Molecular Mechanisms of Bortezomib Action: Novel Evidence for the miRNA−mRNA Interaction Involvement

Bortezomib is an anti-tumor agent, which inhibits 26S proteasome degrading ubiquitinated
proteins. While apoptotic transcription-associated activation in response to bortezomib has been
suggested, mechanisms related to its influence on post-transcriptional gene silencing mediated
regulation by non-coding RNAs remain not fully elucidated. In the present study, we examined
changes in global gene and miRNA expression and analyzed the identified miRNA–mRNA interactions
after bortezomib exposure in human neuroblastoma cells to define pathways affected by this agent in
this type of cells. Cell viability assays were performed to assess cytotoxicity of bortezomib. Global gene
and miRNA expression profiles of neuroblastoma cells after 24-h incubation with bortezomib were
determined using genome-wide RNA and miRNA microarray technology. Obtained results were
then confirmed by qRT-PCR and Western blot. Further bioinformatical analysis was performed
to identify affected biological processes and pathways. In total, 719 genes and 28 miRNAs were
downregulated, and 319 genes and 61 miRNAs were upregulated in neuroblastoma cells treated with
bortezomib. Possible interactions between dysregulated miRNA/mRNA, which could be linked to
bortezomib-induced neurotoxicity, affect neurogenesis, cellular calcium transport, and neuron death.
Bortezomib might exert toxic effects on neuroblastoma cells and regulate miRNA–mRNA interactions
influencing vital cellular functions. Further studies on the role of specific miRNA–mRNA interactions
are needed to elucidate mechanisms of bortezomib action.

Microglia are the resident immune cells of the central nervous system (CNS) and play a major role in the regulation of brain homeostasis. To maintain their cellular protein homeostasis, microglia express standard proteasomes and immunoproteasomes (IP), a proteasome isoform that preserves protein homeostasis also in non-immune cells under challenging conditions. The impact of IP on microglia function in innate immunity of the CNS is however not well described. Here, we establish that IP impairment leads to proteotoxic stress and triggers the unfolded and integrated stress responses in mouse and human microglia models. Using proteomic analysis, we demonstrate that IP deficiency in microglia results in profound alterations of the ubiquitin-modified proteome among which proteins involved in the regulation of stress and immune responses. In line with this, molecular analysis revealed chronic activation of NF-κB signaling in IP-deficient microglia without further stimulus. In addition, we show that IP impairment alters microglial function based on markers for phagocytosis and motility. At the molecular level IP impairment activates interferon signaling promoted by the activation of the cytosolic stress response protein kinase R. The presented data highlight the importance of IP function for the proteostatic potential as well as for precision proteolysis to control stress and immune signaling in microglia function.
Impact of proteostasis and the ubiquitin proteasome system on myeloid cell function in the CNS

Cellular protein homeostasis (proteostasis) maintains a functional proteome and thus proper cell function. Proteostasis is facilitated by the ubiquitin-proteasome system (UPS), an intracellular protein turnover machinery ensuring clearance of damaged, misfolded, old and/or unneeded regulatory proteins. This is particularly important in the central nervous system (CNS), where it is linked to neurodegeneration. Disruptions of the proteostasis systems cause the accumulation of misfolded proteins which are commonly seen in progressive neurodegenerative diseases also linked to neuroinflammation. Proper UPS function can protect cells from the accumulation of defective proteins, neurodegeneration and neuroinflammation. Furthermore, it has been found that loss of function mutations in the genes encoding UPS components are linked to systemic inflammation including neuroinflammation and/or neurodevelopmental disorders. Proteasome defects in patients suffering from these disorders cause decreased proteasome activity, accumulation of proteins, activation of proteotoxic stress responses and systemic inflammation. However, the molecular link between proteotoxic stress and the initiation of inflammatory signalling remained unclear. In Article 2, we summarized the importance of the UPS in immune cell proteostasis and function including activation of innate and adaptive immune responses. Although UPS function is notably important in innate immune signalling, the current understanding of the role of UPS in myeloid cell function in the CNS is limited. We also indicated the involvement of impaired UPS function in sterile systemic inflammation including neuroinflammation as well as tumour diseases and pathogen manipulation of immune cells.
To investigate the molecular link behind proteasome impairment and systemic inflammation in the brain, we focused on microglia cells as the only immune residents of the CNS. In Article 1, we used a pharmacological inhibitor called bortezomib which targets β5 and β5i/LMP7 subunit activities in standard proteasome (SP) and immunoproteasome (IP), respectively. We showed for the first time on the molecular level that inhibition of proteasome activity by bortezomib triggers the accumulation of ubiquitylated proteins, proteotoxic stress responses and innate immune signalling activation depending on the induced proteotoxic stress response called unfolded protein response (UPR) in murine microglia. In particular, activation of the inositol-requiring protein 1α arm of UPR upon bortezomib treatment leads to systemic inflammation as indicated by type I interferon (IFN) response.
IP enhance the proteolytic capacity of UPS by rapid clearance of proteins upon immune signalling activation. Microglia, like other immune cells, exhibit constitutive expression of IP as well as SP to maintain their cellular proteostasis. In Manuscript 3, we studied the particular impact of IP impairment on microglial cellular function. We showed accumulation of ubiquitin-modified proteins and activation of proteotoxic stress responses in IP-impaired mouse and human microglia models. Moreover, we identified possible IP substrates in microglia using β5i/LMP7 knockout mice as an IP deficiency model and, examined how IP deficiency affects microglia function. IP deficient microglia affected the ubiquitylation levels of proteins involved in multiple pathways such as immune responses, energy metabolism, cytoskeleton organisation, cell cycle and ribosome function. Based on the molecular analysis, we confirmed sterile activation of innate immune signalling mechanisms in IP impaired microglia. This is driven by the proteotoxic stress sensor protein kinase R (PKR). In addition, we were able to show that IP impairment altered levels of the microglial activation markers, which are also involved in motility, adhesion and phagocytosis of microglia.
In this thesis, we highlight that UPS function is necessary to maintain microglial proteostasis and, that impairment of proteasome activities triggers sterile inflammation in microglia via activation of proteotoxic stress responses. The described activation of innate immune signalling mechanisms in microglia upon proteasome impairment may be considered as new therapeutic targets for patients suffering from rare protesomapathies or other disorders linked to dysregulated immune signalling.
Der Replikationszyklus der Herpesviren ist sehr komplex und im Detail unzureichend verstanden. Die Funktionen und Eigenschaften einiger viraler Proteine sind bisher kaum charakterisiert. Folglich gibt es wenige Strukturmodelle dieser Proteine, wodurch beispielsweise eine rationale Medikamentenentwicklung kaum möglich war. Die Zielstellung dieser Arbeit war, neun dieser Proteine (pUL4, -7, -11, -16, -21, -26, -26.5, -32 und -33) aus dem pseudorabies virus (PrV) zu charakterisieren und nach Möglichkeit deren Struktur aufzuklären. Hierzu wurden die zur Verfügung gestellten Gensequenzen in geeignete bakterielle Expressionsvektoren umkloniert und in E. coli exprimiert. Lösliche Proteine wurden gereinigt und anschließend Kristallisationsexperimenten unterzogen, während unlösliche Proteine zum Teil auf ihre Renaturierbarkeit getestet wurden. Die Strukturen des kristallisierten N-terminalen Teils von pUL26 (Assemblin) wurden mittels Röntgenkristallographie aufgeklärt. Außerdem wurden alle Proteine in silico auf Signalsequenzen, Phosphorylierungen und Sequenzmuster untersucht. Von der N-terminalen Serinproteasedomäne (Assemblin) von pUL26 wurden drei Strukturen durch Röntgenkristallographie bestimmt: eine native dimere, eine inhibierte dimere, sowie eine native monomere Struktur. Letztere ist das erste bekannte Strukturmodell der monomeren Form eines Assemblins. In Verbindung mit den dimeren Strukturen konnte experimentell bestätigt werden, dass die Aktivierung der Assembline über die Verschiebung eines loop bei der Dimerisierung erfolgt. Die Umlagerung dieses loop basiert darauf, dass sich der in der monomeren Form teilweise flexible Dimerisierungsbereich durch die Dimerisierung etwas verändert und eine weitestgehend starre Konformation einnimmt. Die Helix α8 wird etwas verkürzt und die Helix α7 etwas verlängert und begradigt, wodurch sich der Oxyanionenloch-Loop vom Dimerisierungsbereich entfernt und ein ausgedehntes Wasserstoffbrückenbindungsnetzwerk aufbaut. In dieser Konformation stabilisiert der loop das Oxyanionenloch, wodurch die Protease aktiviert wird. Weiterhin wurde durch small-angle X-ray scattering bestätigt, dass der Dimerisierungsgrad von der Assemblin- und Mg²+;-Ionenkonzentration abhängig ist. Diese Informationen zur Dimerisierung des PrV-Assemblins können dazu beitragen, rationale Medikamentenentwicklung zu betreiben. Daraus resultierende Wirkstoffe können die Dimerisierung und somit die Aktivierung dieses Schlüsselproteins verhindern. Durch die hohe Ähnlichkeit der Assembline in anderen Herpesviren, kann die nun bekannte monomere Struktur des PrV-Assemblins als Modell für die monomere Struktur anderer, zum Teil humanpathogener Herpesviren genutzt werden. Demzufolge könnte dieses Modell auch die Entwicklung von Medikamenten beispielsweise gegen das Epstein-Barr virus oder das herpes simplex virus 1 ermöglichen. Es stellte sich zudem heraus, dass die bisher für das PrV-pUL26 bzw. -pUL26.5 vorhergesagte zweite Assemblinschnittstelle (M-site) vermutlich nicht korrekt ist. Es wurde eine andere M-site vorgeschlagen, welche ebenfalls infrage kommt. Eine Charakterisierung in vitro war bei fünf der neun zu untersuchenden Proteine möglich. Die anderen vier Proteine (pUL7, -16, -21 und -32) konnten aus verschiedenen Gründen nicht erfolgreich exprimiert werden. Die Proteine pUL4, pUL26.5 und pUL33 wurden unlöslich exprimiert, wobei pUL33 renaturiert werden konnte. Der Membrananker pUL11 und die N-terminale Serinproteasedomäne von pUL26 konnten löslich exprimiert werden. Untersuchungen in silico ergaben, dass der Membrananker pUL11 aus dem pseudorabies virus wahrscheinlich ein nukleäres Exportsignal trägt, was bisher nicht bekannt war. Es ist zudem wahrscheinlich, dass pUL11 selbst keine definierte Struktur hat, da es mit 63 Aminosäuren ein sehr kleines Protein ist und über Sequenzmuster mit anderen Proteinen interagiert.
Kunststoffe durchdringen nahezu jeden Bereich unseres alltäglichen Lebens. Zur Herstellung bestimmter Plastikmaterialien (Epoxidharze und Polycarbonate) werden Bisphenole als Grundbaustein benötigt, deren Grundstruktur sich aus zwei Phenolringen zusammensetzt, die über eine substituierte Kohlstoffbrücke miteinander verbunden sind. Die einzelnen Bisphenole unterscheiden sich jeweils durch verschiedene Substituenten an der Kohlenstoffbrücke oder an den aromatischen Ringsystemen. Dabei gehen die großmaßstäbliche Produktion der Bisphenole sowie unser permanenter Kontakt mit bisphenolhaltigen Materialien mit einer verstärkten Nachweisbarkeit dieser Chemikalien in Umweltproben bzw. in menschlichen Geweben und Körperflüssigkeiten einher. Gleichzeitig stehen Bisphenole im Verdacht, aufgrund ihrer Toxizität und hormonähnlichen Wirkung eine Vielzahl von Erkrankungen zu begünstigen. Vor allem die Interaktion mit dem Östrogenrezeptor alpha ist gut verstanden. Daher wurde in dieser Arbeit die bakterielle Transformation von acht verschiedenen Bisphenolen (Bisphenol A, AP, B, C, E, F, PH und Z) durch die Umweltisolate Cupriavidus basilensis SBUG 290 und Bacillus amyloliquefaciens SBUG 1837 untersucht und näher charakterisiert. Beide Bakterienstämme waren in der Lage, alle eingesetzten Bisphenole zu transformieren. Dabei war die Transformation auf die Phenolringe oder daran befindliche Substituenten beschränkt. Ein mikrobieller Angriff an der ringverbindenden Kohlenstoffbrücke wurde nicht nachgewiesen. Während B. amyloliquefaciens die Schadstoffe ungeachtet ihrer Struktur jeweils phosphorylierte, transformierte C. basilensis die Bisphenole in Abhängigkeit von ihrer Struktur zu hydroxylierten Derivaten, Ringspaltungsprodukten, Produkten mit Acetamidstruktur oder zu Dimeren. Neben der Strukturaufklärung der mikrobiell gebildeten Produkte wurden die einzelnen Transformationswege für beide Bakterienstämme näher charakterisiert. Hierfür wurden u.a. die Produkte als Transformationssubstrate eingesetzt, der Einfluss der Kultivierung und Inkubationsmedien auf die Biotransformation untersucht sowie Proteomanalysen durchgeführt. Die Bildung von hydrophileren Transformationsprodukten durch die Bakterienstämme führte zur Detoxifizierung und Reduktion der östrogenen Aktivität der hydrophoben Bisphenole.
Protamine (PRT) is a positively charged protein, which is widely used in medicine as an adjunct to certain preparations of insulin and as a rapidly-acting antidote for heparin, particularly to neutralize the effects of high heparin concentrations needed for anticoagulation during cardiac surgical procedures using cardiopulmonary bypass. It has been demonstrated that PRT and heparin form multimolecular complexes and that these complexes have high immunogenicity in a mouse model. Studies in this thesis provide new insights into the pathophysiology of anti-PRT/heparin antibodies. The results of study I showed that the administration of PRT combined with heparin is responsible for high immunoglobulin G (IgG) immunization after cardiac surgery. A subset of these antibodies was able to induce platelet activation in a way similar to that observed by heparin-induced thrombocytopenia (HIT). Using an animal model, we demonstrated that anti-PRT/heparin antibodies are capable of platelet destruction in the presence of PRT and heparin. Moreover, our data suggests that platelet-activating anti-PRT/heparin antibodies at surgery are potentially associated with postoperative thrombocytopenia and an increased risk for thromboembolic events. In study II, the immune response against PRT/heparin complexes was investigated. This study showed a relatively fast development of IgG with no general preceding IgM formation. In addition, patients undergoing liver transplantation developed anti-PRT/heparin antibodies without previous exposure to PRT. These results suggest that a previous contact with the antigen(s) itself or other antigens with molecular mimicry induced this immune response. In fact, we were able to identify Neutral Protamine Hagedorn (NPH) insulin and core histones (DNA-binding proteins) as potentially antigenic candidates for a previous immunization. Furthermore, the findings of study III demonstrate the ability of anti-PRT/heparin antibodies to activate platelets in the presence of NPH insulin in a heparin-dependent way suggesting that diabetic patients may have an enhanced risk for thromboembolic complications if treated with NPH insulin and possibly while receiving prophylactic heparin. These observations justify further clinical investigations to assess the impact of the interaction between anti-PRT/heparin antibodies and PRT-mimicking antigens, such as NPH insulin or histones.
Chemerin ist ein Adipokin, welches unter anderem vom Fettgewebe exprimiert wird und wichtige Funktionen im Rahmen des Fettstoffwechsels übernimmt. Verschiedene klinische Studien konnten Korrelationen zwischen der Chemerinkonzentration im Blut und diversen metabolischen, renalen sowie kardiovaskulären Phänotypen identifizieren. Aufgrund der Verwendung von sehr unterschiedlichen Patientenpopulationen mit häufig kleinen Kollektiven ist die derzeitige Befundlage insgesamt allerdings noch recht widersprüchlich. Das Ziel der vorliegenden Arbeit war daher, den Zusammenhang zwischen Chemerin und einem breiten Spektrum von inflammatorischen, metabolischen, renalen sowie kardiovaskulären Parametern in einer großen, gut charakterisierten, populationsbasierten Studie zu analysieren und dabei den Einfluss der Fettmasse und anderer Störfaktoren angemessen zu berücksichtigen.
Unter Zuhilfenahme von Methoden der multivariablen linearen und logistischen Regressionsanalyse wurden in dieser Arbeit Daten von 4420 Probanden der SHIP-TREND-Population ausgewertet. Die Analysen zeigten positive Assoziationen zwischen Chemerin und den Inflammationsparametern hsCRP und Fibrinogen. Darüber hinaus konnten positive Zusammenhänge mit dem HbA1c-Wert, dem Nüchterninsulinwert und dem HOMA-IR identifiziert werden. Für den Glukosespiegel ließ sich dagegen kein Zusammenhang mit Chemerin nachweisen. Die Ergebnisse zeigten weiterhin, dass ein hoher Chemerinspiegel mit einem unvorteilhaften Lipid-Profil assoziiert ist. Während eine glockenförmige Assoziation zwischen Chemerin und dem diastolischen Blutdruck vorlag, wurde kein Zusammenhang mit dem systolischen Blutdruck beobachtet. Alle beschriebenen Zusammenhänge waren unabhängig von Unterschieden der Probanden in der individuellen Fettmasse sichtbar. Die Analysen zeigten darüber hinaus, dass ein hoher Chemerinspiegel signifikant mit einer schlechteren Nierenfunktion einhergeht und dass sich dieser Effekt mit zunehmender Fettmasse noch verstärkt. Bezüglich der verschiedenen subklinischen Parameter der Atherosklerose konnte nach vollständiger Modelladjustierung eine inverse Assoziation von Chemerin mit dem ABI sowie eine positive Assoziation mit dem Vorliegen einer Karotisstenose beobachtet werden.
Zusammenfassend zeigen die dargestellten Ergebnisse, dass Chemerin unabhängig von individuellen Unterschieden in der Fettmasse mit einer ganzen Reihe von verschiedenen inflammatorischen, metabolischen, renalen und kardiovaskulären Parametern assoziiert ist. Dementsprechend kann vermutet werden, dass Chemerin auch abseits seiner Funktionen im Fettgewebe wichtige pathophysiologische Prozesse im Körper steuert. Existierende experimentelle Arbeiten liefern erste Hinweise auf die zugrundeliegenden Mechanismen. Das gesamte Funktionsspektrum von Chemerin ist zum jetzigen Zeitpunkt allerdings nicht vollständig geklärt. Die vorliegende Arbeit liefert einen umfassenden Überblick über die epidemiologischen Zusammenhänge der genannten Parameter in einer allgemeinen Bevölkerung und stellt damit eine wichtige Grundlage für weitere Forschungsarbeiten dar. Langfristig ist denkbar, dass Chemerin als diagnostischer Biomarker oder Therapeutikum im klinischen Alltag eingesetzt wird.
Hintergrund International findet die Bedeutung psychischer Störungen seit vielen Jahren zunehmendpolitische und wissenschaftliche Aufmerksamkeit. Depressionen führen nicht nur zu einer Belastung des psychosozialen Befindens, sondern haben auch große Einschränkungen in der Lebensqualität und Arbeitsproduktivität zur Folge [7]. Eine Identifikation von Prädiktoren für die Remission depressiver Symptomatik erscheint aus diesem Grunde besonders wichtig. Material und Methoden Remittierte und Nicht-Remittierte Patienten wurden anhand des in der Literatur verwendeten Cut-off-Wertes 7 in der Hamilton-Depressionsskala in verschiedenen soziodemographischen, klinischen und weiteren Variablen gegenübergestellt. Insgesamt handelt es sich um eine Stichprobe mit 388 Patienten, von denen 205 an der Katamneseuntersuchung teilnahmen. Eingeschlossen wurden Patienten, die die Diagnose einer einzelnen oder rezidivierenden depressiven Episode (F32.x / F33.x / F38.x) oder einer Dysthymie (F34.1) nach ICD-10 erhalten haben. Ausgeschlossen wurden jene, die als komorbide Diagnosen eine Schizophrenie (F2x.x), eine organisch bedingte Störung (F0x.x) oder eine Intelligenzminderung (F7x.x) nach ICD-10 aufwiesen. Es wurden folgende Skalen verwendet: Beck-Depressions-Inventar (BDI), ein Fragebogen zur gesundheitlichen Lebensqualität(WHOQOL-BREF), die Hamilton-Depressionsskala (HAM-D), die globale Erfassung des Funktionsniveaus (GAF) sowie die Clinical Global Scale (CGI). Ein eigens kreierter Fragebogen fand zur Erfassung anamnestischer und weiterer Daten ebenfalls Anwendung. Ergebnisse und Diskussion Insgesamt remittierte die depressive Symptomatik bei 93 (= 45 %) Patienten. Die nicht-remittierte Gruppe war durchschnittlich 49.82 Jahre alt und damit jünger als die remittierten Patienten (55.17 Jahre alt). Fast doppelt so viel remittierte Patienten hatten einen Abschluss am Gymnasium. Dieses spiegelte sich auch in der derzeitigen beruflichen Situation wider: 75.5% der Arbeitslosen oder erwerbs- bzw. berufsunfähigen Patienten waren nicht-remittiert (p= .00, d= 2.578). Es erhielten signifikant (p≤ .0001) häufiger nicht-remittierte Patienten die Diagnose einer rezidivierenden Depression in Verbindung mit einer Komorbidität. Im Vergleich der Medikamentenverordnung wurden signifikant mehr trizyklische Antidepressiva in der nicht-remittierten Gruppe verschrieben. Dies deutet darauf hin, dass diese ältere Generation der Antidepressiva besonders bei einer therapieresistenten Depression als Mittel 2. Wahl zum Einsatz kommt. Hinsichtlich der Psychotherapie kam es zu einem überraschenden Ergebnis, da signifikant (p≤ .0001) häufiger die nicht-remittierten Patienten diese abgeschlossen hatten oder sich noch in einer laufenden Therapie befanden. Einerseits könnte es auf eine selektive Zuweisung in Psychotherapien hindeuten, wenn sich eine auf Medikamente nur unzulänglich ansprechende Symptomatik zeigt. Andererseits könnte man auch von einer Selbstselektion von Patienten ausgehen, welche sich subjektiv besser fühlten und deshalb eine Psychotherapie nicht beanspruchen wollten. Der hohe Stellenwert sozialer Beziehungen wurde anhand der Variablen Freizeitverhalten, sportliche Aktivitäten, Kontakt mit Freunden und Verwandten, Art der Kontakte sowie dem Vorhandensein eines Ansprechpartners bei Problemen deutlich. In der zusammenfassenden binären logistischen Regressionsanalyse wurden die bedeutsamsten Prädiktorvariablen extrahiert. Eine Nicht-Remission demzufolge erhöht die Chance auf die Zuführung in eine Psychotherapie um den Faktor 2.72, was vermuten lässt, dass gerade schwer behandelbare Patienten, wie es auch die Leitlinie [10] empfiehlt, neben der medikamentösen Therapie eine zusätzliche Psychotherapie benötigen. Des Weiteren erhöht eine Nicht-Remission die Chance auf eine höhere Anzahl an stationären Aufenthalten wegen einer Depression um den Faktor 1.34. Es konnten keine Studien gefunden werden, die die Anzahl an stationären Aufenthalten wegen einer Depression als negativen Prädiktor ansah. Es ist jedoch zu vermuten, dass Patienten zwar nicht schwerer erkrankt sind im Sinne der Symptomschwere, aber vielleicht spezifische Bewältigungsmuster zeigen, die immer wieder zu einer Dekompensation und anschließenden Rehospitalisierung führen. Eine stabile berufliche Situation geht mit einer erhöhten Chance auf eine Remission um 4.19 einher und beeinflusst die Remissionswahrscheinlichkeit positiv. Beides erbringt ein gesichertes Einkommen und damit wahrscheinlich auch eine bessere bzw. leichtere soziale Integration. Dass diese Integration eine wichtige Rolle spielt, zeichnete sich auch im Prädiktor „Vorhandensein eines Ansprechpartners bei Problemen“ ab. Die Chance zu remittieren erhöhte sich um 3.58, wenn man bei Problemen einen Ansprechpartner hatte.

Oral mucositis is the most common and severe non-hematological complication associated with cancer radiotherapy, chemotherapy, or their combination. Treatment of oral mucositis focuses on pain management and the use of natural anti-inflammatory, sometimes weakly antiseptic mouth rinses in combination with optimal oral cavity hygiene. To prevent negative effects of rinsing, accurate testing of oral care products is necessary. Due to their ability to mimic realistic in-vivo conditions, 3D models may be an appropriate option in compatibility testing of anti-inflammatory and antiseptically effective mouth rinses. We present a 3D model of oral mucosa based on the cell line TR-146 with a physical barrier, characterized by high transepithelial electrical resistance (TEER) and confirmed cell integrity. Histological characterization of the 3D mucosa model showed a stratified, non-keratinized multilayer of epithelial cells similar to that of human oral mucosa. By means of immuno-staining, tissue-specific expression of cytokeratin 13 and 14 was shown. Incubation of the 3D mucosa model with the rinses had no effects on cell viability, but TEER decreased 24h after incubation in all solutions except ProntOral®. Analogous to skin models, the established 3D model meets the quality control criteria of OECD guidelines and may therefore be suitable for comparing the cytocompatibility of oral rinses.
Aim
Periprosthetic joint infections are a devastating complication after arthroplasty, leading to rejection of the prosthesis. The prevention of septic loosening may be possible by an antimicrobial coating of the implant surface. Poly (hexamethylene) biguanide hydrochloride [PHMB] seems to be a suitable antiseptic agent for this purpose since previous studies revealed a low cytotoxicity and a long-lasting microbicidal effect of Ti6Al4V alloy coated with PHMB. To preclude an excessive activation of the immune system, possible inflammatory effects on macrophages upon contact with PHMB-coated surfaces alone and after killing of S. epidermidis and P. aeruginosa are analyzed.
Methods
THP-1 monocytes were differentiated to M0 macrophages by phorbol 12-myristate 13-acetate and seeded onto Ti6Al4V surfaces coated with various amounts of PHMB. Next to microscopic immunofluorescence analysis of labeled macrophages after adhesion on the coated surface, measurement of intracellular reactive oxygen species and analysis of cytokine secretion at different time points without and with previous bacterial contamination were conducted.
Results
No influence on morphology of macrophages and only slight increases in iROS generation were detected. The cytokine secretion pattern depends on the surface treatment procedure and the amount of adsorbed PHMB. The PHMB coating resulted in a high reduction of viable bacteria, resulting in no significant differences in cytokine secretion as reaction to coated surfaces with and without bacterial burden.
Conclusion
Ti6Al4V specimens after alkaline treatment followed by coating with 5–7 μg PHMB and specimens treated with H2O2 before PHMB-coating (4 μg) had the smallest influence on the macrophage phienotype and thus are considered as the surface with the best cytocompatibility to macrophages tested in the present study.
