Protamine (PRT) is a positively charged protein, which is widely used in medicine as an adjunct to certain preparations of insulin and as a rapidly-acting antidote for heparin, particularly to neutralize the effects of high heparin concentrations needed for anticoagulation during cardiac surgical procedures using cardiopulmonary bypass. It has been demonstrated that PRT and heparin form multimolecular complexes and that these complexes have high immunogenicity in a mouse model. Studies in this thesis provide new insights into the pathophysiology of anti-PRT/heparin antibodies. The results of study I showed that the administration of PRT combined with heparin is responsible for high immunoglobulin G (IgG) immunization after cardiac surgery. A subset of these antibodies was able to induce platelet activation in a way similar to that observed by heparin-induced thrombocytopenia (HIT). Using an animal model, we demonstrated that anti-PRT/heparin antibodies are capable of platelet destruction in the presence of PRT and heparin. Moreover, our data suggests that platelet-activating anti-PRT/heparin antibodies at surgery are potentially associated with postoperative thrombocytopenia and an increased risk for thromboembolic events. In study II, the immune response against PRT/heparin complexes was investigated. This study showed a relatively fast development of IgG with no general preceding IgM formation. In addition, patients undergoing liver transplantation developed anti-PRT/heparin antibodies without previous exposure to PRT. These results suggest that a previous contact with the antigen(s) itself or other antigens with molecular mimicry induced this immune response. In fact, we were able to identify Neutral Protamine Hagedorn (NPH) insulin and core histones (DNA-binding proteins) as potentially antigenic candidates for a previous immunization. Furthermore, the findings of study III demonstrate the ability of anti-PRT/heparin antibodies to activate platelets in the presence of NPH insulin in a heparin-dependent way suggesting that diabetic patients may have an enhanced risk for thromboembolic complications if treated with NPH insulin and possibly while receiving prophylactic heparin. These observations justify further clinical investigations to assess the impact of the interaction between anti-PRT/heparin antibodies and PRT-mimicking antigens, such as NPH insulin or histones.
In dieser retrospektiven Studie wurden die Parameter der Radiojodkinetik: Jod-Uptake (JU), effektive Halbwertzeit (HWZ) und Zeitpunkt der maximalen Jod-Aufnahme (tmax) sowie deren mögliche Beeinflussung durch Erkrankungsart, Schilddrüsenvolumen, Funktionslage, Vortherapien, Patientenalter und Zeitverlauf untersucht. Methodik: Die Studie umfasst Daten von 6156 Radiojodtherapien (RJTH) von Morbus Basedow (MB), unifokalen Autonomien (UFA), multifokalen Autonomien (MFA), disseminierten Autonomien (DISA) und Strumen (STRU), die im Zeitraum von 1996 bis 2008 an der Universitätsklinik für Nuklearmedizin in Greifswald durchgeführt wurden. Die ausgewerteten intratherapeutischen JU-Kurven beruhen auf frequenten Aktivitätsmessungen (automatisierte Patienten-Selbstmessungen, ca. 13/d). Ergebnisse: Für den JU relevant waren Erkrankungsentität und Zielvolumen. In Bereichen äquivalenter Volumen war der JU bei MB etwa 3-mal höher als bei den Strumen. Bei fokalen Autonomien stieg der JU nichtlinear mit dem Zielvolumen; eine Verdoppelung des Zielvolumens im therapierelevanten Bereich (von 10 auf 20 ml) resultierte in einer Zunahme des JU um 30%. Die HWZ war bei Strumen und DISA etwa 20% länger als bei MB und UFA und variierte in den Gruppen um ca. 25%. Mit zunehmender Funktion verkürzte sich die HWZ. Eine Verdopplung des T3-Wertes ergab eine um etwa 20% reduzierte HWZ. Tmax lag im Gesamtkollektiv bei 18h; im Vergleich dazu war tmax bei MB um 20% kürzer und bei STRU um 20% länger. Es bestand eine schwache bis mäßige Korrelation von tmax und Patientenalter sowie eine inverse Korrelation von tmax mit der Funktionslage. Im Zeitverlauf fiel der JU bei abnehmenden Zielvolumina leicht ab (in 12 Jahren ca.1-2%/Jahr). Ein höherer JU bei MB ab 2003 ist annehmbar Folge des veränderten Therapieschemas (Thyreostasepause vor RJTH). Eine frühzeitig bei drohender Unterdosierung durchgeführte Nachapplikation von Radiojod (nötig bei 6% des Kollektivs) führte zu ähnlichen Erfolgsraten (92%) wie in der Normgruppe (91%). Schlussfolgerungen: Die Radiojodkinetik weist relevante Unterschiede zwischen den Erkrankungsgruppen auf. Haupteinflussfaktor auf den JU sind Erkrankungsentität und Zielvolumen. In der Praxis sind für die HWZ die Erkrankungsentität und etwas geringer die Funktionslage von Bedeutung. Bei der Therapieplanung (Radiojodtest) erscheint angesichts der vergleichsweise geringen Streuung der HWZ in den Gruppen die Nutzung von krankheitsspezifischen HWZ (MB, UFA: 5d; MFA: 5,5d, Andere: 6d) vertretbar. Variationen von tmax werden bei großen Messintervallen nicht erfasst, können aber zu fehlerhaften JU- und HWZ-Werten und damit zu falschen Dosisschätzungen führen. Bei Patienten mit drohender Unterdosierung kann durch eine frühzeitige Dosiskorrektur eine Zweittherapie vermieden werden. Dafür ist jedoch eine frequente JU-Messung erforderlich. Ein Teil der divergierenden Literaturangaben zur Radiojodkinetik ist durch Bias (insbesondere Zielvolumenvariation) erklärbar. Orientierende Aktivitätsmessungen per Smartphone sind derzeit nicht geeignet zur Schätzung von Kinetikparametern.
