex reaction networks were investigated by numerical and experimental techniques. Detailed experimental, analytical and computational investigations on the mass and heat transport in the plasma plume were performed: A novel analytical approach to diffusion in jet flows, the non-dispersive path mapping approximation (NDPM) was developed. The method for the first time allows for an estimation of the ambient species density in the near-field of jets that feature a non-homogeneous flow-field. The NDPM approximation was employed for the evaluation of laser induced fluorescence measurements on OH. Through combining measurements and NDPM approximation, this approach yielded an estimation for the ambient species density at the position of the guided streamers, not only in the laminar, but also in the (standard) turbulent operating regime. Accurate measurements of the temporally averaged ambient species density and temperature in the plasma plume were obtained by quantitative Schlieren measurements. The method yields temperature values with sub-Kelvin accuracy and, through combination with computational fluid dynamics (CFD) simulations, allowed for an estimation of the calorimetric power of the jet. In order to obtain a defined environment for the jet to operate in, a shielding gas device was designed in this work, which creates a gas curtain of defined composition around the plasma plume. The plasma dynamics on the ns timescale was investigated by phase resolved optical measurements. The effect of different shielding compositions ranging from pure N2 to pure O2 on guided streamer propagation was investigated. An electrostatic focusing mechanisms was discovered, which promotes the propagation of guided streamers along the channels formed by a noble gas in the plume of plasma jets operating in electronegative gases (such as air or O2). Two zero-dimensional (volume averaged) models were developed: First, the local processes in the guided streamer were modeled using an electron impact reaction kinetic model, which is closely correlated to densities of metastable argon (Ar*) obtained by laser atom absorption measurements. This first model shows that Ar* is the species which dominantly drives the plasma chemistry in the plasma plume. This is exploited in the second plug-flow reaction kinetics model, which is employed to investigate the formation of long-living RONS and uses an Ar* source term as sole energy input. The model uses the previous experimental data on mass and heat transport and temporal dynamics as input and is in turn verified by quantitative FTIR absorption measurements on O3, NO2, N2O, HNO3 and N2O5 in the far-field of the jet, where large absorption lengths can be achieved using a multi pass cell. For the evaluation of the zero-dimensional model, the time-of-flight of RONS from their generation to reaching the multi pass cell was determined using CFD simulations. The insight gained through this combined experimental-modeling approach on the reaction networks revealed relevant control parameters and enabled adjusting the plasma chemistry towards a desired RONS output. Through choosing appropriate feed-gas admixtures and shielding gas compositions, it is possible to generate an NOx-dominated plasma chemistry, although the jet usually produces a strongly O/O3-dominated chemistry. Understanding and controlling the plasma chemistry of cold atmospheric plasma sources for medical applications is not only essential for research, but is also the key for designing future plasma sources for specific medical applications that yield an optimum efficacy and avoid potential side effects of plasma treatment.
The biological decontamination and sterilization is a crucial processing step in producing and reprocessing of medical devices. Since polymer-based materials are increasingly used for the production of medical devices, the application of conventional sterilization processes are restricted to a certain extent. Conventional sterilization techniques on the basis of high temperatures, toxic gases, or ionizing radiation can be detrimental to the functionality and performance of polymeric materials. For this reason, alternative, gentle, and efficient decontamination processes are required. One possible approach is the use of non-thermal physical plasmas. Especially atmospheric pressure plasma is receiving great interest due to the absence of vacuum systems which is highly attractive for the practical applicability. Its mechanisms of action enable the efficient killing and inactivation of micro-organisms which are attributed to the interaction of plasma-generated reactive oxygen and nitrogen species (ROS, RNS) as well as plasma-emitted (V)UV radiation. Owing to the moderate gas temperatures (near or at room temperature) so-called cold plasmas are well-suitable for the treatment of heat-sensitive materials, such as polymers, without affecting their bulk properties. The present work focuses on the investigation of atmospheric pressure plasma processes for the biological decontamination of polymers. The objective is to help elucidate on the one hand the impact of varied plasma process parameters on the inactivation of micro-organisms and on the other hand the influence of plasma on the surface properties of the substrate. The investigations were performed by means of a high-frequency driven plasma jet (from the product line kINPen) operated with argon and argon-oxygen mixtures. Three main aspects were analyzed: 1. The effect of plasma on the viability of micro-organisms dependent on working gas, treatment time, and the sample distance (distance between the jet nozzle and the substrate). 2. The plasma-based removal of microbial biofilms. 3. The effects of the plasma treatment on the surface properties of selected polymers. Additionally to the capability of the applied plasma jet in killing microbes the efficacy of this plasma jet for the removal of complex biological systems (e.g. biofilms) is shown. To model cell constituents of bacteria different synthetic polymers were chosen to gain insight into the decomposition process responsible for biofilm degradation. By investigating the impact of atmospheric pressure plasma on physico-chemical surface properties of various synthetic aliphatic and aromatic polymers the interaction mechanisms between plasma and plasma-exposed material are discussed. These studies are accompanied by applying different optical plasma diagnostic techniques (optical emission spectroscopy and two-photon absorption laser induced fluorescence spectroscopy) to obtain information on the plasma gas phase which contributes to the elucidation of the reaction mechanisms occurring during plasma exposure. Moreover, it is presented to which extent the plasma treatment influences the surface properties of polymers during the plasma-based bio-decontamination process and further, the benefits of surface-functionalized polymers for biomedical application is discussed.