Unlike the native surface of the implant material (Ti6Al4V), oxidation with H2O2 leads to increased binding of the effective antimicrobial agent poly(hexamethylene) biguanide [PHMB]. However, treating with NaOH instead results in an even higher PHMB mass coverage. After oxidation with H2O2, strong differences in the PHMB adsorption capability between polished and corundum-blasted surfaces appear, indicating a roughness dependence. After NaOH treatment, no such effect was observed. The wetting properties of specimens treated with either H2O2 or NaOH prior to PHMB exposure clearly varied. To unravel the nature of this interaction, widespread in silico and in vitro experiments were performed. Methods: By X-ray photoelectron spectroscopy, scanning electron microscopy, water contact angle measurements and MD simulations, we characterized the interplay between the polycationic antimicrobial agent and the implant surface. A theoretical model for PHMB micelles is tested for its wetting properties and compared to carbon contaminated TiO2. In addition, quantitation of anionic functional group equivalents, the binding properties of PHMB with blocked amino end-group, and the ability to bind chlorhexidine digluconate (CHG) were investigated. Ultimately, the capability of osteoblasts to build calcium apatite, and the activity of alkaline phosphatase on PHMB coated specimens, were determined. Results: Simulated water contact angles on carbon contaminated TiO2 surfaces and PHMB micelle models reveal little influence of PHMB on the wetting properties and point out the major influence of remaining and recovering contamination from ambient air. Testing PHMB adsorption beyond the critical micelle concentration and subsequent staining reveals an island-like pattern with H2O2 as compared to an evenly modified surface with NaOH. Both CHG and PHMB, with blocked amino end groups, were adsorbed on the treated surfaces, thus negating the significant influence of PHMB’s terminal groups. The ability of osteoblasts to produce calcium apatite and alkaline phosphatase is not negatively impaired for PHMB mass coverages up to 8 μg/specimen. Conclusion: Differences in PHMB adsorption are triggered by the number of anionic groups and carbon contaminants, both of which depend on the specimen pre-treatment. With more PHMB covering, the implant surface is protected against the capture of new contamination from the ambient air, thus building a robust antimicrobial and biocompatible surface coating.
Objectives
Oral mucositis caused by intensive cancer chemotherapy or radiotherapy frequently results in pronounced damage of the oral mucosa leading to painful oral hygiene. To support oral care, antimicrobial effective mouth rinses may be used. Thus, the efficacy of a hypochlorite-based mouth rinse (Granudacyn®), assumed to be highly biocompatible because of the compounds being part of the natural pathogen defense, as possible antiseptic agent in case of oral mucositis was compared to that of an octenidine based antiseptic mouth rinse (Octenidol® md).
Materials and methods
The study was conducted as monocentric, controlled, randomized, blind cross over comparative study on 20 volunteers. As a proof of principle, we performed the study on orally healthy subjects and not cancer patients. The efficacy was determined as reduction of colony forming units (cfu) on buccal mucosa as well as in saliva. After mouth rinsing for 30 s, samples were taken after 1 min, 15 min, 30 and 60 min. The lg-reduction was calculated as difference between lg-values of cfu pre- and post-treatment.
Results
Both antiseptic mouth rinses induced a significant reduction of cfu on buccal mucosa and in saliva 1 min after mouth rinsing. The effect persisted up to 60 min. The octenidine based rinse was significantly superior to the hypochlorite-based rinse up to the last sample 60 min after rinsing. However, the known cytotoxicity of octenidine argues against its application.
Conclusion
Within the limits of this study, due to its antiseptic efficacy, the hypochlorite-based rinse Granudacyn® can be regarded appropriate to support the oral hygiene in patients with a sensitive oral mucosa during an aggressive cancer chemotherapy and radiation treatment in case of oral mucositis.
Proteom- und Transkriptom-Analysen zur Bestimmung der Immuntoxizität ausgewählter Naturstoffe

Der Einsatz von Tierversuchen in Forschung und Entwicklung nimmt trotz fortschreitender Optimierung von Testmethoden und –verfahren weiter zu. Zeitgleich werden fortwährend neue Substanzen isoliert oder synthetisiert, deren Wirkungen auf den humanen Organismus und speziell das Immunsystem nicht bekannt sind. In vitro Methoden stellen deshalb sowohl eine günstige und schnelle als auch eine ethisch unbedenkliche Alternative zu Tierversuchen dar. In der vorliegenden Arbeit sollten proteom- und transkriptombasierte Methoden dazu dienen, immuntoxische Eigenschaften von Naturstoffen zu identifizieren und diese Verfahren als Alternative zu Tierversuchen zu etablieren. Dazu wurden zwei humane Immunzelllinien mit Naturstoffen behandelt und das intrazelluläre Proteom sowie das Transkriptom spezifischer Biomarker-Gene analysiert. Zusätzlich dienten weitere Methoden wie Metaboliten-, Zellzyklus- und Apoptoseanalysen sowie die Identifizierung intrazellulärer reaktiver Sauerstoffspezies dazu, Ergebnisse zu verifizieren oder zusätzliche Informationen zu erhalten. Wie zu erwarten war, zeigten die Proteomanalysen, dass sowohl immuntoxische als auch nicht-immuntoxische Substanzen eine breite Wirkung auf das intrazelluläre Proteom haben. Vor allem Proteine, die in den allgemeinen Metabolismus, zelluläre Prozesse und Prozesse der Informationsverarbeitung involviert sind, wurden durch die Behandlung mit den Substanzen in ihrer relativen Menge auf den 2D-Gelen verändert. Allein durch die Zuordnung von Proteinen zu Stoffwechselwegen war eine Abgrenzung immuntoxischer und nicht immuntoxischer Substanzen nicht möglich. Dennoch gibt die Methode einen Einblick in die Wirkungsweise der Substanzen, wodurch Wirkmechanismen entschlüsselt und Reaktionen auf das Immunsystem abgeleitet werden können. Dies wird vor allem nach der Behandlung der Zellen mit Tulipalin A und Helenalin deutlich, da auch allgemeine Stoffwechselwege wie die Purinsynthese und die anaerobe Glykolyse einen Einfluss auf das Immunsystem haben. Zusätzlich zu den allgemeinen Stoffwechselwegen wurden einzelne Proteine in ihrer Abundanz verändert, die in Reaktionen des Immunsystems wie der Zytokinbildung oder der Bildung von MHC-Molekülen involviert sind. Außerdem konnten Biomarker für Immuntoxizität auf Proteomebene entwickelt werden. Mit Hilfe dieser Daten war eine Klassifizierung der Substanzen nach ihrer Immuntoxizität möglich. Anhand dieser Analysen wurden die Testsubstanzen Tulipalin A, Helenalin, Vincristin und Cannabidiol als immuntoxisch klassifiziert. Die Klassifizierung der Substanzen als immuntoxisch aufgrund der Biomarker-Proteine und Stoffwechselwege konnte durch die Anwendung von Transkriptom-Biomarkern bestätigt werden. Neben den über 2D-Gelelektrophorese-basierten Proteomanalysen getesteten Substanzen wurden auch Bisphenol A und Ergosterolperoxid aufgrund der Transkriptombiomarker als immuntoxisch klassifiziert. Agaritin und p-Tolylhydrazin sowie der Bisphenol A bis(2,3-dihydroxypropyl) ether haben keine immuntoxische Wirkung. Neben den Proteom-basierten Methoden dient der entwickelte Entscheidungsbaum basierend auf verschiedenen Methoden als Grundlage für die Immuntoxiztätsklassifizierung. Mit dem erstellten Entscheidungsbaum konnten beispielsweise Cyclosporin A, Helenalin und Tulipalin A durch die Anwendung gezielter Tests als immuntoxisch eingestuft werden, während Mannitol als nicht-immuntoxisch bestätigt wurde. Zusammenfassend war es mittels in vitro Methoden möglich, die Immuntoxizität verschiedener Naturstoffe zu identifizieren. Neben Proteom-basierten Methoden wurden auch Transkriptom- sowie funktionelle und Metabolomanalysen genutzt. Eine Validierung der Ergebnisse mit weiteren bekannten immuntoxischen und nicht-immuntoxischen Substanzen würde eine Anwendung als Alternative zu Tierversuchen für eine erstes Screening Testung neuer Substanzen ermöglichen und so sowohl Zeit und Kosten sparen als auch ethische Bedenken minimieren.
Diese Arbeit beschäftigte sich mit den Arthrosen der drei großen Gelenke, der Schulter-, Hüft- und Kniegelenke bei Akromegalie-Patienten. Außerdem wurde die Knochendichte sowohl an der Hüfte als auch an der Wirbelsäule gemessen. Ziel dieser Studie war es, herauszufinden, ob akromegale Patienten vermehrt unter Arthrosen in diesen drei Gelenken leiden und ob eine Häufung von Osteoporose nachgewiesen werden kann. Im Rahmen der Studie wurden bei insgesamt 32 Patienten, die unter Akromegalie leiden, die Bewegungsausmaße beider Schulter-, Hüft- und Kniegelenke nach der Neutral-Null-Methode gemessen, sowie konservative Röntgenaufnahmen dieser Gelenke angefertigt. Des Weiteren wurden mit Hilfe des Lunar Prodigy Advance von GE Healthcare DXA-osteodensitometrische Messungen an der Wirbelsäule (L1 – L4) und beiden Femuren der akromegalen Patienten durchgeführt. Zusammengefasst konnte bei 34,5% der untersuchten Schultergelenke, 72,4% der untersuchten Hüftgelenke und 22,4% der untersuchten Kniegelenke eine Arthrose sowohl im Rahmen der klinischen Untersuchung als auch auf Grund des Röntgenbefunds diagnostiziert werden. Diese Werte liegen besonders für das Schulter- und das Hüftgelenk deutlich über den Werten für die Normalbevölkerung (3-31,3%; 2,1-50%; 10-48,4%). Die Ergebnisse zeigten außerdem deutlich, dass akromegale Patienten im Vergleich zur Normalbevölkerung zu einem früheren Zeitpunkt vor allem von Arthrosen der Hüft- und Kniegelenke betroffen sind. Die Prävalenzrate der Knochensalzminderungen in der untersuchten Gruppe akromegaler Patienten betrug 40,7%, was ebenfalls über den Werten für die Normalbevölkerung liegt (2,7-40%).
Abstract
Background
Duchenne muscular dystrophy (DMD) is a progressive muscle‐wasting disease caused by mutations in the dystrophin gene, which leads to structural instability of the dystrophin–glycoprotein‐complex with subsequent muscle degeneration. In addition, muscle inflammation has been implicated in disease progression and therapeutically addressed with glucocorticosteroids. These have numerous adverse effects. Treatment with human immunoglobulin G (IgG) improved clinical and para‐clinical parameters in the early disease phase in the well‐established mdx mouse model. The aim of the present study was to confirm the efficacy of IgG in a long‐term pre‐clinical study in mdx mice.
Methods
IgG (2 g/kg body weight) or NaCl solution as control was administered monthly over 18 months by intraperitoneal injection in mdx mice beginning at 3 weeks of age. Several clinical outcome measures including endurance, muscle strength, and echocardiography were assessed. After 18 months, the animals were sacrificed, blood was collected for analysis, and muscle samples were obtained for ex vivo muscle contraction tests, quantitative PCR, and histology.
Results
IgG significantly improved the daily voluntary running performance (1.9 m more total daily running distance, P < 0.0001) and slowed the decrease in grip strength by 0.1 mN, (P = 0.018). IgG reduced fatigability of the diaphragm (improved ratio to maximum force by 0.09 ± 0.04, P = 0.044), but specific tetanic force remained unchanged in the ex vivo muscle contraction test. Cardiac function was significantly better after IgG, especially fractional area shortening (P = 0.012). These results were accompanied by a reduction in cardiac fibrosis and the infiltration of T cells (P = 0.0002) and macrophages (P = 0.0027). In addition, treatment with IgG resulted in a significant reduction of the infiltration of T cells (P ≤ 0.036) in the diaphragm, gastrocnemius, quadriceps, and a similar trend in tibialis anterior and macrophages (P ≤ 0.045) in gastrocnemius, quadriceps, tibialis anterior, and a similar trend in the diaphragm, as well as a decrease in myopathic changes as reflected by a reduced central nuclear index in the diaphragm, tibialis anterior, and quadriceps (P ≤ 0.002 in all).
Conclusions
The present study underscores the importance of an inflammatory contribution to the disease progression of DMD. The data demonstrate the long‐term efficacy of IgG in the mdx mouse. IgG is well tolerated by humans and could preferentially complement gene therapy in DMD. The data call for a clinical trial with IgG in DMD.
Die auf der Kola-Halbinsel gelegene Oblast‘ Murmansk war weltweit die erste Territorialeinheit mit polarem Klima, die ab den 1920-er Jahren im Rahmen der sowjetischen Industrialisierung mit Strukturen zur Rohstoffgewinnung ausgestattet wurde. Aufgrund der hohen Bedeutung der hier gefundenen Elemente (Apatit, Buntmetalle, Eisen, Glimmer, Seltene Erden) wurden zahlreiche Städte und Siedlungen in dieser klimatisch sehr schwierigen Region errichtet und bis zum Ende der Sowjetunion weiter ausgebaut. Die Einwohnerzahl wuchs in dieser Zeit um das 50-fache auf rd. 1,2 Mio. an. Zu den Förderindustrien kommen die Funktionen als Verkehrsknotenpunkt für den russischen Außenhandel und die Versorgung nordsibirischer Rohstoffstandorte sowie als Standort der russischen Nordmeerflotte. Obwohl Wirtschaftsprofil und Bedeutung für das Mutterland nach dem Zusammenbruch der Sowjetunion erhalten werden konnten, bestehen schwerwiegende sozioökonomische Probleme, deren deutlichster Ausdruck ein im russischen Vergleich weit überdurchschnittlicher Einwohnerverlust ist. Mit dem Ziel einer Bewertung von Zukunftschancen und -risiken schildert das vorliegende Buch die gegenwärtigen Strukturmerkmale der Region, ihre sozioökonomische Verflechtung sowie ihre Einordnung in administrative, fiskalische und ökonomische Hierarchien Russlands. Die dargelegten Strukturen, Prozesse und darauf fußende Schlussfolgerungen sind beispielhaft für beinahe alle anderen Regionen des russischen „Hohen Nordens“.
Oils and fats from natural origin are sustainable sources for a broad range of economically relevant products in food, feed, fuel, oleochemical, and cosmetic industries. Thereby, a huge variety of lipids or lipid-derived products exist which distinguish themselves by their unique physical properties making them suitable for their individual applications. To obtain such functional lipids in an environmentally friendly manner, enzymes can be employed. In that context, lipases have been proven to be valuable biocatalysts in lipid modification, which are broadly applied in industry. Even though they have been implemented successfully in the dairy, baking, and detergent industries, there is an increasing demand for the expansion of their utilization. New technologies like protein engineering and the implementation of process development are employed in solving this task. Within the enzymes in lipid modification, lipases are the most applied catalysts and in this thesis their utilization was expanded successfully to the implementation of novel separation processes and the production of improved drug delivery matrices.
In einer offenen, kontrollierten, randomisierten, monozentrischen klinischen Studie an 241 Patienten mit geplanter Katarakt-OP wurde untersucht, ob es beim Einsatz von PVP-I-Lösung in der präoperativen Augenantiseptik zu einer Resorption von Iod kommt. Dazu wurden Konjunktiva und/oder periorbitale Haut mit 10 %igem bzw. 1,25 %igem PVP-Iod oder einem iodfreien Mittel behandelt und die Veränderung der Iodausscheidung bestimmt. Von der aufgetragenen verfügbaren Iodmenge wurden zwischen 0,24 und 1,77 % resorbiert. Es konnte gezeigt werden, dass es sowohl bei konjunktivaler als auch bei periorbitaler Applikation einer 10 %igen PVP-I-Lösung zu einer signifikanten Änderung der Iodausscheidung kommt. Bei Verwendung 1,25 %iger PVP-I-Lösung war keine signifikante Änderung der Iodausscheidung festzustellen, allerdings erhöhte sich die Anzahl der Patienten, deren Iodausscheidung im Bereich einer Iodkontamination (>300 µg/g Kreatinin) lag. Aufgrund dieser Ergebnisse ist der Einsatz einer 10 %igen PVP-I-Lösung auf der Konjunktiva abzulehnen. Bei Verwendung einer 1,25 %igen PVP-I-Lösung sind vor jeder präoperativen Augenantiseptik Schilddrüsenerkrankungen auszuschließen.
Multiproxy investigations of lacustrine sediments from Laguna Azul (52 °S) document multi-millennial Holocene influences of Southern Hemispheric Westerlies (SHW) on the hydroclimatic variability of south-eastern Patagonia. During the last 4000 years, this hydroclimatic variability is overprinted by centennial warm/dry periods. A cool/wet period from 11,600 to 10,100 cal. BP is succeeded by an early Holocene dry period (10,100–8300 cal. BP) with a shallow lake, strong anoxia, methanogenesis and high salinity. Between 8300 and 4000 cal. BP the influence of SHW weakened, resulting in a freshwater lake considered to be related to less arid conditions. Since 4000 cal. BP, regional temperature decreased accompanied by re-intensification of SHW reaching full strength since 3000 cal. BP. Centred around 2200, 1000 cal. BP and in the 20th century, Laguna Azul experienced century-long warm/dry spells. Between these dry periods, two pronounced moist periods are suggested to be contemporaneous to the ‘Dark Age Cold Period’ and the ‘Little Ice Age’. Different from millennial SHW variations, centennial fluctuations appear to be synchronous for South America and the Northern Hemisphere. Changes in solar activity, large volcanic eruptions and/or modulations of ocean circulation are potential triggers for this synchronicity.
Microalgae are aquatic, unicellular, eukaryotic organisms, which perform photosynthesis. They have gained interest within the last decades not only for biofuel production due to their high amount of lipids, but also for pharmaceutical and for nutraceutical purposes. Interesting compounds are proteins, carbohydrates, or pigments, such as carotenoids. However, microalgae possess strong and rigid cell walls, which hinder a sufficient and yet, gentle extraction of those valuable compounds. Although standard extraction techniques are available, several shortcomings occur, e.g. high energy demand, use of environmentally harmful solvents or alteration of compounds due to heat or chemicals. Therefore, an alternative method is needed, which is able to address these disadvantages. Physical plasmas were thus studied to answer the question whether they are able to disintegrate the cell walls of microalgae effectively and yet, without degradation of the extractives.
First step of the thesis was to find a suitable plasma source that has an effect on the cell walls because plasma effects, such as electric fields, shockwaves, UV light emission, and the generation of reactive species can be tailored with the respective setup. It was found that spark discharges are most effective for the extraction of Chlorella vulgaris, which was chosen as model organism. All extraction yields were compared to reference methods, whereat microwave radiation was found to be the most effective reference method and were hence, applied for comparative studies.
For the next step, proteins were selected as targets to answer the question, which differences can be determined between plasms-treated and microwave-radiated proteins are observable although the extraction yields were equal. Furthermore, plasma effects, especially the effects of reactive species on the extracted proteins had to be studied. Findings indicate that heat sensitive proteins, such as photosystem-related proteins, or histones are better extractable with spark discharges than with microwave exposure and the effect of reactive species is only minor.
The last step was to determine, which plasma effect is responsible for the observed cell wall disintegration. Therefore, the tensile strength of Chlorella vulgaris was determined and compared to the shockwave pressure, which is generated from the spark channel. It was proven that the shockwave pressure exceeds by far the tensile strength of the microalgae an can be thus held responsible for mechanism for cell wall rupture.
In this thesis, it was found that spark discharges are a promising alternative for the extraction of valuable compounds from microalgae. The discharges are not only effective, but also gentle enough for sensitive compounds, such as proteins or pigments.
Dengue virus (DV) is a positive-strand RNA virus of the Flavivirus genus. It is one of the most prevalent mosquito-borne viruses, infecting globally 390 million individuals per year. The clinical spectrum of DV infection ranges from an asymptomatic course to severe complications such as dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS), the latter because of severe plasma leakage. Given that the outcome of infection is likely determined by the kinetics of viral replication and the antiviral host cell immune response (HIR) it is of importance to understand the interaction between these two parameters. In this study, we use mathematical modeling to characterize and understand the complex interplay between intracellular DV replication and the host cells' defense mechanisms. We first measured viral RNA, viral protein, and virus particle production in Huh7 cells, which exhibit a notoriously weak intrinsic antiviral response. Based on these measurements, we developed a detailed intracellular DV replication model. We then measured replication in IFN competent A549 cells and used this data to couple the replication model with a model describing IFN activation and production of IFN stimulated genes (ISGs), as well as their interplay with DV replication. By comparing the cell line specific DV replication, we found that host factors involved in replication complex formation and virus particle production are crucial for replication efficiency. Regarding possible modes of action of the HIR, our model fits suggest that the HIR mainly affects DV RNA translation initiation, cytosolic DV RNA degradation, and naïve cell infection. We further analyzed the potential of direct acting antiviral drugs targeting different processes of the DV lifecycle in silico and found that targeting RNA synthesis and virus assembly and release are the most promising anti-DV drug targets.
Plus‐strand RNA [(+)RNA] viruses are the largest group of viruses, medically highly relevant human pathogens, and are a socio‐economic burden. The current global pandemic of the severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) shows how a virus has been rapidly spreading around the globe and that– without an antiviral treatment– virus trans mission is solely dependent on human behavior. However, other (+)RNA viruses such as rhino‐, noro‐, dengue‐ (DENV), Zika, and hepatitis C virus (HCV) are constantly spreading and expanding geographically. As in the case of hepatitis C, since its first identification in the 1970s, it took more than 30 years to understand the HCV structure, genome organiza t ion, life cycle, and virus‐host interplay leading to the cure of a chronic and life‐threatening disease. However, no vaccination or antiviral treatment exists for most (+)RNA viruses. Con sequently, a precise and comprehensive analysis of the viruses, their life cycles, and parasitic interactions with their hosts remains an important field of research. In the presented thesis, we use mathematical modeling to study the life cycles of (+)RNA viruses. We analyze replication strategies of closely related (+)RNA viruses, namely HCV, DENV, and coxsackievirus B3 (CVB3), to compare their life cycles in the presence and ab sence of the host’s immune response and antiviral drug treatment and consider different viral spreading mechanisms. Host dependency factors shape the viral life cycle, contribut ing to permissiveness and replication efficiency. Our mathematical models predicted that host dependency factors, such as ribosomes, and thus the virus’ ability to hijack the host cell’s translation machinery play an essential role in the viral genome replication efficiency. Furthermore, our mathematical model suggested that the availability of ribosomes in the vi ral life cycle is a crucial factor in disease outcome: the development of an acute or chronic disease. Even though the host developed strategies to attack the virus, e.g., by degrading the viral genome, blocking the viral protein production, and preventing viral spread, viruses found strategies to countermeasure those so‐called host restriction factors derived from the immune system. Our mathematical models predicted that DENV might be highly effective in blocking the cell’s attempts to recognize the invader. Moreover, we found ongoing HCV RNAreplication even with highly effective antiviral drugs that block processes in the viral life cycle. Furthermore, we found alternative pathways of infection spread, e.g., by HCV RNA carrying exosomes, which may be a possible explanation for reported plasma HCV RNA at the end of treatment, found in a subset of patients. Hence, the mathematical models presented in this thesis provide valuable tools to study the viral replication mechanism in detail. Even though being a simplification of reality, our model predictions confirm and explain known and suggest novel biological mechanisms. In the pre sented thesis, I will summarize and discuss key findings and contextualize model predictions in the broader scientific literature to improve our understanding of the viral dynamics and the virus‐host interplay.
Die Arbeit setzt sich mit dem Begriff des raumbedeutsamen Vorhabens im Sinne des § 3 Abs. 1 Nr. 6 Raumordnungsgesetz (ROG) auseinander und versucht, diesen Begriff zu konkretisieren. Der Autor kommt zu dem Ergebnis, dass die Legaldefinition in § 3 Abs. 1 Nr. 6 ROG kaum zur Konkretisierung beiträgt und der Subsumtion nicht unmittelbar zugänglich ist. Er spricht sich aus diesem Grund dafür aus, dass eine Konkretisierung dieses Begriffs durch die Bildung von vorhabenbezogenen Fallgruppen zu erfolgen hat.
Sodann werden auf Basis der Methode des typisierenden Fallvergleichs Fallgruppen für die Vorhabentypen „Windenergieanlagen“, „Einzelhandelsbetriebe“ und „Abgrabungen“ erarbeitet. Hierzu wurde die einschlägige Rechtsprechung zu dieser Thematik ausgewertet. Die Fallgruppenbildung trägt dazu bei, den Begriff der Raumbedeutsamkeit zu konkretisieren und die Handhabung dieses wertungsoffenen Begriffs in der Praxis zu erleichtern.
Darüber hinaus diskutiert der Autor unter verschiedenen Gesichtspunkten den in Teilen der Literatur vertretenen Ansatz, unter bestimmten Bedingungen den Trägern der Raumplanung zuzugestehen, die Raumbedeutsamkeit eines Vorhabentypus (z.B. Windenergieanlagen) abstrakt-tatbestandlich in Form eines Ziels der Raumordnung festzusetzen. Diesem Ansatz schließt er sich im Ergebnis an.
Background
We investigated the association between low cardiorespiratory fitness and liver fat content (LFC) in the general population.
Materials and Methods
We evaluated data from 2151 adults (51.1% women) from two population-based cohorts of the Study of Health in Pomerania (SHIP-2 and SHIP-TREND-0). We analysed the cross-sectional associations of peak oxygen uptake (VO2peak) with LFC, assessed by magnetic resonance imaging proton density fat fraction, as well as serum gamma-glutamyltransferase (GGT) and aminotransferase concentrations by multivariable regression models.
Results
We observed significant inverse associations of VO2peak with LFC and serum GGT, but not with serum aminotransferase levels. Specifically, a 1 L/min lower VO2peak was associated with a 1.09% (95% confidence interval [CI]: 0.45-1.73; P = .002) higher LFC and a 0.18 μkatal/L (95% CI: 0.09-0.26; P < .001) higher GGT levels. The adjusted odds ratio (OR) for the risk of prevalent hepatic steatosis (HS) by a 1 L/min decrease in VO2peak was 1.61 (95% CI: 1.22-2.13; P = .001). Compared to subjects with high VO2peak, obese and overweight individuals with low VO2peak had 1.78% (95% CI: 0.32-3.25; P = .017) and 0.94% (95% CI: 0.15-1.74; P = .021) higher mean LFC, respectively. Compared to those with high VO2peak, low VO2peak was independently associated with a higher risk of prevalent HS in the obese (adjusted-OR 2.29, 95% CI=1.48-3.56; P < .001) and overweight (adjusted OR 1.57, 95% CI=1.16-2.14; P = .04) groups.
Conclusions
Lower VO2peak was significantly associated with greater LFC and higher serum GGT levels in a population-based cohort of adult individuals. Our results suggest that low VO2peak might be a risk factor for HS.