This thesis deals with thickness optimization of shells. The overall task is to find an optimal thickness distribution in order to minimize the deformation of a loaded shell with prescribed volume. In addition, lower and upper bounds for the thickness are given. The shell is made of elastic, isotropic, homogeneous material. The deformation is modeled using equations from Linear Elasticity. Here, a basic shell model based on the Reissner-Mindlin assumption is used. Both the stationary and the dynamic case are considered. The continuity and the Gâteaux-differentiability of the control-to-state operator is investigated. These results are applied to the reduced objective with help of adjoint theory. In addition, techniques from shape optimization are compared to the optimal control approach. In the following, the theoretical results are applied to cylindrical shells and an efficient numerical implementation is presented. Finally, numerical results are shown and analyzed for different examples.
This thesis draws a comprehensive picture about the radiation and diversification of truncatelloidean gastropods across the south pacific. It covers three more specifc studies focussing on the Truncelloideans from Fiji, Vanuatu and New Caledonia, respectively. And a conclusive analysis that combines the results of the three more specific studies and enhances them using species from the Austral Islands, Lord Howe Island, the Indonesian island Sulawesi as well as several species from New Zealand and Australia. Molecular phylogenies were calculated using four nuclear gene fragments (ITS2; 18S rRNA; 28S rRNA and Histone 3) besides the mitochondrial COI and 16S rRNA. Further molecuular data was used to calculate dated phylogenies, perform ancestral range reconstructions and develop a modified molecular barcoding approach.
Die vorliegende Arbeit befasst sich mit der Verbreitung und Populationsgenetik der invasiven asiatischen Buschmücke Ae. j. japonicus, die sich seit 2008 durch Menschen-vermittelten Transport in Deutschland ausbreitet. Aedes j. japonicus ist unter Laborbedingungen Vektor für verschiedene Viren, unter anderem für das Dengue-Virus und das Chikungunya-Virus, und wurde im Feld mit dem Japanische Enzephalitis-Virus, dem West Nil-Virus und dem La Crosse-Virus infiziert gefunden. 2012 wurde aufgrund mehrerer unabhängiger Mücken-Einsendungen im Rahmen des Citizen-Science-Projekts “Mückenatlas” eine Population der Asiatischen Buschmücke in Westdeutschland entdeckt. Das Verbreitungsgebiet dieser Population befand sich weit nördlich der bisher angenommenen nördlichen Verbreitungsgrenze der Art in Süddeutschland. Das Ausmaß der Population wurde nach einem zeitlich begrenzten Monitoring auf eine Fläche von ca. 2000 km2 bestimmt. Aus dieser Population wurden Individuen von fünf Orten populationsgenetischen Analysen unterzogen, um verwandtschaftliche Beziehungen innerhalb der Population und im Vergleich zu anderen europäischen Populationen aufzudecken. Hierzu wurden sieben Mikrosatelliten-Loci untersucht. Zusätzlich wurde ein Teil der mitochondrialen nad4-Genregion der Individuen auf Nukleotid-Polymorphismen untersucht. Die Ergebnisse wurden mit bereits zuvor erhobenen Daten von Populationen aus der Schweiz, aus Österreich/Slowenien und Belgien verglichen. Die Mikrosatellitensignatur der westdeutschen Population unterschied sich deutlich von der der anderen europäischen Populationen. Weiterhin wurden verschiedene nad4-Haplotypen gefunden, die zuvor nirgendwo sonst in Europa aufgetreten waren. Demnach ist zu vermuten, dass diese Population auf eine unabhängige Einschleppung von Individuen aus Übersee zurückgeht. Der genaue Ursprung – USA oder Ostasien – konnte nicht bestimmt werden. 2013 wurden zwei weitere Ae. j. japonicus-Populationen in Europa entdeckt: eine in Norddeutschland und eine weitere in den Niederlanden. Die genetischen Signaturen von Individuen dieser Populationen wurden wie beschrieben analysiert. Zusätzlich wurde das genetische Material einer größeren Menge von Individuen aus Slowenien sowie von Individuen aus Kroatien und Süddeutschland untersucht. Die Ergebnisse wurden mit denen aus der vorigen Studie verglichen und zeigten aufgrund einer ähnlichen Mikrosatellitensignatur und gleicher nad4-Haplotypen klar, dass die norddeutsche Population eine Subpopulation der westdeutschen ist. Die geringe Populationsdichte und die vergleichsweise kleine Ausdehnung der norddeutschen Population deuten außerdem darauf hin, dass die Abspaltung nicht lange zurückliegt. Die niederländische Population scheint hingegen auf einer weiteren Einschleppung von Individuen aus Übersee zu basieren. Im Spätsommer 2015 wurde die bisher letzte deutsche Ae. j. japonicus-Population in Oberbayern und dem angrenzenden Österreich entdeckt. Populationsgenetischen Analysen zufolge ist diese Population eng mit der früher beschriebenen österreichisch-slowenischen Population verwandt und unterscheidet sich von allen anderen deutschen Populationen, was darauf schließen lässt, dass es sich bei ihr um eine Abspaltung von der österreichisch-slowenischen Population handelt. Die Ver- und Ausbreitung von Ae. j. japonicus in West- und Norddeutschland wurde vom Zeitpunkt der Entdeckung in 2012/2013 bis 2015 beobachtet. In dieser Periode erweiterte die westdeutsche Population ihr Verbreitungsgebiet beträchtlich, während die norddeutsche überhaupt nicht zu expandieren schien. Dies ist möglicherweise darauf zurückzuführen, dass die norddeutsche Population jünger als die westdeutsche ist, das Verschleppungsereignis noch nicht so weit zurückliegt und die Population sich noch in der Gründerphase befindet. Die passive weltweite Verschleppung von Stechmücken wird in der Zukunft vermutlich zunehmen, und die Etablierung und Ausbreitung invasiver Spezies, inklusive der Asiatischen Buschmücke und anderer potenzieller Überträger von Krankheitserregern, werden Europa und Deutschland weiterhin vor herausfordernde Probleme stellen. Das Monitoring der Ausbreitung von Populationen und die Durchführung populationsgenetischer Analysen zur Ermittlung von geographischen Ursprüngen sowie von Wanderungs- und Transportrouten werden helfen, weitere Einschleppungs- und Ausbreitungsereignisse nachzuvollziehen und zu unterbinden und sind daher essenzielle Instrumente des Managements von Mückenvektoren.