In dieser retrospektiven Studie wurden die Parameter der Radiojodkinetik: Jod-Uptake (JU), effektive Halbwertzeit (HWZ) und Zeitpunkt der maximalen Jod-Aufnahme (tmax) sowie deren mögliche Beeinflussung durch Erkrankungsart, Schilddrüsenvolumen, Funktionslage, Vortherapien, Patientenalter und Zeitverlauf untersucht. Methodik: Die Studie umfasst Daten von 6156 Radiojodtherapien (RJTH) von Morbus Basedow (MB), unifokalen Autonomien (UFA), multifokalen Autonomien (MFA), disseminierten Autonomien (DISA) und Strumen (STRU), die im Zeitraum von 1996 bis 2008 an der Universitätsklinik für Nuklearmedizin in Greifswald durchgeführt wurden. Die ausgewerteten intratherapeutischen JU-Kurven beruhen auf frequenten Aktivitätsmessungen (automatisierte Patienten-Selbstmessungen, ca. 13/d). Ergebnisse: Für den JU relevant waren Erkrankungsentität und Zielvolumen. In Bereichen äquivalenter Volumen war der JU bei MB etwa 3-mal höher als bei den Strumen. Bei fokalen Autonomien stieg der JU nichtlinear mit dem Zielvolumen; eine Verdoppelung des Zielvolumens im therapierelevanten Bereich (von 10 auf 20 ml) resultierte in einer Zunahme des JU um 30%. Die HWZ war bei Strumen und DISA etwa 20% länger als bei MB und UFA und variierte in den Gruppen um ca. 25%. Mit zunehmender Funktion verkürzte sich die HWZ. Eine Verdopplung des T3-Wertes ergab eine um etwa 20% reduzierte HWZ. Tmax lag im Gesamtkollektiv bei 18h; im Vergleich dazu war tmax bei MB um 20% kürzer und bei STRU um 20% länger. Es bestand eine schwache bis mäßige Korrelation von tmax und Patientenalter sowie eine inverse Korrelation von tmax mit der Funktionslage. Im Zeitverlauf fiel der JU bei abnehmenden Zielvolumina leicht ab (in 12 Jahren ca.1-2%/Jahr). Ein höherer JU bei MB ab 2003 ist annehmbar Folge des veränderten Therapieschemas (Thyreostasepause vor RJTH). Eine frühzeitig bei drohender Unterdosierung durchgeführte Nachapplikation von Radiojod (nötig bei 6% des Kollektivs) führte zu ähnlichen Erfolgsraten (92%) wie in der Normgruppe (91%). Schlussfolgerungen: Die Radiojodkinetik weist relevante Unterschiede zwischen den Erkrankungsgruppen auf. Haupteinflussfaktor auf den JU sind Erkrankungsentität und Zielvolumen. In der Praxis sind für die HWZ die Erkrankungsentität und etwas geringer die Funktionslage von Bedeutung. Bei der Therapieplanung (Radiojodtest) erscheint angesichts der vergleichsweise geringen Streuung der HWZ in den Gruppen die Nutzung von krankheitsspezifischen HWZ (MB, UFA: 5d; MFA: 5,5d, Andere: 6d) vertretbar. Variationen von tmax werden bei großen Messintervallen nicht erfasst, können aber zu fehlerhaften JU- und HWZ-Werten und damit zu falschen Dosisschätzungen führen. Bei Patienten mit drohender Unterdosierung kann durch eine frühzeitige Dosiskorrektur eine Zweittherapie vermieden werden. Dafür ist jedoch eine frequente JU-Messung erforderlich. Ein Teil der divergierenden Literaturangaben zur Radiojodkinetik ist durch Bias (insbesondere Zielvolumenvariation) erklärbar. Orientierende Aktivitätsmessungen per Smartphone sind derzeit nicht geeignet zur Schätzung von Kinetikparametern.
Diese retrospektive Studie untersucht klinische und funktionelle Ergebnisse der Radiojodtherapie (RJTH; 1991-2006) großer, inoperabler Strumen ≥ 100ml. Methode: Die Studie umfasste 456 Patienten (7% blande Strumen, 85% Autonomien, 7% Autoimmunhyperthyreosen) mit einem medianen Schilddrüsenvolumen von 128 ml (100;455) und einer Nachbeobachtungszeit von 7,4 Monaten (1,2;161,9). Die verabreichte Aktivität betrug 1373 MBq (422;3150) bzw. 10,63 MBq/g, die Dosis lag bei 116 Gy (30;217). Die Schilddrüsenvolumetrie erfolgte auf Grundlage von Sonografie und Szintigrafie, sowie teilweise mit CT oder MRT. Einflussfaktoren wurden mittels Subgruppenvergleichen, Kaplan-Meier-Analysen und Regressionsanalysen untersucht. Ergebnisse: Die Schilddrüsenvolumenreduktion betrug 1, 2 und 3 Jahre nach RJTH 47% und jeweils 56%. Eine signifikante Zunahme der Volumenreduktion war bis 2 Jahre nach RJTH nachweisbar. In Subgruppenvergleichen zeigten euthyreote Strumen eine geringere Volumenreduktion als hyperthyreote Strumen. Blande Strumen reduzierten weniger Volumen als Autonomien, die wiederum weniger Reduktion erreichten als Autoimmunhyperthyreosen. Retrosternale Strumen und voroperierte Patienten erreichten vergleichsweise geringere Volumenreduktionen. Die Kaplan-Meier-Analyse bestätigte diese Beobachtungen. Zudem zeigte sich die Tendenz zu einer Dosis -Wirkungsbeziehung. Die Hypothyreoserate betrug nach 7 Jahren 11%, Autoimmunhyperthyreosen wiesen mit 25% eine deutlich höhere Rate auf. Die Beseitigung der Überfunktion gelang mit einmaliger RJTH bei Autonomien vollständig, bei Autoimmunhyperthyreosen nur zu 40%. Im Verlauf zeigten 90% der Patienten eine Beschwerdeabnahme, 3 Jahre nach RJTH waren 65% beschwerdefrei. Insbesondere Patienten mit Lokalsymptomen profitierten. Eine signifikante Beschwerdeabnahme zeigte sich bis 1 Jahr nach RJTH. Therapieversager wurden in 3% der Patienten beobachtet, therapierelevante Nebenwirkungen bei ca. 1%. Schlussfolgerung: Die Schilddrüsenoperation ist für große Strumen die Therapie der Wahl, bei Inoperabilität oder Ablehnung einer Operation ist die RJTH eine in vielen Fällen geeignete, komplikationsarme Alternative. Nach einem Jahr wird hier ca. eine Halbierung des Strumavolumens und im Verlauf eine Beschwerdeabnahme bei 90% der Patienten erreicht. Die Rate der Therapieversager beträgt dabei 3%.
Staphylococcus aureus has acquired resistance to antibiotics since their first use. The S. aureus protein NorA, an efflux pump belonging to the major facilitator superfamily (MFS), contributes to resistance to fluoroquinolones (e.g., ciprofloxacin), biocides, dyes, quaternary ammonium compounds, and antiseptics. Different compounds have been identified as potential efflux pump inhibitors (EPIs) of NorA that result in increased intracellular concentration of antibiotics, restoring their antibacterial activity and cell susceptibility. However, none of the currently known EPIs have been approved for clinical use, probably due to their toxicity profiles. In the present study, we screened approved drugs for possible efflux pump inhibition. By screening a compound library of approximately 1200 different drugs, we identified nilotinib, a tyrosine kinase inhibitor, as showing the best efflux pump inhibitory activity, with a fractional inhibitory concentration index of 0.1875, indicating synergism with ciprofloxacin, and a minimum effective concentration as low as 0.195 μM. Moreover, at 0.39 μM, nilotinib, in combination with 8 μg/mL of ciprofloxacin, led to a significant reduction in biofilm formation and preformed mature biofilms. This is the first description of an approved drug that can be used as an efflux pump inhibitor and to reduce biofilms formation at clinically achievable concentrations.
Ziel: In der retrospektiven Studie soll anhand der BI-RADS-Klassifikation die diagnostische Wertigkeit von Mammographie, Mammasonographie und Palpation bei Fibroadenomen unter Berücksichtigung der Variablen Dichte der Brust, Alter der Patientin, sonographischer Größe und Palpierbarkeit des Befundes dargestellt werden. Material und Methoden: Es wurden im interdisziplinären Brustzentrum der UFK Greifswald 339 für diese Studie verwendbare Fibroadenome diagnostiziert und histologisch gesichert. Die Dignitätsbeurteilung nach BI-RADS wurde in benigne vs. maligne vereinfacht und mit der Histologie verglichen. Die Spezifitäten wurden in Abhängigkeit von den o.g. Variablen getrennt beurteilt. Für den Chi2-Test wurde ein p-Wert < 0,05 als signifikant angenommen. Die Übereinstimmung von Mammographie und Sonographie wurde mit der Kappa-Statistik nach Cohen berechnet. Ergebnisse: Bei den Gesamtspezifitäten von 100% für Palpation, 83,9% für Mammographie und 88,2% für Sonographie zeigten sich auch unter Berücksichtigung der Variablen deutliche Vorteile der Sonographie gegenüber der Mammographie. Es konnte für die Sonographie eine Abhängigkeit zwischen Dignitätseinstufung und Alter sowie Palpierbarkeit dargestellt werden (p = 0,001 und p = 0,025). Größte Übereinstimmungen in der Dignitätseinstufung zeigten Mammographie und Sonographie bei ACR 1 - 2 sowie Befunden > 1,9 cm. Schlussfolgerung: Für die Diagnostik von Fibroadenomen zeigt die Sonographie auch unter Berücksichtigung von den Variablen eine höhere Spezifität als die Mammographie und sollte daher vor allem bei jüngeren Patientinnen der Vorrang gegeben werden. Bei der Sonographie ist die Abhängigkeit bei der Dignitätseinstufung von Alter und Palpierbarkeit des Befundes zu beachten; in angezeigten Fällen sollte somit trotzdem die Mammographie als ergänzende Methode in Erwägung gezogen werden.
Die unterschiedlichen Ergebnisse einer Vielzahl von Studien über eventuelle Alters- und Geschlechtsabhängigkeiten des sekretorischen Immunglobulin A (slgA) lassen aufgrund unterschiedlich angewandter Methodik keine einheitlichen Schlussfolgerungen zu. Die hier vorgelegte Studie untersucht Unterschiede in den slgA-Konzentrationen in den drei Speichelarten Parotis-, Submandibularis- u. Mischspeichel, in Abhängigkeit von Alter und Geschlecht. In Übereinstimmung mit früheren Studien zeigen unsere Resultate bezüglich des Parotis- u. Submandibularisspeichels keine altersabhängigen Unterschiede, weder für Männer noch fur Frauen. Bezüglich des Mischspeichel zeigt sich jedoch ein Anstieg der slgA-Konzentration mit fortschreitendem Alter. Es ergeben sich signifikant höhere Werte in der Gruppe der 26-50jährigen Männer im Vergleich zur Gruppe der 18-25jährigen. Ebenfalls signifikant erhöht sind die Werte der Männer über 50 Jahre verglichen mit der Gruppe der 26-50jährigen. Die über 50jährigen Männer zeigen höchst signifikant ansteigende slgA-Spiegel im Vergleich zur Gruppe der 18-25jährigen. Für die Gruppe der über 50jahrigen Frauen ergibt sich im Mischspeichel eine höchst signifikante Zunahme der slgA-Konzentrationen sowohl im Vergleich zur Gruppe der 18- 25jährigen als auch im Vergleich zu den 26-50jährigen. Bezüglich der Geschlechtsabhängigkeit haben die Vergleiche der slgA-Konzentrationen im Parotisspeichel in keiner Altersstufe signifikante Unterschiede aufdecken können. Für Submandibularisspeichel sind bei Männern in allen Altersstufen signifikant höhere slgA-Werte ermittelt worden verglichen mit den jeweils gleichaltrigen Frauen. Andererseits liegen die slgA-Spiegel im Mischspeichel bei den 26-50jährigen Frauen sowie bei den über 50jährigen signifikant höher im Vergleich zu den Werten der entsprechenden männlichen Altersklassen. Fur die Gruppe der 18-25jährigen Frauen zeigt sich eine hoch signifikante Zunahme der slgA-Konzentrationen gegenüber den Werten der gleichaltrigen Männer.
Der Herzinfarkt stellt weiterhin in den Industrienationen eine der häufigsten Todesursachen dar. In der Akutsituation wird schnellst möglich versucht entweder mechanisch oder medikamentös eine Wiederherstellung des Koronarflusses im betroffenen Ischämiegebiet zu erreichen. Dennoch gibt es verschiedene Ansatzpunkte und Mechanismen, die zum Myokardschutz nach einer Ischämie/Reperfusion führen können. In der hier vorliegenden Arbeit wurden zwei unterschiedliche Wege des Myokardschutzes am ex vivo perfundierten Rattenherz im Ischämie/Reperfusionsmodell untersucht. Die notwendige Reperfusion nach einer Ischämie führt meist zu einer zusätzlichen Schädigung des Myokardgewebes. Es konnte jedoch gezeigt werden, dass kurze Intervalle von Ischämie und Reperfusion nach Wiedereröffnung eines Koronargefäßes die Infarktgröße drastisch senken. Dieses als IPost bezeichnete Verfahren zeigt ein hohes Potenzial zur Reduktion der Myokardschädigung nach einer Ischämie und es konnte demonstriert werden, dass es sich sowohl mechanisch als auch durch exogen zugeführte pharmakologische Substanzen auslösen lässt. Daher wurde im ersten Teil dieser Arbeit im Rahmen der IPost das kardioprotektive Potenzial des A2bAR Agonisten Bay 60-6583 und deren Signalwege untersucht. Im Ischämie/Reperfusionsmodell des ex vivo perfundierten Rattenherzens konnte eine deutliche Reduktion der Infarktgröße in der Bay 60-6583-Gruppe im Vergleich zur Kontrollgruppe dargestellt werden. Des Weiteren konnte eine Beteiligung der PKG und der eNOS am schützenden Signalweg demonstriert werden, denn sowohl bei Inhibition von PKG als auch von eNOS, wurde der Bay 60-6583 vermittelte Myokardschutz aufgehoben. Bei Inhibition der PKC konnte gezeigt werden, dass der Bay 60-6583 vermittelte Myokardschutz weiterhin existiert. Der zweite Teil der Arbeit beschäftigt sich mit transgenen Ratten, die Exon(2-9)Renin überexprimieren. Eine Ischämie am Herzen führt zum Zelluntergang der als Nekrose bezeichnet wird. Mögliche Folgen sind Fibrose, Remodelling und Herzinsuffizienz. Das RAS induziert sowohl Hypertrophie als auch Fibrose am Herzen. Renin ist das Schlüsselenzym des RAS, welches ein sekretorisches Glykoprotein darstellt und von der Niere hergestellt, gespeichert und sezerniert wird. Es konnte vor kurzem ein alternatives Renintranskript vom selben Reningen identifiziert werden, das Exon(2-9)Renin. Dieses Transkript kodiert für ein intrazelluläres, zytosolisches Renin, welches ausschließlich im Rattenherz exprimiert wird und zur Steigerung der Expression nach einen Myokardinfarkt führt. Des Weiteren konnte gezeigt werden, dass es in H9c2 Kardiomyozyten vor Nekrose schützt und Apoptose fördert. Daher wurde im zweiten Teil dieser Arbeit das kardioprotektive Potenzial von transgenen Ratten die Exon(2-9)Renin überexprimieren und mögliche Signalwege der zellschützenden Kinasen, die der IPost identisch sind, geprüft. Im Ischämie/Reperfusionsmodell des ex vivo perfundierten Rattenherzens konnte gezeigt werden, dass kürzlich neu entdecktes zytosolisches Renin in den beiden verwendeten Exon(2-9)Renin überexprimierten Linien zu einer deutlichen Reduktion in der Infarktgröße im Vergleich zu beiden Kontrollgruppen geführt hat. Dies zeigt einen deutlichen Unterschied zum sekretorischen Renin, welches über eine Angiotensin II Synthese zu Entzündungen und Fibrose am Herzen führt [108, 109]. Des Weiteren konnte über Western Blot Analysen eine Beteiligung, der bei der IPost mitwirkenden zellschützenden Kinasen Akt und Erk 1/2, ausgeschlossen werden. Es muss daher eine andere schützende Signalkaskade aktiviert werden. In dieser Arbeit konnte im Ischämie/Reperfusionsmodell des ex vivo perfundierten Rattenherzens der Myokardschutz sowohl medikamentös unter Verwendung des A2bAR Agonisten Bay 60-6583 als auch in transgenen Ratten, die Exon(2-9)Renin überexprimieren gezeigt werden.
Infektionen des Zentralnervensystems (ZNS) können durch unterschiedliche Erreger verursacht werden, wobei Viren das Hauptpotential bilden. Bei der Abklärung der Ätiologie von Infektionen des ZNS nimmt die Labordiagnostik eine zentrale Rolle ein. Die Kenntnis des ätiologischen Agents ist von hoher prognostischer und therapeutischer Relevanz und für die Optimierung des Patientenmanagements bedeutend. Es wurden molekularbiologische Methoden zur Identifizierung und Charakterisierung ZNS-assoziierter Viren etabliert und zur Gewinnung aktueller Prävalenzdaten eingesetzt. Enteroviren (EV) waren mit 21,8% das häufigste Pathogen, gefolgt von Adenoviren. HSV und VZV spielten nur eine untergeordnete Rolle. Eine Bedeutung von West Nil-Virus bei ZNS-Infektionen in der Region Vorpommern konnte ausgeschlossen werden. Die genotypische Charakterisierung zirkulierender Stämme zeigte für EV Cluster mit hoher Homologie zur Gruppe der Coxsackie B-Viren. Weiterhin wurden Vertreter von Coxsackievirus A und von Echovirus identifiziert. Isolierte EV-Stämme wiesen gegenüber Pleconaril eine hohe Empfindlichkeit auf. Ein unerwartet hoher Anteil wurde für Adenoviren gefunden. Die identifizierten Serotypen waren ADV-2, ADV-5 und ADV-41. Untersuchungen zum Proteinprofil EV-infizierter Zellen zeigten signifikante Veränderungen in der Expression für Proteine des Zytoskeletts, für Bestandteile von metabolischen Prozessen und für Proteine, die in Signal- und Transportprozesse sowie die Stress-Abwehr involviert sind und bieten Ansätze für die Entwicklung neuer therapeutischer Strategien.
Im Fokus der dieser Arbeit zugrundeliegenden Untersuchung steht die Entwicklung von Schülern, deren Erleben von bzw. Aufwachsen in der Ganztagsschule2 über acht Jahre hinweg (Klasse 5 bis nach dem Abitur) im Zwei-Jahres-Rhythmus erhoben und ausgewertet wurde. Der Datensatz basiert auf vier Fragebogenerhebungen während und einer abschließenden Interviewbefragung nach Beendigung der Schulzeit. Als forschungsmethodischer Anknüpfungspunkt und Datengrundlage dient die Studie der Forschungsgruppe „Schulentwicklung in Mecklenburg-Vorpommern“ (FoSE). Eine wichtige Vergleichsbasis bieten zudem die Ergebnisse der Studie zur Entwicklung der Ganztagsschule (StEG). Unter Heranziehung und Modifizierung des Pekrunschen Rahmenmodells zur Persönlichkeitsentwicklung von Schülern (1991) und dem Dimensionenmodell der Ganztagsschule von Prüß (2009) werden die Erkenntnisse aus den wissenschaftlichen Diskursen zur Schülerbiographie und zur Ganztagsschule verortet und mit den eigenen Untersuchungsergebnissen in Beziehung gesetzt. Es wird untersucht, inwieweit sich der Ganztagsschulbesuch auf die Entwicklung von jungen Menschen auswirkt und wie diese den Besuch sowie seine Wirkung zu verschiedenen Zeitpunkten ihrer Schullaufbahn bewerten. Ebenso war von Interesse, inwiefern sich durch die aktive Mitgestaltung und Partizipation an der schulischen Umwelt sowie dem Nutzungsverhalten bezüglich der Ganztagsangebote, Chancen und Impulse für den jeweiligen Bildungsweg der (ehemaligen) Schüler ergeben haben und welche Rolle die Familie in diesem Entwicklungsabschnitt einnimmt. Einen besonderen Schwerpunkt stellt dabei die retrospektive Betrachtung der Schulzeit nach dem Abitur, insbesondere hinsichtlich ihrer Bedeutung für die eigene Persönlichkeitsentwicklung, dar.
Purpose: To (1) describe the prevalence of abnormal sleep quality in patients with hip abductor tears (HAT), to (2) determine whether sleep quality improves after open HAT repair, and to (3) to report clinical short-term outcomes in patients undergoing open HAT repair. Methods: The data of 28 patients (29 hips) who underwant open HAT repair were prospectively analyzed at midterm follow-up. The Pittsburgh Sleep Quality Index (PSQI), modified Harris Hip Score (mHHS), the University of California, Los Angeles activity scale (UCLA), and Visual Analog Scale (VAS) for pain were determined via questionnaire. Paired t-tests were applied to compare preoperative and post-operative Patient-reported Outcome Measures (PROMs). Logistic regression was performed to determine the association between PSQI improvement achievement and demographic variables (laterality, sex, age, body-mass-index (BMI), and preoperative mHHS). The minimal clinically important difference (MCID) was calculated for the mHHS. Results: A total of 28 patients were included. Four patients (14.3%) suffered post-operative complications after open HAT repair. The predominance of patients was female (77.4%), with a mean age of 60 ± 13 years. The average follow-up was 30.35 ± 16.62 months. Preoperatively, 27 (96.4%) patients experienced poor sleep quality (PSQI > 5); at follow-up, 7 (25%) patients experienced poor sleep quality. Univariate logistical regression analysis demonstrated no significant association between preoperative demographic data and achieving postoperative PSQI < 5. The MCID of mHHS was calculated to be 12.5. Overall, 90% of patients achieved MCID for mHHS. Conclusion: Preoperative sleep quality was impaired in 96.4% of HAT patients (PSQI > 5). However, these patients showed an improvement in sleep disturbances after open HAT repair in the early postoperative period. Ninety percent of patients showed significant improvements in mHHS and achieved the corresponding MCID. Level of Evidence: Case series; Level IV.
In der vorliegenden Studie wurde die bei der Arthroskopie gewonnene Lavageflüssigkeit von28 Patienten mit unterschiedlich stark ausgeprägter Cranio-Mandibulärer Dysfunktion anfand der Parameter Interleukin-lß (DL-lß), lnterleukin-6 (IL-6) als Marker für die Regulation entzündlicher Vorgänge, Nitrotyrosin (Ntyr) und Peroxidase (PER) als Marker oxidativer Veränderungen N-Acethyl-ß-D-Giucosaminidase und Kollagenase als Marker des Abbaus der Knorpelmatrix sowie Gesamtprotein untersucht. Mit diesen Messungen sollte der Frage nachgegangen werden, ob eine biochemische Analyse der Kiefergelenklavageflüssigkeit zur Differentialdiagnose, Therapiekontrolle und Prognoseverbesserung bei Kiefergelenkerkrankungen beitragen kann. Die Bewertung der Gelenke nach dem klinisch-arthroskopischen Befund konnte durch die biochemischen Resultate nur teilweise bestätigt werden. Die Ergebnisse weisen darauf hin, dass Kiefergelenke mit Dysfunktion sich unterteilen lassen in Gelenke mit und ohne biochemische Marker einer Entzündung in der Synovialflussigkeit. Die Parameter Eiweiß und Peroxidase erwiesen sich als geeignet, subakut chronische Entzündungen im Kiefergelenk anzuzeigen.
Background:
Arthroscopic treatment of femoroacetabular impingement syndrome (FAIS) has become a common procedure. However, meaningful long-term clinical outcomes have not been defined.
Purpose:
To define the minimal clinically important difference (MCID), substantial clinical benefit (SCB), and patient acceptable symptomatic state (PASS) for the modified Harris Hip Score (mHHS) at a minimum 10-year follow-up in patients undergoing arthroscopic treatment for FAIS and identify preoperative predictors for achievement of the MCID, SCB, and PASS.

Methods:
A consecutive series of patients undergoing arthroscopic treatment for FAIS between 2007 and 2009 with a minimum 10-year follow-up was analyzed. Patient data included patient characteristics, radiographic parameters, and the pre- and postoperative mHHS and visual analog scale (VAS) for pain score. Paired t tests were used to compare the patient-reported outcome measures (PROMs). The MCID was determined by calculating half of the standard deviation, and SCB and PASS were calculated by the anchor method. Correlation and logistic regression analyses were conducted to identify predictors for the achievement of the MCID, SCB, and PASS.
Results:
A total of 44 patients (27 men, 17 women) were included. The mean age and body mass index were 42.2 years (range, 16-67 years) and 22.3 kg/m2 (range, 16.76-29.78 kg/m2), respectively. The MCID, absolute SCB, net change SCB, and PASS of the mHHS were calculated to be 19.6, 90.1, 31.5, and 84.4 points, respectively. Preoperative symptom duration was identified as an independent predictor for the achievement of meaningful clinical outcomes. The median symptom durations for patients who achieved the MCID, absolute SCB, net change SCB, and PASS were 11.7, 9.1, 9.0, and 10.8 months, respectively. The median symptom duration for patients who did not achieve the MCID, absolute SCB, net change SCB, and PASS were 15.8, 17.4, 17.3, and 18.4 months, respectively. No other statistically significant correlations were found.
Conclusion:
The preoperative duration of symptoms was identified as an independent predictor for achievement of the MCID, SCB, and PASS. These findings can be helpful in accelerating the transition to surgical treatment of FAIS.
The pore forming alpha-toxin (hemolysin A, Hla) of Staphylococcus aureus (S. aureus) is a major virulence factor with relevance for the pathogenicity of this bacterium, which is involved in many cases of pneumonia and sepsis in humans. Until now, the presence of Hla in the body fluids of potentially infected humans could only be shown indirectly, e.g., by the presence of antibodies against Hla in serum samples or by hemolysis testing on blood agar plates of bacterial culture supernatants of the clinical isolates. In addition, nothing was known about the concentrations of Hla actually reached in the body fluids of the infected hosts. Western blot analyses on 36 samples of deep tracheal aspirates (DTA) isolated from 22 hospitalized sepsis patients using primary antibodies against different epitopes of the Hla molecule resulted in the identification of six samples from five patients containing monomeric Hla (approx. 33 kDa). Two of these samples showed also signals at the molecular mass of heptameric Hla (232 kDa). Semiquantitative analyses of the samples revealed that the concentrations of monomeric Hla ranged from 16 to 3200 ng/mL. This is, to our knowledge, the first study directly showing the presence of S. aureus Hla in samples of airway surface liquid in human patients.
Die positive Assoziation zwischen Testosteron, Blutdruck und arterieller Hypertonie bei Frauen

Die aHT stellt einen der wichtigsten Risikofaktoren für kardiovaskuläre Erkrankungen dar. Der Einfluss der Androgene auf den Blutdruck bei Frauen ist durch bisherige Studien, welche die Assoziation zwischen der Testosteron Serumkonzentration und dem Blutdruck bei Frauen untersuchten, aufgrund von widersprüchlichen Ergebnissen noch unzureichend geklärt. Daher war es Ziel dieser Studie, die Assoziation von Testosteron, Androstendion, freiem Testosteron, Sexualhormon-bindendem Globulin und Blutdruck bei Frauen sowohl im Quer- als erstmals auch im Längsschnitt zu untersuchen. Grundlagen für die Analysen waren die Daten von 1428 Teilnehmerinnen der Study of Health in Pomerania (SHIP), einer bevölkerungsbasierten Studie, die seit 1996 in der Studienregion Nord-Ostvorpommern durchgeführt wird. Um Assoziationen feststellen zu können, wurden multivariate Regressionsmodelle sowohl an der Gesamtpopulation als auch an prä- und postmenopausalen Untergruppen durchgeführt. In der Gesamtpopulation war die Testosteron Serumkonzentration im Querschnitt betrachtet positiv mit Blutdruck assoziiert, allerdings bedingt durch die starke Assoziation in der postmenopausalen Subgruppe. Längsschnittanalysen konnten die im Querschnitt festgestellten Assoziationen bestätigen. Weiterhin waren im Längsschnitt niedrigere Testosteron Serumkonzentrationen mit einem erniedrigten Risiko von prävalenter aHT vergesellschaftet. Niedrige SHBG-Konzentrationen waren mit erhöhtem Risiko von prävalenter aHT in der postmenopausalen Untergruppe und einem erhöhtem Risiko von inzidenter aHT in der Gesamtpopulation assoziiert. Obwohl die genauen Wirkmechanismen der Beeinflussung des Blutdruckes durch Testosteron noch nicht abschließend geklärt sind, scheinen Androgene bei Frauen eine Steigerung des Blutdrucks zu bewirken. Die Kausalität dieser Beziehung muss noch überprüft werden und daher kann Testosteron zum jetzigen Zeitpunkt nur als Risikomarker und nicht als Risikofaktor für erhöhten Blutdruck und aHT bezeichnet werden.
This thesis deals with thickness optimization of shells. The overall task is to find an optimal thickness distribution in order to minimize the deformation of a loaded shell with prescribed volume. In addition, lower and upper bounds for the thickness are given. The shell is made of elastic, isotropic, homogeneous material. The deformation is modeled using equations from Linear Elasticity. Here, a basic shell model based on the Reissner-Mindlin assumption is used. Both the stationary and the dynamic case are considered. The continuity and the Gâteaux-differentiability of the control-to-state operator is investigated. These results are applied to the reduced objective with help of adjoint theory. In addition, techniques from shape optimization are compared to the optimal control approach. In the following, the theoretical results are applied to cylindrical shells and an efficient numerical implementation is presented. Finally, numerical results are shown and analyzed for different examples.
This thesis draws a comprehensive picture about the radiation and diversification of truncatelloidean gastropods across the south pacific. It covers three more specifc studies focussing on the Truncelloideans from Fiji, Vanuatu and New Caledonia, respectively. And a conclusive analysis that combines the results of the three more specific studies and enhances them using species from the Austral Islands, Lord Howe Island, the Indonesian island Sulawesi as well as several species from New Zealand and Australia. Molecular phylogenies were calculated using four nuclear gene fragments (ITS2; 18S rRNA; 28S rRNA and Histone 3) besides the mitochondrial COI and 16S rRNA. Further molecuular data was used to calculate dated phylogenies, perform ancestral range reconstructions and develop a modified molecular barcoding approach.
Die vorliegende Arbeit befasst sich mit der Verbreitung und Populationsgenetik der invasiven asiatischen Buschmücke Ae. j. japonicus, die sich seit 2008 durch Menschen-vermittelten Transport in Deutschland ausbreitet. Aedes j. japonicus ist unter Laborbedingungen Vektor für verschiedene Viren, unter anderem für das Dengue-Virus und das Chikungunya-Virus, und wurde im Feld mit dem Japanische Enzephalitis-Virus, dem West Nil-Virus und dem La Crosse-Virus infiziert gefunden. 2012 wurde aufgrund mehrerer unabhängiger Mücken-Einsendungen im Rahmen des Citizen-Science-Projekts “Mückenatlas” eine Population der Asiatischen Buschmücke in Westdeutschland entdeckt. Das Verbreitungsgebiet dieser Population befand sich weit nördlich der bisher angenommenen nördlichen Verbreitungsgrenze der Art in Süddeutschland. Das Ausmaß der Population wurde nach einem zeitlich begrenzten Monitoring auf eine Fläche von ca. 2000 km2 bestimmt. Aus dieser Population wurden Individuen von fünf Orten populationsgenetischen Analysen unterzogen, um verwandtschaftliche Beziehungen innerhalb der Population und im Vergleich zu anderen europäischen Populationen aufzudecken. Hierzu wurden sieben Mikrosatelliten-Loci untersucht. Zusätzlich wurde ein Teil der mitochondrialen nad4-Genregion der Individuen auf Nukleotid-Polymorphismen untersucht. Die Ergebnisse wurden mit bereits zuvor erhobenen Daten von Populationen aus der Schweiz, aus Österreich/Slowenien und Belgien verglichen. Die Mikrosatellitensignatur der westdeutschen Population unterschied sich deutlich von der der anderen europäischen Populationen. Weiterhin wurden verschiedene nad4-Haplotypen gefunden, die zuvor nirgendwo sonst in Europa aufgetreten waren. Demnach ist zu vermuten, dass diese Population auf eine unabhängige Einschleppung von Individuen aus Übersee zurückgeht. Der genaue Ursprung – USA oder Ostasien – konnte nicht bestimmt werden. 2013 wurden zwei weitere Ae. j. japonicus-Populationen in Europa entdeckt: eine in Norddeutschland und eine weitere in den Niederlanden. Die genetischen Signaturen von Individuen dieser Populationen wurden wie beschrieben analysiert. Zusätzlich wurde das genetische Material einer größeren Menge von Individuen aus Slowenien sowie von Individuen aus Kroatien und Süddeutschland untersucht. Die Ergebnisse wurden mit denen aus der vorigen Studie verglichen und zeigten aufgrund einer ähnlichen Mikrosatellitensignatur und gleicher nad4-Haplotypen klar, dass die norddeutsche Population eine Subpopulation der westdeutschen ist. Die geringe Populationsdichte und die vergleichsweise kleine Ausdehnung der norddeutschen Population deuten außerdem darauf hin, dass die Abspaltung nicht lange zurückliegt. Die niederländische Population scheint hingegen auf einer weiteren Einschleppung von Individuen aus Übersee zu basieren. Im Spätsommer 2015 wurde die bisher letzte deutsche Ae. j. japonicus-Population in Oberbayern und dem angrenzenden Österreich entdeckt. Populationsgenetischen Analysen zufolge ist diese Population eng mit der früher beschriebenen österreichisch-slowenischen Population verwandt und unterscheidet sich von allen anderen deutschen Populationen, was darauf schließen lässt, dass es sich bei ihr um eine Abspaltung von der österreichisch-slowenischen Population handelt. Die Ver- und Ausbreitung von Ae. j. japonicus in West- und Norddeutschland wurde vom Zeitpunkt der Entdeckung in 2012/2013 bis 2015 beobachtet. In dieser Periode erweiterte die westdeutsche Population ihr Verbreitungsgebiet beträchtlich, während die norddeutsche überhaupt nicht zu expandieren schien. Dies ist möglicherweise darauf zurückzuführen, dass die norddeutsche Population jünger als die westdeutsche ist, das Verschleppungsereignis noch nicht so weit zurückliegt und die Population sich noch in der Gründerphase befindet. Die passive weltweite Verschleppung von Stechmücken wird in der Zukunft vermutlich zunehmen, und die Etablierung und Ausbreitung invasiver Spezies, inklusive der Asiatischen Buschmücke und anderer potenzieller Überträger von Krankheitserregern, werden Europa und Deutschland weiterhin vor herausfordernde Probleme stellen. Das Monitoring der Ausbreitung von Populationen und die Durchführung populationsgenetischer Analysen zur Ermittlung von geographischen Ursprüngen sowie von Wanderungs- und Transportrouten werden helfen, weitere Einschleppungs- und Ausbreitungsereignisse nachzuvollziehen und zu unterbinden und sind daher essenzielle Instrumente des Managements von Mückenvektoren.
Adaptation mechanisms within the B cell composition for successful human and murine pregnancies.

Introduction
A well-balanced immune maternal status is essential for favourable outcome of pregnancy. Due to their complexities, not all immune adaptations that promote tolerance during pregnancy are known. To understand the adaptation of the B cell compartment, we analysed and compared B cell lymphopoiesis in different lymphoid tissues in a number of murine models.
Furthermore, we focused on the humoral immune response during pregnancy. We analysed immunoglobulin profiles in human subjects and mice during pregnancy.
These cellular alterations are subject to the influence of chemokines, among others. Therefore, we assessed serum levels of B cell activation factor to clarify its effects during pregnancy.
Methods
For analysis of the human peripheral B cell compartment, peripheral blood samples from age-matched non-pregnant and pregnant women without pregnancy complications, immunological disease or acute/chronic inflammation were collected and sub-classified into four different groups: non-pregnant, and first, second, or third trimester of pregnancy. The experiments, based on a mouse model, were performed with 8-week-old female mice: clinically healthy non-pregnant (CBA/J (H2k)), pregnant mice with normal gestation (BALB/c (H2d) x CBA/J (H2k)), and mice with pregnancy loss (DBA/2J (H2d) x CBA/J (H2k)). Subsequently, peripheral blood mononuclear cells from blood and lymphatic organs were isolated following standard protocols. The B cell analysis was performed by flow cytometry. The immunoglobulin serum levels of the human and murine subgroups were quantitated using Bio-Plex isotyping assay and analysed by a Bio-Plex reader. To quantify B cell activating factor (BAFF) in serum of pregnant and non-pregnant mice a BAFF enzyme-linked immunosorbent assay was used. The concentrations were determined by using a FLUOstar OPTIMA microplate reader. All statistical analyses were performed using the Kruskal–Wallis test with Dunn’s post-test in GraphPad Prism software. P values of < 0.05 were considered statistically significant.
Results
We were able to demonstrate B cell lymphopenia in mice bone marrow downstream of pre-pro B cells, irrespective of pregnancy outcome. The mature bone marrow B cells did not show this adjustment mechanism during normal gestation.
Closer inspection of the splenic tissue revealed expansion and activation of marginal zone B cells in mice with a normal pregnancy. However, this was not observed in mice suffering from pregnancy disturbances. Natural antibodies secreted from marginal zone B cells were also present at higher concentrations in serum of pregnant mice, compared to non-pregnant animals.
We also found significantly higher levels of natural antibodies in serum of pregnant women compared to non-pregnant age-matched controls. Analysis showed significantly lower levels of BAFF in mice with normal pregnancy as compared to non-pregnant mice.
Conclusions
We are able to show mechanisms within the B cell compartment as well as the change within the natural antibodies that might be crucial for successful pregnancy in both humans and mice. Furthermore, BAFF seems to play a central role as a mediator of peripheral B cell compartment and B cell lymphopoiesis in the bone marrow for successful pregnancy.
Bats (Chiroptera) form the second largest order of mammals and with over 1,250 species, they represent about 20% of all mammalian species worldwide. They are the only mammals with true and sustained flight and distributed all over the world except the arctic regions. Moreover, bats entered specific ecological niches and with their food spectra, they reduce different arthropod populations as well as disperse seeds and pollen of plant species in various regions and habitats.
Bats also have a crucial role in spreading high-pathogenic and zoonotic viruses, harbor in general more viruses (zoonotic and non-zoonotic), and, related to the species, number even more than rodents. However, clinical symptoms of viral diseases are rarely reported in bat communities. Also seroconversions after infection were not reported for a variety of viruses found in bats. Since the incidence of virus-positive bats estimated in passive surveillance studies is usually very low, it is a question how such viruses can use bats as reservoir hosts. There is obviously a special evolutionary relationship between the pathogens and bats as hosts, which are based on possibly physiologic adaptations also in resistance and immunity.
In this thesis, the two lyssaviruses, European Bat Lyssavirus 1 and 2 (EBLV-1 and -2) were chosen as a model to investigate the immune response of European bats against viral infection in vitro. Lyssaviruses are the causative agents of rabies, a fatal zoonotic disease with neurotropic characteristics.
One main question to investigate was in which way bats act as reservoir host and developed a high disease resistance. The present thesis is based on three hypotheses about innate immune response against lyssavirus infection:
A) In bats specific peripheral resistance mechanisms evolved which reduce the risk of systemic viral infection after a hypothesized airborne transmission and infection via nasal epithelium supported by the social structure of and communication within bat communities.
B) The co-evolution of EBLV and the innate resistance of bats resulted in a very effective type I interferon response to inhibit a systemic lyssavirus infection.
C) The specific physiology of body temperature of bats with daily torpor depresses the viral replication but favours the type I interferon response.
To analyze the interferon-based resistance mechanisms, the type I interferon (IFN) genes of two European bats species (Eptesicus serotinus and Myotis myotis) were cloned and sequenced. Using established cell lines from the respiratory nasal epithelium (MmNep), olfactory nasal epithelium (MmNol), and Bulbus olfactorius brain (MmBr), the type I IFN response along a possible airborne infection route was investigated. The anti-viral effects and induction of IFNs/interferon stimulated genes (ISGs) in each cell line were also investigated in detail after infection in vitro. Finally, the influence of different temperatures on lyssavirus replication was analyzed in cell culture experiments.
The results indicated that (a) along the hypothesized airborne infection route the susceptibility for lyssavirus infections is decreased, (b) the type I IFN activity in contrast is increased contributing to a limitation of lyssavirus replication and (c) an obvious influences of varying cultivation temperatures on the resistance against lyssavirus infections, which favor the IFN response and repressing lyssavirus replication.
The result from these in vitro studies supports the hypothesis of a special co-evolution between lyssaviruses and bats. However, in vivo studies on the relevance in infected animals are missing so far. This model could also explain the generally limited pathogenicity of bat-associated viruses.
The vast majority of RNA splicing in today‘s organisms is achieved by the highly regulated and precise removal of introns from pre-mRNAs via the spliceosome. Here we present a model of how RNA splicing may have occurred in earlier life forms. We have designed a hairpin ribozyme derived spliceozyme that mediates two RNA cleavages and one ligation event at specific positions and thus cuts a segment (intron) out of a parent RNA and ligates the remaining fragments (exons). The cut-out intron then performs a downstream function, acting as a positive regulator of the activity of a bipartite DNAzyme. This simple scenario shows how small RNAs can perform complex RNA processing dynamics, involving the generation of new phenotypes by restructuring segments of given RNA species, as well as delivering small RNAs that may play a functional role in downstream processes.
Planning Modes for Major Transportation Infrastructure Projects (MTIPs): Comparing China and Germany

Endothelial dysfunction (ED) comes with age, even without overt vessel damage such as that which occurs in atherosclerosis and diabetic vasculopathy. We hypothesized that aging would affect the downstream signalling of the endothelial nitric oxide (NO) system in the vascular smooth muscle (VSM). With this in mind, resistance mesenteric arteries were isolated from 13-week (juvenile) and 40-week-old (aged) mice and tested under isometric conditions using wire myography. Acetylcholine (ACh)-induced relaxation was reduced in aged as compared to juvenile vessels. Pretreatment with L-NAME, which inhibits nitrix oxide synthases (NOS), decreased ACh-mediated vasorelaxation, whereby differences in vasorelaxation between groups disappeared. Endothelium-independent vasorelaxation by the NO donor sodium nitroprusside (SNP) was similar in both groups; however, SNP bolus application (10−6 mol L−1) as well as soluble guanylyl cyclase (sGC) activation by runcaciguat (10−6 mol L−1) caused faster responses in juvenile vessels. This was accompanied by higher cGMP concentrations and a stronger response to the PDE5 inhibitor sildenafil in juvenile vessels. Mesenteric arteries and aortas did not reveal apparent histological differences between groups (van Gieson staining). The mRNA expression of the α1 and α2 subunits of sGC was lower in aged animals, as was PDE5 mRNA expression. In conclusion, vasorelaxation is compromised at an early age in mice even in the absence of histopathological alterations. Vascular smooth muscle sGC is a key element in aged vessel dysfunction.
Metabolic syndrome is a significant worldwide public health challenge and is inextricably linked to adverse renal and cardiovascular outcomes. The inhibition of the transient receptor potential cation channel subfamily C member 6 (TRPC6) has been found to ameliorate renal outcomes in the unilateral ureteral obstruction (UUO) of accelerated renal fibrosis. Therefore, the pharmacological inhibition of TPRC6 could be a promising therapeutic intervention in the progressive tubulo-interstitial fibrosis in hypertension and metabolic syndrome. In the present study, we hypothesized that the novel selective TRPC6 inhibitor SH045 (larixyl N-methylcarbamate) ameliorates UUO-accelerated renal fibrosis in a New Zealand obese (NZO) mouse model, which is a polygenic model of metabolic syndrome. The in vivo inhibition of TRPC6 by SH045 markedly decreased the mRNA expression of pro-fibrotic markers (Col1α1, Col3α1, Col4α1, Acta2, Ccn2, Fn1) and chemokines (Cxcl1, Ccl5, Ccr2) in UUO kidneys of NZO mice compared to kidneys of vehicle-treated animals. Renal expressions of intercellular adhesion molecule 1 (ICAM-1) and α-smooth muscle actin (α-SMA) were diminished in SH045- versus vehicle-treated UUO mice. Furthermore, renal inflammatory cell infiltration (F4/80+ and CD4+) and tubulointerstitial fibrosis (Sirius red and fibronectin staining) were ameliorated in SH045-treated NZO mice. We conclude that the pharmacological inhibition of TRPC6 might be a promising antifibrotic therapeutic method to treat progressive tubulo-interstitial fibrosis in hypertension and metabolic syndrome.
Over the past 10 years, the crisis of sepsis has remained a great challenge. According to data from 2016, the sepsis-related mortality rate remains high. In addition, sepsis consumes extensive medical resources in intensive care units, and anti-inflammatory agents fail to improve sepsis-associated hyperinflammation and symptoms of immunosuppression. The specific immune mechanism of sepsis remains to be elucidated. Reactive oxygen species (ROS) are triggered by energy metabolism and respiratory dysfunction in sepsis, which not only cause oxidative damage to tissues and organelles, but also directly and indirectly promote NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome activation. NLRP3 inflammasomes enlarge the inflammatory response and trigger apoptosis of immune cells to exacerbate sepsis progression. Inhibiting the negative effects of ROS and NLRP3 inflammasomes therefore provides the possibility of reversing the excessive inflammation during sepsis. In this review, we describe the interaction of ROS and NLRP3 inflammasomes during sepsis, provide prevention strategies, and identify fields that need further study.
Recent climate change has affected the forest system comprehensively. Northern hemisphere elevational treelines are considered as a key environment for monitoring the effects of current anthropogenic climate change. Moreover, trees from these areas are also widely employed in paleo-climate reconstructions. The stability of the tree growth climate relationship under current scenario is crucial for all tree ring based climate researches. It is important to investigate how trees respond to this rapid environmental change at altitudinal treelines. Tree cores from 21 treeline sites of three species (Pinus tabulaeformis, Picea crassifolia, and Sabina przewalskii) from Northeastern Tibetan have been conducted in this thesis. The instable correlations between tree growth and climate are the general response pattern of trees from all study sites in NE Tibetan Plateau. Picea crassifolia shows the most instable response to climate factors (mean monthly temperature and total monthly precipitation). Pinus tabulaeformis and Sabina przewalskii just showed instable and divergent responses to their main limiting climate factors but no clear trend was found which is limited by the few sample sites. Corresponding to divergent responses of Picea crassifolia to mean monthly temperature, most radial growth of Picea crassifolia were inhibited by this climate change type drought, only few trees within same sites grew faster due to temperature increasing during recent decades. The divergence response mainly started in last 30 years in six of eleven sample sites over the Northeastern Tibetan Plateau. North-westerly drier sites showed a large percentage of trees per site with a negative correlation to temperature and mostly southerly moister sites showed more mixed responses with both negatively and positively responding trees within site. Concurrent with the regional pattern, low elevation sites show mostly negative correlations with temperature and high elevation sites show more mixed responses. As the hydrothermal conditions of the investigation area changed to a drier and warmer combination, drought stress on tree growth have been intensifying over time and expanding spatially from the middle to most of our study area during the last half century. The Picea crassifolia tree growth climate relationship conducted on an elevational gradient with four different levels from upper treeline to lower treeline at the NE Tibetan Plateau. Results show that upper treeline trees show divergent growth trends and divergent responses in recent decades. Trees from lower treeline show a strengthening drought stress signal over time and no divergent growth trends within sites. This potential ecological reaction of tree populations to changing environmental conditions shows an implications for using trees to reconstruct climate, since the indiscriminate use of tree ring data from sites showing opposite responses to increasing warming could cause mis-calibration of tree ring based climate reconstructions, and over- or underestimation of carbon sequestration potential in biogeochemical models. The physiological response of Sabina przewalskii tree growth to major limiting climate factors based on the Vaganov-Shashkin (VS) model indicated that precipitation during the early growing season, especially in May and June, has significant effect on tree growth, while temperature mainly affects tree growth by warming-induced drought and by extending the growing season in the NE Tibetan Plateau. Under current and projected climate scenarios, modeling results predict an increase in radial growth of Sabina przewalskii around the Qaidam Basin, with the potential outcome that regional forests will increase their capacity to sequester carbon. However, most Picea crassifolia trees growing at lower elevations than Sabina przewalskii might be continue stressed by the warming induced drought and might decrease radial growth in future.
In dieser Studie wurde die Wirkung der Kombination der Akupunktur-Therapie und der kliniküblichen analgetischen Therapien auf postoperative Schmerzen bei gelenkersetzenden operativen Eingriffen am Kniegelenk untersucht. Die Akupunkturbehandlung in der Verum-Akupunktur-Gruppe (VAG) erfolgte in 10 Sitzungen, beginnend vom ersten postoperativen Tag bis zum zehnten postoperativen Tag. In der Sham-Akupunktur-Gruppe (SAG) bekamen die Probanden im gleichen Zeitraum jeweils eine Sham-Akupunktur. Folgende Punkte wurden bei der Verum-Akupunktur-Gruppe (VAG) genadelt: Di 4, Ma 36, Mi 6, Ni 3, Gb 32, Le 3, Lg 20. Diese Punkte können Schmerzen lindern, die Psyche beruhigen, die Muskulatur entspannen, Übelkeit und Erbrechen lindern das Qi stärken und regulieren, gestautes Blut beleben und befreien und die Harmonie von Yin und Yang wieder herstellen. Als objektive Untersuchungskriterien und Kontrollparameter für den Verlauf der Therapie dienten die Messungen des Beinumfangs, Kniegelenkergusses und der lokalen Überwärmungen am Knie und der Knee-Society-Score nach Insall et al. (1989). Der Status der einzelnen Probanden wurde vor und nach der Behandlung standardisiert erfasst. Ein Vergleich der Untersuchungsergebnisse ergab folgende Erkenntnisse: Die Schmerzintensität verbesserte sich in der Verum-Akupunktur-Gruppe deutlich im Vergleich zur Sham-Akupunktur-Gruppe. So lag der Mittelwert bei 46,5 Punkten in der Verum-Akupunktur-Gruppe nach der Behandlung, wobei in der Kontrollgruppe der Mittelwert lediglich bei 40,97 Punkten lag. Dieser signifikante Unterschied ist auch statistisch von immenser Bedeutung p = 0,001. Auch der Vergleich des Kniescore und Funktionsscore zeigten ähnliche Ergebnisse. Die Mittelwerte des Kniescore der SAG lag nach der Behandlung bei 77,87 und in der VAG bei 84,23. Bei der Untersuchung des Funktionsscores betrug der Mittelwert in der SAG nach der Behandlung 45,48 Punkte und 55,81 Punkte in der VAG. Diese Unterschiede in den Gruppen sind statistisch signifikant (p = 0,005 Kniescore; p = 0,002 Funktionsscore). Zusätzlich wurden in dieser Studie die Ergebnisse innerhalb der VAG zwischen männlichen und weiblichen Probanden verglichen. Hierbei konnten keine signifikanten Unterschiede zwischen den Geschlechtern in der VAG festgestellt werden. Die Ergebnisse der Wundheilungsuntersuchungen zeigten keine signifikanten Unterschiede zwischen beiden Gruppen. So waren die Werte bei der Gelenkergussuntersuchung in beiden Gruppen identisch, so dass keine Unterschiede beobachtet werden konnten. Beim Vergleich der lokalen Überwärmung am Kniegelenk lag nur ein minimaler Unterschied vor, der nicht von statistischer Bedeutung ist p = 0,69. Auch die Ergebnisse der Beinumfanguntersuchungen zeigten keine statistisch signifikanten Unterschiede. Die Ergebnisse der Sham-Akupunkturbehandlung sind deutlich schlechter als die der Akupunkturbehandlung, was darauf schließen lässt, dass die Akupunkturbehandlung nicht einer Sham-Akupunkturbehandlung gleich zu setzen ist.