All types of muscles use Ca2+ as their main intracellular messenger. In skeletal muscle fibers abnormal levels of intracellular calcium result in altered contractile properties, altered energy metabolism, and altered gene expression. Moreover, long term failure of normal Ca2+ homeostasis can lead to cell death of muscle fibers by necrosis and apoptosis. Elevations of intracellular Ca2+ levels are more and more regarded as the reason for pathological changes and muscle fiber damage in Duchenne Muscular Dystrophy (DMD). DMD is a severe recessive x-linked muscle disease caused by mutations in the dystrophin gene. The characteristics of DMD are muscle tissue wasting and fibrosis. Both muscle wasting and intracellular Ca2+ are to be reflected in changes of muscle force. Several Ca2+ conducting channels including transient receptor potential (TRP) channels are supposed to account for the abnormal Ca2+ homeostasis in DMD. Gene expressions of TRP channels have been studied in human and mouse skeletal muscle and among others TRPC3, TRPC6 and TRPV4 channels were found to occur in skeletal muscles. The present study followed the hypothesis that TRPC3, TRPC6 and TRPV4 are functional in skeletal muscle fibers and that they contribute to muscular Ca2+ homeostasis. Further, it was assumed that dysfunction of the mentioned TRP channels contributes to abnormal contractile properties and pathology and of dystrophin-deficient muscle. To study Ca2+ changes in mouse skeletal muscle fibers the fluorescent calcium indicator Fura-2 was used. Further, the technique of Mn2+ quench of Fura-2 fluorescence was applied. Muscle force measurements of mouse soleus and diaphragm strips were performed. To elucidate abnormalities of TRP channel function in dystrophin-deficient muscle, muscles and muscle fibers of mdx mice were studied. Hyperforin, an activator of TRPC6 channels elicited increases of calcium levels in wildtype muscle fibers. These increases were partly inhibited by the TRPC6 inhibitor 1-(5-chloronaphthalenesulfonyl) homopiperazine hydrochloride (ML-9). The TRPC3/TPRC6 activator 1-oleoyl-2-acetyl-sn-glycerol (OAG) resulted in increased calcium entry, which was attenuated by ML-9. 2-aminoethoxydiphenylborane (2-APB), an unspecific TRP channel inhibitor, suppressed calcium entry in muscle fibers under basal conditions. In addition, the specific TRPC3 inhibitor Pyr3, strongly inhibited background calcium entry. The TRPV4 activator 4α-phorbol 12,13-didecanoate (4α-PDD) induced significant increased calcium entry and this increase could be inhibited by the TRPV4 inhibitor HC 067047. During muscle force recordings ML-9 significantly inhibited twitches and tetani and accelerated muscle fatigue during sustained repetitive stimulation. The results indicate that TRPC3, TRPC6 and TRPV4 are functionally expressed in mouse muscle fibers. TRPC3 stays active under the basal conditions and contributes to background calcium entry. In contrast, TRPC6 and TRPV4 did not seem to be active at resting conditions, but could be pharmacologically activated. TRPC6 may play a role to counteract the calcium loss under long-term muscle fatigue. Though TRPC3 and C6 play a role for muscular Ca2+ homeostasis, it is unclear whether and how the two channels associate and cross-talk with each other in skeletal muscle cells. In mdx fibers Pyr3 inhibited background calcium influx stronger that in WT fibers, implying a possible over-activation of TRPC3 channels in mdx muscle fibers. At later stages mdx muscle showed marked decrease in force reflecting muscle wasting. Soleus showed moderate decrease and diaphragm showed severe decrease (more than 60%) in force. Resistance to muscle fatigue was shown in mdx soleus muscle when compared with WT soleus muscle. Diaphragm segments of mdx mice showed very strong resistance to muscle fatigue. The results indicate a substantial loss of muscle mass, an increase in oxidative fiber types and a reduction of fast fatigable muscle fibers. It is concluded that the hypothesis of functional expression of TRPC3, TRPC6 and TRPV4 in mouse skeletal muscle has been confirmed. The results give improved knowledge about the relation of Ca2+ homeostasis, mdx pathology and TRP channels. Diaphragms of old mdx mice show severe muscle weakness but the remaining fibers of the diaphragm showed strong fatigue-resistance. The application of a TRPC3 inhibitor may be a promising treatment to prevent high Ca2+ mediated muscle damage in muscular dystrophy.
In der vorliegenden Untersuchung konnte bestätigt werden, dass die bisher aus dem Schrifttum bekannten Risikofaktoren für eine Kolonisation/Infektion von Patienten durch MRSA bei Krankenhausaufnahme nach wie vor zutreffen.