All types of muscles use Ca2+ as their main intracellular messenger. In skeletal muscle fibers abnormal levels of intracellular calcium result in altered contractile properties, altered energy metabolism, and altered gene expression. Moreover, long term failure of normal Ca2+ homeostasis can lead to cell death of muscle fibers by necrosis and apoptosis. Elevations of intracellular Ca2+ levels are more and more regarded as the reason for pathological changes and muscle fiber damage in Duchenne Muscular Dystrophy (DMD). DMD is a severe recessive x-linked muscle disease caused by mutations in the dystrophin gene. The characteristics of DMD are muscle tissue wasting and fibrosis. Both muscle wasting and intracellular Ca2+ are to be reflected in changes of muscle force. Several Ca2+ conducting channels including transient receptor potential (TRP) channels are supposed to account for the abnormal Ca2+ homeostasis in DMD. Gene expressions of TRP channels have been studied in human and mouse skeletal muscle and among others TRPC3, TRPC6 and TRPV4 channels were found to occur in skeletal muscles. The present study followed the hypothesis that TRPC3, TRPC6 and TRPV4 are functional in skeletal muscle fibers and that they contribute to muscular Ca2+ homeostasis. Further, it was assumed that dysfunction of the mentioned TRP channels contributes to abnormal contractile properties and pathology and of dystrophin-deficient muscle. To study Ca2+ changes in mouse skeletal muscle fibers the fluorescent calcium indicator Fura-2 was used. Further, the technique of Mn2+ quench of Fura-2 fluorescence was applied. Muscle force measurements of mouse soleus and diaphragm strips were performed. To elucidate abnormalities of TRP channel function in dystrophin-deficient muscle, muscles and muscle fibers of mdx mice were studied. Hyperforin, an activator of TRPC6 channels elicited increases of calcium levels in wildtype muscle fibers. These increases were partly inhibited by the TRPC6 inhibitor 1-(5-chloronaphthalenesulfonyl) homopiperazine hydrochloride (ML-9). The TRPC3/TPRC6 activator 1-oleoyl-2-acetyl-sn-glycerol (OAG) resulted in increased calcium entry, which was attenuated by ML-9. 2-aminoethoxydiphenylborane (2-APB), an unspecific TRP channel inhibitor, suppressed calcium entry in muscle fibers under basal conditions. In addition, the specific TRPC3 inhibitor Pyr3, strongly inhibited background calcium entry. The TRPV4 activator 4α-phorbol 12,13-didecanoate (4α-PDD) induced significant increased calcium entry and this increase could be inhibited by the TRPV4 inhibitor HC 067047. During muscle force recordings ML-9 significantly inhibited twitches and tetani and accelerated muscle fatigue during sustained repetitive stimulation. The results indicate that TRPC3, TRPC6 and TRPV4 are functionally expressed in mouse muscle fibers. TRPC3 stays active under the basal conditions and contributes to background calcium entry. In contrast, TRPC6 and TRPV4 did not seem to be active at resting conditions, but could be pharmacologically activated. TRPC6 may play a role to counteract the calcium loss under long-term muscle fatigue. Though TRPC3 and C6 play a role for muscular Ca2+ homeostasis, it is unclear whether and how the two channels associate and cross-talk with each other in skeletal muscle cells. In mdx fibers Pyr3 inhibited background calcium influx stronger that in WT fibers, implying a possible over-activation of TRPC3 channels in mdx muscle fibers. At later stages mdx muscle showed marked decrease in force reflecting muscle wasting. Soleus showed moderate decrease and diaphragm showed severe decrease (more than 60%) in force. Resistance to muscle fatigue was shown in mdx soleus muscle when compared with WT soleus muscle. Diaphragm segments of mdx mice showed very strong resistance to muscle fatigue. The results indicate a substantial loss of muscle mass, an increase in oxidative fiber types and a reduction of fast fatigable muscle fibers. It is concluded that the hypothesis of functional expression of TRPC3, TRPC6 and TRPV4 in mouse skeletal muscle has been confirmed. The results give improved knowledge about the relation of Ca2+ homeostasis, mdx pathology and TRP channels. Diaphragms of old mdx mice show severe muscle weakness but the remaining fibers of the diaphragm showed strong fatigue-resistance. The application of a TRPC3 inhibitor may be a promising treatment to prevent high Ca2+ mediated muscle damage in muscular dystrophy.
Liu–II coal pit is a typical example of China’s deep coal mines which is seriously threatened by groundwater inrush from the underlying carboniferous Taiyuan limestone formation. An exhaustive data set of this confined aquifer exists. The aquifer lies 45 m∼ 60 m below the major coal seam. A traditional artesian aquifer test has been performed in order to assess the hydraulic properties, e.g. transmissivity (T) and storage coefficient (S). This artesian aquifer test is conducted with four simultaneously operating production wells while the discharge of each production well varied with time. The results of this test suggest that the aquifer is heterogeneous. Therefore, the according problems are: (1) how to analyze the artesian aquifer test with linearly declining discharge; (2) how to deal with multiple production wells in an aquifer test; (3) how to adequately consider aquifer heterogeneity. Thus, the objective of this thesis is to solve these problems. 1) As opposed to classical above-ground pumping tests, it is difficult to control the discharge rate of the production well in a deep mine artesian aquifer test since the hydraulic pressure is extraordinary high. Moreover the discharge rate won’t descend rapidly to zero, thus the analytical solution of Jacob and Lohman (1952) type curve for the artesian aquifer test will not be applicable. It is more reasonable to analyze the test as a pumping test with variable discharge. It is considered to rebuild a hydrogeological conceptual model which is similar with Theis (1935) model but with the variable discharge. A general equation for any discharge variability is given. Its application for the linearly declining discharge is presented subsequently, and a type curve of this equation with linearly declining discharge is given as well. After that, a simple numerical model is built by FEFLOW to simulate an artificial pumping test with the linearly declining discharge by assigning different parameter sets for transmissivity and storage coefficient. The type curve method is applied to evaluate transmissivity and storage coefficient for the linearly declining discharge well. The deviation between the given values of transmissivity and storage coefficient in FEFLOW and the values of those calculated by matching point are sufficiently small. Thus, when the discharge of production well declines linearly, a type curve method as an empirical method is reasonable and gives satisfactory values of these hydrogeological parameters. 2) In some cases, it is necessary to conduct a pumping test (or an artesian aquifer test) with several pumping wells (or production wells) which work simultaneously in order to discharge maximum quantity of groundwater. Normally, the superposition method or numerical simulation is applied to analyze the test result. However, a new approach called “Well Generalization Method” is defined and analyzed in this thesis. It is an easy–to–use approach for hydrogeologist to estimate the aquifer parameters while conducting an aquifer test. Since the key point of this approach is using a generalization well to substitute the pumping (or production) wells, it is obvious that this approach will generate the estimated error of parameters. Accordingly, several scenarios are analyzed and discussed based on the artificial type aquifer designed in FEFLOW. A homogeneous aquifer and a heterogeneous aquifer which is generated by geostatistical stochastic simulation technique (see 3)) are discussed separately. As a result, this approach is feasible and applicable under some conditions when the calculated observation well is arranged more than about 2.5 times the scale of the multi–pumping–wells field away from the center of the multi–pumping–wells field, furthermore, the maximum deviation of drawdown resulting from these observation wells will be less than 0.5 m, and the estimated value of transmissivity will be 0.44% smaller than real value. 3) Finally aquifer heterogeneity is addressed, in order to check the introduced method for applicability under realistic conditions. It has been described that aquifer heterogeneity plays a major role in hydrodynamic processes (e.g. de Marsily et al., 1998). Geostatistics which is considered as a useful tool for characterizing the spatial variability of transmissivity is applied to solve this problem. Based on the results of the artesian aquifer test conducted in Liu–II coal pit, a model of spatial variability of transmissivity is developed. Sequentially, the variogram model is applied in ordinary kriging to interpolate the transmissivity distribution, and in sequential Gaussian simulation to simulate a random field of transmissivity data in order to reflect its small scale variability. A comparison of the results of estimation and simulation of transmissivity indicates that the simulated values better reflect the spatial variability, reversely, the estimated values are much smoother.
Bats belong to the most gregarious and diverse mammals with highly complex social behaviors. Despite extensive research on their ecology and social behavior in some bat species, gained insights are restricted to only few of the more than 1300 species. In the recent past, bats have also become a central topic of a different branch of research: Since the 1990s bats came to the fore of virologists and immunologists due to the bats’ apparent importance as reservoir hosts and vectors of several (mostly tropical) diseases. While this research is focused mainly on emerging infectious diseases linked to bats, and their zoonotic potential, little has been invested regarding the link between disease transmission and bat social systems.
In my work, I aim at filling this gap by merging automated daily roosting observations, social network analysis, and a virological screening in Natterer’s bats (Myotis nattereri). In a collaborative approach, my co-workers and I analyzed the social structure of individually marked Natterer’s bats, their astrovirus detection rate and transmission pathways within their colony, as well as roosting interactions between different co-occurring con- and heterospecific bat colonies.
We discovered Natterer’s bats to display a very divergent social network structure that contradicts the findings of previous studies on large fission-fusion groups. Contrary to the modular social network structure found in e.g. primates or other bats species, the social network of Natterer’s bats consists of only one highly interconnected community. Moreover, although the close proximity between bat hosts in the colony should strongly promote direct transmission, we found indications that astrovirus infections follow at least partly an indirect transmission pathway via contaminated roost use. Lastly, our results prove that co-occurring con- and heterospecific bat colonies, e.g. as in this study Natterer’s bats, brown long-eared bats and Bechstein’s bats, can influence each other in their roost use by avoiding conspecific roosts and by being attracted towards those of heterospecifics. This holds implication for the transmission of parasites and pathogens within and between different colonies with opportunities for spillovers. To conclude, this multidisciplinary study led to valuable insights in the hitherto hidden mechanisms within and among bat colonies.
Tapire wurden bislang im Gegensatz zu ihren Verwandten, den Nashörnern und Pferden bei Studien zur Kommunikation deutlich weniger beachtet. Ziel der vorliegenden Studie war es daher zu überprüfen, welche Reize Informationen für die Kommunikation bei Tapiren bergen. Zu diesem Zweck wurden die Reaktionen von Tapiren auf olfaktorische (Kotproben männlicher Tapire), akustische (Playback verschiedener Tierstimmen) und optische Reize (Plakate mit bearbeiteten Tapirsilhouetten) untersucht sowie das Pflegepersonal zur Wahrnehmung und Kommunikation bei Tapiren befragt. Die Forschungsaufenthalte fanden während der Jahre 2004, 2005 und 2006 in den Zoologischen Einrichtungen der Städte Berlin, Dortmund, Heidelberg, München, Nürnberg, Osnabrück und Mulhouse (Frankreich) statt. Insgesamt wurden 30 Individuen, davon 13 (8.5) Schabrackentapire (Tapirus indicus) und 17 (7.10) Flachlandtapire (Tapirus terrestris) in die Versuche einbezogen.
In this thesis we have revisited the formation of the excitonic insulator (EI), which realizes an exciton condensate. In contrast to optically created exciton condensates, the EI forms in thermal equilibrium and is solely driven by the Coulomb attraction between electrons and holes. The EI phase is anticipated to occur near the semimetal-semiconductor (SM-SC) transition at low temperatures. Depending from which side the EI is approached, it forms due to a BCS-type condensation of electron-hole pairs or a Bose-Einstein condensation (BEC) of excitons. The extended Falicov-Kimball model (EFKM) is the minimal model the EI can be described with. This model describes spinless fermions in two dispersive bands (f band and c band), that interact via a local Coulomb repulsion. The EFKM is also used to describe electronic ferroelectricity (EFE). Both phases, the EI and EFE-type ordering, are characterized by a spontaneous f-c hybridization in the EFKM. We have presented the EI phase, the EFE phase, and the orderings they compete with. Moreover, we have determined the ground-state phase diagram of the EFKM. We have focused particularly on the anticipated BCS-BEC crossover within the EI and have analyzed the formation scenarios. The exciton spectrum and the exciton density in the normal phase close to the critical temperature give information about relevant particles and therefore the nature of the transition. We have demonstrated that the whole EI is surrounded by a halo", that is, a phase composed of electrons, holes and excitons. However, on the SM side, only excitons with a finite momentum exist. These excitons appear only in a small number and barely influence the SM-EI transition. This phase transition is driven by critical electron-hole fluctuations, generated by electrons and holes at the Fermi surface. On the SC side, excitons with arbitrary momenta exist. Most notably, we have found the number of zero-momentum excitons to diverge at the SC-EI transition, signaling the BEC of these particles. Within the EI phase, there is a smooth crossover from the BCS regime to the BEC regime. One of the promising candidates to observe the EI experimentally, is the transition-metal dichalcogenide 1T-TiSe2. Strong evidences were found favoring an EI scenario of the charge-density-wave (CDW) formation in this material. However, some aspects point to a lattice instability to drive the CDW transition. We have addressed this issue by analyzing the recently discovered chiral property of the CDW in 1T-TiSe2. We have found that the EI scenario is insufficient to explain a stable, long range chiral charge ordering. Lattice degrees of freedom must be taken into account. In particular, nonlinear electron-phonon coupling and phonon-phonon interaction are crucial. By estimating appropriate model parameters for 1T-TiSe2, we have suggested a combination of excitonic and lattice instability to drive the CDW transition in this material. Experiments in 1T-TiSe2 and other materials suggest that the coupling to the lattice is non-negligible. We have extended therefore the model by an explicit exciton-phonon interaction, and have analyzed crucial effects of this interaction. While the single-particle spectrum is not modified qualitatively, the electron-hole pair spectrum changes significantly. The inclusion of the phonons lead to a massive collective mode in the ordered ground state in contrast to the case for vanishing exciton-phonon coupling, where the mode is acoustic. We have suggested that a gapless collective mode leads to off-diagonal long range order. This questions that the ground state for finite exciton-phonon coupling represents a condensate.
The region surrounding the excitonic insulator phase is a three-component plasma composed of electrons, holes, and excitons. Due to the extended nature of the excitons, their presence influences the surrounding electrons and holes. We analyze this correlation. To this end, we calculate the density of bound electrons, the density of electrons in the correlated state, the momentum-resolved exciton density, and the momentum-resolved density of electron-hole pairs that are correlated but unbound. We find qualitative differences in the electron-hole correlations between the weak-coupling and the strong-coupling regime.
Problemstellung Nach dem Einzug in ein Verwaltungsgebäude traten nach ca. 8 Wochen in rekonstruierten Räumen sowie in Räumen der neugebauten Gebäudeteile Geruchsbelästigungen auf. Die Angestellten klagten über Befindlichkeitsstörungen, insbesondere über Schleimhautreizungen, Hautrötungen, Kopfschmerzen und Atembeschwerden. Auf Grund dieser Beschwerden wurden Raumluftmessungen veranlasst. Die Bewertung der Belastungen mit flüchtigen organischen Verbindungen (VOC's) in der Raumluft bereitet Schwierigkeiten, da es sich hier um eine Vielzahl chemisch zum Teil unterschiedlicher Verbindungen handelt und bis heute hinsichtlich der Belastung mit VOC's verschiedene Empfehlungen herausgegeben wurden, aber keine rechtsverbindlichen Grenzwerte existieren. Für die Beurteilung der Meßergebnisse wurde das Richtwertkonzept der ad-hoc-Arbeitsgruppe der Innenraumlufthygiene-Kommission des UBA (IRK) und der Arbeitsgemeinschaft oberer Landesgesundheitsbehörde (AOLG) zu Grunde gelegt. Methodik Zur Klärung der Frage, ob zwischen der Raumluftbelastung in den Arbeitsräumen des Verwaltungsgebäudes und den bei den Angestellten aufgetretenen Befindlichkeitsstörungen und gesundheitlichen Beschwerden ein Zusammenhang besteht, wurden standardisierte Befragungen mittels eines Fragebogens der Raumnutzer und im Vergleich mit einer Kontrollgruppe durchgeführt. Um die Inimissionssituation in den Arbeitsräumen beurteilen zu können, wurden qualitative und quantitative Messungen der Innenraumluft vorgenommen. Ergebnisse 2 Monate nach Bezug des Gebäudes wurden VOC-Gesamtkonzentrationen von 1700 nachgewiesen, nach 42 Monaten war die Belastung durch verschiedene Sanierungsmaßnahmen und intensives Lüften auf ca. 700 \i/m3 zurückgegangen. Bei der Betrachtung der Raumluftergebnisse konnte festgestellt werden, dass die Emission der Schadstoffe in dem Zeitraum von 3,5 Jahren nur langsam abnahm und sich das Schadstoffprofil (Aldehyderhöhung) zwischenzeitlich veränderte. Die Ergebnisse der umweltmedizinischen Befragungen korrellieren mit den Messergebnissen der umwelthygienischen Untersuchungen. Schlußfolgerungen Aufgrund der anhaltenden gesundheitlichen Beschwerden, die sich in Einzelbefunden und bei den Fragebogenaktionen zeigten, ist es erforderlich, durch geeignete Maßnahmen die Innenraumluft zu verbessern und den Gesundheitszustand der Betroffenen weiter zu kontrollieren. Insbesondere sind weiterführende Untersuchungen der Personen auf allergische Reaktionen und Entwicklung eines Belastungsasthmas notwendig. Für eine erfolgreiche Sanierung muss eine gezielte Materialauswahl und eine Bewertung deren Prüfzertifikate und - berichte erfolgen.
For many years, rangeland ecologists have debated about whether the state of semi-arid and arid rangelands is the expression of an ecological equilibrium or non-equilibrium dynamics reached in response to grazing livestock. Since the problem has been considered at different spatial scales, it is recognised that the competing concepts of equilibrium and non-equilibrium dynamics need to be integrated. Furthermore, the role of environmental variables as vegetation driving factors has long been ignored in the discussion on grazing effects on ecosystems. Present thesis, examines the dependence of plant communities on environmental in particular site-ecological conditions in three ecosystems of Western Mongolia established along a precipitation gradient to detect the vegetation-driving ecological factors involved. Furthermore, grazing impact is exemplary assessed in a desert steppe at additional spatial scales of plant communities and population. At the landscape level, a classification of plant communities in dependence on environmental conditions is carried out. Additionally, the investigations focused on the impact of grazing on soil and on the occurrence of grazing-mediated plant communities. Data were sampled along an altitudinal gradient between 1150 m to 3050 m a.s.l. from arid lowland with desert steppe via semi-arid mountain steppe to humid alpine belt. Within each altitudinal belt, data sampling was carried out along grazing gradients, established from grazing hot spots to areas distant from them. By means of an environmentally based vegetation classification, factors with highest explanation values for largest variation in vegetation were identified and considered as most responsible for vegetation patterns. To validate and affirm the classification, three different statistical methods are applied: environmentally adjusted table work of vegetation relevés supported by cluster analysis of species distribution, detrended correspondence analysis of vegetation data separately from environmental data, and the principle component analysis of only environmental data. Vegetation-driving factors change along the altitudinal gradient from abiotic forces in the desert steppe, as e.g. altitude and soil texture, to abiotic and biotic forces in the alpine belt represented by soil texture, soil nutrients and grazing. Vegetation and soil of all ecosystems respond to grazing but with different patterns and to a different extent. While desert steppe does not indicate grazing communities, mountain steppe demonstrates grazing communities at fertilised sites and alpine belt at nutrients depleted sites. Thus, the grazing sensitiveness of the ecosystems is assumed to be linked with plant productivity and the role of vegetation as site-determining factor (Chapter 2). To examine grazing impact at lower spatial scales on desert steppe as the ecosystem with lowest grazing sensitiveness at the landscape scale, at community scale the total number of species, the total vegetation cover, the percentage of annual species, the cover of annual species, and properties of soil nutrient along gradients of grazing intensity within three different communities were assessed. Vegetation parameters respond to grazing in different ways, and the responses of the same parameters vary between plant communities. Correlations with grazing intensity indicate only partly statistical significance. Significant correlations of grazing intensity with concentrations of soil nutrient point to eutrophication in two communities. A comparison of vegetation and soil properties refers to a greater indirect influence of grazing via increased soil nutrients than the direct effect on vegetation (Chapter 4). At the population level, data about stand density, aboveground biomass, individual plant weight, and the proportion of flowering plants of the dominant dwarf semi-shrub Artemisia xerophytica were collected along a grazing gradient. Soil data were used to distinguish between grazing and edaphic influences. All parameters of Artemisia xerophytica reflect the assumed gradient of grazing intensity up to 800 m distance from the grazing hot spot. As grazing pressure decreases, plant density and total biomass per plot increase. The average shrub weight, an indicator of plant vitality, is related to both: distance from the grazing hot spot and stand density, which may be explained by additional intraspecific competition at higher densities. At a longer distance, these effects are masked by variations in soil parameters determining water availability, leading to quite similar degradation forms. These results are in contrast to other studies carried out at the scale of plant communities which did not detect significant changes along a grazing gradient. One explanation is the different map scale: the study took place only within a single plant community comparing populations of one species (Chapter 3). The comparative study demonstrates that even arid desert steppes of western Mongolia display equilibrial and non-equilibrial properties, depending on the observational scale: while no grazing mediated plant communities could be identified at the landscape scale as predicted by the non-equlilibrium model, at the community level vegetation parameters imply an intermediate position between equilibrium and non-equilibrium system. At the population level, the results clearly reflect the grazing gradient as predicted by the equilibrium model (Chapter 4). As a consequence, the assessment of vegetation dynamics and grazing impact in rangelands requires a multiple-scale approach that duly considers different vegetation properties responding differently to grazing, climatic and edaphic variability at different spatial scales. It is further suggested, that future research should draw comparisons between landscapes that co-evolved with herbivory, and those that did without (Chapter 4).
Die vorliegende Studie basiert auf der Untersuchung von 89 verschiedenen Säugetierschädeln und 14 menschlichen Schädeln aus dem Institut für Anatomie und Zellbiologie der Universität Greifswald.
Die Arbeit vergleicht die Morphologie der knöchernen Schädel der Säugetiere mit denen des Menschen. Dabei wird von kieferorthopädischen Messpunkten, wie sie beim Menschen bereits seit längerer Zeit angewendet werden, ausgegangen. Sämtliche Messpunkte, die in der Kieferorthopädie herangezogen werden, finden sich auch an den Säugetierschädeln, sodass von einem gemeinsamen Schädelbauplan ausgegangen werden kann. Systematische Untersuchungen an einer größeren Gruppe von Säugetieren mit kieferorthopädischem Ansatz wurden, soweit ersichtlich, bisher noch nicht durchgeführt. Ziel dieser Arbeit ist es damit auch, eine dementsprechende Datengrundlage zu schaffen. Die dreidimensionale Vermessung erfolgte mit dem MicroScribe 3DX Digitalisierer (Immersion Corp., San Jose, CA) sowie einem digitalen Messschieber. Die Schädel wurden entsprechend der in der Literatur angewandten Taxonomie in Gruppen eingeteilt. Aus der Ordnung der Primaten wurden die Hominoidea, Platyrrhini, Cercopithecoidea und Lemuriformes untersucht, bei den Carnivora die Feliformia, Canidae, Ursidae und Pinnipedia. Bei den Cetartiodactyla wurden Ruminantia und Suidae vermessen, bei den Mesaxonia die Equidae.
In einem ersten Teil der Arbeit kommt die klassische Morphometrie zu Anwendung. Dabei werden klassische KFO- Messpunkte wie Nasion, Menton, Gonion, Pogonion, Zygion, Spina nasialis antetior, etc. verwendet, ebenso klassische Indizes wie Bolton-Analyse, Pont- Index und Izard- Index. In einem zweiten Teil kommt die geometrische Morphometrie zur Anwendung. Diese in der Biologie und Anthropologie bereits häufiger angewandte Methode wird jetzt auch vermehrt in der Kieferorthopädie angewandt. Durch die sogenannte Procrustes Transformation können dabei die vermessenen Schädel in Form und Gestalt unabhängig von der Größe verglichen werden. Bei sämtlichen Messungen werden die Unterschiede bzw. Gemeinsamkeiten zwischen Mensch und den einzelnen Säugetierarten herausgearbeitet und tabellarisch und graphisch dargestellt.
So können die beim Menschen nachgewiesenen kieferorthopädischen Indizes auch teilweise bei den Säugetieren gefunden werden. Sowohl beim Izard- Index als auch bei der Bolton- Analyse und der Tonn- Relation können Gemeinsamkeiten festgestellt werden. Größere Abweichungen gibt es dagegen beim Pont- Index, dem Gaumenhöhen- Index und dem Gaumen- Index. Auch der Jugomandibularindex zeigt wenig Übereinstimmung mit dem Menschen.
Dass die Primaten und hier insbesondere die Hominoidea und Cercopithecoidea dem Menschen schädelbezüglich am ähnlichsten sind, war zu erwarten und kann durch die hier vorliegenden Ergebnisse auch bestätigt werden. Allerdings weichen die Primaten bei der Gaumenform stärker vom Menschen ab, während bei der Gesichts- und Kieferform eine weniger zu erwartende Übereinstimmung mit Feliformia und Canidae festgestellt werden kann.
Das evolutionsbedingt stärkste Unterscheidungsmerkmal zu den Säugetieren ist der beim Menschen im Vergleich zur Schädellänge relativ kurze Gesichtsschädel.
Die in der Literatur beschriebene Taxonomie der Säugetiere, die durch eine Vielzahl verschiedener Untersuchungen hervorgegangen ist, kann hier sowohl mit Hilfe der klassischen Morphometrie als auch insbesondere durch die geometrische Morphometrie bestätigt werden. Bei der letzteren wird nach der Procrustes Transformation durch Clusterbildung sowohl nach deutlich voneinander abweichenden, als auch nach in sich homogenen Gruppen differenziert.
Durch die in dieser Studie differenziert herausgearbeiteten anatomische Strukturen in verschiedenen Schädelbereichen würde sich zusätzlich die Möglichkeit ergeben, bei kieferorthopädischen Tierversuchen in kraniofazialen Schädelregionen das anatomisch geeignete Tiermodell zu bestimmen, d.h. nicht eine einzelne Tierart ist für alle Versuche geeignet, sondern je nach Fragestellung müssen in unterschiedlichen Schädelbereichen verschiedene Tierarten herangezogen werden.
A slice is an intersection of a hyperplane and a self-similar set. The main purpose of this work is the mathematical description of slices. A suitable tool to describe slices are branching dynamical systems. Such systems are a generalisation of ordinary discrete dynamical systems for multivalued maps. Simple examples are systems arising from Bernoulli convolutions and beta-representations. The connection between orbits of branching dynamical systems and slices is demsonstrated and conditions are derived under which the geometry of a slice can be computed. A number of interesting 2-d and 3-d slices through 3-d and 4-d fractals is discussed.
This study deals with attitudes and opinions of the population on the Ukrainian Black Sea coast in 2020/2021, a good year before the Russian invasion of Ukraine. The basis is a sociological survey conducted in the regions of Odesa, Mykolajiv and Kherson with 1,200 respondents. These regions are a clear target of the Russian offensive against Ukraine, on the grounds of an alleged ideological and linguistic affinity of these areas with Russia. The attitudes and opinions sought in the study cover three domains: firstly, attitudes and opinions on language policy, secondly, on Ukrainian autonomy and identity and the role played by languages in this context, and thirdly, Ukraine’s geopolitical orientation. Respondents were grouped according to their “ethnic/national” self-identification and language preferences, and a comparison was made. Overall, the attitudes of the respondents showed a clear identification with Ukraine and the Ukrainian language and a skepticism towards the Russian state, but by no means a hostile attitude towards the Russian language. A picture emerges that clearly contradicts Russian propaganda and also puts into perspective the difference between southern Ukraine and the center and the west, which has often been emphasized in the past.
Ziel der vorliegenden Arbeit war es, durch den Vergleich von zwei unterschiedlichen Systemen in der Kariesentfernung, die möglichen Vor- und Nachteile zwischen diesen Systemen zu ermitteln. Verglichen wurden die konventionellen Hartmetallrosenbohrer mit neuentwickelten kunststoffbeschichteten SmartPrep™ Bohrern. Als Vergleichsparameter sollten die verbliebene Restkaries nach der Exkavation (Schliff- und Zahnebene), die benötigte Zeit des kompletten Exkavationsvorgangs sowie eine Abschätzung der wirtschaftlichen Nutzung dienen. Für die Untersuchung wurden 30 extrahierte, permanente menschliche Zähne mit Dentinkaries und möglichst gleichen Defekten verwendet. Diese wurden in zwei Gruppen mit jeweils 15 Zähnen aufgeteilt. Zur Optimierung und Schaffung möglichst gleicher Bedingungen, wurde unbedingt darauf geachtet, dass auch die Kariesentfernung mit den konventionellen Rosenbohrern nach der Gebrauchsanweisung der SmartPrep™ Instrumente erfolgte. Im Gegensatz zur klassischen Vorgehensweise begann die Kariesexkavation im Zentrum der Kavitäten. Ein zügiges überkappen und verschließen bei Pulpenexposition wäre hier nicht möglich gewesen. Nach der Kariesentfernung wurde bei beiden Systemen die Behandlung bei ausreichender Sondierungshärte beendet. Im Anschluss daran wurden die Zähne eingebettet und Dünnschliffpräparate von 400 mm Dicke angefertigt. Diese Präparate wurden dann mit einem Kariesdedektor angefärbt und 60 Sekunden später mit Wasser abgespült. Mit Hilfe des Auflichtmikroskops wurden die Präparate mit einer Videokamera als Standbilder in das PC-Programm AnalySIS übertragen. Danach wurden die angefärbten Flächen und die Längen der behandelten Kavitätenränder beider Versuchsgruppen vermessen und miteinander ins Verhältnis gesetzt. Daraus konnte die Tiefe der verbliebenen Restkaries ermittelt werden. Bewertet wurden die maximale und die durchschnittliche Länge, Fläche und Tiefe der Restdentinkaries auf Schliff- und auf Zahnebene. Alle ermittelten Ergebnisse zeigten keine signifikanten Unterschiede zwischen beiden Systemen. Aus wirtschaftlicher Sicht ist die Verwendung von Hartmetallrosenbohrern primär günstiger. Das schonendere Verfahren in Bezug auf eine mögliche Überexkavation lag aber im SmartPrep™ System.
We present classical and hybrid modeling approaches for genetic regulatory networks focusing on promoter analysis for negatively and positively autoregulated networks. The main aim of this thesis is to introduce an alternative mathematical approach to model gene regulatory networks based on piecewise deterministic Markov processes (PDMP). During somitogenesis, a process describing the early segmentation in vertebrates, molecular oscillators play a crucial role as part of a segmentation clock. In mice, these oscillators are called Hes1 and Hes7 and are commonly modeled by a system of two delay differential equations including a Hill function, which describes gene repression by their own gene products. The Hill coefficient, which is a measure of nonlinearity of the binding processes in the promoter, is assumed to be equal to two, based on the fact that Hes1 and Hes7 form dimers.However, by standard arguments applied to binding analysis, we show that a higher Hill coefficient is reasonable. This leads to results different from those in literature which requires a more sophisticated model. For the Hes7 oscillator we present a system of ordinary differential equations including a Michaelis-Menten term describing a nonlinear degradation of the proteins by the ubiquitinpathway. As demonstrated by the Hes1 and Hes7 oscillator, promoter behavior can have strong influence on the dynamical behavior of genetic networks. Since purely deterministic systems cannot reveal phenomenons caused by the inherent random fluctuations, we propose a novel approach based on PDMPs. Such models allow to model binding processes of transcription factors to binding sites in a promoter as random processes, where all other processes like synthesis, degradation or dimerization of the gene products are modeled in deterministic manner. We present and discuss a simulation algorithm for PDMPs and apply it to three types of genetic networks: an unregulated gene, a toggle switch, and a positively autoregulated network. The different regulation characteristics are analyzed and compared by numerical means. Furthermore, we determine analytical solutions of the stationary distributions of one negatively, and three positively autoregulated networks. Based on these results, we analyze attenuation of noise in a negative feedback loop, and the question of graded or binary response in autocatalytic networks.
The six extraocular muscles (EOMs) are arranged around the eyeball as agonist-antagonist pairs performing the eye movements. The EOMs comprise a distinct muscle group that is fundamentally different from other skeletal muscle, which is reflected on many levels, such as functionality, anatomy as well as in their molecular make-up. Physiologically EOMs are considered superfast, high endurance muscles that are continuously active. In addition, EOMs contain unusual slow-tonic fibers that share features with amphibian and avian slow-tonic fibers. EOMs also express slow/cardiac isoforms of proteins and genes along with the typical isoforms of fast muscle fibers. Another striking hallmark of EOM is their differential involvement in a number of diseases. For instance, EOMs are preferentially spared in Duchenne Muscular Dystrophy (DMD). DMD is the most common fatal, genetic disease in males clinically characterized by progressive muscle wasting. Mutations in the dystrophin gene result in a destabilization of the muscle membrane causing muscle fiber damage. While all other skeletal muscles deteriorate the EOMs remain morphologically and functionally healthy. In the pathogenesis of DMD elevated Ca2+ levels are believed to be an early event and it has been shown that EOMs are protected from pharmacologically induced Ca2+ damage. The goal of this study was to characterize the spared EOMs, in particular their Ca2+ homeostasis, in the context of DMD pathology to reveal new potential therapeutic targets for the disease. A combination of physiological, molecular and biochemical methods was used to investigate the Ca2+ homeostasis of EOMs to demonstrate clear differences compared with the fast limb muscle tibialis anterior (TA). Ca2+ handling of stimulated cultured EOM myotubes suggested more efficient Ca2+ removal from the cytoplasm after induced Ca2+ influx compared with cultured myoblasts from TA. Subsequent mRNA and protein expression analyses of myoblasts and adult muscle tissue revealed high expression levels of many key Ca2+ regulating and buffering proteins in rodent EOMs compared with TA. Among these Ca2+ proteins were slow/cardiac proteins, which normally are not found in fast muscles. For instance, the sarcoplasmic Ca2+ ATPase SERCA2 was elevated along with its regulator phospholamban (PLN). Further, PLN was preferentially endogenously phosphorylated at Thr17 suggesting continuous activation of SERCA2 and possibly the fast isoform SERCA1, the main Ca2+ pumps responsible for removing Ca2+ from the cytoplasm after muscle contraction. Furthermore, Ca2+ buffers, such as calsequestrin (CASQ2) and parvalbumin (PARV) were elevated. These results suggest that EOMs are endowed with a unique and superior Ca2+ homeostasis that facilitates efficient Ca2+ buffering and removal from the cytoplasm. This is in agreement with their continuous and fast activation cycles, as well as with a potential protective mechanism in prevention of Ca2+ overload in DMD. The extreme activity patterns of EOM suggested that a high activity of store-operated Ca2+ entry (SOCE) plays a critical part to replenish Ca2+ for rapid and continuous cycles of contractions. To extend the data on general Ca2+ homeostasis and because of possible implications of store-operated Ca2+ influx and other Ca2+ influx pathways in DMD, the expression patterns of group 1 transient receptor potential (TRP) channels and the proteins Orai1 and STIM1 were studied. The TRP channels, TRPC1, TRPC6 and TRPV4 channel proteins in addition to STIM1 showed higher expression in EOM compared with TA. High TRPC1, TRPV4 and STIM1 levels could play a significant role in the high fatigue resistance, muscle differentiation and SOCE in EOM. In addition, tissue from the mdx mouse model of DMD was investigated. The only channels differentially expressed in mdx EOM compared with normal EOM were TRPM4 and TRPM7 (decreased in mdx EOM) and TRPV4 (increased in mdx EOM). Although, these changes in mdx EOM were of small magnitude, they could point toward subtle compensatory changes related to the disease process. In general, EOMs seem to be unaffected by the disease and inherently protected. In conclusion, the results in this thesis have improved the understanding of the Ca2+ homeostasis in EOMs and suggest that EOM may be better able to prevent prolonged elevation of cytoplasmic Ca2+ levels. These data may help to design new therapeutic approaches targeting Ca2+ handling proteins to ameliorate muscular dystrophy.
Das Fach Rechtsmedizin unterlag in den letzten Jahren gewissen Fehlentwicklungen. Ganze Institute zu schließen und die Versorgung der Gebiete mit rechtsmedizinischen Dienstleistungen aus der Ferne zu betreiben, ist als nicht tragbar einzustufen. Als theoretischer Hintergrund für diese Arbeit wurde sich der betriebswirtschaftlichen Krisentheorie bedient. Das Ziel dieser Arbeit bestand darin, ursächliche Faktoren für die derzeitige Entwicklung des Faches und des Institutes für Rechtsmedizin Greifswald aufzuzeigen. Die betriebswirtschaftliche Krise des Institutes für Rechtsmedizin Greifswald hat folgende Ursachen: Die abgegrenzten Versorgungsgebiete und damit die Festsetzung der Institute zu regionalen Monopolen, haben sich als nicht optimal herausgestellt. Aufgrund eines annähernd gleichen Qualitätsstandards müssen die Institute unabhängig von ihrer Versorgungsgröße eine festgelegte Anzahl an Personal und an Geräten vorhalten. Das führt dazu, dass die kleineren Institute aufgrund ihrer Versorgungsgebiete nur unwirtschaftlich arbeiten können. Private Anbieter im Dienstleistungsbereich der Forensischen Labore dringen in die monopolisierten Märkte der jeweiligen Standorte ein und bieten ihre Dienstleistungen kostengünstig an. Sie können durch die beschriebene Markterweiterung Economies of Scale erzielen. Ein weiterer Krisenfaktor ist in der Zusammensetzung der Kundenstruktur zu sehen. Bisher werden von der Rechtsmedizin in hohem Maß nur öffentliche Auftraggeber bedient. Der Markt mit Privatkunden kann als nahezu unerschlossen definiert werden. In einem nächsten Schritt wurde auf die Dienstleistungserstellung unter Berücksichtigung des Distanzreibungseffektes eingegangen. Die Laborbereiche haben sich als standortungebunden herausgestellt. Die Inanspruchnahme der Leistungen der Labore ist weitestgehend unabhängig vom Standort. Hingegen ist der Bereich der Forensischen Medizin als standortgebunden zu betrachten, da der Kunde in den Dienstleistungserstellungsprozess direkt eingebunden werden muss. Auf die standortgebundenen Dienstleistungen wirkt der Distanzreibungseffekt, da mit der Entfernung vom Institut die Inanspruchnahme der Dienstleistungen abnimmt. Als Empfehlung aus den Erkenntnissen des Distanzreibungseffektes ist zu sagen, dass zusätzliche Außenstellen gegründetwerden sollten, vor allem in Flächenländern wie Schleswig-Holstein, Mecklenburg- Vorpommern oder Brandenburg. In diesen Ländern stehen große Versorgungsräume einer geringen Anzahl an Einwohnern gegenüber. Aufgrund der großen Distanzen müssen die Rechtsmediziner direkt vor Ort erreichbar sein und ihre Dienstleistungen den verschiedenen Kundenkreisen anbieten. Hier sei der Aufbau von Opferambulanzen (körperliche Untersuchungen) und Besichtigungen von Leichen (Leichenschau) erwähnt. Die weiteren Punkte kritische Personalausstattung, Vergütungsstruktur, Leasingkosten und Cherry Picking sollen an dieser Stelle nur erwähnt werden. Die Krisensymptome äußern sich im Anstieg der Fixkosten, der Abnahme der Aufträge und einem negativen Betriebsergebnis der Forensischen Labore. Anhand der identifizierten Krisenursachen konnten im Anschluss auf Grundlage der generischen Wettbewerbsstrategien sogenannte Krisenbewältigungsstrategien entwickelt werden. Die wichtigsten Modelle bestehen in der gezielten Gründung von Außenstellen in rechtsmedizinisch unterversorgten Regionen, der Erlaubnis von Fusionen mit anderen Universitäts- bzw. Landesinstituten und der Gründung von strategischen Allianzen. Die verschiedenen Modelle bergen jeweils Vor- und Nachteile gegenüber dem Status Quo. Als Ergebnis ist jedoch festzuhalten, dass der Erhalt der universitären Institute Vorrang vor deren Schließung haben sollte. Nur so kann die Vielfalt in der Forschung und das hohe Niveau in der Lehre, der Facharztweiterbildung und der Dienstleistungserbringung der deutschen Rechtsmedizin erhalten bleiben. Es besteht jedoch der Bedarf, sich mit den vorgestellten betriebswirtschaftlichen Modellen auseinanderzusetzen, damit in Zukunft eine effiziente Dienstleistungserbringung unter Ausnutzung der eingesetzten Faktoren möglich ist. Aus diesem Grund sind die Nachteile, die sich durch die Modelle für die Rechtsmedizin gegenüber dem Status Quo im Einzelnen ergeben, zu prüfen.
In the present thesis, a systematic study of beam driven Alfvén eigenmodes in high-density and low-temperature plasmas of the W7-AS stellarator is performed. The device went out of operation in 2002 and the study is based on stored experimental data. Alfvén instabilities can roughly be divided into ideal MHD Alfvén eigenmodes and those existing due to kinetic effects. The spectrum of ideal MHD Alfvén waves in toroidal fusion devices consists of a continuum of stable waves that are strongly localized. Weakly damped, discrete eigenmodes can exist in gaps of the continuous spectrum which are formed by plasma inhomogeneities and the coupling of Alfvén continua. This allows an identification of ideal MHD Alfvén eigenmodes in terms of their frequency and mode numbers. Kinetic effects can modify this spectrum and cause additional types of eigenmodes, the kinetic Alfvén eigenmodes (KAE) and energetic particle modes (EPM). The goal of this thesis is twofold: (I) identification and description of fast particle driven Alfvén instabilities in W7-AS, and (II) study of energetic particle losses induced by Alfvén instabilities. The reconstruction of the ideal MHD plasma equilibrium for each discharge with sufficient accuracy is the very foundation of all subsequent steps. This is achieved, based on measured plasma parameter profiles that are further refined by validating them to the measurements of other, independent plasma diagnostics. The applied scheme is inspired by an approach of Integrated Data Analysis (IDA) to combine different diagnostic data and provide combined uncertainties. After mode number analysis and eigenmode identification, the theoretically expected, linear growth rate of the instability is calculated where possible, and the various contributions of the fast particle drive to the instability of the mode are identified. Alfvénic activity recorded by the Mirnov diagnostic is analyzed, which consists of a set of spatially distributed coils that measure magnetic fluctuations. On W7-AS, the probes are arranged in three poloidal arrays at different toroidal positions. The spacing between the probes is non-equidistant. In addition, the signals of one probe array are digitized with a different sample rate. These characteristics prohibit the straight-forward use of standard tools available for harmonic analysis. Instead, a new tool has been developed and thoroughly tested. It is a multi-dimensional extension of the Lomb periodogram, able to provide reliable time-resolved frequency and mode number spectra in the case of uneven datapoint spacing. Numerical studies of this periodogram show a good performance with respect to mode number resolution given the low number of available probes, and robustness against perturbations of the signal. Only two of the probe arrays can be used for the analysis of eigenmodes with frequencies >70 kHz, such that for high-frequency phenomena insufficient information about the mode numbers is available. A total of 133 different Alfvén eigenmodes is studied in discharges from different experimental campaigns. A restriction to discharges from various high-beta campaigns with neutral beam heating is required to allow for a realistic reconstruction of plasma equilibrium and velocity distribution functions of energetic particles. The discharges are characterized by high density, ne = 5 x 1019 m-3 to 2.5 x 1020 m-3 at relatively low temperatures of Te = Ti = 150 ... 600 eV. Alfvén eigenmodes often appear transiently in the startup phase of these discharges, where density and heating power are being ramped up. Occasionally, Alfvén eigenmodes are seen in the stationary, high-beta phase in the presence of considerable neutral beam heating. Most of the Alfvén eigenmodes are successfully classified as ideal MHD eigenmodes. 19 global, 47 toroidicity-induced and 8 ellipticity-induced Alfvén eigenmodes (GAEs, TAEs, and EAEs, respectively) are unambiguously identified by their mode numbers and frequencies. Excellent agreement between experimentally observed mode number spectra and theoretically calculated eigenmode structure is shown for a TAE example. Additional 13 events are found to have frequencies inside the EAE gap and could possibly be EAEs. Evidence for high-frequency Alfvén eigenmodes (mirror- and helicity-induced Alfvén eigenmodes) is seen, but can not be proven rigorously due to uncertain mode numbers and the complexity of the Alfvén continuum. The remaining 41 Alfvén eigenmodes can not be classified to be one of the above cases. Reasons are either high frequencies, mode numbers obscured by far-field effects, or mode numbers that could not be related to ideal MHD Alfvén eigenmodes. A selection of these shows indications of strong non-linear wave-particle interactions and are assumed to be EPMs. Kinetic Alfvén eigenmodes are not expected to exist in the experimental conditions that were studied. The radially resolved velocity distribution function is used to describe the parameter regimes in which the modes are observed in terms of the dimensionless parameters vb/vA (beam velocity normalized to the Alfvén velocity) and ßfast/ßth, where beta is the ratio of plasma pressure to magnetic pressure. The first parameter describes through which of the possible resonance velocities particles can interact with the eigenmode. A peculiarity of the fast particle dynamics in fusion devices is that they can resonantly interact with Alfvén eigenmodes through sideband resonances even if v < vA. The second parameter describes the energy content of the destabilizing fast particle population compared to the potentially stabilizing thermal plasma component. These parameters contain relevant information about the instability of an eigenmode and such diagrams are given for all observed modes. In addition to that, the expected linear growth rate of gap modes is calculated based on a theoretical model that extends the ideal MHD by a perturbative, drift-kinetic description of the energy exchange between waves and circulating particles, neglecting the effects of trapped particles. For the discharges under consideration the thermal electron speed is comparable to vA and the electrons provide a significant Landau damping contribution. Due to strong density gradients near the plasma boundary in most of the discharges, the thermal ions can provide a small drive via the spatial inhomogeneity which does not overcome the electron damping, however. The drive by spatial inhomogeneity of thermal ions requires a certain propagation direction of the mode and is equally stabilizing for opposite mode numbers. The fast particles also contribute to the growth rate via spatial inhomogeneity, velocity gradients and velocity anisotropy terms are negligible in W7-AS. Most of the observed GAE or EAE modes have negative mode numbers, which correspond to a propagation direction for which the spatial inhomogeneity of thermal and beam ions is predicted to be stabilizing. A fast particle drive of these modes is not confirmed, whereas the TAEs are found to be strongly destabilized by neutral beam injection. The distribution of plasma parameters for discharges showing TAEs in terms of the dimensionless stability parameters suggests an instability threshold that is qualitatively confirmed by an exploration of the parameter space with the theoretical model. Wave-induced, resonant losses of energetic ions scale linearly with the wave amplitude. To identify them, correlations between ion loss probe signals and wave amplitudes are searched, where correlation times in the order of the slowing-down time of energetic particles are expected. Significant correlations can be established only exceptionally for 3 of the identified ideal MHD Alfvén eigenmodes. Those Alfvén eigenmodes, however, which are assumed to be EPMs frequently show severe losses of energetic ions that are visible in the time traces of the plasma energy as well.
ABSTRACT
The Upper Pleistocene geoarchives in the south‐eastern Carpathian Basin are represented predominantly by loess–palaeosol records. In 2015, a 10 m sediment core composed of clay‐rich lacustrine sediments was recovered by vibracoring a dry lake basin located between the Vršac Mountains (Serbia) and the Banat Sands in the south‐eastern Carpathian Basin; a location relevant for placing regional archaeological results in a palaeoenvironmental context. Here, we present results from geoelectrical prospection and a lithostratigraphic interpretation of this sequence supported by a detailed granulometric study supplemented by ostracod analysis. An age model based on luminescence dating is discussed against sedimentological proxy data and its implication for palaeoenvironmental change. The cores show a stratigraphy of lighter ochre‐coloured and darker greyish sediment, related to the deposition of clay and silt trapped in an aquatic environment. Geophysical measurements show ~20 m thick lacustrine sediments. The grain‐size distributions including the variability in fine clay are indicative of a lacustrine environment. Fine particles were brought into the depositional environments by aquatic input and settled from suspension; also, direct dust input is constrained by grain‐size results. Riverine input and aeolian dust input interplayed at the locality.
Als ein hochwirksamer Mechanismus zur Generierung und Aufrechterhaltung einer lokalen Toleranz des adaptiven Immunsystems wurde die durch inflammatorische Stimuli, wie IFN-gamma, TNF-alpha oder Lipopolysaccharide, induzierbare Enzymaktivität der Indoleamin 2,3- Dioxygenase (IDO) erkannt, welche durch den Abbau der essentiellen Aminosäure Tryptophan die Bildung toxischer/immunregulativer Metabolite (Kynurenine) initiiert, und dadurch adaptive Immunreaktionen durch Bildung eines tolerogenen Milieus am Ort ihrer Expression wirkungsvoll unterdrückt. Das in der Sepsis beobachtete Versagen des Immunsystems wird als Immunparalyse bezeichnet. Darunter wird die anhaltende Hypoinflammation bei persistierender Infektion im septischen Syndrom verstanden. Der funktionelle Immunzelldefekt, durch den die septische Immunparalyse gekennzeichnet ist, scheint maßgeblich für die Mortalität in der hypoimmunen Phase des septischen Syndroms verantwortlich zu sein. Die zuvor genannten Beobachtungen führen zu der Frage, inwieweit eine Steigerung des durch IDO eingeleiteten Kynureninabbauweges im septischen Syndrom beobachtet werden kann, und ob ein Zusammenhang mit dem Schweregrad des Syndroms und damit mit der Pathophysiologie der Sepsis besteht. Die frühzeitigere und richtige Erkennung einer Sepsis bei gefährdeten Patienten würde einen bedeutenden Vorteil bei der erfolgreichen Behandlung gewähren. Ein besseres Verständnis der immunregulativen Mechanismen in der Sepsis könnte von entscheidender Relevanz für die Beeinflussung der mortalitätsdeterminierenden Immunparalyse und damit für die erfolgreiche Behandlung der Sepsis sein. Aus diesem Verständnis ließen sich immunmodulatorische / immunrekonstitutive Therapieansätze ableiten. Zur Bestimmung der Kynureninstoffwechselaktivität bei septischen Patienten wurde eine Methode zur simultanen Messung von Schlüsselmetaboliten dieses Stoffwechsels mittels quantitativer Tandem Massenspektrometrie etabliert. Diese ermöglichte die näherungsweise Bestimmung der IDO Aktivität sowie der Aktivität des durch sie eingeleiteten Stoffwechselweges aus humanen Plasmaproben. Im Rahmen einer klinischen Studie an 100 intensivtherapierten chirurgischen Patienten wurde der Verlauf der Plasmakonzentrationen der Schlüsselmetabolite während des Intensivaufenthaltes gemessen und mit intensivmedizinischen Routinelaborparametern, immunologischen Laborparametern sowie klinischen Parametern und Scores verglichen. In septischen Patienten ist der durch IDO eingeleitete Kynureninabbauweg deutlich aktiviert. Die Aktivierung des Abbauweges erfolgt sehr früh im septischen Syndrom und differenziert in dem untersuchten Patientengut septische von nicht septischen Patienten besser als Procalcitonin (PCT). In einer multivariaten Varianzanalyse konnte ein Zusammenhang besonders hoher Plasmakonzentrationen der Kynurenine mit dem Zustand des septischen Schocks sowie mit dem septischen Nierenversagen nachgewiesen werden. Die plasmatische Akkumulation der Kynurenine in der Niereninsuffizienz wurde im Rahmen einer weiteren Studie an Patienten in verschiedenen Stadien einer Niereninsuffizienz näher untersucht. Ein Anstieg der Plasmakonzentrationen der Kynurenine in Abhängigkeit von der Schwere der Niereninsuffizienz konnte in dieser Studie auch unabhängig von dem Vorliegen einer Sepsis gezeigt werden. Niedrige Tryptophanspiegel im Plasma von kritisch kranken und septischen Patienten sprechen für eine verminderte Verfügbarkeit von Tryptophan für abhängige Stoffwechselwege, wie die Synthese von Indoleaminen und als Substratquelle für den zellulären Energie- stoffwechsel. Die beobachteten Konzentrationen der Kynurenine erreichen Werte, welche in vitro und in vivo im Mausmodell durch eine zytotoxische Wirkung auf natürliche Killerzellen, B-Lymphozyten und insbesondere T-Lymphozyten als immunregulatorisch wirksam nachgewiesen wurden. Es kann somit festgehalten werden, dass bei septischen Patienten ein immunsuppressiver Mechanismus zu einem sehr frühen Zeitpunkt aktiviert wird und eine systemische Ausprägung erfährt, die sich experimentell als hoch wirksam erwiesen hat. Die Beobachtung der erhöhten Konzentrationen der Kynurenine und deren Korrelation mit Indikatoren chronischer Entzündungsprozesse hsCRP (High Sensitivity C Reactive Protein) und sTNFR-1 (Soluble Tumor Necrose Factor Receptor-1) bei Patienten mit chronischer Niereninsuffizienz stützen die These sowie neuere Erkenntnisse einer chronischen Inflammation als zugrundeliegenden Pathomechanismus des Nierenversagens. Die vorliegenden Ergebnisse liefern neue Erkenntnisse über einen immunsuppressiven Mechanismus, welcher in der Sepsis bereits zu einem sehr frühen Zeitpunkt des Syndroms aktiviert wird und eine mögliche Bedeutung für die Entstehung der septischen Immunparalyse sowie des akuten Nierenversagens im septischen Multiorganversagen haben könnte.
Genetic variants in α-actinin-2 (ACTN2) are associated with several forms of (cardio)myopathy. We previously reported a heterozygous missense (c.740C>T) ACTN2 gene variant, associated with hypertrophic cardiomyopathy, and characterized by an electro-mechanical phenotype in human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs). Here, we created with CRISPR/Cas9 genetic tools two heterozygous functional knock-out hiPSC lines with a second wild-type (ACTN2wt) and missense ACTN2 (ACTN2mut) allele, respectively. We evaluated their impact on cardiomyocyte structure and function, using a combination of different technologies, including immunofluorescence and live cell imaging, RNA-seq, and mass spectrometry. This study showed that ACTN2mut presents a higher percentage of multinucleation, protein aggregation, hypertrophy, myofibrillar disarray, and activation of both the ubiquitin-proteasome system and the autophagy-lysosomal pathway as compared to ACTN2wt in 2D-cultured hiPSC-CMs. Furthermore, the expression of ACTN2mut was associated with a marked reduction of sarcomere-associated protein levels in 2D-cultured hiPSC-CMs and force impairment in engineered heart tissues. In conclusion, our study highlights the activation of proteolytic systems in ACTN2mut hiPSC-CMs likely to cope with ACTN2 aggregation and therefore directs towards proteopathy as an additional cellular pathology caused by this ACTN2 variant, which may contribute to human ACTN2-associated cardiomyopathies.