Ferner konnte in der vorliegenden Untersuchung nachgewiesen werden, dass die Verlegungshäufigkeit MRSA positiver Patienten von 2007-2010 abnahm. Damit nahm die Wahrscheinlichkeit einer nosokomialen Weiterverbreitung auf Patienten sowie der Übertragung von MRSA auf das Personal ab. In wie weit das und die vermutlich angestiegene Compliance bei der Einhaltung der SAA durch Hygieneschulungen und Supervision zur Abnahme einer MRSA- Kolonisation bei den Mitarbeitern führte, kann anhand der vorliegenden Daten nicht beantwortet werden.
Da die MRSA-Last im Jahr 2010 im Vergleich zum Jahr 2007 deutlich angestiegen ist, müssen die Präventionsmaßnahmen in ihrer Gesamtheit gegriffen haben. Zur Aufklärung der Bedeutung einzelner Präventionsmaßnahmen innerhalb der Multibarrieren-Strategie der MRSA- Prävention sind weitere Untersuchungen notwendig, um die Präventionsstrategie gegebenenfalls zu adjustieren.
Die vorliegende Arbeit ist im Bereich der parameterfreien Statistik anzusiedeln und beschäftigt sich mit der Anwendung von ordinalen Verfahren auf Zeitreihen und Bilddaten. Die Basis bilden dabei die sogenannten ordinalen Muster in ein bzw. zwei Dimensionen. Der erste Hauptteil der Arbeit gibt einen Überblick über die breiten Einsatzmöglichkeiten ordinaler Muster in der Zeitreihenanalyse. Mit ihrer Hilfe wird bei simulierten gebrochenen Brownschen Bewegungen der Hurst-Exponenten geschätzt und anhand von EEG-Daten eine Klassifikationsaufgabe gelöst. Des Weiteren wird die auf der Verteilung der ordinalen Muster beruhende Permutationsentropie eingesetzt, um in Magnetresonanztomographie (MRT)-Ruhedaten Kopfbewegungen der Probanden zu detektieren. Der zweite Hauptteil der Arbeit befasst sich mit der Erweiterung der ordinalen Muster auf zwei Dimensionen, um sie für Bilddaten nutzbar zu machen. Nach einigen Betrachtungen an fraktalen Oberflächen steht eine automatisierte und robuste Einschätzung der Qualität struktureller MRT-Daten im Vordergrund.
Background: Abdominal obesity is a major driver for adverse medical conditions. While an interaction between adipose tissue and thyroid function is thought to exist, to our knowledge, no study has examined the effect of thyroid-stimulating hormone (TSH) on visceral adipose tissue (VAT) in a population-based context. Objective: We determined an association between serum TSH levels and VAT. Methods: A sample of 1,021 female and 956 male adults aged 20-79 years was drawn from registry offices in the cross-sectional, population-based Study of Health in Pomerania Trend (SHIP Trend) in Northeast Germany from 2008 to 2012. Our main exposure was serum TSH levels. Our main outcome was VAT measured using magnetic resonance imaging. The possibly mediating role of leptin on the TSH-VAT association was also assessed. Results: A total of 1,719 participants (87.9%) had serum TSH levels within the reference range. The mean volume of VAT was 5.33 liters for men and 2.83 liters for women. No association between TSH and VAT (β = 0.06, 95% CI: -0.02, 0.14) was observed, and there were no differences detected between sexes. VAT was strongly associated with leptin with a greater effect in women than in men. Leptin was strongly associated with TSH. Conclusions: No association between TSH and VAT was observed. Other biomarkers such as leptin may play a role in the relationship between thyroid function and metabolic risk.
Mithilfe der hochauflösenden funktionellen Magnetresonanztomographie (fMRT) wurde die neuronale Respräsentation des Schluckens in zwei Altersgruppen von insgesamt 51 gesunden ProbandInnen (SeniorInnen: Durchschnittsalter 64 Jahre; junge Erwachsene: Durchschnittsalter 24 Jahre) untersucht. Zwei statistische Vergleiche (klassischer und Bayesscher Rückschluss) zeigten keine signifikanten Unterschiede zwischen den beiden Altersgruppen. Einzig eine erhöhte kortikale Aktivierung im Frontalpol 1 des Brodmann-Areals 10 bei den SeniorInnen konnte mittels der Bayesschen Statistik gezeigt werden. SeniorInnen wiesen im Vergleich mit den jungen ProbandInnen zudem eine längere Reaktionszeit und eine höhere Hautleitfähigkeitsreaktion (SCR) während des Schluckens auf. Gleichzeitig konnte nur bei der Gruppe der alten Gesunden eine positive Assoziation der SCR mit fMRT-Aktivierung in Arealen gezeigt werden, welche sensomotorische Leistung, Erregung und emotionale Wahrnehmung verarbeiten. Die Ergebnisse deuten darauf hin, dass das hoch automatisierte Schlucknetzwerk auch im Alter seine Funktionalität beibehält. Allerdings scheinen SeniorInnen mit einer erhöhten Hautleitfähigkeit während des Schluckens kortikale Areale in Anspruch zu nehmen, welche bei der Aufmerksamkeitskontrolle und Emotionsregulation eine Rolle spielen. Dies kann möglicherweise eine erhöhte Aufmerksamkeit und emotionale Anforderung während der Erfüllung der Schluckaufgabe nahelegen.
Interactions between bacteria and the human body are manifold and happen constantly. Most parts of the skin and gastrointestinal tract, the saliva, the oral mucosa, the conjunctiva and the vaginal mucosa are colonized with a multitude of bacterial species forming the human microbiota. Strikingly, the estimated amount of bacterial cells outnumbers the human body by 10 to 1. However, most of these bacteria colonize the human body without positive or negative effects and are regarded as commensals. Staphylococcus aureus a Gram positive bacterium is such a commensal bacterium of 25 % to 30 % of the world population. It is also an opportunistic pathogen and is able to cause infections in the lung, skin and heart and to induce sepsis. Its pathogenicity is mainly facilitated by the secretion of a broad spectrum of virulence factors which interact with the host. Some are distracting the immune system, others are targeting the host cell membrane or degrade macromolecular structures of the host in order to provide nutrients. Furthermore S. aureus is able to invade the host cell and to survive and replicate in the host cell cytosol or other compartments. The Gram negative proteobacterium Burkholderia pseudomallei is an environmental bacterium but still has the ability to enter the human body via body orifices or skin wounds. In a very efficient way it penetrates the host cell, replicates intracellular and the uses host structures to spread from cell to cell thereby causing the disease melioidosis often with fatal outcomes. Since the natural habitats of B. pseudomallei are wet soils, the change to the environment in the human body is drastic and requires a high degree of flexibility of the bacterium. Environmental stress conditions such as temperature, pH, nutrient limitation or presence of antibiotics induce a switch of colony morphology which is a special characteristic of this bacterium. Since it is assumed, that changes in colony morphology are connected to adaptive processes to the environmental changes, these morphology switches might also be important during infection. The host organism and the host cell on the other side try to kill and remove the bacterial threat by activating the immune system and cellular defence mechanisms. This includes generation of reactive oxygen and nitrogen species, production of antimicrobial peptides and cellular processes such as phagocytosis, autophagy, apoptosis and activation of the immune response. The actions and reactions on both, the pathogen side and the host side, are summarized as host-pathogen interactions. In the field of functional genomics, methods were developed to understand various levels of host-pathogen interactions. The holistic analysis of the mRNA (the transcriptome) or translated proteins (the proteome) were already very useful tools to describe important cellular processes on the host and the pathogen site. The level of metabolites with regard to host-pathogen interactions however, has been neglected so far. In this dissertation the metabolic composition in the intracellular and extracellular space of the host and the pathogen was analyzed. For this matter biochemical analytical tools were used such as 1H-nuclear magnetic resonance spectroscopy and chromatographic methods (GC and HPLC) coupled to mass spectrometry. The combination of these methods allows a broad coverage of physicochemical diverse metabolites. In accordance to the above mentioned biological levels like mRNA and proteins, the sum of all metabolites is referred as the metabolome. Consequently to transcriptomics and proteomics the analysis of the metabolome is referred as metabolomics. To gain insights into the infection relevant metabolome of the host-pathogen relationship between S. aureus and human lung cells several approaches were developed. First the distribution of the recently identified bacillithiol in different S. aureus strains was investigated with regard to its role during the infection. For that matter a HPLC-methodology was used with fluorescence based detection of labelled low molecular weight thiols (article I: Distribution and infection-related functions of bacillithiol in Staphylococcus aureus). After that the next aim was to reveal the effect of S. aureus on the host cell metabolism. To reduce the complexity of effects on the host cells an artificial model was chosen in a first approach. The lung cells were treated with the staphylococcal virulence factor alpha-hemolysin, a pore forming toxin and a holistic metabolomics approach was performed (article II: Staphylococcus aureus Alpha-Toxin Mediates General and Cell Type-Specific Changes in Metabolite Concentrations of Immortalized Human Airway Epithelial Cells). Using this approach, a protocol for cell culture metabolomics was established and first changes in the host cell metabolome that could be caused by S. aureus were described. However, this only describes specific changes caused by one single virulence factor and does not necessarily describes the reality during a S. aureus infection. Therefore in a next approach, an infection model using a human lung epithelial cell line and the S. aureus strain USA300 was established and used for metabolome analysis. Furthermore a combination of inhibitor treatment and metabolic labelling was used to clarify the metabolic activity in the host cell after exposure to S. aureus (article III: Metabolic features of a human airway epithelial cell line infected with Staphylococcus aureus revealed by a metabolomics approach). Finally this thesis deals with the host-pathogen interaction of B. pseudomallei and its host with a focus on the role of the switch in colony morphology in basic metabolism. Various morphotypes of two strains were generated by nutrient limitation and their uptake of nutrients was monitored. Furthermore the morphotypes were used in in vitro and in vivo infections and subsequently isolated out of the cell line and mice respectively. After isolation, the colony morphology was determined and again the nutrient uptake profile was monitored (article IV: Burkholderia pseudomallei morphotypes show a synchronized metabolic pattern after acute infection). The information provided by this thesis adds a new complexity to the knowledge about the host-pathogen interactions of S. aureus and B. pseudomallei and their hosts. It furthermore lays the groundwork for future studies, which will deal with these and other bacterial host-pathogen interactions in order to understand the interdependencies of infection and metabolism.