Abstract
Ecosystems with highly pulsed water supply must be better understood as climate change may increase frequency and severity of intense storms, droughts and floods. Here we collected data over 3 years (2016–2018) in the episodic wetland outflow channel (Aluize), Banhine National Park, in which the system state changed from dry to wet to dry. Field sampling included vegetation records, small‐scale vegetation zoning, the seed bank and water and soil quality. The same main plant species were found in both dry and wet conditions across the riverbed of the outflow channel. We found only very few diaspores of plants in the soil after prolonged drought. In the subsequent flooded state, we examined very dense vegetation on the water surface, which was dominated by the gramineous species Paspalidium obtusifolium. This species formed a compact floating mat that was rooted to the riverbed. The Cyperaceae Bolboschoenus glaucus showed high clonal growth in the form of root tubers, which likely serve as important food reservoir during drought. Soil and water analyses do not indicate a limitation by nutrients. We outline how resident people may change the plant community structure with an increasing practice of setting fire to the meadows in the dried‐up riverbed to facilitate plant regrowth as food for their livestock.
MRP4 (multi drug resistance protein 4) ist ein Transporter aus der ATP-binding-cassette Familie (ABCC4) und verfügt über ein sehr breites Substratspektrum. Dieses beinhaltet Xenobiotika u.a. Cephalosporine, Diuretika und Zytostatika sowie endogene Substanzen wie Gallensäuren, Steroide, Eicosanoide und auch Nukleotide, wie den second messenger zyklisches AMP (cAMP). MRP4 wird in einer Vielzahl von Geweben exprimiert u.a. in der Niere, in der Leber, im Gehirn, in Blutzellen und in vaskulären glatten Muskelzellen. Über die Regulation von MRP4 ist weniger bekannt, daher war es das Ziel dieser Arbeit die transkriptionelle Regulation von MRP4 näher zu betrachten. Zur Bearbeitung dieser Zielstellung wurde die Promoter-Region des ABCC4/MRP4-Gens in ein Luciferase-Reporter-System kloniert. Anschließend wurde in Zellversuchen mit HeLa und HepG2-Zellen der Einfluss verschiedener Substanzen auf die Transkription von MRP4 untersucht. Zusätzlich wurde mittels Western Blot und quantitativer RealTime-PCR die Expression von MRP4 auf Protein- und mRNA-Ebene betrachtet. Keine bzw. nur geringe Effekte auf die Promoteraktivität wurden in den erwähnten Zelllinien u.a. nach Behandlung mit Glucocorticoiden, Zytokinen, cGMP, Nrf2-/AhR-Aktivatoren und unter dem Einfluss verschiedener Glucose-Konzentrationen beobachtet. Dagegen zeigte sich ein signifikanter Einfluss des zellulären cAMP-Spiegels. MRP4 vermittelt physiologisch einen Auswärtstransport von cAMP. Bei Inkubation mit dem membranpermeablen cAMP-Analogon Dibutyryl-cAMP zeigte sich eine signifikante Steigerung der Promoter-Aktivität von MRP4. Auch auf mRNA- und Protein-Ebene konnte eine signifikante Steigerung der Expression nach Behandlung mit cAMP detektiert werden. Durch Ko-Inkubation mit Inhibitoren der Proteinkinase A (PKA) und der Extracellular signal-regulated kinases 1/2 (Erk1/2) konnte weiterhin gezeigt werden, dass dieser Effekt nicht über die PKA, dem häufigsten Effektor von cAMP, sondern über Erk1/2 vermittelt wird. Die Induktion von MRP4 durch cAMP, welches selbst Substrat des Transporters ist, könnte eine Art Feedback-Mechanismus darstellen und darauf hinweisen, dass MRP4 eine größere Rolle im zellulären cAMP-Stoffwechsel zukommt, als bisher angenommen. Der zelluläre Export stellt neben dem Abbau von cAMP durch Phosphodiesterasen einen alternativen Mechanismus der Signalregulation dar, der entweder nur als Ventilmechanismus bei hohen cAMP-Spiegeln oder Zelltyp- und Kompartiment-abhängig möglicherweise auch als hauptsächlicher Eliminationsweg dient. MRP4 könnte somit neben der Inhibition der Phosphodiesterasen einen weiteren Angriffspunkt zur Erhöhung zellulärer cAMP-Spiegel z.B. im Rahmen der Therapie von Erkrankungen wie der pulmonalen Hypertonie darstellen.
Technological advances in light microscopy have always gone hand in hand with unprecedented biological insight. For microbiology, light microscopy even played a founding role in the conception of the entire discipline. The ability to observe pathogens that would otherwise evade human observation makes it a critical necessity and an indispensable tool to infectious disease research. Thus, the aim of this thesis was to optimize, extend, and functionally apply advanced light microscopy techniques to elucidate spatio-temporal and spatio-morphological components of bacterial and viral infection in vitro and in vivo.
Pathogens are in a constant arms race with the host’s immune system. By finding ways to circumvent host-mediated immune responses, they try to evade elimination and facilitate their own propagation. The first study (publication I) demonstrated that the obligate intracellular pathogen Coxiella burnetii is not just able to infect natural killer (NK) cells, but is actually capable of surviving the harsh degradative conditions in the cytotoxic lymphocyte’s granules. Using live-cell imaging of reporter-expressing Coxiella burnetii, the transient NK cell passage was closely monitored to provide detailed spatio-temporal information on this dynamic process in support of a range of static analyses. Bacterial release from NK cells was pinpointed to a time frame between 24 to 48 hours post-infection and the duration of release to about 15 minutes.
The second approach (publications II-V) aimed at shedding light on the greater spatio-morphological context of virus infection. Thus far, most studies investigating the distribution or tropism of viruses in vivo have used conventional immunohistochemistry in thin sections. Omitting the native spatial context of the infection site in vivo inherently bears the risk of incomplete description. While the microscopic tools and sample preparation protocols needed for volumetric 3D immunofluorescence imaging have recently been made available, they had not gained a foothold in virus research yet. An integral part of this thesis was concerned with the assessment and optimization of available tissue optical clearing protocols to develop an immunofluorescence-compatible 3D imaging pipeline for the investigation of virus infection inside its intact spatio-morphological environment (publication II). This formed the basis for all subsequent volumetric analyses of virus infection in vivo presented here. Consequently, this thesis provided a valuable proof of concept and blueprints for future virus research on the mesoscopic scale of host-pathogen interactions in vivo (publications II-V), using rabies virus (RABV; publications II-IV) and the newly-emerged severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2; publication V) as infection models for the nervous system and the respiratory tract, respectively.
Applying and further improving this volumetric 3D imaging workflow enabled unprecedented insights into the comprehensive in vivo cell tropism of RABV in the central (CNS) (publication III) and peripheral nervous system (PNS) (publication IV). Accordingly, differential infection of CNS-resident astrocytes by pathogenic and lab-attenuated RABV was demonstrated (publication III). While either virus variant showed equal capacity to infect neurons, as demonstrated by quantitative image analysis, only pathogenic field RABVs were able to establish non-abortive infection of astrocytes via the natural intramuscular inoculation route. A combined 3D LSFM-CLSM workflow further identified peripheral Schwann cells as a relevant target cell population of pathogenic RABV in the PNS (publication IV). This suggested that non-abortive infection of central and peripheral neuroglia by pathogenic RABV impairs their immunomodulatory function and thus represents a key step in RABV pathogenesis, which may contribute significantly to the establishment of lethal rabies disease.
Finally, utilizing the full volumetric acquisition power of LSFM, a further refined version of the established 3D imaging pipeline facilitated a detailed mesoscopic investigation of the distribution of SARS-CoV-2 in the respiratory tract of the ferret animal model (publication V). Particularly for this newly-emerged pathogen of global concern, in-depth knowledge of host-pathogen interactions is critical. By preserving the complete spatio-morphological context of virus infection in the ferret respiratory tract, this thesis provided the first specific 3D reconstruction of SARS-CoV-2 infection and the first report of 3D visualization of respiratory virus infection in nasal turbinates altogether. 3D object segmentation of SARS-CoV-2 infection in large tissue volumes identified and emphasized a distinct oligofocal infection pattern in the upper respiratory tract (URT) of ferrets. Furthermore, it corroborated a preferential replication of SARS-CoV-2 in the ferret URT, as only debris-associated virus antigen was detected in the lower respiratory tract of ferrets, thus providing crucial information on the spatial distribution of SARS-CoV-2.
The collisionless tearing mode is investigated by means of the delta-f PIC code EUTERPE solving the gyrokinetic equation. In this thesis the first simulations of electromagnetic non-ideal MHD modes in a slab geometry with EUTERPE are presented. Linear simulations are carried out in the cases of vanishing and finite temperature gradients. Both cases are benchmarked using a shooting method showing that EUTERPE simulates the linearly unstable tearing mode to a very high accuracy. In the case of finite diamagnetic effects and values of the linear stability parameter Delta of order unity analytic predictions of the linear dispersion relation are compared with simulation results. The comparison validates the analytic results in this parameter range. Nonlinear single-mode simulations are performed in the small- to medium-Delta range measuring the dependency of the saturated island half width on the equilibrium current width. The results are compared with an analytic prediction obtained with a kinetic electromagnetic model. In this thesis the first simulation results in the regime of fast nonlinear reconnection~(medium- to high-Delta range) are presented using the standard gyrokinetic equation. In this regime a nonlinear critical threshold has been found dividing the saturated mode from the super-exponential phase for medium-Delta values. This critical threshold has been proven to occur in two slab equilibria frequently used for reconnection scenarios. Either changing the width of the equilibrium current or the wave number of the most unstable mode makes the threshold apparent. Extensive parameter studies including the variation of the domain extensions as well as the equilibrium current width are dedicated to a comprehensive overview of the critical threshold in a wide range of parameters. Additionally, a second critical threshold for high-Delta equilibria has been observed. A detailed comparison between a compressible gyrofluid code and EUTERPE is carried out. The two models are compared with each other in the linear regime by measuring growth rates over wave numbers of the most unstable mode for two setups of parameters. Analytical scaling predictions of the dispersion relation relevant to the low-Delta regime are discussed. Employing nonlinear simulations of both codes the saturated island half width and oscillation frequency of the magnetic islands are compared in the small-Delta range. Both models agree very well in the limit of marginal instability and differ slightly with decreasing wave vector. Recently, the full polarisation response in the quasi-neutrality equation was implemented in EUTERPE using the Padé approximation of the full gyrokinetic polarisation term. Linear simulation results including finite ratios of ion to electron temperature are benchmarked with the dispersion relation obtained from a hybrid model. Finite temperature effects influence the saturated island width slightly with increasing ion to electron temperature ratio which has been verified by both models.
Abstract
Climate change is increasing the frequency and intensity of drought events in many boreal forests. Trees are sessile organisms with a long generation time, which makes them vulnerable to fast climate change and hinders fast adaptations. Therefore, it is important to know how forests cope with drought stress and to explore the genetic basis of these reactions. We investigated three natural populations of white spruce (Picea glauca) in Alaska, located at one drought‐limited and two cold‐limited treelines with a paired plot design of one forest and one treeline plot. We obtained individual increment cores from 458 trees and climate data to assess dendrophenotypes, in particular the growth reaction to drought stress. To explore the genetic basis of these dendrophenotypes, we genotyped the individual trees at 3000 single nucleotide polymorphisms in candidate genes and performed genotype–phenotype association analysis using linear mixed models and Bayesian sparse linear mixed models. Growth reaction to drought stress differed in contrasting treeline populations. Therefore, the populations are likely to be unevenly affected by climate change. We identified 40 genes associated with dendrophenotypic traits that differed among the treeline populations. Most genes were identified in the drought‐limited site, indicating comparatively strong selection pressure of drought‐tolerant phenotypes. Contrasting patterns of drought‐associated genes among sampled sites and in comparison to Canadian populations in a previous study suggest that drought adaptation acts on a local scale. Our results highlight genes that are associated with wood traits which in turn are critical for the establishment and persistence of future forests under climate change.
Forests influence the climate of our Earth and provide habitat and food for many species and resources for human use. These valuable ecosystems are threatened by fast environmental changes caused by human-induced climte change. Negative growth responses and higher tree mortality rates were associated with increasing physiological stress induced by global warming. Especially boreal forests at high latitudes in the arctic region are threatened, a region predicted to undergo the highest increase in temperature during the next decades. Therefore, it is important to assess the adaptation potential in trees. For this purpose, I studied natural populations of white spruce (Picea glauca (Moench) Voss) in Alaska. In this thesis, I present three scientific papers in which my co-authors and I studied the phenotypic plasticity and genetic basis of tree growth, wood anatomy and drought tolerance as well as the genetic structure of white spruce populations in contrasting environments. We established three sites representing two cold-limited treelines and one drought-limited treeline with a paired plot design including one plot located at the treeline and one plot located in a closed-canopy forest, respectively. Additionally, the study design included one forest plot as reference. Within the entire project, in total 3,000 trees were measured, genotyped and dendrochronological data was obtained. I used several approaches to estimate the neutral and adaptive genetic diversity and phenotypic plasticity of white spruce as a model organism to explore the adaptation potential of trees to climate change.
In the first chapter, I combined neutral genetic markers with dendrochronological and climatic data to investigate population structure and individual growth of white spruce. Several individual-based dendrochronological approaches were applied to test the influence of genetic similarity and microenvironment on growth performance. The white spruce populations of the different sites showed high gene flow and high genetic diversity within and low genetic differentiation among populations, rather explained by geographic distance. The individual growth performances showed a high plasticity rather influenced by microenvironment than genetic similarity.
In the second chapter, I investigated the populations of the drought and cold-limited treeline sites to decipher the underlying genetic structure of drought tolerance using different genotype-phenotype association analyses. Based on tree-ring series and climatic data, growth declines caused by drought stress were identified and the individual reaction to the drought stress event was determined. A subset of 458 trees was genotyped, using SNPs in candidate genes and associated with the individual drought response. Most of the associations were revealed by an approach which took into account small-effect size SNPs and their interactions. Populations of the contrasting treelines responded differently to drought stress events. Populations further showed divergent genetic structures associated with drought responsive traits, most of them in the drought-limited site, indicating divergent selection pressure.
In the third chapter, my co-authors and I studied xylem anatomical traits at one of the cold-limited treeline sites to investigate whether genetic or spatial grouping affected the anatomy and growth of white spruce. Annual growth and xylem anatomy were compared between spatial groups and between genetic groups and individuals. Overall, wood traits were rather influenced by spatial than genetic grouping. Genetic effects were only found in earlywood hydraulic diameter and latewood density. Environmental conditions indirectly influenced traits related to water transport.
In conclusion, white spruce showed a high genetic diversity within and a low genetic differentiation among populations influenced by high gene flow rates. Genetic differences among populations are rather caused by geographical distance and therefore genetic drift. Differing selection pressure at the treeline ecotones presumably lead to divergent genetic structures underlying drought-tolerant phenotypes among the populations. Thus, adaptation to drought most likely acts on a local scale and involves small frequency shifts in several interacting genes. The identified genes with adaptive growth traits can be used to further exlore local adaptation in white spruce. Tree growth and wood anatomical traits are rather influenced by the environment than genetics and showed a high phentoypic plasticity. The high genetic diverstiy and phenotypic plasticity of white spruce may help the species to cope with rapid environmental changes. Still, additional work is needed to further explore adaptation processes to estimate how tree species reacted to rapid climate change. The presented thesis shed some light on the adaptation potential of trees by the example of white spruce using several approaches.
Ziel der Arbeit war es zu untersuchen, in wie weit die Akustische Rhinometrie und die Rhinoresistometrie miteinander korrelieren. Durch die Strömungsbeobachtung verschiedener Nasenformen sollten neue Erkenntnisse zur Physiologie und Pathophysiologie der Nase gewonnen werden. Gleichzeitig sollten die funktionsdiagnostischen Befunde der ARM der RRM und die der Strömungsbilder verglichen werden. Fünf Probanden wurden vor und nach dem Abschwellen der Nasenschleimhaut mittels Akustischer Rhinometrie und der Rhinoresistometrie untersucht. Somit ergaben sich 7 verschiedene Nasenkonfigurationen. " Die Normale Nase, die Nase mit rundem Isthmus, Sattelnase, Nase mit SD konvexe und konkave Seite, sowie auch die konkave Seite mit kompensatorischen Muschelhyperplasie, und letztlich die Spalltseite einer Nase mit einseitigen LKGS. Für die Modellherstellung wurden präoperativ Silikonabdrücke aus dem Naseninneren der Probanden entnommen. Die 7 Nasenmodelle wurden rhinoresistometrisch, akustisch rhinometrisch sowie auch strömungsphysikalisch mittels Wasser untersucht. Durch den Vergleich der rhinoresistometrischen Befunde bei Modell und Proband zeigt sich beim Modell ein geringerer Atemwiderstand als beim Probanden. Gleichzeitig findet man beim Modell einen größeren Hydraulischen Durchmesser sowie einen späteren Turbulenzeinsatz als beim Probanden. Die Befunde der ARM der Modell zeigen im hinteren Bereich des Cavum nasi eine kleinere Querschnittsflächenzunahme im Vergleich zum Probanden. Zusammenfassung Die RRM mißt die Druck-Fluss-Beziehung in der Nase und ermöglicht genauere Aussagen über die Nasenfunktion. Weiterhin erfaßt sie die von der Luft durchströmten Anteile der Nase. Eventuell vorhandene Toträume werden hierbei aber nicht erfaßt. Die ARM mißt die Weite der gesamten Nasenhöhle und ermöglicht somit die Aussage zur Nasengeometrie. Im hinteren Nasenbereich ist sie mit großen Meßfehler behaftet. Engstellen im vorderen Bereich des Cavum nasi lassen sich gut lokalisieren und quantifizieren. Aus der Kombination mit der RRM ergeben sich Hinweise auf funktionelle Störungen der Nase. Schlußfolgerungen Beide Messmethoden ergänzen sich in ihrer diagnostischen Aussagekraft und gut miteinander korrelieren. Die Kombination beider Methoden ist deshalb empfehlenswert. Es lassen sich leider aufgrund der kleinen Fallzahl der untersuchten Modelle nur tendenzielle Aussagen über die Strömung bei den Nasendeformationen treffen.
Durch das CEREC®-Verfahren mit dem Intraoralscanner Omnicam (Fa. Dentsply Sirona, York, USA) ist es möglich, digitale Abformungen zu erstellen und anschließend digital Restaurationen zu konstruieren. Für die korrekte Herstellung von prothetischen Restaurationen ist die Okklusion essenziell. Diese wird während der digitalen Aufzeichnung durch den Bukkalscan registriert. Im Anschluss wird das digitale okklusale Kontaktpunktmuster sichtbar.
In dieser Studie wurde die Genauigkeit der digitalen Okklusionsbestimmung im Vergleich zu Bissregistraten und intraoralen Kontaktpunktmuster untersucht.
Getestet wurden dazu 30 Probanden einer Altersgruppe zwischen 18 - 30 Jahren. Es handelte sich um 15 Frauen und 15 Männer mit nahezu naturgesundem Gebiss. Es wurden bei allen Probanden zwei zeitlich versetzte Bukkalscans durchgeführt und die durch Artikulationspapier markierten Kontaktpunkte auf den Kauflächen fotografiert. Anschließend erfolge die Herstellung eines Bissregistrates aus A-Silikon. Es erfolgten zeitgleich Elektromyogrammmessungen, um Muskelaktivität während der unterschiedlichen Registrierungsmethoden festzustellen.
Es konnte festgestellt werden, dass beim Vergleich der digitalen Okklusion zum intraoralen Kontaktpunktbild über Artikulationspapier zu 6,69 % weniger Kontaktpunkte durch CEREC® errechnet wurden. Die Kontaktpunktanzahl von der digitalen Okklusion zu den Bissregistraten unterschied sich zu 39,45 % deutlich. Mit dem Korrelationskoeffizienten nach Pearson wurde belegt, dass eine große Korrelation von r = 0,79 zwischen dem ersten Bukkalscan und dem klinischen Kontaktpunktmuster besteht. Beim Vergleich zum Bissregistrat lag der Korrelationskoeffizient bei r = 0,59. Das Ergebnis der Studie war, dass die Übereinstimmung bei der Anzahl der digitalen zu den intraoralen Kontaktpunkten größer war als zur Kontaktpunktanzahl bei Bissregistraten.
Auch bei den Ergebnissen des Elektromyogramms ist ein Unterschied zu erkennen. Es konnte festgestellt werden, dass während der Herstellung der Bissregistrate eine wesentlich höhere Muskelaktivität aufgezeichnet wurde. So wurden 22,99 % der maximalen Muskelaktivität während der Herstellung des Bissregistrates aufgebracht, während die Aktivität beim Bukkalscan im Durchschnitt (vor und nach dem Scannen der Zahnreihen) bei 14,38 % lag. Generell konnte kein Zusammenhang zwischen Muskelaktivität und Kontaktpunktanzahl erkannt werden.
Diese Studie ergab, dass die digitale Okklusion (generiert durch die CEREC® Omnicam) im Vergleich zum klinischen Kontaktpunktmuster verlässlich und für die Herstellung von Restaurationen im digitalen Workflow geeignet ist. Dem Verwender der CEREC® Omnicam wird empfohlen, die Kontaktpunkte vor dem Scan mittels Artikulationspapier zu markieren. So kann die digitale Okklusion am Computer überprüft und auf die Restauration übertragen werden.
Objective: To compare the effectiveness and complications of intraligamentary anesthesia
(ILA) with conventional inferior alveolar nerve block (IANB) during injection and dental
treatment of mandibular posterior teeth.
Materials and Methods: In this randomized, prospective clinical trial, 72 patients (39 males, 33
females) patients scheduled for dental treatment of mandibular posterior teeth, were randomly
allocated to ILA group (n=35) received ILA injection or IANB group (n=37) received the
conventional IANB. Our primary outcome was to assess pain and stress (discomfort) during the
injection and dental treatment, using the Numeric Rating Scale (NRS) from 0 to 10 (0 = no
pain, 10= the worst pain imaginable). Whereas; recording 24 hours postoperative complications
were our Secondary outcomes.
Results: Patients in ILA group reported significantly less pain during injection when compared
with IANB group (p=0.03). While pain during dental treatment was similar in both groups
(p=0.2). Patients in both groups also reported similar low values of discomfort during treatment
(p= 0.7). Although no signs of nerve contact or any other postoperative complications were
observed, five patients in IANB group (none in ILA group) reported temporary irritations
Conclusion: This study showed equivalent effectiveness of both intraligamentary anesthesia
and conventional inferior alveolar nerve block, for pain control during routine dental treatment
of mandibular posterior teeth. Nevertheless, ILA showed significantly less pain during
injection. No major postoperative complications in both groups were observed.
Clinical Relevance: ILA could be considered as an effective alternative for routine dental
treatment.
Die hier vorgestellte Pilotstudie erfolgte in einem Zeitraum von 19 Monaten basierend auf Notarzteinsätzen im Raum Nienburg. Es wurden insgesamt 10 Patienten mit frisch aufgetretenen Schlaganfällen untersucht. Dafür wurde die Nahinfrarotspektroskopie als Methode verwendet. Das Ziel dieser Arbeit war es herauszufinden, wie sich die rSO2 bei Patienten mit akutem Schlaganfall verhält und ob es einen Zusammenhang zwischen dem arteriellen Blutdruck und der SpO2 mit der rSO2 gibt. Um die regionale zerebrale Sauerstoffsättigung zu dokumentieren, wurde das NIRS-Gerät verwendet. Zwei der untersuchten Fälle waren intrazerebrale Blutungen. Ein Fall wurde im Krankenhaus als epileptischer Anfall mit Schlaganfallsymptomatik dargestellt. Die übrigen Fälle waren akute ischämische Insulte. Es wird die Hypothese aufgestellt, dass durch die NIRS-Methode möglicherweise ein Insult schon in der Prähospitalphase erkannt werden kann. Von den zehn beobachteten Fällen zeigte die rSO2 -Messung in sechs Fällen (Patienten 1, 4, 6, 7, 8 und 9) niedrigere rSO2- Werte auf der aktuell betroffenen Hemisphäre an als die nicht betroffene Hemisphäre. In allen sechs Fällen war in der Anamnese kein alter Schlaganfall bekannt. Es handelte sich jeweils um ein Erstereignis. In einem Fall konnte eine massive Hirnblutung aufgrund einer Ruptur der A. basilaris diagnostiziert werden (Patient 5). Hier verliefen die beiden rSO2 - Kurven fast parallel. In drei Fällen (Patienten 2,3 und 10) waren die rSO2-Werte auf der betroffenen Hemisphäre aktuell niedriger als auf der nicht betroffenen Hemisphäre. In diesen drei Fällen hatten die Patienten in der Anamnese einen alten Schlaganfall. Es wird daher vermutet, dass es bei diesen Fällen nach dem Erstereignis zu einer ischämischen Präkonditionierung und zu Ausbildung von Gefäßkollateralen gekommen sein könnte. Ein direkter Zusammenhang zwischen dem arteriellen Blutdruck und der rSO2 konnte nur in einem Fall (Patient 5) dokumentiert werden. Diese Patientin musste intubiert werden. Der Blutdruck sank durch die Narkose. Es reduzierten sich auf beiden Hemisphären die rSO2- Werte. In den übrigen Fällen waren die Blutdruckwerte hypertensiv. Es konnte kein direkter Zusammenhang beobachtet werden. Vermutlich ist durch den Schlaganfall die Autoregulation außer Kraft gesetzt worden. Es wird angenommen, dass die Blutdruckwerte dennoch ausreichten, um einen ausreichenden CBF zu gewährleisten. Es ist aber davon auszugehen, dass bei sinkenden Blutdrücken auch die rSO2-Werte sinken würden wie bei Zusammenfassung 66 der 5. Patientin. Um präzisere Aussagen zu treffen, sind jedoch weitere Untersuchungen notwendig. Durch den Anstieg des SpO2-Wertes durch Sauerstoffgabe sind in fast allen Fällen (Patienten 1, 2, 3, 6, 7, 8 und 9) auch die rSO2-Werte angestiegen. Hier sind ebenfalls noch weitere Untersuchungen notwendig, um die Hypothese zu untermauern, dass ein Anstieg der SpO2 durch Sauerstoffgabe die rSO2 -Werte verbessert. Die NIRS-Methode eignet sich möglicherweise in Zukunft dafür, dass einerseits ein Schlaganfall schon prähospital im Rettungsdienst erkannt werden kann und anderseits durch die rSO2-Werte die zerebrale Blutversorgung beeinflusst werden kann. Die Ungenauigkeit der Messungen durch Artefakte könnte in Zukunft durch die Hybridtechnologie verbessert werden (Vgl. Winther Schytz, H., 2015). Durch die Regulierung des Blutdruckes anhand der rSO2-Werte und der SpO2 könnte einer weiteren Hirnschädigung bei akutem Schlaganfall schon im Rettungsdienst entgegengesteuert werden.
Proteasomes comprise a family of proteasomal complexes essential for maintaining protein homeostasis. Accordingly, proteasomes represent promising therapeutic targets in multiple human diseases. Several proteasome inhibitors are approved for treating hematological cancers. However, their side effects impede their efficacy and broader therapeutic applications. Therefore, understanding the biology of the different proteasome complexes present in the cell is crucial for developing tailor-made inhibitors against specific proteasome complexes. Here, we will discuss the structure, biology, and function of the alternative Proteasome Activator 200 (PA200), also known as PSME4, and summarize the current evidence for its dysregulation in different human diseases. We hereby aim to stimulate research on this enigmatic proteasome regulator that has the potential to serve as a therapeutic target in cancer.
Polyethylene terephthalate (PET) is a mass-produced petroleum-based non-biodegradable plastic that contributes to the global plastic pollution. Recently, biocatalytic degradation has emerged as a viable recycling approach for PET waste, especially with thermophilic polyester hydrolases such as a cutinase (LCC) isolated from a leaf-branch compost metagenome and its variants. To improve the enzymatic PET hydrolysis performance, we fused a chitin-binding domain (ChBD) from Chitinolyticbacter meiyuanensis SYBC-H1 to the C-terminus of the previously reported LCCICCG variant, demonstrating higher adsorption to PET substrates and, as a result, improved degradation performance by up to 19.6% compared to with its precursor enzyme without the binding module. For compare hydrolysis with different binding module, the catalytic activity of LCCICCG-ChBD, LCCICCG-CBM, LCCICCG-PBM and LCCICCG-HFB4 were further investigated with PET substrates of various crystallinity and it showed measurable activity on high crystalline PET with 40% crystallinity. These results indicated that fusing a polymer-binding module to LCCICCG is a promising method stimulating the enzymatic hydrolysis of PET.
The utilization of fluorescein-guided biopsies has recently been discussed to improve and expedite operative techniques in the detection of tumor-positive tissue, as well as to avoid making sampling errors. In this study, we aimed to report our experience with fluorescein-guided biopsies and elucidate distribution patterns in different histopathological diagnoses in order to develop strategies to increase the efficiency and accuracy of this technique. We report on 45 fluorescence-guided stereotactic biopsies in 44 patients (15 female, 29 male) at our institution from March 2016 to March 2021, including 25 frame-based stereotactic biopsies and 20 frameless image-guided biopsies using VarioGuide®. A total number of 347 biopsy samples with a median of 8 samples (range: 4–18) per patient were evaluated for intraoperative fluorescein uptake and correlated to definitive histopathology. The median age at surgery was 63 years (range: 18–87). Of the acquired specimens, 63% were fluorescein positive. Final histopathology included glioblastoma (n = 16), B-cell non-Hodgkin lymphoma (n = 10), astrocytoma, IDH-mutant WHO grade III (n = 6), astrocytoma, IDH-mutant WHO grade II (n = 1), oligodendroglioma, IDH-mutant and 1p/19q-codeleted WHO grade II (n = 2), reactive CNS tissue/inflammation (n = 4), post-transplantation lymphoproliferative disorder (PTLD; n = 2), ependymoma (n = 1), infection (toxoplasmosis; n = 1), multiple sclerosis (n = 1), and metastasis (n = 1). The sensitivity for high-grade gliomas was 85%, and the specificity was 70%. For contrast-enhancing lesions, the specificity of fluorescein was 84%. The number needed to sample for contrast-enhancing lesions was three, and the overall number needed to sample for final histopathological diagnosis was five. Interestingly, in the astrocytoma, IDH-mutant WHO grade III group, 22/46 (48%) demonstrated fluorescein uptake despite no evidence for gadolinium uptake, and 73% of these were tumor-positive. In our patient series, fluorescein-guided stereotactic biopsy increases the likelihood of definitive neuropathological diagnosis, and the number needed to sample can be reduced by 50% in contrast-enhancing lesions.
Ribozymes for Aminoacylation

Aminoacyl-tRNA synthetases (aaRS) are at the heart of modern translation, catalyzing the accurate biosynthesis of aminoacyl-tRNAs. According to the RNA world hypothesis, the early translation system should have aminoacylation ribozymes for RNA aminoacylation. For this, an aaRS ribozyme system, consisting of the KK13 ribozyme and the C3a ribozyme was successfully designed, which can perform both amino acid activation and aminoacyl transfer reaction. Generation of such aminoacylation ribozyme system would fill up the gap between the RNA world and the modern biological world. In addition, two types of diversified aminoacylation ribozymes, symmetrical ribozymes and self-assembling ribozymes were successfully developed, which may have great meaning in the origin of life.
Sturgeons are among the most ancient linages of actinopterygians. At present, many sturgeon species are critically endangered. Surrogate production could be used as an affordable and a time-efficient method for endangered sturgeons. Our study established a method for identifying and isolating type A spermatogonia from different developmental stages of testes using flow cytometric cell sorting (FCM). Flow cytometric analysis of a whole testicular cell suspension showed several well-distinguished cell populations formed according to different values of light scatter parameters. FCM of these different cell populations was performed directly on glass slides for further immunocytochemistry to identify germ cells. Results showed that the cell population in gate P1 on a flow cytometry plot (with high forward scatter and high side scatter parameter values) contains the highest amount of type A spermatogonia. The sorted cell populations were characterized by expression profiles of 10 germ cell specific genes. The result confirmed that setting up for the P1 gate could precisely sort type A spermatogonia in all tested testicular developmental stages. The P2 gate, which was with lower forward scatter and side scatter values mostly, contained type B spermatogonia at a later maturing stage. Moreover, expressions of plzf, dnd, boule, and kitr were significantly higher in type A spermatogonia than in later developed germ cells. In addition, plzf was firstly found as a reliable marker to identify type A spermatogonia, which filled the gap of identification of spermatogonial stem cells in sterlet. It is expected to increase the efficiency of germ stem cell culture and transplantation with plzf identification. Our study thus first addressed a phenotypic characterization of a pure type A spermatogonia population in sterlet. FCM strategy can improve the production of sturgeons with surrogate broodstock and further the analysis of the cellular and molecular mechanisms of sturgeon germ cell development.
The glioblastoma multiforme (GBM) not only presents the most common tumor of the central nervous system in adults, it is also the most aggressive brain tumor. Although patients suffering from GBM standardly receive a combination of multiple treatments including surgery, radiotherapy and chemotherapy, its prognosis is still poor with a median survival time of only 12-15 months. Therefore, new and effective treatment methods are urgently needed.
A signaling molecule which is both involved in proliferation, migration and invasion of a broad range of healthy and malignant cells is the lipid mediator sphingosine-1-phosphate (S1P). Previous studies have confirmed that sphingosine-1-phosphate (S1P) receptor 1 (S1PR1) is involved in the regulation of proliferation, invasion, metastasis, vascular maturation and angiogenesis of GBM cells, and is closely related to the occurrence and development of tumors. Thus, ACT-209905 (provided by Actelion Pharmaceuticals) as a selective S1PR1 modulator was applied to gain insights into the molecular processes activated by S1PR1 in GBM cells using two human (LN18, U87MG) and one murine (GL261) GBM cell line.
In our in vitro cell viability analyses, we found that ACT-209905 significantly reduced viability of LN18 cells in a concentration dependent manner. A combined administration of ACT-209905 with S1PR2 inhibitors (Compound 16, Compound 16ME – both provided by ONO Pharmaceuticals, and JTE-013 – commercially available) showed a stronger effect than the single administration demonstrating that both S1PR1 and S1PR2 are involved in growth of GBM cells and may interact with each other. Our results also demonstrated that ACT-209905 can induce apoptosis in GBM cells since caspase 3 activity was induced by the S1PR1 modulator which might therefore play an important role in inhibiting the proliferation of GBM cells. Further, we found a significant inhibitory effect of ACT-209905 on the migration and invasion of LN18 and U87MG GBM cells arguing for a participation of S1PR1 signaling in migration and invasion of GBM cells, too. Stimulation of S1P receptors results in the activation of several kinases such as AKT1 and ERK1/2, correspondingly our immunoblot analyses showed a strong activation of both kinases by S1P which was reduced by ACT-209905 in LN18 cells but not in GL261 cells suggesting that different pathways are activated by S1P in these GBM cell lines. Further studies have to be performed to clarify the role of AKT1 and ERK1/2 in the inhibitory effects of ACT-209905 on GBM proliferation, migration and invasion.
Currently, GBM stem cells are discussed as a reason for resistance against the radiochemotherapy and the recurrence of the tumor. Our immunoblot analyses showed that Nestin and CD133, two marker proteins for GBM stem cells, were higher expressed in GBM cells treated with ACT-209905 compared to control or S1P treated LN18 cells. Further investigations in the future might contribute to the elucidation of an involvement of the S1P receptors in the stem cell behavior of GBM cells. Paradoxically to the up-regulation of CD133 and Nestin by ACT-209905, treatment of LN18 stem-like neurospheres with ACT-209905 showed a significant cytotoxic effect of the compound which was even more pronounced in the stem-like neurosphere cells compared to the adherent parental LN18 cells.
Overall, the studies of this work improve our understanding of the complex mechanisms of S1P signaling in GBM cells and might drive the development of its pharmacological modulation as a new therapeutic principle in GBM. Furthermore, an extended knowledge about the molecular effects of ACT-209905 on GBM cells will broaden the understanding for possible future applications and clinical indications.
With the aim to discover and create suitable biocatalysts for the synthesis of chiral amines in a faster and more efficient way, this thesis includes protein engineering studies (Article I), explores transaminase substrate specificities (Articles II and IV), and an ultrahigh-throughput growth system-based for the directed evolution of amine-forming enzymes (Article III).
The protein engineering studies described in Article I deal with the creation of a (R)-amine transaminase activity in the α-amino acid transaminase scaffold to expand our knowledge of the evolutionary relationship between amine transaminase and α-amino acid transaminase. Article II describes the broadening of the limited substrate scope of transaminases to enable the conversion of bulky substrates. In Article III, a growth selection system is described for an ultra-high throughput screening strategy to accelerate the identification of desired mutants, which can be widely applied to the directed evolution of amine-forming enzymes.
Global climate change is occurring all over the world, but in the Arctic the climate is changing more rapidly and drastically than in many other parts of our planet. Many species that are already at their climatic limit need to adapt to recent climate conditions or migrate in order to not go extinct. The possibilities of adaption include phenotypic plasticity and adaptation to various extents. This is also the case for white spruce P. glauca, which belongs to the conifers and thus in the largest group of gymnosperms still living today. Among the approx. 600 extant conifer species white spruce is one of the most widespread trees in North American boreal forests. Its range extends from 69° N in the Canadian Northwest Territories to the Great Lakes at about 44° N, where it occurs from sea level to an altitude of about 1520 m (Burns and Honkala, 1990). Site related, climate-dependent differences in white spruce reproduction can be seen as a strategy to survive under the harsh climatic conditions at Alaska's treelines: Besides sexual reproduction, the vegetative propagation occurs in the white spruce as an additional reproductive mechanism. This can be realized by "layering" when the lower branches of the tree crown touch the ground and develop roots to later grow as a separate individual with or without a connection to the mother tree. Known as other mechanisms of vegetative propagation are also the rooting of fallen trees which were not completely uprooted, and the "root suckering", in which new shoots sprout from the roots of the tree. However, the latter was not yet observed in the genus Picea. With the help of short, repetitive, non-coding sequences in the genome, which are therefore not subject to selection and are called microsatellites, these clones can be determined by genotyping.
For this purpose, using different polymorphic microsatellites, an individual multilocus genotype is created for each tree, by means of which it can be compared with all other trees of the same species.
In the first part of this work (article I), the occurrence of clones in three study areas at Alaskan treelines are examined and the reasons for their appearance in variable numbers are discussed. For this purpose, 2571 white spruces (P. glauca) were genotyped and their position was determined via differential GPS in the field. The percentage of clonal trees is higher in areas with harsh climatic conditions and correlates with the height of the lowest branches of the tree crown. This suggests that the vegetative propagation of white spruce is a backup strategy for times when climatic conditions hamper sexual reproduction. The correlation between clone numbers and tree crown height suggests "layering" as the main mechanism for cloning whereas selection for vegetative reproduction seems to be very unlikely shown by the results for genetic differentiation between the clonal and the singleton trees in this study.
In the second part of this work (articles II and III), the influence of environmental factors and phenotypic traits on the mycobiome of the needles (including all fungi living on (epiphytic) and in (endophytic) the needles) in our study areas in Alaska was investigated. The mycobiome of the white spruce needles was chosen as a proxy for the parasite infection rate by fungi and thus serves as a fitness parameter. For this purpose, all epiphytic and endophytic fungal species were analyzed by a metabarcoding analysis.
In article II, 48 trees of one study area at Alaska’s northern treeline (Brooks Range) were examined for differences in mycobiome due to genetic differentiation, phenotypic characteristics and / or habitat characteristics. The trees used for this study were sampled from two adjacent plots on a south-facing mountain slope with an elevation gradient from 875 to 950 meters above sea level. It could be shown that, in contrast to the trees genotype, the height above sea level, the mountain slope, as well as the height and age of the trees have a significant impact on the mycobiome. The genetic differentiation between the tree individuals, however, showed no significant effect.
Based on article II we examined the mycobiome composition of a total of 96 trees in 2 plots (16 trees each) at three sites in Alaska over a distance of 500 kilometers. Additionally, we sampled needles of two different ages for each tree (current year and three years old needles) summing up to 192 samples in total. The incentive of this study (article III) was to investigate the influence of origin and age of spruce needles on their mycobiome and if there is a genetic predisposition that is related to the fungal species community. In addition, the sampling design was improved by collecting needles from all four orientations (North, South, East and West) and sampling trees at a standardized distance to each other to avoid systematic errors. Comparable to article II the influence of the trees genetics on the species community of the epiphytic and endophytic fungi of the white spruce needles seems to be very unlikely. In contrast, a significant influence of the geographic origin and the needle age on the species structure of the needle inhabiting fungal species was found. The phenotypic tree traits height and dbh (diameter at breast height) had only minor influence and did in fact explain less than 2% of the mycobiome variance. Using Illumina sequencing, 10.2 million reads from the nucleotide sequence between the internal transcribed spacer (ITS) genes could be obtained, which yielded in 1575 ribotypes (called operational taxonomic unit, OTU) for the fungi. These were compared with a reference database to compare and assign them to known fungal species. For example, 942 OTUs with >95% similarity could be identified as known species, with 1975 samples identified on genus level and 2683 when determined to family level. The most pronounced difference between the two studies (article II and III) were due to the fungal species of the class of Pucciniomycetes, more specifically the genus Chrysomyxa which belongs to the rust fungi and is plant pathogenic. In the study of article II (sampling in 2012), Pucciniomycetes accounted for only a minor portion of the assigned DNA sequences. In the second study (article III, sampling in 2015) they accounted for more than half of all basidiomycetes found, which in turn contain 20.0% of all DNA sequences, the second largest phylum found beside Ascomycetes (51.4%).
GEGENSTAND: Bisphosphonate spielen derzeit eine herausragende Rolle bei der Behandlung von Erkrankungen mit erhöhter Knochenresorption wie Osteoporose, tumorassoziierte Osteolyse und Multiples Myelom. Bisphosphonate hemmen die Knochenresorption über komplexe Mechanismen, vor allem durch ihre zytotoxische Wirkung auf Osteoklasten. Ein Zusammenhang zwischen Bisphosphonat-Therapie und dem Auftreten von Kieferosteonekrosen ist weithin anerkannt (Bisphosphonate-related osteonecrosis of the jaw = BRONJ). Diese Nebenwirkung ist zu einem ernstzunehmenden klinischen Problem geworden. PATIENTEN & METHODEN: In der vorliegenden retrospektiven Arbeit wurden die Daten von 51 Patienten mit ossären Metastasen (n=40) oder Multiplem Myelom (n=11) unter intravenöser Amino-Bisphosphonat-Therapie hinsichtlich möglicher Risikofaktoren für die Entwicklung einer BRONJ untersucht. Es wurden das klinische Bild, die Bedeutung präventiver Maßnahmen, das kieferchirurgische Therapiemanagement und die Therapieergebnisse analysiert und mit der derzeit verfügbaren Literatur verglichen. ERGEBNISSE: Der Beobachtungszeitraum für das Gesamtkollektiv war im Median 26,1 Monate. Das Durchschnittsalter der Gruppe betrug 65,8 Jahre. 17 Patienten zeigten eine klinisch manifeste Kiefernekrose, wobei Unterkiefernekrosen signifikant häufiger gesehen wurden (p=0,008). Das BRONJ-Risiko stieg mit der Dauer der Bisphonatbehandlung (p=0,010). Am häufigsten traten Nekrosen nach Zahnextraktionen auf. Die Ergebnisse der kieferchirurgischen Therapie waren bei Patienten ohne Osteonekrose signifikant besser (p<0,001) im Vergleich zu Patienten mit manifester Osteonekrose. Patienten mit präventiven Maßnahmen vor Bisphosphonat-Therapiebeginn hatten im Vergleich zu Patienten ohne präventive Maßnahmen ein signifikant niedrigeres Risiko für die Entwicklung einer BRONJ (p=0,002). SCHLUSSFOLGERUNG: Die Daten der vorliegenden Arbeit unterstreichen die Bedeutung präventiver Maßnahmen zur Vermeidung von Osteonekrosen bei Bisphosphonat-Therapie. Da die Pathogenese der BRONJ nicht geklärt und damit eine kausale Therapie derzeit nicht verfügbar ist, muss die Standardisierung und Durchführung präventiver Maßnahmen als conditio sine qua non gefordert werden. Die Aussagen der vorliegenden Studie weisen durch ihre retrospektive Natur und die kleine Fallzall der einzelnen Gruppen Grenzen auf. So konnten für Alter, Geschlecht und Allgemeinzustand, die Art der Grund- und Zweiterkrankungen, die tumorspezifischen Ko-Therapien, Bisphosphonat-Typ, Dosis und Applikationsform keine statistisch signifikanten Unterschiede nachgewiesen werden. Weitere Forschungsarbeiten durch große, prospektiv randomisierte Studien sind erforderlich.
Demenznetzwerke versuchen mithilfe regionaler Kooperationen eine moderne, sektorenübergreifende sowie multidisziplinär ausgerichtete Demenzversorgung aufzubauen. Diese Kooperationsmodelle werden seit einigen Jahren politisch und gesellschaftlich gefordert. Obwohl es in Deutschland bereits eine Vielzahl von Demenznetzwerken gibt, ist wenig über sie bekannt. Mit dem Ziel, den MmD so lange wie möglich ein selbstständiges Leben zu ermöglichen, fordern nationale Demenzleitlinien eine integrierte Versorgung von Menschen mit Demenz (MmD). Die vorliegende Dissertation untersucht, wie häufig MmD eine integrierte, spezialisierte Demenzversorgung in deutschen Demenznetzwerken erhalten und mit welchen Faktoren diese auf Seiten des MmD und Netzwerkes assoziiert ist. Die Dissertation geht im Kern der Frage nach, inwieweit Demenznetzwerke die Versorgung von Menschen mit Demenz verbessern und ob Demenznetzwerke ein Zukunftsmodell zur Versorgung von MmD darstellen könnten. In der multizentrischen, interdisziplinären Evaluationsstudie von Demenznetzwerken in Deutschland (DemNet-D) wurden 560 Bezugspersonen von Menschen mit Demenz in n=13 Demenznetzwerken über sozio-demografische und klinische Parameter sowie die Inanspruchnahme von medizinischen, pharmazeutischen, nicht-pharmakologischen Therapien sowie die Hilfsmittelversorgung des MmD befragt. Die Daten wurden mit deskriptiven und multivariaten Regressionsmodellen analysiert. Im Ergebnis erreichen MmD in Demenznetzwerken eine hohe Quote in der Konsultation von Hausärzten (93%) und Fachärzten für Neurologie/ Psychiatrie (74%). Mit einem Antidementivum werden 52% der MmD in den untersuchten NW versorgt. Bei nicht-pharmakologischem Therapien erhalten 24% der MmD Physiotherapie und 15% der MmD Ergotherapie. Die häusliche Versorgungssituation wird u.a. mit Mobilitätshilfen bei 59% der Nutzer von Demenznerzwerken unterstützt. Weibliches Geschlecht ist innerhalb der neurologischen/ psychiatrischen Facharztversorgung mit einer geringeren Versorgungsquote (OR 0.6) assoziiert. Die Antidementivaversorgung ist u.a. signifikant positiv mit der formalen Diagnose (OR 16.8) assoziiert. Bei nicht-pharmakologischen Therapien (OR 1.2) und Hilfsmitteln (OR 1.4) sind Komorbiditäten signifikant mit der Versorgung assoziiert. Eine medizinische Affiliation des Netzwerkes zeigt sich als positiver Einflussfaktor u.a. bei der neurologischen/ psychiatrischen Facharztversorgung (8.4) und der Versorgung mit Antidementiva (OR 3.7). Die vorliegenden Ergebnisse zeigen, dass MmD in Demenznetzwerken im Vergleich zu primärärztlicher Versorgung höhere Inanspruchnahme-Quoten erreichen. Sie zeigen nicht, ob MmD bedarfsgerecht versorgt werden oder auch ob sich Demenznetzwerke auf Institutionalisierungsraten auswirken. Die Ergebnisse weisen jedoch darauf hin, dass Nutzer von Demenznetzwerken häufiger eine demenzspezifische Versorgung erhalten. Demenznetzwerke adressieren somit zentrale Forderungen u.a. der WHO.  
Mass spectrometry-based Proteome analysis of porcine cells infected with African swine fever virus

ASFV, a highly contagious, pathogenic and lethal pathogen of swine, poses a major threat to domestic and wild suids worldwide as neither vaccines nor treatments are available. Compared to other well-characterized similarly complex viruses like herpesviruses or adenoviruses, the understanding of ASFV biology is poor.
To improve the understanding of ASFV biology, following the establishment of a robust protocol for the isolation of primary monocyte-derived porcine macrophages (moMΦ) and their infection with ASFV for mass spectrometry (MS)-based proteome analysis was performed.
Under both conditions, naïve and infected, the isolated cells showed cell type-specific characteristics like phagocytosis and antigen presentation and protein expression patterns, including the expression of swine leucocyte antigens and CD markers. Furthermore, moMΦ could be reproducibly infected with ASFV isolates of different genotypes and pathogenicity.
The ASFV protein expression patterns in moMΦ correlate well with those observed in established cell lines at transcript and protein level. The expression of 27 ASFV proteins was confirmed at the protein level. Among them, 9 members of multi-gene families (MGF) and 12 novel open reading frames (nORFs) were recently predicted based on transcription start site mapping.
The direct comparison of closely related ASFV genotype II isolates revealed no virulence-associated protein expression patterns beyond those expected based on the genome sequences of the isolates.
Using different MS quantification strategies, it was shown that ASFV affects both static protein expression levels and protein synthesis. These changes in protein expression impact proteins and pathways known to be targeted by ASFV, including CD-markers, ER-stress and cell death pathways, and cellular antiviral responses. Beyond these observations that further validated the moMΦ infection model, novel effects of the ASFV infection on the cellular proteome were noticed.
These effects include the decreased expression levels of cathepsins, especially cathepsins D (CTSD), H (CTSH) and L (CTSL) as well as the transient activation of MAPK14/p38 prior to its strong downregulation. In addition to MAPK14/p38 further members of the MAPK14/p38 signaling pathway, like MAPKAPK2, were affected by ASFV infection.
As these modulations of the cellular proteome would in general result in decreased pro-inflammatory responses, it did stand out that the synthesis of interferon-response related genes including MX1 and ISG15 evaded the ASFV-induced global reduction of protein synthesis. In contrast, the synthesis of genes involved in RNA processing and splicing was significantly impaired. In total, the regulations of individual host proteins assessed in the context of the whole cellular proteome integrate well with each other and other cellular responses to ASFV infection and may help to improve the understanding of host-virus interactions.
Overall, this thesis provides novel insights into the expression of ASFV-encoded ORFs of different isolates and the host response to ASFV infection. It points out that the current knowledge of the ASFV coding capacity, temporal protein expression patterns, protein functionality, post-translational modifications and host interactions is still sketchy as many aspects of ASFV replication have yet to be understood. The established moMΦ-model to study ASFV infections in vitro provides a powerful tool for future applications to increase the understanding of ASFV biology.
Cerebral palsy (CP) remains one of the most common debilitating diseases in the world.
Factors such as neuromuscular incoordination, use of soft diet and delayed oral
clearance of food make maintenance of optimum oral hygiene a challenging task in
these children. The compromised oral hygiene in turn make these children highly
vulnerable to dental disease. Maintaining optimal oral/dental health in CP children is of
utmost importance as these children usually suffer from several associated general
health problems in addition to their primary condition. Poor dental health further
compromises their general health. Nevertheless, CP children often suffer from dental
diseases including dental caries more than healthy children. This underscores the need
for improvement in oral health of the CP children. The CP children are dependent on
those around them for their oral hygiene maintenance and dietary intake. CP children
spend most of their time with health care workers in special centers for them or with
their parents/care takers at home. Therefore, the attitude of special health care workers
and parents towards their dental health is of prime importance. The purpose of this
project included to determine the attitude of special health care workers towards their
dental health and oral health comprehension by parents of CP children. In addition, a
study on caries experience of CP children and associated risk factors was also completed.
The final objective was to identify various potential areas of improvement with ultimate
goal of improving dental health of the CP children.
The study involving special health care workers in a Disabled Children’s Center revealed
that oral health knowledge and practices among the workers could generally be labeled
as satisfactory, however some weak areas (such as trend towards symptom-oriented
utilization of oral health care and lack of knowledge about fluoridated water) were
identified. There is a need to enhance the workers’ knowledge in these areas.
The study involving parents of the CP children showed mixed results. Some areas of
strong oral health comprehension (such as importance of good dental health, harmful
effects of sweetened foods), others satisfactory (such as need for routine dental visits,
use of fluoride) and some weak areas (use of flavored fizzy drinks, bottled/canned juices
and sweetened/flavored milks, seeking early treatment) were identified among parents
of CP children. The results strongly indicated a need for enhanced efforts towards
improvement of oral health comprehension in the parents of the CP children.
The study about caries experience and risk factors in CP children has provided useful
information in an area where literature has been scarce. Almost all the studied children
had clinical dental caries, and most of the caries were untreated. Very few children in
the present study had optimal oral hygiene. This study confirmed the strong correlation
between high dental caries experience and poor oral hygiene. Routine dental check-up
visits and topical fluoride application clearly resulted in lower caries experience. Bottled
juices and crispy potato chips were the two predictors for high caries experience.
As stated before, the goal of the project was to identify various potential areas of
improvement in oral health attitude of special health care workers, oral health
comprehension of the parents of CP children and risk factors for dental caries in CP
children. It is contemplated that by addressing the identified weak areas, an
improvement in dental health and care of the CP children is expected. It is also expected
that the collected information will help in improvement of clinical preventive as well as
restorative services for the CP children. CP children and their parents lead a challenging
life. This project was an effort to assist these children and their parents/care takers with
ultimate goal of improving their oral health and in turn their overall quality of life.