Häufig sind Pathologien an der Aorta mit Veränderungen des Aortendurchmessers verbunden. Kenntnisse über Referenzwerte des Aortendurchmessers und deren Einflussfaktoren sind wichtige Aspekte der Prävention und optimalen Versorgung von potenziell lebensbedrohlichen Erkrankungen in diesem Abschnitt des Gefäßsystems. Die Ziele dieser Arbeit waren die Bestätigung einer ausreichenden Validität von Messungen des Durchmessers der thorakalen und abdominalen Aorta in nativen VIBE-Sequenzen der MRT, die Beschreibung von bevölkerungsrelevanten Referenzwerten des Durchmessers der abdominalen Aorta und die Untersuchung des Einflusses von kardiovaskulären Risikofaktoren auf diesen Durchmesser. Für die Analyse der Validität standen 50 männliche Probanden zur Verfügung, bei welchen im Rahmen der SHiP-Studie sowohl eine CE-MRA, als auch eine native MRT in der VIBE-Sequenz durchgeführt wurde. Die Vermessung der Aorta erfolgte dabei in der CE-MRA in orthogonaler und in der VIBE-Sequenz in axialer Darstellung. Zur Überprüfung der Validität wurde im Anschluss der Korrelationskoeffizient r nach Bravais und Pearson berechnet, wobei sich starke Korrelationen ergaben für die Aorta Ascendens mit r = 0,95 (P<0,0001), für die Aorta Descendens mit r = 0,88 (P<0,0001), für die suprarenale Aorta mit r = 0,92 (P<0,0001) und die infrarenale Aorta mit r = 0,87 (P<0,0001). Beim Arcus Aortae zeigte sich eine moderate Korrelation mit r = 0,78 (P<0,0001). Bei einer durchgeführten Bland-Altman-Analyse lag der größte mean bias bei 5,55% für die infrarenale Aorta, was einer Abweichung von unter 0,1cm entspricht. Bei allen weiteren Abschnitten war der mean bias geringer. Die 95% LOA lagen für alle Abschnitte unter 15% mit maximal 14,29% bei der infrarenalen Aorta, was weniger als 0,5cm entspricht. Die Analyse der Intraoberservervariabilität erbrachten einen maximalen mean bias von 1,32% und die 95% LOA immer unter 13%. Bei der Interobservervariabilität ergab sich ein maximaler mean bias von 1,61% und die 95% LOA lagen stets unter 10%. Demensprechend besitzt die axiale Durchmesserbestimmung der Aorta in der nativen VIBE-Sequenz eine ausreichende Validität und Reliabilität und kann für epidemiologische Studien genutzt werden. Für die Erstellung von Referenzwerten des Durchmessers der abdominalen Aorta konnten Bilddaten von 1759 Probanden der SHiP-Studie genutzt werden. Die erstellten Referenzwerte sind nach Geschlecht aufgeteilt, an die Körperoberfläche angepasst und beinhalten den Median und die 5. und 95. Perzentile des Durchmessers der subphrenischen, suprarenalen und infrarenalen Aorta. Zur Untersuchung des Einflusses von kardiovaskulären Risikofaktoren wurden Modelle der linearen Regression genutzt. Hier zeigten sich konstante Zunahmen des angepassten Aortendurchmessers in Verbindung mit dem Alter, dem Rauchverhalten, dem diastolischen Blutdruck und dem HDL-Cholesterin. Konstant negative Korrelationen ergaben sich bei der Untersuchung des Einflusses des systolischen Blutdrucks, des HbA1c, des LDL-Cholesterins und der Triglyzeride. Das männliche Geschlecht war lediglich bei der infrarenalen Aorta mit einer Durchmesserzunahme assoziiert und der angepasste Durchmesser war bei Männern im Bereich der subphrenischen und suprarenalen Aorta kleiner als bei Frauen. Durch die in dieser Arbeit beschriebenen Referenzwerte und die Untersuchung des Einflusses von kardiovaskulären Risikofaktoren wird eine individuellere Beurteilung des Durchmessers der abdominalen Aorta ermöglicht. Zusätzlich können durch die Validierung der Durchmesserbestimmung in nativen MRT-Aufnahmen zukünftige Studien auf diesem Gebiet erleichtert werden.
Introduction: Inhibition of androgen synthesis by abiraterone acetate (AA) entails enhanced overall survival rates and clinical benefit for patients with locally advanced and metastasized prostate cancer (PC). The expression of heat shock protein 27 (HSP27) is generally associated with cytoprotection and was demonstrated to mediate chemoresistance under cytostatic therapy, for instance, docetaxel treatment. In this study, we investigated the impact of AA treatment on HSP27 expression and PC cell growth. Materials and Methods: HSP27 expression levels in docetaxel and AA-treated PC cell lines LNCaP and PC-3 were determined by SDS PAGE and Western blot analysis. Proliferation assays were performed using a CASY Cell Counter and Analyzer Model TT (Roche Applied Science). Results: Despite significantly increased HSP27 expression in PC cells incubated with docetaxel, Western blot analysis implicated a significant reduction of the cytoprotective HSP27 in AA-treated PC cells. Notably, HSP27 stably overexpressed in PC-3-HSP27 cells did not appear as an HSP27-mediated proliferation benefit in the presence of AA as shown in docetaxel incubation studies. Conclusion: In contrast to repeatedly demonstrated HSP27-driven chemoresistance related to chemotherapeutics, our results may constitute a broader molecular mode of action of AA chemotherapy. AA efficacy may exert an HSP27 suppressive role that goes beyond the primarily assumed inhibition of androgen biosynthesis.
Comprehensive study of the discharge mode transition in inductively coupled radio frequency plasmas

In this contribution, the mode transition of an inductively coupled radio frequency plasma at low pressure is investigated. Therefore, a comprehensive set of plasma diagnostics were applied to determine plasma and processing parameters. Therewith, the plasma kinetics and especially the important elementary processes were studied. Hence, the reason for the mode transition was identified.
In der Querschnittsanalyse der Daten der bevölkerungsrepräsentativen Studie „Study of Health in Pomerania“ (SHIP 0) konnten allgemein anerkannte und gut erforschte Zusammenhänge zwischen Parodontalerkrankungen und deren Risikofaktoren belegt werden. Die hohe Probandenanzahl von 2208 Teilnehmern ermöglichte es, die Prävalenz von Zahnfehlstellungen als Risikofaktor bezüglich parodontaler Erkrankungen in Mecklenburg-Vorpommern zu berechnen. Ziel dieser Arbeit war es, einen Überblick über die soziodemographischen Aspekte des definierten Probandenguts zu geben. Neben den allgemeinen Faktoren sollten prävalenzbezügliche Aussagen über parodontale und kieferorthopädische Parameter getroffen werden. Dabei wurden Dysgnathiesymptome des Frontzahnbereiches und des Gesamtgebisses separat untersucht und eugnathen Gebissverhältnissen gegenüber gestellt. Darauf folgte das Auffinden möglicher signifikanter Zusammenhänge parodontaler Faktoren mit zunächst allgemeinen Faktoren und im Weiteren mit Zahn- und Kieferfehlstellungen. Es wurden Regressionsmodelle erstellt, mit deren Hilfe mögliche Risikofaktoren für parodontale Erkrankungen bestimmt werden konnten. Letztendlich wurden durch rückwärts bedingte Regression mögliche Assoziationen zwischen Parodontalerkrankungen und Malokklusionen untersucht. Durch die Analyse der Daten konnte belegt werden, dass neben gut untersuchten statistischen Zusammenhängen mit allgemeinen Risikofaktoren wie Geschlecht, Schulbildung, Alter, Rauchverhalten, Familienstand sowie Plaque- und Blutungsindizes, auch ziemlich sichere Zusammenhänge zwischen parodontalen Erkrankungen und bestimmten Zahn- und Kieferfehlstellungen bestehen. Es fanden sich deutliche Hinweise, dass insbesondere eine vergrößerte sagittale Schneidekantendistanz (SKD) als Risikofaktor für Parodontalerkrankungen gesehen werden muss (p < 0,001). Hier fiel eine Dosis-Wirkungsbeziehung auf, da mit der Größe des Abstands der Schneidekanten die statistische Signifikanz in jeder der vier Betrachtungen zunahm. Des Weiteren schienen Lücken im Frontzahnbereich sowie extreme Engstände in Verbindung mit parodontalen Erkrankungen zu stehen. Es stellte sich die Frage nach der Kausalität, also ob diese Fehlstellungen, insbesondere eine lückige Zahnstellung im Frontzahnbereich, aber auch eine vergrößerte sagittale Schneidekantendistanz sowie extremer Engstand, Auslöser oder Folge einer Parodontitis marginalis chronica sind. Klarheit über die Frage nach Ursache und Wirkung kann hier nur eine Longitudinalstudie schaffen. Zusätzlich zeigte sich, dass neben allgemeinen Faktoren wie beispielsweise guter Schulbildung, Nichtrauchen und dem weiblichen Geschlecht anzugehören, auch spezielle Zahn- und Kieferfehlstellungen einer Parodontalerkrankung entgegenwirken können. Ein tiefer Biss ohne Gingivakontakt war hoch signifikant (p < 0,001) mit deutlich weniger Attachmentverlust assoziiert. Auch ein tiefer Biss mit Gingivakontakt und ein Kopfbiss schienen signifikant protektiv auf Attachmentverluste zu wirken. Zusammenfassend lässt sich sagen, dass insgesamt wenige Malokklusionen mit höheren Attachmentverlusten und Sondierungstiefen assoziiert waren. Somit ist eine kieferorthopädische Behandlung mit dem Ziel der Verbesserung der parodontalen Situation nur bei den Malokklusionen mit erhöhtem Risiko für Attachmentverluste und höheren Sondierungstiefen indiziert. Eine gute Mundhygiene und die Vermeidung von Nikotin waren und sind für die Parodontitisprophylaxe wesentlichere Faktoren.
Diese Arbeit widmet sich der funktionellen und strukturellen Untersuchung von SCO3201, einem Protein aus der Klasse der TetR-Repressoren, dessen Struktur bisher unbekannt war und das eine geringe sequenzielle Ähnlichkeit zu anderen Mitgliedern seiner Familie besitzt. SCO3201 wurde als Repressorprotein identifiziert, das durch Überexpression sowohl die Antibiotikaproduktion, als auch die morphologische Differenzierung von Streptomyces coelicolor unterdrückt. In früheren Arbeiten wurde gezeigt, dass SCO3201 an mindestens 16 verschiedene Promotor-Sequenzen binden kann. Das Protein konnte in E. coli exprimiert und anschließend isoliert werden. Wegen des Fehlens geeigneter Strukturmodelle gelang eine Strukturlösung mittels Molekularem Ersatz nach erfolgreicher Kristallisation zunächst nicht. Mittels Single-Wavelength-Anomalous-Dispersion-Methode konnte die Struktur des teilweise induzierten Proteins jedoch aufgeklärt werden. Zudem wurde eine Apo-Form des Proteins kristallisiert und ebenfalls strukturell aufgeklärt. Dies erlaubte die Lokalisation der Ligandenbindungstasche und ließ Rückschlüsse auf die Domänenbewegungen zu, die durch den Prozess der Induktion ausgelöst werden. Daneben wurde mittels Röntgenkleinwinkelstreuung die Struktur von SCO3201 in Lösung untersucht, um eventuelle Kristallisationsartefakte auszuschließen. Durch den Electrophoretic Mobility Shift Assay (EMSA) wurde außerdem die Interaktion zwischen dem Regulator SCO3201 zu seinen Operatoren untersucht.
Staphylococcus aureus is present in around a third of the human population as a constant commensal in the anterior nares, in a third as an intermittent commensal, and a third are non-carriers. However, S. aureus is also a dangerous pathogen, responsible for many types of infections. Recently, the emerging of methicillin-resistant S. aureus strains has aggravated the health problem. Treating infections caused by the invasive strains has become ineffective with conventional antibiotics. Noticeably, transmission of S. aureus has occurred not only in healthcare settings but also in the community; furthermore, transmission between humans and domestic animals has been reported. Although studies about host-pathogen interactions of S. aureus have advanced our knowledge in the last decades, we still have not fully understood mechanisms of the immune system in responses to S. aureus. The aim of this study is to unravel interactions of the human adaptive immune system to selected S. aureus virulence factors. In particular, the study focuses on two aspects: the reaction of human antibodies to the bacterial extracellular proteins in S. aureus-induced furunculosis with an emphasis on Panton-Valentine Leukocidin and responses of the adaptive immune system to membrane-bound lipoproteins of S. aureus. Furunculosis is a variety of hair follicle infection in which S. aureus is one of the chief causal pathogens involved. The corresponding bacterial strains are generally capable of producing of a pore-forming toxin, known as Panton-Valentine Leukocidin (PVL). Recently, the emerging of pvl-positive methicillin-resistant S. aureus has become a problem for treating the bacterially caused furuncles. Colonization with the bacteria is a risk factor for development of chronic or recurrent boils. It is not yet known why furunculosis patients are largely infants or young adults. In this context, we untangled the responses of antibody IgG antibodies to S. aureus extra-cellular factors, notably the PVL toxin, in families in which the patients were children. Multiplex PCR demonstrated that S. aureus clones, isolated from the patients’ wounds but also from the nares of family members, harbored genes coding for PVL toxin. Spa-typing highlighted that bacterial genotypes were very similar in each family. This suggests that transmission of pvl-positive S. aureus took place between family members. The finding also raises the question why only the young patients but not family members who were colonized by the same S. aureus clones suffered from furunculosis. 2D immune proteomics procedures showed a tendency of higher IgG titers against bacterial virulence factors in family healthy members than in patients. PVL-specific antibodies were measured using ELISA, in which patients’ PVL-specific IgG titers were low. This supports the idea that antibodies, probably in conjunction with T cells, might contribute to clinical protection in furunculosis. This research will serve as a foundation for future studies, in which our results should be validated in a larger cohort. Among S. aureus’ virulence factors are lipoproteins, which are anchored in the bacterial cell membrane. Lipoproteins perform various functions in colonization, immune evasion, and immunomodulation. These proteins are potent activators of the complex of innate immune receptors termed Toll-like receptors (TLR) 2 and 6. This study addressed the specific B-cell and T-cell responses to lipoproteins in human S. aureus carriers and non-carriers. 2D immune proteomics and ELISA approaches revealed that titers of serum antibody (IgG) binding to the S. aureus lipoproteins were very low or even unmeasurable in healthy individuals except for the lipoprotein SaeP. Only patients with cystic fibrosis or epidermolysis bullosa who were heavily exposed to the bacteria, generated an antibody response also to lipoproteins. Proliferation assays and cytokine profiling data showed only subtle responses of T cells in healthy individuals; three out of eight tested lipoproteins did not elicit proliferation. Hence, the robust activation of the innate immune system by S. aureus lipoproteins does not translate into a strong adaptive immune response. Reasons for this may be inaccessibility of lipoproteins for B cells as well as ineffective processing and presentation of the antigens to T cells. The main findings implicate that family members can serve as S. aureus reservoirs causing recurrent furunculosis in young patients and that antibodies may provide partial protection from such infections by S. aureus. We have found that, different from proteins that are secreted by S. aureus, lipoproteins which anchored in the bacterial cell membrane, do not trigger strong responses from the human adaptive immune system. This suggests that these proteins remain mostly hidden in the bacterial cell-wall.
Neue robuste Methoden zur Herzschlagerkennung und zur Quantifizierung der Herzfrequenzvariabilität

Für die Analyse der Herzfrequenz ist eine genaue Detektion des Herzschlags aus Rohdaten unerlässlich. Standardmethoden der Herzschlagerkennung sind für elektrische Biosignale konfiguriert worden, die in einem standardisierten klinischen Umfeld erhoben wurden, insbesondere für das Elektrokardiogramm. Im Zuge neuer Möglichkeiten zur Erfassung der Vitalparameter (über Smartphone, drahtlose Möglichkeiten) und zur Reduktion von Falschalarmen im Krankenhaus werden zunehmend robuste Methoden benötigt. Im ersten Kapitel haben wir einen neuen Algorithmus eingeführt, welcher in der Lage ist, unterschiedliche Wellenformen zu verarbeiten und die Informationen aus mehreren gleichzeitig erhobenen Biosignalen zu bündeln. Die Leistungsfähigkeit wurde im Vergleich mit anderen Methoden an freien Datensätzen überprüft und wir konnten uns von der vielfältigen Anwendbarkeit und der Störungsresistenz überzeugen. Im zweiten Kapitel haben wir uns mit der Quantifizierung der Herzfrequenzvariabilität (HRV) beschäftigt und ein neues leicht verständliches Maß eingeführt. Das dafür notwendige Konzept von relativen RR-Abständen wurde diskutiert und die Nutzung zur Artefaktfilterung und zur Klassifikation von Arrhythmiearten aufgezeigt. Vor- und Nachteile klassischer Methoden der HRV haben wir durch einige mathematische Eigenschaften begründet. Im dritten Kapitel der Dissertation haben wir das neue Maß an realen Daten angewendet und die Abhängigkeit der HRV vom Alter der Probanden und von der Herzfrequenz untersucht. Zudem haben wir periodische Strukturen des Streudiagramms von relativen RR-Abständen betrachtet, für die die Atmung ursächlich ist. Als wissenschaftliche Transferleistung wurde abschließend ein freies Programm geschaffen, welches die neuen robusten Methoden umsetzt.
